GIMAP (GTPase of the immunity-associated protein family) proteins are a family of putative GTPases believed to be regulators of cell death in lymphomyeloid cells. GIMAP1 was the first reported member of this gene family, identified as a gene up-regulated at the RNA level in the spleens of mice infected with the malarial parasite, Plasmodium chabaudi. Methods A monoclonal antibody against mouse GIMAP1 was developed and was used to analyse the expression of the endogenous protein in tissues of normal mice and in defined sub-populations of cells prepared from lymphoid tissues using flow cytometry. It was also used to assess the expression of GIMAP1 protein after infection and/or immunization of mice with P. chabaudi. Real-time PCR analysis was employed to measure the expression of GIMAP1 for comparison with the protein level analysis. Results GIMAP1 protein expression was detected in all lineages of lymphocytes (T, B, NK), in F4/80+ splenic macrophages and in some lymphoid cell lines. Additional evidence is presented suggesting that the strong expression by mature B cells of GIMAP1 and other GIMAP genes and proteins seen in mice may be a species-dependent characteristic. Unexpectedly, no increase was found in the expression of GIMAP1 in P. chabaudi infected mice at either the mRNA or protein level, and this remained so despite applying a number of variations to the protocol. Conclusion The model of up-regulation of GIMAP1 in response to infection/immunization with P. chabaudi is not a robustly reproducible experimental system. The GIMAP1 protein is widely expressed in lymphoid cells, with an interesting increase in expression in the later stages of B cell development. Alternative approaches will be required to define the functional role of this GTPase in immune cells.

We present the first measurement of photoproduction of J/psi and of two-photon production of high-mass e(+)e(-) pairs in electromagnetic (or ultra-peripheral) nucleus-nucleus interactions, using Au + Au data at root s(NN) = 200 GeV. The events are tagged with forward neutrons emitted following Coulomb excitation of one or both Au* nuclei. The event sample consists of 28 events with m(e+e-) > 2 GeV/c(2) with zero like-sign background. The measured cross sections at midrapidity of d sigma/dy (J/psi + Xn, y = 0) = 76 +/- 33 (stat) +/- 11 (syst) pb and d(2)sigma /dm dy (e(+) e(-) + Xn, y = 0) = 86 +/- 23(stat) +/- 16(syst) mu b/ (GeV/c(2)) for m(e+e-) epsilon vertical bar 2.0, 2.8 vertical bar GeV/c(2) have been compared and found to be consistent with models for photoproduction of J/psi and QED based calculations of two-photon production of e(+)e(-) pairs. PHENIX has measured the electron-positron pair mass spectrum from 0 to 8 GeV/c(2) in p + p collisions at root s = 200 GeV. The contributions from light meson decays to e(+)e(-) pairs have been determined based on measurements of hadron production cross sections by PHENIX. Within the systematic uncertainty of similar to 20% they account for all e(+)e(-) pairs in the mass region below similar to 1 GeV/c(2). The e(+)e(-) pair yield remaining after subtracting these contributions is dominated by semileptonic decays of charmed hadrons correlated through flavor conservation. Using the spectral shape predicted by PYTHIA, we estimate the charm production cross section to be 544 +/- 39(stat) +/- 142(syst) +/- 200(model) pb. which is consistent with QCD calculations and measurements of single leptons by PHENIX. Algal blooms are a management concern in shallow water bodies. This project investigated the use of artificial substrates to enhance biofilm growth and shift primary production from the open water to artificial surfaces. This resulted in a shift from algal dominated wetland back to a clear water macrophyte dominated wetland.

The thesis examined systemic factors that placed children in stepfamilies at greater risk of physical abuse and neglect by comparing differences between biological- and step-families on a number of known risk factors. A model describing the pathway of child abuse and neglect for children living in stepfamilies was developed to aid in prevention.The portfolio presents four case studies which demonstrate the impact of disruptions to attachment experiences and the sequelae on psychological functioning and how the associated affect dysregulation may be linked to attachment disruptions.

