Investigations on formation and specification of neural precursor cells in the central nervous system of the Drosophila melanogaster embryoSpecification of a unique cell fate during development of a multicellular organism often is a function of its position. The Drosophila central nervous system (CNS) provides an ideal system to dissect signalling events during development that lead to cell specific patterns. Different cell types in the CNS are formed from a relatively few precursor cells, the neuroblasts (NBs), which delaminate from the neurogenic region of the ectoderm. The delamination occurs in five waves, S1-S5, finally leading to a subepidermal layer consisting of about 30 NBs, each with a unique identity, arranged in a stereotyped spatial pattern in each hemisegment. This information depends on several factors such as the concentrations of various morphogens, cell-cell interactions and long range signals present at the position and time of its birth. The early NBs, delaminating during S1 and S2, form an orthogonal array of four rows (2/3,4,5,6/7) and three columns (medial, intermediate, and lateral) . However, the three column and four row-arrangement pattern is only transitory during early stages of neurogenesis which is obscured by late emerging (S3-S5) neuroblasts (Doe and Goodman, 1985; Goodman and Doe, 1993). Therefore the aim of my study has been to identify novel genes which play a role in the formation or specification of late delaminating NBs.In this study the gene anterior open or yan was picked up in a genetic screen to identity novel and yet unidentified genes in the process of late neuroblast formation and specification. I have shown that the gene yan is responsible for maintaining the cells of the neuroectoderm in an undifferentiated state by interfering with the Notch signalling mechanism. Secondly, I have studied the function and interactions of segment polarity genes within a certain neuroectodermal region, namely the engrailed (en) expressing domain, with regard to the fate specification of a set of late neuroblasts, namely NB 6-4 and NB 7-3. I have dissected the regulatory interaction of the segment polarity genes wingless (wg), hedgehog (hh) and engrailed (en) as they maintain each other’s expression to show that En is a prerequisite for neurogenesis and show that the interplay of the segmentation genes naked (nkd) and gooseberry (gsb), both of which are targets of wingless (wg) activity, leads to differential commitment of NB 7-3 and NB 6-4 cell fate. I have shown that in the absence of either nkd or gsb one NB fate is replaced by the other. However, the temporal sequence of delamination is maintained, suggesting that formation and specification of these two NBs are under independent control.

Zusammenfassung der Dissertation von Gerd Jochen ArnoldThema: Aufstieg und Fall eines Gebirges - Konvektionsmodelle zur variscischen Orogenese Diese Arbeit befaßt sich mit der Entwicklung der variscischen Gebirgswurzel. Numerische Modellierung wurden mit dem Finite-Differenzen-Programm FDCON durchgeführt. Dieses berechnet auf einem Gitter die Entwicklung von Konvektion unter Berücksichtigung von Massen-, Impuls- und Energieerhaltung. Der Modellansatz eignet sich für die Untersuchung der Orogenese unter Miteinbeziehung der thermo-mechanischen Wechselwirkung von Kruste und Mantel. Kapitel 1 beschreibt wesentliche Aspekte der variscischen Gebirgsbildung. Die tektonostratigraphische Gliederung erlaubt Rückschlüsse auf das plattentektonische Szenarium während der Gebirgsbildung. Die Kollision involvierte einige Terranes, die sukzessive an Laurasia angebaut wurden und die finale Konvergenz von Gondwana mit den konsolidierten Einheiten. Diese bewirkte den maximalen Aufbau der Varisciden, der durch Hochdruck-Gesteine dokumentiert ist. Diese wurden kurz nach maximaler Krustenverdickung innerhalb weniger Millionen Jahre in geringe Tiefen angehoben. Darauf folgte eine Aufheizung der Kruste, die durch Granite dokumentiert ist. Eine zentrale Frage der Variscidenforschung ist die Ursache für die erhöhten Temperaturen in der späten orogenen Phase. Geringe und gleichförmige Krustenmächtigkeiten unter den heutigen europäischen Varisciden legen die Vermutung nahe, daß sich Teile der Mantelwurzel des Gebirges abgelöst und abgesunken sind.Kapitel 2 beschreibt physikalische Aspekte von gebirgsbildenden Prozessen, insbesondere den thermischen Zustand kontinentaler Lithosphäre und die Folgen von Krustenkonvergenz. Der Einfluß von kompositioneller Schichtung und extrinsischen Parametern auf die Rheologie und das Wechselspiel zwischen auf- und abwärts gerichteten Kräften in verschiedenen orogenen Szenarien werden diskutiert.Kapitel 3 stellt die Modelle zur variscischen Gebirgsbildung vor. Ausgehend von Kontinent-Kontinent-Kollision werden Konsequenzen von rheologischer Schichtung und tiefreichenden Schwächezonen auf das Deformationsverhalten der Kruste untersucht. Außerdem wird ein Modell zum Abriß von Mantellithosphäre während Kollision vorgestellt. Abschließend werden die thermischen Konsequenzen von Subduktion junger ozeanischer Lithosphäre diskutiert.Kapitel 4 versucht eine Synthese der Kollisionsmodelle mit Modellen zur spätorogenen variscischen Entwicklung. Die mechanischen Eigenschaften der Delaminationsmodelle sollten während der Aufbauphase des Orogens angelegt worden sein. Außerdem wird eine Fallstudie vorgestellt, bei der die Modellparameter auf die Entwicklungsgeschichte erzgebirgischer Einheiten angepaßt worden sind. Schließlich werden Modelle zur thermischen Entwicklung nach Aufstieg von Asthenosphäre an die Krustenwurzel diskutiert. Die vulkanische Aktivität während der spätorogenen variscischen Phase kann plausibel erklärt werden.Kapitel 5 faßt die Ergebnisse zusammen und interpretiert diese bezüglich des plattentektonischen Szenariums der variscischen Orogenese. Durch die sukzessive Anlegung der Suturzonen während der Konsolidierung der Terranes sind die für Delamination nötigen Schwächezonen sukzessive ins variscische Orogen eingebaut worden.

