低能离子束的诱变效应首先由我国科学家发现并将其广泛应用于育种实践,但是离子注入诱导DNA变异的研究结果主要是以微生物离体质粒DNA为材料获得的,以活体高等生物为材料的研究尚未见报道。 我们以30 keV N+(注入剂量80×1015 ions/cm2)注入拟南芥后获得的稳定突变体T80II为实验材料,对突变体植株进行了RAPD标记,并将T80II和对照部分RAPD特异条带进行克隆测序和DNA序列分析。结果显示,在可分辨的总计397个RAPD条带中,T80II株系中有52个条带表现出差异,包括条带的缺失和增加,条带变异率为13.1%;克隆的T80II序列中,平均每16.8个碱基出现一个碱基变异位点,表现出较高频率的碱基突变。碱基突变的类型包括碱基的颠换、转换、缺失、插入等。在检测到的275个碱基突变中,主要是单碱基置换(97.09%),碱基缺失或者插入的比例较小(2.91%)。在碱基置换中,转换的频率(66.55%)高于颠换的频率((30.55%)。此外,构成DNA的四种碱基均可以被离子束辐照诱发变异,而且每一种碱基都可以被其它三种碱基所替换,但是胸腺嘧啶(T)的辐射敏感性要高于其它三种碱基。通过分析突变碱基周边序列,对低能N+离子注入拟南芥突变体引发的碱基突变热点进行了讨论。 另外,低能离子注入诱变获得的突变体特异表达基因的克隆方面也没有报道。我们以突变体T80II作为实验材料,用PCR增效的减法杂交技术构建了T80II特异表达的cDNA减法文库,克隆特异表达的cDNA片段,并对其中1个与14-3-3 protein GF14 nu (GRF7) gene有部分同源性、长712 bp的cDNA片段进行了讨论。我们的研究证明通过减法杂交技术克隆低能离子诱发的突变体特异表达的cDNA是可能的,这为低能离子注入技术在分子生物学上的应用开辟了一个新思路。

Previous studies have shown that gonads were the second target organ of microcystins (MCs), and that MCs exposure exerted obvious toxic effects on male reproductive system of mammals. However, relevant molecular evidences are still lacking. Fas-signaling pathway plays a key role in toxicant-induced germ cell apoptosis. This study was to evaluate the responses of Fas/FasL system related genes and proteins in testes of rats injected intravenously with MCs. Enhanced apoptosis of germ cells in the testes of MCs-treated rats was detected by the terminal deoxynucleotidyl transferase-mediated deoxy-UTP nick end labeling (TUNEL) associated with up-regulation of the Fas/FasL system. Both Fas and FasL protein expression were induced evidently from I h post-injection, and this high expression level maintained throughout the experiment. In addition, the activation of caspase-8 and caspase-3 protein was also observed, which were indicators of apoptosis. These results suggested the likely involvement of Fas/FasL system in the MCs-induced germ cell apoptosis. It is also suggested that MCs can cause damage to Sertoli cells directly. Extracellular polymeric substances (EPS) from four filamentous cyanobacteria Microcoleus vaginatus, Scytonema javanicum, Phormidium tenue and Nostoc sp. and a coccoid single-cell green alga Desmococcus olivaceus that had been separated from desert algal crusts of Tegger desert of China, were investigated for their chemical composition, structure,and physical properties. The EPS contained 7.5-50.3% protein (in polymers ranging from 14 to more than 200 kD, SDS-PAGE) and 16.2-46.5% carbohydrate (110-460 kD, GFC). 