While the phenomenon of sexual fantasy has been researched extensively, little contemporary inquiry has investigated the structural properties of sexual fantasy within the context of sexual offending. In this study, a qualitative analysis was used to develop a descriptive model of the phenomena of sexual fantasy during the offence process. Twenty-four adult males convicted of sexual offences provided detailed retrospective descriptions of their thoughts, emotions and behaviours—before, during and after their offences. A data-driven approach to model development, known as Grounded Theory, was undertaken to analyse the interview transcripts. A model was developed to elucidate the structural properties of sexual fantasy in the process of sexual offending, as well as the physiological and psychological variables associated with it. The Sexual Fantasy Structural Properties Model (SFSPM) comprises eight categories that describe various properties of sexual fantasy across the offence process. These categories are: origin, context, trigger, perceptual modality, clarity, motion, intensity and emotion. The strengths of the SFSPM are discussed and its clinical implications are reviewed. Finally, the limitations of the study are presented and future research directions discussed.

The apical cytoplasm of airway epithelium (AE) contains abundant labile zinc (Zn) ions that are involved in the protection of AE from oxidants and inhaled noxious substances. A major question is how dietary Zn traffics to this compartment. In rat airways, in vivo selenite autometallographic (Se-AMG)-electron microscopy revealed labile Zn-selenium nanocrystals in structures resembling secretory vesicles in the apical cytoplasm. This observation was consistent with the starry-sky Zinquin fluorescence staining of labile Zn ions confined to the same region. The vesicular Zn transporter ZnT4 was likewise prominent in both the apical and basal parts of the epithelium both in rodent and human AE, although the apical pools were more obvious. Expression of ZnT4 mRNA was unaffected by changes in the extracellular Zn concentration. However, levels increased 3-fold during growth of cells in air liquid interface cultures and decreased sharply in the presence of retinoic acid. When comparing nasal versus bronchial human AE cells, there were significant positive correlations between levels of ZnT4 from the same subject, suggesting that nasal brushings may allow monitoring of airway Zn transporter expression. Finally, there were marked losses of both basally-located ZnT4 protein and labile Zn in the bronchial epithelium of mice with allergic airway inflammation. This study is the first to describe co-localization of zinc vesicles with the specific zinc transporter ZnT4 in airway epithelium and loss of ZnT4 protein in inflamed airways. Direct evidence that ZnT4 regulates Zn levels in the epithelium still needs to be provided. We speculate that ZnT4 is an important regulator of zinc ion accumulation in secretory apical vesicles and that the loss of labile Zn and ZnT4 in airway inflammation contributes to AE vulnerability in diseases such as asthma.

Poli(butadieno) foi reticulado, em solução, com 4,4'-(4,4'-difenilmetileno)-bis-1 ,2,4-triazolina- 3, 5 - diona (BPMTD). Para monitoramento da reação foi medida a absorbância de uma transição n -> pi * do sistema 1,2,4-triazolina-3,5-diona. Segundo os estudos realizados, para baixas conversões a reação segue um esquema cinético de pseudo-primeira ordem e o processo não é controlado pela difusão, pois independe da viscosidade inicial do sistema. Para altas conversões, o processo torna-se dependente do peso molecular e da concentração do polímero inicial, devido à ocorrência de uma difusão impedida a nível de segmentos de cadeia. Estudos preliminares da reação de reticulação da borracha butílica com a BPMTD mostraram que, como o número de ligações duplas por cadeia polimérica é menor neste caso, ocorre formação de um menor número de ligações intramoleculares e a quantidade de BPMTD necessária para que ocorra gelificação é muito menor do que no caso do poli(butadieno).

