Four trials of identical experimental design were conducted to determine the effects of temperature, dietary Lys level, and dietary Arg:Lys ratios on performance and carcass yield of male broilers. Birds of a commercial strain were grown from 21 to 42 d of age in wire-floored finishing batteries placed in environmental chambers. The chambers were programmed to provide either a constant thermoneutral temperature (21.1 C), a constant cold temperature (15.5 C), or a cycling hot diurnal temperature (25.5 to 33.3 C). Within each environment there was a factorial arrangement of three Lys levels (1.0, 1.1, and 1.2%) with four Arg:Lys ratios (1.1:1, 1.2:1, 1.3:1, and 1.4:1). Environmental temperature significantly influenced virtually every characteristic examined. Hot cyclic temperatures reduced weight gain, feed intake, and breast meat yield, and increased feed conversion, dressing percentage, leg quarter yield, and abdominal fat content. The cold environment promoted increased feed intake and mortality. Ascites and cardiomyopathy were the leading causes of death under cold exposure and thermoneutral conditions, whereas complications arising from heat exposure were the main cause of death under hot cyclic conditions. Levels of Lys affected leg quarter yield and abdominal fat content over all environments but increased breast meat yield only under cold conditions. Increasing Arg: Lys ratios improved feed conversion and dressing percentage and reduced abdominal fat content; it could not be determined whether these responses were consistent with Arg per se or were due to a nonspecific N response. As increasing Lys levels or Arg:Lys ratios did not improve weight gain, increase breast meat yield, or attenuate adverse effects due to heat or cold exposure, it is concluded that the levels of Lys and Arg suggested for 21 to 42 d by the NRC are adequate for birds of this age under the environmental conditions encountered.

Pigs are quite sensitive to high environmental temperatures and the thermoregulation mechanisms represent great expenses in energy for heating loss, reducing animal well-being and production performance, and altering carcass quality. The aim of this study was to assess the effects of sex and dietary energy level in growing-finishing pigs submitted to characteristic seasonal variation of temperature in subtropical humid climate, and to propose a mathematical model to predict growth performance and carcass characteristics. Twenty-eight crossbred growing-finishing pigs were randomly allotted to twelve treatments, in a 2x2x3 factorial trial (2 sex; 2 environmental conditions, and 3 energy levels). Heat stress condition (climatic chamber) showed temperatures of 31 oC at 7:00 and 22 oC at 17:00 (maximum of 33 °C) and thermal comfort condition (stall) showed temperatures of 18 °C at 7:00 and 24 °C (maximum of 27 °C). Pigs were fed ad libitum with diets containing 12.2 (low), 13.6 (medium) and 15.0 (high) MJ ME/ kg DM. Voluntary feed intake, daily weight gain, and final body weight were higher (P<0.01) at thermal comfort condition and were influenced by sex (P<0.01) in growing pigs. Feed to gain ratio decreased as the energy level increased (P<0.01), with values of 2.67, 2.59, and 2.32 (12.2, 13.6, and 15.0 MJ ME/kg DM, respectively). There was energy level and sex interaction only for daily weight gain. Regarding finishing pigs, environmental conditions also showed effects (P<0.01) on voluntary feed intake, daily weight gain, and final body weight. Performance of pigs was better at thermal comfort condition. Feed to gain ratio values were 3.55, 3.42, and 2.95 for low, medium, and high energy level, respectively. Interactions between energy level and sex were observed for voluntary feed intake, daily weight gain, and final body weight (P<0.05). Carcass yield and quality were affected by environmental condition and dietary energy level. Both hot and cold carcass weight increased as energy of ration increased. Cold carcass weight increased by 1.142 kg/MJ EM whereas backfat thickness was up to 252 mm/MJ EM. Longissimus thoracis muscle thickness was around 16 mm smaller in pigs under heat stress, but lean content was 2.68% higher in those animals. Regression equations were proposed to predict the performance values in the different situations studied.