6-12 kinds of monosaccharides, including 2-O-methyl rhamnose, 2-O-methyl glucose, and N-acetyl glucosamine were found. The main carbohydrate chains from M. vaginatus and S. javanicum consisted mainly of equal proportion of Man, Gal and Glc, that from P. tenue consisted mainly of arabinose, glucose and rhamnose. Arabinose was present in pyranose form, mainly alpha-L 1 --> 3 linked, with branches on C4 of almost half of the units. Glucose was responsible for the terminal units, in addition of having some units as beta1 --> 3 and some as beta1 --> 4 linked. Rhamnose was mainly 1 --> 3 linked with branches on C2 on half of the units. The carbohydrate polymer from D. olivaceus was composed mainly of beta1 --> 4 linked xylose, galactose and glucose. The galactose part was present both in beta-pyranose and -furanose forms. Arabinose in alpha-L-furanose form was mainly present as 1 --> 2 and 1 --> 2, 5 linked units, rhamnose only as alpha 1 --> 3 and xylose as beta 1 --> 4. The backbone of the polysaccharide from Nostoc sp. was composed of beta-1 --> 4 linked xylose, galactose and glucose. Most of the glucose was branched on position C6, terminal glucose and 2-O-methyl glucose units are also present. 从四川攀枝花钒铁矿周围的土壤中分离出一株具有较高乙炔还原活性的固氮菌,经鉴定为棕色固氮菌(Azotobacter vinelandii)。在Asby's选择性培养基上产生黄绿色萤光色素,还原乙炔,当培养基中加入lUMN_aVO_3·2H_2O时生长旺盛,产生深绿色萤光,具有较高的乙炔还原作用,实验表明,该菌株含有第二固氮酶,用Tn5对该菌进行接合诱变,获得了大量突变株,但进一步筛选nifH-突变株遇到了困难,可能是由于接合诱变效率太低的原因。用含有克氏肺炎杆菌nifH基因的质粒PPC1201对该菌进行定点整合诱变,得到了29株抗氨苄菁老素的菌株。进一步乙炔还原活性测定表明,其中可能含有14株第一固氮酶nifH基因突变的菌株,10株在第二固氮酶nifH突变的菌株,2株可能在第三固氮酶的nifH-like基因或在三个固氮酶结构基因之处的区域突变的菌株,3株可能在第一、第二固氮酶所共有的基因区域发生突变的菌株。

本文通过胶州湾围隔实验、微藻培养实验研究了海洋微藻产生溶解有机物的三维荧光特性,并初步利用经验正交函数对三维荧光光谱图进行了主成分分析,通过东海取样研究了东海荧光溶解有机物的时空分布特征,并对荧光溶解有机物的来源进行了分析。得到了一些初步结论: 胶州湾围隔实验中不同营养盐培养结果表明浮游植物可产生类蛋白和类腐殖质荧光,类蛋白荧光峰由类酪氨酸(tyrosine-like)荧光峰和类色氨酸(tryptophan-like)荧光峰组成,主要位置为Exmax/Emmax=270nm/290~310nm,Exmax/Emmax= 270~290/320~350的荧光峰强度比较弱;在Exmax/Emmax=250~260/380~480nm(A峰)、Exmax/Emmax=310~320/380~420nm(C峰)和Exmax/Emmax=330~350/420~480nm(M峰)位置均出现零散的类腐殖质荧光峰,其中以A峰为主。类酪氨酸荧光强度明显高于类腐殖质荧光强度。浮游植物量降低时,类酪氨酸荧光强度与叶绿素a浓度呈明显的负相关。硅藻和甲藻产生的类酪氨酸和类色氨酸荧光强度之间具有较好的相关性,两者来源相似, 并且甲藻与硅藻相比能够产生更多的类蛋白荧光物质。不同环境下类腐殖质混合物的组分比例不同,甲藻生长环境下相对于硅藻具有较低的A:C比值。 