A região oeste da Bahia passa por acelerado processo de desenvolvimento agrícola, o que tem gerado problemas de infraestrutura nos municípios. A falta de saneamento básico e de coleta dos resíduos são alguns desses problemas, os quais têm papel importante no aumento de criadouros disponíveis aos culicídeos. Assim, considerando as possibilidades de contato entre vetor e população humana, o objetivo deste trabalho foi estudar a fauna de culicíeos adultos em uma área urbana no oeste da Bahia e verificar a presença de espécies de interesse em saúde pública. As amostragens de mosquitos adultos foram realizadas no município de Barreiras, de fevereiro de 2009 a janeiro de 2010, das 17h30min às 21h00min. Ao total foram capturados 1.744 mosquitos e oito gêneros foram identificados: Aedes, Coquilletidia, Culex, Limatus, Mansonia, Ochlerotatus, Psorophora e Uranotaenia. Culex quinquefasciatus foi a espécie mais frequente e abundante. Outras espécies encontradas, e que apresentam importância epidemiológica, foram Aedes aegypti e Ochlerotatus scapularis. Considerando que muitas espécies coletadas em Barreiras possuem importância epidemiológica e ocorrem mesmo durante a estação seca devido à ausência de saneamento básico, é importante que os municípios da região oeste da Bahia sejam alvo constante das atividades de vigilância epidemiológica.

Apresentamos os resultados de observação sobre o ciclo circadiano de atividade hematofágica dos mosquitos, em Granja Calábria, Jacarepaguá, na planície litorânea do Rio de Janeiro, onde realizamos, em isca humana, ao ar livre, capturas semanais, de 8 às 10, de 13 às 15 e 18 às 20 horas, de agosto de 1981 a julho de 1982, além de três capturas horárias de 24 horas seguidas. A maioria das espécies locais revelou caráter crepuscular vespertino e noturno. Contudo Limatus durhami, Phoniomyia davisi, Wyeomyia leucostigma e Wyeomyia (Dendromyia) sp. foram essencialmente diurnas, enquanto Anopheles albitarsis, Culex chidesteri e Culex quinquefasciatus foram obtidas somente no crepúsculo vespertino e à noite. Embora Anopheles aquasalis, Culex coronator, Culex saltanensis, Culex crybda e Coquillettidia venezuelensis fossem preponderantemente noturnas e Phoniomyia deanei e Phoniomyia theobaldi principalmente diurnas, obtivemô-las algumas vezes, fora do horário preferencial, sendo que Phoniomyia deanei teve nítido incremento pré-crepuscular vespertino. Aedes scapularis, Aedes taeniorhynchus e Mansonia titillans, espécies mais ecléticas, picaram durante todo o nictêmero, mas com flagrante acentuação crepuscular vespertina.

Apresentamos os resultados de observações sobre os criadouros dos mosquitos, que realizamos numa fazenda - Granjas Calábria, da Baixada de Jacarepaguá, Rio de Janeiro, no período de agosto de 1981 a julho de 1983. A maioria das espécies locais preferiu coleções líquidas no solo, particularmente as de caráter natural, não deixando, entretanto, de procurar aquelas propiciadas pelas atividades humanas. Os criadouros transitórios foram mais freqüentados por Culex saltanensis e pelas espécies da tribo Aedini, como Aedes scapularis, Aedes taeniorhynchus, Psorophora ciliata e Psorophora confinnis, enquanto os de caráter permanente foram mananciais de formas imaturas de Mansonia titillans, Culex amazonensis, Culex chidesteri, Culex bidens, Culex declarator, Culex nigripalpus e Culex plectoporpe. Algumas espécies foram coletadas em recipientes naturais: Culex ocellatus, os Culex (Microculex), Phoniomyia davisi, Phoniomyia deanei e Wyeomyia forcipenis, em bromélias; Aedes terrens, Culex gairus e Culex imitador, em buraco em árvore; e Wyeomyia leucostigma, em axilas submersas das folhas de taboas (Thypha dominguensis). Culex gairus foi encontrado pela primeira vez criando em recipientes artificiais, locais também preferidos por Culex corniger, Culex quinquefasciatus e Limatus durhami.

