Genotypic diversity in Fusarium pseudograminearum and F. graminearum from Australia and the relationship between diversity and pathogen aggressiveness for head blight and/or crown rot of wheat were examined. Amplified fragment length polymorphism (AFLP) analysis revealed a high level of genotypic diversity within each species. Sixty-three of the 149 AFLP loci were significantly different between the two species and 70 of 72 F. pseudograminearum and 56 of 59 F. graminearum isolates had distinct haplotypes. When head blight and crown rot severity data from a recently published work on isolates representing the entire range of aggressiveness were used, only the genotypic diversity of F. pseudograminearum was significantly associated with its aggressiveness for the two diseases. Cluster analyses clearly demonstrated the polyphyletic structures that exist in both pathogen populations. The spatial diversity within F. graminearum was high within a single field, while frequent gene flow (N-m similar to 14) and a low fixation index (G(st) = 0.03) were recorded among F. pseudograminearum isolates from the adjacent states of New South Wales and Queensland. The differences in population structure between the heterothallic F. pseudograminearum (teleomorph G. coronicola) and the homothallic F. graminearum (teleomorph G. zeae) were not as pronounced as expected given their contrasting mating systems. Neither species was panmictic or strictly clonal. This points to sexual recombination in F. pseudograminearum, suggesting that ascospores of G. coronicola may also play a role in its biology and epidemiology.

In vitro experimental environments are used to study interactions between microorganisms, and predict dynamics in natural ecosystems. This study highlights that experimental in vitro environments should be selected to closely match the natural environment of interest during in vitro studies to strengthen extrapolations about aflatoxin production by Aspergillus and competing organisms. Fungal competition and aflatoxin accumulation was studied in soil, cotton wool or tube (water-only) environments, for Aspergillus flavus competition with Penicillium purpurogenum, Fusarium oxysporum or Sarocladium zeae within maize grains. Inoculated grains were incubated in each environment at two temperature regimes (25oC and 30oC). Competition experiments showed interaction between main effects of aflatoxin accumulation and environment at 25oC, but not so at 30oC. However, competition experiments showed fungal populations were always interacting with their environments. Fungal survival differed after the 72-hour incubation in different experimental environments. Whereas, all fungi incubated within the soil environment survived; in the cotton-wool environment, none of the competitors of A. flavus survived at 30 oC. With aflatoxin accumulation, F. oxysporum was the only fungus able to interdict aflatoxin production at both temperatures. This occurred only in the soil environment and fumonisins accumulated instead. Smallholder farmers in developing countries face serious mycotoxin contamination of their grains, and soil is a natural reservoir for the associated fungal propagules, and a drying and storage surface for grains on these farms. Studying fungal dynamics in the soil environment and other environments in vitro can provide insights into aflatoxin accumulation post harvest.

In the context of this work we evaluated a multisensory, noninvasive prototype platform for shake flask cultivations by monitoring three basic parameters (pH, pO2 and biomass). The focus lies on the evaluation of the biomass sensor based on backward light scattering. The application spectrum was expanded to four new organisms in addition to E. coli K12 and S. cerevisiae [1]. It could be shown that the sensor is appropriate for a wide range of standard microorganisms, e.g., L. zeae, K. pastoris, A. niger and CHO-K1. The biomass sensor signal could successfully be correlated and calibrated with well-known measurement methods like OD600, cell dry weight (CDW) and cell concentration. Logarithmic and Bleasdale-Nelder derived functions were adequate for data fitting. Measurements at low cell concentrations proved to be critical in terms of a high signal to noise ratio, but the integration of a custom made light shade in the shake flask improved these measurements significantly. This sensor based measurement method has a high potential to initiate a new generation of online bioprocess monitoring. Metabolic studies will particularly benefit from the multisensory data acquisition. The sensor is already used in labscale experiments for shake flask cultivations.

This study presents an application of the geographical information system technology on plant disease involving a multidisciplinary teamwork of geoprocessing and physiopathology specialists. The spatial analysis tools in a GIS were used to evaluate the spatial distribution of two diseases of maize in Brazil: polysora rusl caused by Puccinia polysora and tropical rust caused by Physopella zeae. A database of cIimate variables (mean temperature. relative humidity. and leaf wetness duration) of cIimatological normal from 1961-1990 was obtained and then related it to a mathematical model of disease development (polysora rust) and to the cIimate intervals (tropical rust) in order to obtain the maps. The choice of the model or the favorable climate interval is the important chalIenge of the method because the difficulty of adequacy to the spatial and temporal scales for the specific application. The major incidence of both disease occurred in almost alI the North region from January to June. although this region has traditionalIy a low production of maize. Considering the biggest producers regions. for both the diseases, favorable areas are located in part of Mato Grosso, Tocanlins. Minas Gerais; Mato Grosso do Sul. and coastal areas of São Paulo, Paraná, and Santa Catarina. varying among the dilferent months from January to June. The method allowed making an adequate distinction of the states and the months considered.

A presente pesquisa tem o objetivo de avaliar a incidência e evolução das principais pragas e doenças do cafeeiro em agroecossistemas sob manejo convencional, organo-mineral, orgânico e agroflorestal nos municípios sul mineiros de Machado e Poço-Fundo. Foram realizados monitoramentos mensais do bicho-mineiro (Leucoptera coffeella), broca-do-café (Hypothenemus hampei), ferrugem (Hemileia vastatrix) e da cercosporiose (Cercospora coffeicola) por um período de um ano (dez/2007 a nov/2008) em lavouras da variedade Mundo Novo sob diferentes manejos. O monitoramento do bichomineiro e das doenças do cafeeiro foi realizado através da coleta de dez folhas do terceiro ou quarto par em todos os lados do cafeeiro, sendo amostradas vinte plantas por agroecossistema, totalizando duzentas folhas coletadas para avaliação dessas moléstias em cada sistema. Para o monitoramento da brocado- café selecionou-se 32 plantas, seis pontos diferentes em cada planta (10 frutos agrupados por ponto estabelecido) e realizou-se avaliação nãodestrutiva. Em todos os agroecossistemas monitorados, ambos manejados pela agricultura familiar, a infestação da broca-do-café e do bicho-mineiro não atingiram nível de dano econômico em nenhuma avaliação. Com relação às doenças avaliadas, observou-se que tanto a ferrugem como a cercosporiose atingiram nível de dano econômico em todos os sistemas de manejo. No entanto, nos sistemas organo-mineral, orgânico (monocultivo) e agroflorestal (diversificado) a ocorrência da ferrugem foi crítica, atingindo índices elevados nos meses de abril a outubro de 2008, registrando-se valores acima de 60% de incidência em quatro meses de avaliação. A incidência da cercosporiose foi relativamente menor no sistema convencional e orgânico (monocultivo), cujos picos não ultrapassaram 41% e 55%, consecutivamente. Já no sistema organomineral e agroflorestal (orgânico) a ocorrência da cercosporiose atingiu níveis muito elevados, chegando a 76% e 67%, respectivamente, no mês de julho. Apesar dos elevados níveis de incidência da ferrugem e cercosporiose encontrados nos agroecossistemas cafeeiros organo-mineral e orgânico, a produtividade média alcançada por esses sistemas nos últimos três anos foi superior ao convencional, mesmo esse último sistema investindo pesado em agrotóxicos e fertilizantes químicos.