在实验室培养中肋骨条藻(Skeletonema costatum)、塔玛亚历山大藻(Alexandrium tamarense)、微小亚历山大藻(Alexandrium mimutum)、锥状斯氏藻(Scrippsiella trochoidea)、东海原甲藻(Prorocentrum donghaiense)及海洋原甲藻(Prorocentrum micans)的实验结果表明,微藻在生长过程中产生出荧光溶解有机物,中肋骨条藻为代表的硅藻主要产生类腐殖质荧光物质,而甲藻在指数增长期主要产生类蛋白荧光物质。进入消亡期后类蛋白荧光和类腐殖质荧光强度迅速增大,原因可能是衰老、死亡藻细胞的破碎释放出大量的荧光有机物质所致,此外还有细菌对非荧光有机物进一步降解。塔玛亚历山大藻、微小亚历山大藻、东海原甲藻及海洋原甲藻的类蛋白荧光强度在消亡后期由于细菌降解或光降解等因素而降低。同属微藻产生的荧光物质相似,例如塔玛亚历山大藻与微小亚历山藻、东海原甲藻与海洋原甲藻,但具体荧光峰位置有所不同。利用经验正交函数能够对三维荧光光谱谱图进行主成份分析。 在利用三维荧光光谱法研究长江口海域台风前后不同站位荧光溶解有机物荧光特性及分布特征的结果表明,长江口海区主要的荧光溶解有机物荧光峰为T峰、S峰和A峰。风前和风后的类色氨酸分别来源于相似物质。风前,在表层浮游植物能够产生类蛋白荧光物质,而底层类蛋白荧光物质不受浮游植物的影响,长江冲淡水能够带来部分类蛋白荧光物质;表层的类腐殖质不受浮游植物的影响,而底层的浮游植物在降解过程中能够产生一部分类腐殖质,并且长江冲淡水对表层和底层的类腐殖质来源均有很大贡献。风后,表层的类色氨酸与叶绿素a浓度不呈相关性,而底层却呈正相关,另外表层和底层的类色氨酸均受到长江冲淡水的影响;表层和底层的类腐殖质与叶绿素a均没有相关性,但受陆源影响显著,长江冲淡水能带来类腐殖质。

Thymic graft-versus-host disease (tGVHD) can contribute to profound T cell deficiency and repertoire restriction after allogeneic BM transplantation (allo-BMT). However, the cellular mechanisms of tGVHD and interactions between donor alloreactive T cells and thymic tissues remain poorly defined. Using clinically relevant murine allo-BMT models, we show here that even minimal numbers of donor alloreactive T cells, which caused mild nonlethal systemic graft-versus-host disease, were sufficient to damage the thymus, delay T lineage reconstitution, and compromise donor peripheral T cell function. Furthermore, to mediate tGVHD, donor alloreactive T cells required trafficking molecules, including CCR9, L selectin, P selectin glycoprotein ligand-1, the integrin subunits alphaE and beta7, CCR2, and CXCR3, and costimulatory/inhibitory molecules, including Ox40 and carcinoembryonic antigen-associated cell adhesion molecule 1. We found that radiation in BMT conditioning regimens upregulated expression of the death receptors Fas and death receptor 5 (DR5) on thymic stromal cells (especially epithelium), while decreasing expression of the antiapoptotic regulator cellular caspase-8-like inhibitory protein. Donor alloreactive T cells used the cognate proteins FasL and TNF-related apoptosis-inducing ligand (TRAIL) (but not TNF or perforin) to mediate tGVHD, thereby damaging thymic stromal cells, cytoarchitecture, and function. Strategies that interfere with Fas/FasL and TRAIL/DR5 interactions may therefore represent a means to attenuate tGVHD and improve T cell reconstitution in allo-BMT recipients.