Studying the physical environment of a watershed is the basic condition for a successful planning of the riparian forest preservation, and for water production and conservation. The aims of the present study were to analyze and quantify the spatial and temporal evolution (1984 and 2010) using Landsat-5 satellite images of Cintra Stream sub-watershed, Botucatu, São Paulo State, Brazil, processed by the software IDRISI Andes, as well as to analyze the water quality through the parameters pH, EC, DO and BOD5 at 4 different sites in the years 1999, 2008 and 2009. Considering the 1076.48ha area of the sub-watershed, the pasture class of 1984 was reduced by 25.55% in 2010, resulting in an increase in the remaining classes. The most important class was native forest and reforestation since it had an increase of 5.08%, which indicates recovery of the riparian forest. Degraded areas were identified close to the inferior limit of the sub-watershed (P3 and P4), as well as local contamination (P1 and P2) with worsening of the water quality in the remaining sites in the periods 2008 and 2009. Recovery and management of the ecological succession of degraded areas and water quality monitoring at 1 and 2 sites will be necessary to reestablish the natural condition of the area studied.

O trabalho em questão, desenvolvido em um assentamento de trabalhadores rurais no município de Parauapebas/PA, objetivou: (01) identificar, no assentamento Palmares, os principais problemas decorrentes da implantação de um modelo de reprodução econômica, agroindustrial, estranho à região sudeste do Pará (importado do sul do país); (02) verificar alguns resultados, principalmente econômicos, mas também sociais, decorrentes da atual estrutura de organização da produção existente no referido assentamento e, finalmente; (03) analisar o suposto conflito que há entre o modelo de produção coletiva, proposto pelo MST, e a reprodução econômica individualizada, priorizada pelos agricultores. Foi identificado que os maiores entraves na tentativa de inserção forçada, de um modelo econômico que não contemplava as diferenças infra-estruturais e tecnológicas entre estas duas regiões, apresentaram-se na falta de conhecimentos técnicos e administrativos por parte da quase totalidade dos assentados, bem como dos dirigentes da Associação de Produção e Comercialização do Assentamento Palmares. Em relação aos principais resultados econômicos obtidos, verificou-se que as culturas temporárias mais trabalhadas são: arroz, milho e mandioca (comercializada in natura e na forma de farinha), comuns na região. No que se refere ao conflito entre a produção coletiva e a reprodução individualizada, observouse que a estrutura de organização e gerenciamento dos agricultores precisa ser melhor desenvolvida, para que este modelo econômico coletivo realmente tenha êxito. O trabalho considerou um universo de 517 famílias, oficialmente cadastradas pelo Instituto Nacional de Colonização e Reforma Agrária, sendo abordadas um total de 105 pessoas responsáveis pelas famílias e, como forma de garantir a representatividade do estudo, tendo sido entrevistadas no mínimo uma e no máximo três famílias por cada núcleo de produção. Foram identificados 38 núcleos em funcionamento, quantidade encontrada após levantamento junto à Associação e aos coordenadores destes núcleos, aplicando-se questionário a todos estes dirigentes locais. Como um dos resultados da pesquisa foi verificado que mais de 95% das famílias, vinculadas a algum dos núcleos de produção, trabalham unicamente de forma individualizada e que, quase 50% dos coordenadores de núcleo informaram que não existe, ou existe pouquíssima cooperação entre os componentes das estruturas que dirigem. O estudo indica ainda ser necessário avançar nas investigações aqui apresentadas, principalmente na definição e na construção de modelos econômicos que possam acomodar em seu interior tanto atividades produtivas individuais, quanto coletivas, ou de cooperação no campo.