In the United States, composites accounted for nearly 70% of the 173.2 million composite and amalgam restorations placed in 2006 (Kingman et al., 2012), and it is likely that the use of composite will continue to increase as dentists phase out dental amalgam. This trend is not, however, without consequences. The failure rate of composite restorations is double that of amalgam (Ferracane, 2013). Composite restorations accumulate more biofilm, experience more secondary decay, and require more frequent replacement. In vivo biodegradation of the adhesive bond at the composite-tooth interface is a major contributor to the cascade of events leading to restoration failure. Binding by proteins, particularly gp340, from the salivary pellicle leads to biofilm attachment, which accelerates degradation of the interfacial bond and demineralization of the tooth by recruiting the pioneer bacterium Streptococcus mutans to the surface. Bacterial production of lactic acid lowers the pH of the oral microenvironment, erodes hydroxyapatite in enamel and dentin, and promotes hydrolysis of the adhesive. Secreted esterases further hydrolyze the adhesive polymer, exposing the soft underlying collagenous dentinal matrix and allowing further infiltration by the pathogenic biofilm. Manifold approaches are being pursued to increase the longevity of composite dental restorations based on the major contributing factors responsible for degradation. The key material and biological components and the interactions involved in the destructive processes, including recent advances in understanding the structural and molecular basis of biofilm recruitment, are described in this review. Innovative strategies to mitigate these pathogenic effects and slow deterioration are discussed.

The literature reports dealing with the dietary electrolyte ratio (K+Cl)/ Na are rare, although the concept has been proposed by Mongin in 1981. Thus, its application appears to be limited as a nutritional strategy in feed formulation, which usually meets only the minimum nutritional recommendations in Na, K and Cl. The objective of this study was to evaluate the performance of broilers submitted to different dietary electrolyte balances (DEB) Na+K-Cl and dietary electrolyte ratio (DER) from 1 to 21 d of age. A total of 1575 male 1-d old broiler chicks were randomly assigned to 5 treatments with 9 replicates of 35 chicks each. The treatments consisted of diets with 5 relation of electrolytes, arising from combinations DEB/DER 150/3, 250/2, 250/3, 250/4 and 350/3, with use of NaCl, NaHCO3, KCl, K2SO4 and CaCl2. All diets were corn-soybean meal based and formulated to meet or exceed the NRC (1994) requirements. Chicks had ad libitum access to feed and water in floor pens with wood shavings as litter. Body weight, feed intake and feed conversion ratio were measured at 21 d of age. It was found that only the feed conversion was significantly affected (P = 0.0142) by the combinations of relation of electrolytes (DEB and DER). The supplementation levels in DEB and DER were made to fit the data into a surface analysis to allow increasing levels of DEB (150–350 mEq/kg) and narrow and broad ratio of DER (2–4). For the canonical analysis of response surface was obtained the stationary point for body weight (DEB = 255.77 mEq/kg, and DER = 2.73:1) the value was 942.02 g; for feed intake (DEB = 251.69 mEq / kg and DER = 3.51:1), the value was 1200.02 g; and for feed conversion (DEB = 254.62 mEq/kg and DER = 3.06:1) the value was 1.35. The results of this experiment indicated that the best performance was obtained with combinations of relation of electrolytes for DEB between 251 and 255 mEq/kg and DER between 2.73:1 and 3.5:1.

Studies have suggested a relationship between drug abuse and compulsive behaviors. The present experiments investigated the relationship between schedule-induced polydipsia (SIP) and self-administration (SA) of ethanol and sucrose. SIP served as a model of compulsive behavior. and oral self-administration on a progressive-ratio (PR) schedule of reinforcement assessed the reinforcing value of either a 10% ethanol solution or an isocaloric sucrose solution. Rats first were exposed to PR sessions in which break points were the dependent variable and then switched to SIP sessions. with number of licks as the dependent variable. Results showed a positive relationship between PR and SIP for sucrose but not for ethanol: higher and lower PRs for sucrose were associated with higher and lower SIP levels. The order of the sessions then was reversed, such that SIP sessions were run before PR sessions. An opposite relationship was observed in which high and low SIP animals exhibited low and high PR break points, respectively. The relationship between SIP and SA was dependent on the reinforcing value of the substance and on prior SIP exposure. These results may reflect a common dopaminergic substrate and suggest that prior experience in coping with stress may reduce vulnerability to substance abuse behavior. Background: The prevalence of overweight individuals has increased in recent years. Moreover, the importance of a healthy diet is associated with the practice of physical activity and attempt to verify the achievement of physical exercise influences on food choice. However, it relationship between food intake and physical activity have not been studied. Aim: To evaluate if the period in which the trainings are conducted, morning and nocturne, interfere qualitatively and quantitatively in food consumption as well as verify possible associations between anthropometric profiles and dietary habits. Methods: We collected data from 33 adult volunteers, between men and women, practitioners of bodybuilding. Results: A total of 33 volunteers were interviewed (18 (54.5%) were men and 15 (45.5%) were women). Regarding anthropometric data, it turns out that that the volunteers of the two periods had similar characteristics, differentiating only weight. The consumption of nutritional supplements was observed in 30.77% of the practitioners in the morning period vs. 35% for the nocturne. Considering macronutrient intake, there was a significant difference in the consumption of protein between the periods. The consumption during nocturne period was greater (126 +/- 5% of the daily requirement) than the morning period (115.7 +/- 2%). As for micro-nutrients, calcium intake was greater among men when compared to women. There was a positive correlation between the BMI, and arm circumference for practitioners of the morning period. Conclusion: This study show that the practitioners who train in the morning have quietly better eating habits than those in the nocturne period, however both are inappropriate.