Histidine is a naturally occurring amino acid with antioxidant properties, which is present in low amounts in tissues throughout the body. We recently synthesized and characterized histidine analogues related to the natural dipeptide carnosine, which selectively scavenge the toxic lipid peroxidation product 4-hydroxynonenal (HNE). We now report that the histidine analogue histidyl hydrazide is effective in reducing brain damage and improving functional outcome in a mouse model of focal ischemic stroke when administered intravenously at a dose of 20 mg/kg, either 30 min before or 60 min and 3 h after the onset of middle cerebral artery occlusion. The histidine analogue also protected cultured rat primary neurons against death induced by HNE, chemical hypoxia, glucose deprivation, and combined oxygen and glucose deprivation. The histidine analogue prevented neuronal apoptosis as indicated by decreased production of cleaved caspase-3 protein. These findings suggest a therapeutic potential for HNE-scavenging histidine analogues in the treatment of stroke and related neurodegenerative conditions.

The genetic diversity of liver fluke populations in three different countries from Eastern Europe (Greece, Bulgaria, and Poland) in comparison with available data from other countries was determined. Specifically, SNPs from regions of two nuclear genes, 28S rDNA, ß-tubulin 3 and an informative region of the mitochondrial genome were examined. Two major lineages for the 28S rDNA gene based on the highly polymorphic 105th nucleotide position were found. These lineages were widely and almost equally spread not only through the countries studied but also in other investigated geographical areas. Two basic lineages and additional haplotypes were defined for the mtDNA gene region, consisting of the cytochrome c oxidase subunit III gene, transfer RNA histidine gene and cytochome b gene. The basic lineages were observed within Greek, Bulgarian, and Polish Fasciola hepatica populations but the distribution of additional haplotypes differed between the populations from the three countries. For the ß-tubulin 3 gene multiple polymorphic sites were revealed but no explicit clades. The SNPs were spread unequally in all studied geographical regions with an evident distinction between the Greek and Polish specimens. Additional genotypes for the 28S rDNA region as well as haplotypes of the mtDNA region that were typical for the Greek or Polish populations were observed. Significant polymorphisms for ß-tubulin 3 gene were displayed with decreasing percentage of presence within populations from Greece to Poland. There was an amino acid substitution in ß-tubulin 3 protein found only among Polish specimens. It is hypothesized that genotypic differences between Greek, Bulgarian, and Polish liver fluke populations are due to territorial division and genetic drift in past epochs.

In ecotoxicology a major focus is in the aquatic environment, not only because it presents a great economic value to man but it is an ecosystem widely affected by the growing anthropogenic pollution. Most of the studies performed relate to adverse effects in development, reproductive or endocrine disruption but little is known about the possible effects in bone formation and skeletal development. In this study, we set out to evaluate the effects of 8 aquatic pollutants on the skeletal development using an in vivo system, the zebrafish larvae aged 20 days post-fertilization, through chronic exposure. Several endpoints were considered such as the cumulative mortality, total length, occurrence of skeletal deformities and marker gene expression. We were able to establish LD50 values for some pollutants, like 3-methylcholanthrene, lindane, diclofenac, cobalt and vanadate and found that the total length was not affected by any of the pollutants tested. Cobalt was the most harmful chemical to affect hatching time, severely affecting the ability of the zebrafish embryos to hatch and overall the number of deformities increased upon exposure to tested chemicals but no patterns of deformities were identified. We also propose that 3-methylcholanthrene has an osteogenic effect, affecting osteoblast and osteoclast function and that op levels can act as a mediator of 3-methylcholanthrene toxic stress to the osteoblast. In turn we found naphthalene to probably have a chondrogenic effect. Our results provided new insights into the potential osteotoxicity of environmental pollutants. Future studies should aim at confirming these preliminary data and at determining mechanisms of osteotoxicity.