Background: Heavy-flavor production in p + p collisions is a good test of perturbative-quantum-chromodynamics (pQCD) calculations. Modification of heavy-flavor production in heavy-ion collisions relative to binary-collision scaling from p + p results, quantified with the nuclear-modification factor (R-AA), provides information on both cold-and hot-nuclear-matter effects. Midrapidity heavy-flavor R-AA measurements at the Relativistic Heavy Ion Collider have challenged parton-energy-loss models and resulted in upper limits on the viscosity-entropy ratio that are near the quantum lower bound. Such measurements have not been made in the forward-rapidity region. Purpose: Determine transverse-momentum (p(T)) spectra and the corresponding R-AA for muons from heavy-flavor meson decay in p + p and Cu + Cu collisions at root s(NN) = 200 GeV and y = 1.65. Method: Results are obtained using the semileptonic decay of heavy-flavor mesons into negative muons. The PHENIX muon-arm spectrometers measure the p(T) spectra of inclusive muon candidates. Backgrounds, primarily due to light hadrons, are determined with a Monte Carlo calculation using a set of input hadron distributions tuned to match measured-hadron distributions in the same detector and statistically subtracted. Results: The charm-production cross section in p + p collisions at root s = 200 GeV, integrated over p(T) and in the rapidity range 1.4 < y < 1.9, is found to be d(sigma e (e) over bar)/dy = 0.139 +/- 0.029 (stat)(-0.058)(+0.051) (syst) mb. This result is consistent with a perturbative fixed-order-plus-next-to-leading-log calculation within scale uncertainties and is also consistent with expectations based on the corresponding midrapidity charm-production cross section measured by PHENIX. The R-AA for heavy-flavor muons in Cu + Cu collisions is measured in three centrality bins for 1 < p(T) < 4 GeV/c. Suppression relative to binary-collision scaling (R-AA < 1) increases with centrality. Conclusions: Within experimental and theoretical uncertainties, the measured charm yield in p + p collisions is consistent with state-of-the-art pQCD calculations. Suppression in central Cu + Cu collisions suggests the presence of significant cold-nuclear-matter effects and final-state energy loss.

We report STAR measurements of the longitudinal double-spin asymmetry A(LL), the transverse singlespin asymmetry A(N), and the transverse double-spin asymmetries A(Sigma) and A(TT) for inclusive jet production at mid-rapidity in polarized p + p collisions at a center-of-mass energy of root s = 200 GeV. The data represent integrated luminosities of 7.6 pb(-1) with longitudinal polarization and 1.8 pb(-1) with transverse polarization, with 50%-55% beam polarization, and were recorded in 2005 and 2006. No evidence is found for the existence of statistically significant jet A(N), A(Sigma), or A(TT) at mid-rapidity. Recent model calculations indicate the A(N) results may provide new limits on the gluon Sivers distribution in the proton. The asymmetry A(LL) significantly improves the knowledge of gluon polarization in the nucleon.

Neutral-pion pi(0) spectra were measured at midrapidity (vertical bar y vertical bar < 0.35) in Au + Au collisions at root s(NN) = 39 and 62.4 GeV and compared with earlier measurements at 200 GeV in a transverse-momentum range of 1 < p(T) < 10 GeV/c. The high-p(T) tail is well described by a power law in all cases, and the powers decrease significantly with decreasing center-of-mass energy. The change of powers is very similar to that observed in the corresponding spectra for p + p collisions. The nuclear modification factors (RAA) show significant suppression, with a distinct energy, centrality, and p(T) dependence. Above p(T) = 7 GeV/c, R-AA is similar for root sNN = 62.4 and 200 GeV at all centralities. Perturbative-quantum-chromodynamics calculations that describe R-AA well at 200 GeV fail to describe the 39 GeV data, raising the possibility that, for the same p(T) region, the relative importance of initial-state effects and soft processes increases at lower energies. The p(T) range where pi(0) spectra in central Au + Au collisions have the same power as in p + p collisions is approximate to 5 and 7 GeV/c for root sNN = 200 and 62.4 GeV, respectively. For the root sNN = 39 GeV data, it is not clear whether such a region is reached, and the x(T) dependence of the x(T)-scaling power-law exponent is very different from that observed in the root sNN = 62 and 200 GeV data, providing further evidence that initial-state effects and soft processes mask the in-medium suppression of hardscattered partons to higher p(T) as the collision energy decreases.