Die vorliegende Arbeit gewährte neue Einblicke in zwei fundamentale Vorgänge der frühen Neurogenese von Drosophila melanogaster. Der erste Teil untersuchte die zeitliche Spezifizierung der Neuroblastenidentitäten. Durch die Expression verschiedener Gene entlang der Dorsoventral- und der Anterioposteriorachse wird ein kartesisches Koordinatensystem aufgebaut, indem ein Neuroblast (NB), der in einem bestimmten Quadranten entsteht, eine spezifische Identität erhält. Die Delamination der NBs erfolgt in fünf Segregationswellen, wobei in jeder Welle die gleiche Population NBs gebildet wird. In dieser Arbeit konnte nun gezeigt werden, dass es nicht nur einen räumlichen, sondern auch einen zeitlichen Aspekt bei der Entstehung der NBs gibt: So zeigten Transplantationsexperimente, dass sowohl im frühen als auch im späten Neuroektoderm extrinsische induktive Signale an der Spezifizierung der Neuroblastenidentität beteiligt sind. Die Natur dieser Signale bleibt noch unklar. Allerdings stellen die Segmentpolaritätsgene aufgrund ihrer dynamischen Expression eine potenzielle Kandidatengruppe dar. Der zweite Teil beschäftigte sich mit der segmentalen Spezifizierung der Neuroblasten. Für diesen Prozess zeigten frühere Genexpressionsstudien, dass NBs, die zwar an korrespondierenden Positionen innerhalb des kartesischen Systems, aber in unterschiedlichen Segmenten gebildet werden, die gleichen Genexpressionsmuster aufweisen und fast identische Zellstammbäume hervorbringen. Einige dieser seriell homologen NBs generieren jedoch segmentspezifische Zellstammbäume – ein solches Beispiel ist der NB6-4, der als Modellsystem benutzt wurde. Für die thorakale Variante dieses NBs konnte ich zeigen, dass die Homöotischen Gene zur Spezifizierung nicht notwendig sind – thorakales Schicksal ist eine Grundidentität. Diese wird in abdominalen Segmenten jedoch durch die Funktion der Homöotischen Gene abdominal-A (abd-A) und Abdominal-B (Abd-B) in abdominales Schicksal transformiert. Dieser segmentale Unterschied wird durch die Regulation des Zellzyklusgens CycE bewerkstelligen. Genauer: CycE ist notwendig, um neurogliales Schicksal in thorakalen Segmenten zu generieren und ausreichend, dieses Schicksal ebenfalls in abdominalen Segmenten zu erzeugen. Eine direkte Inhibierung der Expression von CycE durch Abd-A in abdominalen Segmenten führt dagegen zu einer differenziellen Expression von CycE im neuronalen thorakalen Anteil des Zellstammbaums. Weiterhin konnten in einem Enhancerelement, das für die Expression von CycE im Nervensystem verantwortlich ist, mehrere Bindestellen für Abd-A und Abd-B gefunden werden. Die gewonnen Daten legen – in Verbindung mit bereits bekannten Ergebnissen – den Schluss nahe, dass diese neuronspezifizierende Funktion von CycE unabhängig von seiner Rolle im Zellzyklus ist.