L'hypertension artérielle est l'une des principales causes de morbidité et de mortalité dans le monde. La compréhension des mécanismes qui sont à la base du développement de l'hypertension offrira de nouvelles perspectives pour un meilleur contrôle de l'hypertension. Nous avons précédemment montré que le niveau des protéines Giα-2 et Giα-3 est augmenté chez les rats spontanément hypertendus (SHR) avant l'apparition de l'hypertension. Le traitement avec les inhibiteurs de l’enzyme de conversion de l’Angiotensine (IEC) est associé à une diminution de l’expression des protéines Gi. De plus, l'injection intrapertoneale de la toxine de la coqueluche inactive les deux protéines Giα et empêche le développement de l'hypertension chez les SHR. Cependant, la contribution spécifique des protéines Giα-2 et Giα-3 dans le développement de l'hypertension n'est pas encore connue. Dans la présente étude, l’Anti-sens oligodésoxynucléotide (AS-ODN) de Giα-2 et Giα-3 (1mg/Kg en poids corporel) encapsulé dans des liposomes cationiques PEG / DOTAP/ DOPE ont été administrés par voie intraveineuse aux SHR pré-hypertendus âgé de trois semaines et aux Wistar Kyoto (WKY) rats de même âge. Les contrôles des WKY et SHR non traités ont été injectés avec du PBS stérile, liposomes vides ou oligomères sens. La pression artérielle (PA) a été suivie chaque semaine en utilisant la technique manchon caudal. Les rats ont été sacrifiés à l'âge de six semaines et neuf semaines. Le coeur et l'aorte ont été utilisés pour étudier l'expression des protéines Gi. Le knockdown des protéines Giα-2 par l’injection de Giα-2-AS a empêché le développement de l'hypertension à l'âge de six semaines. Par la suite, la PA a commencé à augmenter rapidement et a atteint le niveau que l'on retrouve dans les groupes témoins à l'âge de neuf semaines. D'autre part, la PA du groupe traité avec le Giα-3-AS a commencé à augmenter à l'âge de quatre semaines. Dans le groupe des SHR-Giα-3-AS, la PA a augmenté à l’âgé de six semaines, mais moins que celle de SHR-CTL. Le coeur et l'aorte obtenues des SHR Giα-2-AS et Giα-3-AS à partir de l’âgé de six semaines ont eu une diminution significative de l’expression des protéines Giα-2 et Giα-3 respectivement. Dans le groupe des WKY Giα-2-AS et Giα-3-AS l'expression des protéines Giα-2 et Giα-3 respectivement a diminué malgré l'absence de changement dans la PA par rapport aux WKY CTL. À l'âge de neuf semaines, les SHR traités avec du Giα-2-AS et Giα-3-AS avaient la même PA et expression des protéines Gi que le SHR CTL. Ces résultats suggèrent que les deux protéines Giα-2 et Giα-3 sont impliqués dans le développement de l'hypertension chez les SHR, mais le knockdown de Giα-2 et pas de Giα-3 a empêché le développement de l'hypertension.

In a recent study, the serotype 3 Dearing strain of mammalian orthoreovirus was adapted to Vero cells; cells that exhibit a limited ability to support the early steps of reovirus uncoating and are unable to produce interferon as an antiviral response upon infection. The Vero cell-adapted virus (VeroAV) exhibits amino acids substitutions in both the σ1 and μ1 outer capsid proteins but no changes in the σ3 protein. Accordingly, the virus was shown not to behave as a classical uncoating mutant. In the present study, an increased ability of the virus to bind at the Vero cell surface was observed and is likely associated with an increased ability to bind onto cell-surface sialic acid residues. In addition, the kinetics of μ1 disassembly from the virions appears to be altered. The plasmid-based reverse genetics approach confirmed the importance of σ1 amino acids substitutions in VeroAV's ability to efficiently infect Vero cells, although μ1 co-adaptation appears necessary to optimize viral infection. This approach of combining in vitro selection of reoviruses with reverse genetics to identify pertinent amino acids substitutions appears promising in the context of eventual reovirus modification to increase its potential as an oncolytic virus.