The cardiac voltage-gated Na(+) channel Na(v)1.5 generates the cardiac Na(+) current (INa). Mutations in SCN5A, the gene encoding Na(v)1.5, have been linked to many cardiac phenotypes, including the congenital and acquired long QT syndrome, Brugada syndrome, conduction slowing, sick sinus syndrome, atrial fibrillation, and dilated cardiomyopathy. The mutations in SCN5A define a sub-group of Na(v)1.5/SCN5A-related phenotypes among cardiac genetic channelopathies. Several research groups have proposed that Na(v)1.5 may be part of multi-protein complexes composed of Na(v)1.5-interacting proteins which regulate channel expression and function. The genes encoding these regulatory proteins have also been found to be mutated in patients with inherited forms of cardiac arrhythmias. The proteins that associate with Na(v)1.5 may be classified as (1) anchoring/adaptor proteins, (2) enzymes interacting with and modifying the channel, and (3) proteins modulating the biophysical properties of Na(v)1.5 upon binding. The aim of this article is to review these Na(v)1.5 partner proteins and to discuss how they may regulate the channel's biology and function. These recent investigations have revealed that the expression level, cellular localization, and activity of Na(v)1.5 are finely regulated by complex molecular and cellular mechanisms that we are only beginning to understand.

Although death receptors and chemotherapeutic drugs activate distinct apoptosis signaling cascades, crosstalk between the extrinsic and intrinsic apoptosis pathway has been recognized as an important amplification mechanism. Best known in this regard is the amplification of the Fas (CD95) signal in hepatocytes via caspase 8-mediated cleavage of Bid and activation of the mitochondrial apoptosis pathway. Recent evidence, however, indicates that activation of other BH3-only proteins may also be critical for the crosstalk between death receptors and mitochondrial triggers. In this study, we show that TNF-related apoptosis-inducing ligand (TRAIL) and chemotherapeutic drugs synergistically induce apoptosis in various transformed and untransformed liver-derived cell lines, as well as in primary human hepatocytes. Both, preincubation with TRAIL as well as chemotherapeutic drugs could sensitize cells for apoptosis induction by the other respective trigger. TRAIL induced a strong and long lasting activation of Jun kinase, and activation of the BH3-only protein Bim. Consequently, synergistic induction of apoptosis by TRAIL and chemotherapeutic drugs was dependent on Jun kinase activity, and expression of Bim and Bid. These findings confirm a previously defined role of TRAIL and Bim in the regulation of hepatocyte apoptosis, and demonstrate that the TRAIL-Jun kinase-Bim axis is a major and important apoptosis amplification pathway in primary hepatocytes and liver tumor cells.
Plectin interacts with the rod domain of type III intermediate filament proteins desmin and vimentin

Plectin is a versatile cytolinker protein critically involved in the organization of the cytoskeletal filamentous system. The muscle-specific intermediate filament (IF) protein desmin, which progressively replaces vimentin during differentiation of myoblasts, is one of the important binding partners of plectin in mature muscle. Defects of either plectin or desmin cause muscular dystrophies. By cell transfection studies, yeast two-hybrid, overlay and pull-down assays for binding analysis, we have characterized the functionally important sequences for the interaction of plectin with desmin and vimentin. The association of plectin with both desmin and vimentin predominantly depended on its fifth plakin repeat domain and downstream linker region. Conversely, the interaction of desmin and vimentin with plectin required sequences contained within the segments 1A-2A of their central coiled-coil rod domain. This study furthers our knowledge of the interaction between plectin and IF proteins important for maintenance of cytoarchitecture in skeletal muscle. Moreover, binding of plectin to the conserved rod domain of IF proteins could well explain its broad interaction with most types of IFs.