The aim of this Thesis is to investigate the effect of heterogeneities within the subducting plate on the dynamics of subduction. In particular, I study the motion of the trench for oceanic and continental subduction, first, separately, and, then, together in the same system to understand how they interact. The understanding of these features is fundamental to reconstruct the evolution of complex subduction zones, such as the Central Mediterranean. For this purpose, I developed 2D and 3D numerical models of oceanic and continental subduction where the rheological, geometrical and compositional properties of the plates are varied. In these models, the trench and the overriding plate move self-consistently as a function of the dynamics of the system. The effect of continental subduction on trench migration is largely investigated. Results from a parametric study showed that despite different rheological properties of the plates, all models with a uniform continental crust share the same kinematic behaviour: the trench starts to advance once the continent arrives at the subduction zone. Hence, the advancing mode in continental collision scenarios is at least partly driven by an intrinsic feature of the system. Moreover, the presence of a weak lower crust within the continental plate can lead to the occurrence of delamination. Indeed, by changing the viscosity of the lower crust, both delamination and slab detachment can occur. Delamination is favoured by a low viscosity value of the lower crust, because this makes the mechanical decoupling easier between crust and lithospheric mantle. These features are observed both in 2D and 3D models, but the numerical results of the 3D models also showed that the rheology of the continental crust has a very strong effect on the dynamics of the whole system, since it influences not only the continental part of plate but also the oceanic sides.