Scientists throughout the world are in search of a better methodology to reduce the use of environmentally hazardous chemicals common in industries .A significant contribution in this field is given by different redox catalysts in oxidation reactions. The oxidation of organic substrates represents one of the most important industrial chemical reactions, explaining the significant efforts invested in the research and development of new heterogeneous catalysts with increased activities and selectivities in these type reactions[l-4|. Hence liquid phase reactions like epoxidation of cylcohexene and hydroxylation of phenol were carried out with a new outlook in the challenge using CeO2/TiO;; and CuO/TiO2 catalysts denoted as TiO2-Ce as TiO2-Cu respectively in this work. Also different wt% of metals incorporated titania catalysts like 3, 6, 9 wt% CeO2/TiO; and CuO/TiO;were subjected to the present study .The interaction between metal oxides and the oxide supports have attracted much attention because of the wide applications of supported metal oxide systems[7,8]. It is well known that supported oxides of transition metals are widely used as catalysts for various reactions. Titania as well its metal modified catalysts systems afford high activity and selectivity in the liquid phase epoxidation of cyclohexene[9]. Cyclohexene epoxide is obtained as the major product during the reaction with small amounts of allylic substitution products.This chapter gives an idea about the liquid phase oxidation reactions like epoxidation of cylcohexene and hydroxylation of phenol in which many industrially important products are formed. Here discusses about the redox properties of the ceria and copper incorporated titania catalysts.The epoxidation of cyclohcxene is carried out efficiently over the prepared systems with the selective formation of cyclohexane epoxide. This reaction hints that it might be possible to create cleaner nylon chemistry. The total acidity of the prepared systems plays an important role in determining the catalytic activity in the dehydrogenation of cyclohexane and cyclohexene. Lignocellulosic biomass is probably the best alternative resource for biofuel production and it is composed mainly of cellulose, hemicelluloses and lignin. Cellulose is the most abundant among the three and conversion of cellulose to glucose is catalyzed by the enzyme cellulase. Cellulases are groups of enzymes act synergistically upon cellulose to produce glucose and comprise of endoglucanase, cellobiohydrolase and β-glucosidase. β -glucosidase assumes great importance due to the fact that it is the rate limiting enzyme. Endoglucanases (EG) produces nicks in the cellulose polymer exposing reducing and non reducing ends, cellobiohydrolases (CBH) acts upon the reducing or non reducing ends to liberate cellobiose units, and β - glucosidases (BGL) cleaves the cellobiose to liberate glucose completing the hydrolysis. . β -glucosidases undergo feedback inhibition by their own product- β glucose, and cellobiose which is their substrate. Few filamentous fungi produce glucose tolerant β - glucosidases which can overcome this inhibition by tolerating the product concentration to a particular threshold. The present study had targeted a filamentous fungus producing glucose tolerant β - glucosidase which was identified by morphological as well as molecular method. The fungus showed 99% similarity to Aspergillus unguis strain which comes under the Aspergillus nidulans group where most of the glucose tolerant β -glucosidase belongs. The culture was designated the strain number NII 08123 and was deposited in the NII culture collection at CSIR-NIIST. β -glucosidase multiplicity is a common occurrence in fungal world and in A.unguis this was demonstrated using zymogram analysis. A total 5 extracellular isoforms were detected in fungus and the expression levels of these five isoforms varied based on the carbon source available in the medium. Three of these 5 isoforms were expressed in higher levels as identified by the increased fluorescence (due to larger amounts of MUG breakdown by enzyme action) and was speculated to contribute significantly to the total _- β glucosidase activity. These isoforms were named as BGL 1, BGL3 and BGL 5. Among the three, BGL5 was demonstrated to be the glucose tolerant β -glucosidase and this was a low molecular weight protein. Major fraction was a high molecular weight protein but with lesser tolerance to glucose. BGL 3 was between the two in both activity and glucose tolerance.121 Glucose tolerant .β -glucosidase was purified and characterized and kinetic analysis showed that the glucose inhibition constant (Ki) of the protein is 800mM and Km and Vmax of the enzyme was found to be 4.854 mM and 2.946 mol min-1mg protein-1respectively. The optimumtemperature was 60°C and pH 6.0. The molecular weight of the purified protein was ~10kDa in both SDS as well as Native PAGE indicating that the glucose tolerant BGL is a monomeric protein.The major β -glucosidase, BGL1 had a pH and temperature optima of 5.0 and 60 °C respectively. The apparent molecular weight of the Native protein is 240kDa. The Vmax and Km was 78.8 mol min-1mg protein-1 and 0.326mM respectively. Degenerate primers were designed for glycosyl hydrolase families 1, 3 and 5 and the BGL genes were amplified from genomic DNA of Aspergillus unguis. The sequence analyses performed on the amplicons results confirmed the presence of all the three genes. Amplicon with a size of ~500bp was sequenced and which matched to a GH1 –BGL from Aspergillus oryzae. GH3 degenerate primers producing amplicons were sequenced and the sequences matched to β - glucosidase of GH3 family from Aspergillus nidulans and Aspergillus acculateus. GH5 degenerate primers also gave amplification and sequencing results indicated the presence of GH5 family BGL gene in the Aspergillus unguis genomic DNA.From the partial gene sequencing results, specific as well as degenerate primers were designed for TAIL PCR. Sequencing results of the 1.0 Kb amplicon matched Aspergillus nidulans β -glucosidase gene which belongs to the GH1 family. The sequence mainly covered the N-Terminal region of the matching peptide. All the three BGL proteins ie. BGL1, BGL3 and BGL5 were purified by chromatography an electro elution from Native PAGE gels and were subjected to MALDI-TOF mass spectrometric analysis. The results showed that BGL1 peptide mass matched to . β -glucosidase-I of Aspergillus flavus which is a 92kDa protein with 69% protein coverage. The glucose tolerant β -glucosidase BGL5 mass matched to the catalytic C-terminal domain of β -glucosidase-F from Emericella nidulans, but the protein coverage was very low compared to the size of the Emericella nidulans protein. While comparing the size of BGL5 from Aspergillus unguis, the protein sequence coverage is more than 80%. BGL F is a glycosyl hydrolase family 3 protein.The properties of BGL5 seem to be very unique, in that it is a GH3 β -glucosidase with a very low molecular weight of ~10kDa and at the same time having catalytic activity and glucose 122 tolerance which is as yet un-described in GH β -glucosidases. The occurrence of a fully functional 10kDA protein with glucose tolerant BGL activity has tremendous implications both from the points of understanding the structure function relationships as well as for applications of BGL enzymes. BGL-3 showed similarity to BGL1 of Aspergillus aculateus which was another GH3 β -glucosidase. It may be noted that though PCR could detect GH1, GH3 and GH5 β-glucosidases in the fungus, the major isoforms BGL1 BGL3 and BGL5 were all GH3 family enzymes. This would imply that β-glucosidases belonging to other families may also co-exist in the fungus and the other minor isoforms detected in zymograms may account for them. In biomass hydrolysis, GT-BGL containing BGL enzyme was supplemented to cellulase and the performances of blends were compared with a cocktail where commercial β- glucosidase was supplemented to the biomass hydrolyzing enzyme preparation. The cocktail supplemented with A unguis BGL preparation yielded 555mg/g sugar in 12h compared to the commercial enzyme preparation which gave only 333mg/g in the same period and the maximum sugar yield of 858 mg/g was attained in 36h by the cocktail containing A. unguis BGL. While the commercial enzyme achieved almost similar sugar yield in 24h, there was rapid drop in sugar concentration after that, indicating probably the conversion of glucose back to di-or oligosaccharides by the transglycosylation activity of the BGl in that preparation. Compared this, the A.unguis enzyme containing preparation supported peak yields for longer duration (upto 48h) which is important for biomass conversion to other products since the hydrolysate has to undergo certain unit operations before it goes into the next stage ie – fermentation in any bioprocesses for production of either fuels or chemicals.. Most importantly