Gegenstand und Ziel der vorliegenden Arbeit war die Synthese und Charakterisierung einer Hydrogelmatrix, welche für die Herstellung eines vielseitig verwendbaren Sensors, der mehrere Analyten (Proteine, DNA etc.) in hoher Verdünnung (c0 < 50 fM) aus kleinsten Probenmengen (Volumina <20 nl) schnell (t < 1 min) und parallel nachweisen kann, Verwendung finden soll. Der Fokus der Arbeit lag dabei in der Synthese und Charakterisierung von Copolymeren als Hydrogelmatrix, welche geeignetes temperaturabhängiges Verhalten zeigen. Die Copolymere wurden in eine dreidimensionale Netzwerkstruktur überführt und auf einer Goldoberfläche kovalent angebunden, um Delamination zu vermeiden und die Untersuchung mittels Oberflächenplasmonenresonanz-Spektroskopie (SPR) und Optischer Wellenleiter-Spektroskopie (OWS) zu erlauben. Weiterhin wurde das temperaturabhängige Verhalten der Polymernetzwerke in Wasser mittels optischen Messungen (SPR/OWS) untersucht, um Erkenntnisse über die Quell- und Kollabiereigenschaften des Hydrogels zu gewinnen. Um temperaturschaltbare Hydrogele herzustellen, wurden auf N-Isopropylacrylamid (NIPAAm) basierende Polymere synthetisiert. Es wurde sowohl die für Hydrogele übliche Methode der freien radikalischen Vernetzungspolymerisation in Wasser, wie eine neue, auf Benzophenoneinheiten basierende Syntheseroute, welche die freie radikalische Polymerisation in organischem Medium nutzt, verwendet. Die synthetisierten Polymere sind Copolymere aus N‑Isopropylacrylamid (NIPAAm) und 4-Methacryloyloxybenzophenon (MABP). NIPAAm ist dabei für das temperaturschaltbare Verhalten der Gele verantwortlich und MABP dient als Photovernetzer. Weitere Copolymere, die neben den genannten Monomeren noch andere Funktionen, wie z.B. ionische Gruppen oder Aktivesterfunktionen enthalten, wurden ebenfalls synthetisiert. Das temperaturabhängige Quellverhalten in Bezug auf die chemische Zusammensetzung wurde mit der Oberflächenplasmonenresonanz-Spektroskopie (SPR) und Optischen Wellenleiter-Spektroskopie (OWS) untersucht. Es zeigte sich, dass die Anwesenheit von Salz im Hydrogel (Natriumacrylat als Monomer, P4S) Inhomogenität, in Form eines Brechungsindexgradienten senkrecht zur Substratoberfläche, hervorruft. Dies ist nicht der Fall, wenn statt des Salzes die Säure (Methacrylsäure als Monomer, P4A) verwendet wird. Durch die Inhomogenität lassen sich die Filme mit dem Natriummethacrylat nicht mehr mit dem, üblicherweise zur Auswertung genutzten, Kastenmodell beschreiben. Die Anwendung der Wentzel-Kramers-Brillouin-Näherung (WKB) auf die Messdaten führt hingegen zu dem gewünschten Ergebnis. Man findet ein kastenähnliches Brechungsindexprofil für das Hydrogel mit der Säure (P4A) und ein Gradientenprofil für das Gel mit dem Salz (P4S). Letzteres ist nicht nur hydrophiler und insgesamt stärker gequollen, sondern ragt auch weiter in die überstehende Wasserphase hinein. Anhand eines säurehaltigen Hydrogels (P8A) konnte der quellungshemmende Einfluss von hohen Salzkonzentrationen gezeigt werden. Weiterhin wurde während des Quellvorgangs eine gewisse Anisotropie gefunden, die aber im vollständig gequollenen und vollständig kollabierten Zustand nicht mehr vorliegt. Anhand eines Hydrogels ohne ionisierbare Gruppen (P9) wurde die Reversibilität des Quell- und Kollabiervorgangs gezeigt. Bei einem Vergleich zwischen einem säurehaltigen Hydrogel (P8A, Quellgrad von 7,3) und einem ohne ionisierbare Gruppen (P9, Quellgrad von 6,1), hat die Anwesenheit der 8 mol% Säuregruppen eine leichte Verstärkung der Quellung um den Faktor 1,2 bewirkt. Rasterkraftmikroskopische Untersuchungen (AFM) an diesen beiden Hydrogelen im getrockneten Zustand, haben gezeigt, dass nach dem Quellen, Kollabieren und Trocknen bei beiden Gelen Porenstrukturen sehr unterschiedlicher Ausmaße vorliegen.

The present work aims for investigate the influence of electrospun Nylon 6,6 nanofibrous mat on the behavior of composite laminates. The main idea is that nanofibrous interleaved into particular ply-to-ply interfaces of a laminate can lead to significant improvements of mechanical properties and delamination/damage resistance. Experimental campaigns were performed to investigate how nanofibers affect both the static and dynamic behavior of the laminate in which they are interleaved. Fracture mechanics tests were initially performed on virgin and 8 different configuration of nanomodified specimens. The purposes of this first step of the work are to understand which geometrical parameters of the nanointerleave influence the behavior of the laminate and, to find the optimal architecture of the nanofibrous mat in order to obtain the best reinforcement. In particular, 3 morphological parameters are investigated: nanofibers diameter, nanofibers orientation and thickness of the reinforce. Two different values for each parameter have been used, and it leads to 8 different configurations of nanoreinforce. Acoustic Emission technique is also used to monitor the tests. Once the optimum configuration has been found, attention is focused on the mechanism of reinforce played by the nanofibers during static and dynamic tests. Low velocity impacts and free decay tests are performed to attest the effect of nanointerlayers and the reinforce mechanism during the dynamic loads. Bump tests are performed before and after the impact on virgin and two different nanomodified laminates configurations. The authors focused their attention on: vibrational behavior, low velocity impact response and post-impact vibration behavior of the nano-interleaved laminates with respect to the response of non-nanomodified ones. Experiments attest that nanofibers significantly strength the delamination resistance of the laminates and increase some mechanical properties. It is demonstrated that the nanofibers are capable to continue to carry on the loads even when the matrix around them is broken.

La mancanza di procedure standard per la verifica delle strutture in compositi, al contrario dei materiali metallici, porta all’esigenza di una continua ricerca nel settore, al fine di ottenere risultati significativi che culminino in una standardizzazione delle procedure. In tale contesto si colloca la ricerca svolta per la stesura del presente elaborato, condotta presso il laboratorio DASML del TU Delft, nei Paesi Bassi. Il materiale studiato è un prepreg (preimpregnated) costituito da fibre di carbonio (M30SC) e matrice epossidica (DT120) con la particolare configurazione [0°/90°/±45°/±45°/90°/0°]. L’adesivo utilizzato per l’incollaggio è di tipo epossidico (FM94K). Il materiale è stato assemblato in laboratorio in modo da ottenere i provini da testare, di tipo DCB, ENF e CCP. Due differenti qualità dello stesso materiale sono state ottenute, una buona ottenuta seguendo le istruzione del produttore, ed una povera ottenuta modificando il processo produttivo suggerito, che risulta in un incollaggio di qualità nettamente inferiore rispetto al primo tipo di materiale. Lo scopo era quello di studiare i comportamenti di entrambe le qualità sotto due diversi modi di carico, modo I o opening mode e modo II o shear mode, entrambi attraverso test quasi-statici e a fatica, così da ottenere risultati comparabili tra di essi che permettano in futuro di identificare se si dispone di un materiale di buona qualità prima di procedere con il progetto dell’intera struttura. L’approccio scelto per lo studio dello sviluppo della delaminazione è un adattamento della teoria della Meccanica della Frattura Lineare Elastica (LEFM)

Several CFCC (Continuous Fiber Composite Ceramics) production processes were tested, concluding that PIP (Polymer Impregnation, or Infiltration, Pyrolysis) and CBC (Chemically Bonded Ceramics) based procedures have interesting potential applications in the construction and transportation fields, thanks to low costs to get potentially useful thermomechanical performances. Among the different processes considered during the Doctorate (from the synthesis of new preceramic polymers, to the PIP production of SiC / SiC composites) the more promising results came from the PIP process with poly-siloxanes on basalt fabrics preforms. Low processing time and costs, together with fairly good thermomechanical properties were demonstrated, even after only one or two PIP steps in nitrogen flow. In alternative, pyrolysis in vacuum was also tested, a procedure still not discussed in literature, but which could originate an interesting reduction of production costs, with only a moderate detrimental effect on the mechanical properties. The resulting CFCC is a basalt / SiCO composite that can be applied for continuous operation up to 600°C, also in oxidant environment, as TG and XRD demonstrated. The failure upon loading is generally pseudo-plastic, being interlaminar delamination the most probable rupture mechanism. . The strength depends on several different factors (microstructure, polymer curing and subsequent ceramic phase evolution, fiber pull-out, fiber strength, fiber percentage) and can only be optimized empirically. In order to be open minded in selecting the best technology, also CBC (Chemically Bonded Ceramics) matrixes were considered during this Doctorate, making some preliminary investigations on fire-resistant phosphate cements. Our results on a commercial product evidenced some interesting thermomechanical capabilities, even after thermal treatments. However the experiments showed also phase change and possible cracking and deformations even on slow drying (at 130°C) and easy rehydration upon exposure to environmental humidity.

Moderate to severe slipped capital femoral epiphysis leads to premature osteoarthritis resulting from femoroacetabular impingement. We believe surgical correction at the site of deformity through capital reorientation is the best procedure to fully correct the deformity but has traditionally been associated with high rates of osteonecrosis. We describe a modified capital reorientation procedure performed through a surgical dislocation approach. We followed 40 patients for a minimum of 1 year and 3 years from two institutions. No patient developed osteonecrosis or chondrolysis. Slip angle was corrected to 4 degrees to 8 degrees and the mean alpha angle after correction was 40.6 degrees. Articular cartilage damage, full-thickness loss, and delamination were observed at the time of surgery, especially in the stable slips. This technique appears to have an acceptable complication rate and appears reproducible for full correction of moderate to severe slipped capital femoral epiphyses with open physes.