the Aspergillus unguis BGL preparation yields approximately 1.6 fold increase in the sugar release compared to the commercial BGL within 12h of time interval and 2.25 fold increase in the sugar release compared to the control ie. Cellulase without BGL supplementation. The current study therefore leads to the identification of a potent new isolate producing glucose tolerant β - glucosidase. The organism identified as Aspergillus unguis comes under the Aspergillus nidulans group where most of the GT-BGL producers belong and the detailed studies showed that the glucose tolerant β -glucosidase was a very low molecular weight protein which probably belongs to the glycosyl hydrolase family 3. Inhibition kinetic studies helped to understand the Ki and it is the second highest among the nidulans group of Aspergilli. This has promoted us for a detailed study regarding the mechanism of glucose tolerance. The proteomic 123 analyses clearly indicate the presence of GH3 catalytic domain in the protein. Since the size of the protein is very low and still its active and showed glucose tolerance it is speculated that this could be an entirely new protein or the modification of the existing β -glucosidase with only the catalytic domain present in it. Hydrolysis experiments also qualify this BGL, a suitable candidate for the enzyme cocktail development for biomass hydrolysis

A novel topical codrug, naproxyl–dithranol (Nap-DTH), in which dithranol and naproxen are linked via an ester in a 1:1 ratio to form a single chemical entity, was synthesized. The antiproliferative, anti-inflammatory and toxic effects of Nap-DTH were assessed, at the cellular level, using various in vitro methods. Cultured HaCaT keratinocytes were treated with Nap-DTH, and the cellular effects were compared with those of the parent compounds, individually and as a 1:1 mixture of naproxen:dithranol to mimic 1:1 in situ liberation from Nap-DTH. The results demonstrate that Nap-DTH did not modify proliferation and only exhibited slight toxic effects after 24 h at concentrations >21 μM. At a lower concentration (3.4 μM), Nap-DTH did not alter cell proliferation or inflammation, which suggests that the codrug is therapeutically inert. Relating to this, the 1:1 mixture of naproxen:dithranol exhibited the lowest toxic effect and the highest antiproliferative effect on HaCaT keratinocytes compared to dithranol at the same concentration. Moreover, the 1:1 mixture exhibited a reduced inflammatory effect compared to dithranol alone, as reflected by the upregulation of cyclooxygenase-2 by 45% and 136%, respectively. In spite of the 1:1 mixture showing a greater downregulation of Ki-67 and a 2-fold reduction of proliferating cell nuclear antigen (both cellular markers of proliferation) than dithranol, dithranol showed a much greater induction of cleaved caspase-3 protein expression (upregulated by 287%, compared to 85% for the 1:1 mixture). This suggests that when dithranol was administered with naproxen, inhibition of cell growth plays a more important role in the antiproliferation effects than the induction of apoptotic cell death. These results confirm that the codrug would lead to a better therapeutic profile and fewer adverse effects compared to its parent compounds.

Araucaria angustifolia is an endangered Brazilian native conifer tree. The aim of the present work was to identify differentially expressed proteins between mature and germinated embryos of A. angustifolia, using one and two dimensional gel electrophoresis approaches followed by protein identification by tandem mass spectrometry. The identities of 32 differentially expressed protein spots from two dimensional gel maps were successfully determined, including proteins and enzymes involved in storage mobilization such as the vicilin-like storage protein and proteases. A label free approach, based on spectral counts, resulted in detection of 10 and 14 mature and germinated enriched proteins, respectively. Identified proteins were mainly related to energetic metabolism pathways, translational processes. oxidative stress regulation and cellular signaling. The integrated use of both strategies permitted a comprehensive protein expression overview of changes in germinated embryos in relation to matures, providing insights into the this process in a recalcitrant seed species. Applications of the data generated on the monitoring and control of in vitro somatic embryos were discussed. Published by Elsevier Ltd.