Regulation of tissue size requires fine tuning at the single-cell level of proliferation rate, cell volume, and cell death. Whereas the adjustment of proliferation and growth has been widely studied [1, 2, 3, 4 and 5], the contribution of cell death and its adjustment to tissue-scale parameters have been so far much less explored. Recently, it was shown that epithelial cells could be eliminated by live-cell delamination in response to an increase of cell density [6]. Cell delamination was supposed to occur independently of caspase activation and was suggested to be based on a gradual and spontaneous disappearance of junctions in the delaminating cells [6]. Studying the elimination of cells in the midline region of the Drosophila pupal notum, we found that, contrary to what was suggested before, Caspase 3 activation precedes and is required for cell delamination. Yet, using particle image velocimetry, genetics, and laser-induced perturbations, we confirmed [ 6] that local tissue crowding is necessary and sufficient to drive cell elimination and that cell elimination is independent of known fitness-dependent competition pathways [ 7, 8 and 9]. Accordingly, activation of the oncogene Ras in clones was sufficient to compress the neighboring tissue and eliminate cells up to several cell diameters away from the clones. Mechanical stress has been previously proposed to contribute to cell competition [ 10 and 11]. These results provide the first experimental evidences that crowding-induced death could be an alternative mode of super-competition, namely mechanical super-competition, independent of known fitness markers [ 7, 8 and 9], that could promote tumor growth.

We report the Sr, Nd and Pb isotopic compositions (1) of 66 lava flows and dikes spanning the circa 15 Myr subaerial volcanic history of Gran Canaria and (2) of five Miocene through Cretaceous sediment samples from DSDP site 397, located 100 km south of Gran Canaria. The isotope ratios of the Gran Canaria samples vary for 87Sr/86Sr: 0.70302-0.70346, for 143Nd/144Nd: 0.51275-0.51298, and for 206Pb/204Pb: 18.76-20.01. The Miocene and the Pliocene-Recent volcanics form distinct trends on isotope correlation diagrams. The most SiO2-undersaturated volcanics from each group have the least radiogenic Sr and most radiogenic Pb, whereas evolved volcanics from each group have the most radiogenic Sr and least radiogenic Pb. In the Pliocene-Recent group, the most undersaturated basalts also have the most radiogenic Nd, and the evolved volcanics have the least radiogenic Nd. The most SiO2-saturated basalts have intermediate compositions within each age group. Although the two age groups have overlapping Sr and Nd isotope ratios, the Pliocene-Recent volcanics have less radiogenic Pb than the Miocene volcanics. At least four components are required to explain the isotope systematics of Gran Canaria by mixing. There is no evidence for crustal contamination in any of the volcanics. The most undersaturated Miocene volcanics fall within the field for the two youngest and westernmost Canary Islands in all isotope correlation diagrams and thus appear to have the most plume-like (high 238U/204Pb) HIMU-like composition. During the Pliocene-Recent epochs, the plume was located to the west of Gran Canaria. The isotopic composition of the most undersaturated Pliocene-Recent volcanics may reflect entrainment of asthenospheric material (with a depleted mantle (DM)-like composition), as plume material was transported through the upper asthenosphere to the base of the lithosphere beneath Gran Canaria. The shift in isotopic composition with increasing SiO2-saturation in the basalts and degree of differentiation for all volcanics is interpreted to reflect assimilation of enriched mantle (EM1 and EM2) in the lithosphere beneath Gran Canaria. This enriched mantle may have been derived from the continental lithospheric mantle beneath the West African Craton by thermal erosion or delamination during rifting of Pangaea. This study suggests that the enriched mantle components (EM1 and EM2) may be stored in the shallow mantle, whereas the HIMU component may have a deeper origin.

The book deals with results of complex geological and geophysical studies in the Doldrums and Arkhangelsky Fracture Zones of the Central Atlantic. Corrosion of a reinforcement bar leads to expansive pressure on the surrounding concrete that provokes internal cracking and, eventually, spalling and delamination. Here, an embedded cohesive crack 2D finite element is applied for simulating the cracking process. In addition, four simplified analytical models are introduced for comparative purposes. Under some assumptions about rust properties, corrosion rate, and particularly, the accommodation of oxide products within the open cracks generated in the process, the proposed FE model is able to estimate time to surface cracking quite accurately. Moreover, emerging cracking patterns are in reasonably good agreement with expectations. As a practical case, a prototype application of the model to an actual bridge deck is reported.

This research focused on the evaluation of damage formation on ±45º carbon fiber laminates subjected to tensile tests. The damage was evaluated by means of X-ray tomography. A high density of cracks developed during the plateau of the stress-strain curve and were qualitatively analyzed, showing that the inner plies eventually developed a higher crack concentration than the outer plies. Delamination started to occur in the outermost ply interface when the slope after the plateau of the stress-strain curve began to increase.