Along 24 consecutive months, from January 1984 to December 1986, mosquito captures were performed in a rural area of said municipality. Aiming to evaluate the comparative frequency of the mosquito species inside houses and in the immediate vicinity and far from houses, the captures were made in two types of domiciles - one permanently and the other only sporadically inhabited - as well as in surrounding woods. Variations in temperature, relative humidity and rainfall were locally recorded. A tendency for domiciliation became evident by the presence of Aedes scapularis in the human domiciles, mainly in those permantely inhabited. Culex quinquefasciatus showed to be adapted to live with humans also in rural areas, in which, in some instances, insecticides had to be used to keep in under control. Such observations, mainly in terms of Ae. scapularis, reinforce the possibility of those mosquitoes, under favourable conditions, becoming carries of arboviroses to humans in rural environments.

A study of peridomestic man-biting culicines in the Amazon Basin was conducted from January through December, 1987. Fifteen species of mosquitoes from six genera were collected by volunters in all-night human-bait indoor and outdoor collections at five hoses in and near the town of Costa Marques, Rondônia, Brazil. Culex quinquefasciatus and members of the Mansonia titillans/indubitans Group comprised 61 and 33%, respectively, of all culcines collected from human-bait outside houses and 62 and 35%, respectively, of those collected from volunteers inside houses in the town. In rural areas, Cx. quinquefasciatus was less abundant and only comprised 2 and 5% of the culcines, respectively, collected inside and outside houses. Mansonia titillans/indubitans Group comprised 75% and 86% of the culicines collected inside and outside houses, respectively, from rural residences. Culex quinquefasciatus and members of Mn. titillans/indubitans Group were more endophilic than other culcines collected. Hexaflumuron, an insect growth regulator (IGR), was found to greatly affect the development of immatures and emergence of adults of three species of vector mosquitoes, Culex quinquefasciatus, Aedes aegypti and Anopheles stephensi, when larvae were subjected to short time exposure of < or = 1h. This IGR could completely prevent adult emergence even at a minimum exposure time of 10 min at 0.001, 0.01 and 0.1 mg/l. On treatment, larval and pupal mortality as well as varying degrees of morphogenetic abnormalities were induced in immatures and adults of the three species. Four weeks of control achieved in a slow moving sullage canal breeding Culex quinquefasciatus indicates that this IGR can be of use in such breeding habitats.

Characterization of the insecticidal and hemolytic activity of solubilized crystal proteins of Bacillus thuringiensis (Bt) subsp. medellin (Btmed) was performed and compared to solubilized crystal proteins of isolates 1884 of B. thuringiensis subsp. israelensis (Bti) and isolate PG-14 of B. thuringiensis subsp. morrisoni (Btm). In general, at acid pH values solubilization of the Bt crystalline parasporal inclusions (CPI) was lower than at alkaline pH. The larvicidal activity demonstrated by the CPI of Btmed indicated that optimal solubilization of CPI takes place at a pH value of 11.3, in Bti at pH values from 5.03 to 11.3 and in Btm at pH values from 9.05 to 11.3. Hemolytic activity against sheep red blood cells was mainly found following extraction at pH 11.3 in all Bt strains tested. Polyacrylamide gel electrophoresis under denaturing conditions revealed that optimal solubilization of the CPI in all Bt strains takes place at the alkaline pH values from 9.05 to 11.3. An enriched preparation of Btmed crystals was obtained, solubilized and crystal proteins were separated on a size exclusion column (Sephacryl S-200). Three main protein peaks were observed on the chromatogram. The first peak had two main proteins that migrate between 90 to 100 kDa. These proteins are apparently not common to other Bt strains isolated to date. The second and third peaks obtained from the size exclusion column yielded polypeptides of 68 and 28-30 kDa, respectively. Each peak independently, showed toxicity against 1st instar Culex quinquefasciatus larvae. Interestingly, combinations of the fractions corresponding to the 68 and 30 kDa protein showed an increased toxicity. These results suggest that the 94 kDa protein is an important component of the Btmed toxins with the highest potency to kill mosquito larvae. When crystal proteins of Bti were probed with antisera raised independently against the three main protein fractions of Btmed, the only crystal protein that showed cross reaction was the 28 kDa protein. These data suggest that Btmed could be an alternative bacterium for mosquito control programs in case mosquito larval resistance emerges to Bti toxic proteins.

Wuchereria bancrofti in Pernambuco was first documented in 1952 (Azevedo & Dobbin 1952), and since then it has been reported in surveys carried out in selected areas of Recife. Several surveys were carried out from 1981 to 1991 by SUCAM. In the 1985 SUCAM's report the disease is considered under control. The CPqAM Filariasis Research Program was established in 1985 and a filarial survey was carried out in the town of Olinda, Greater Recife. In order to verify the real epidemiological situation, a study was conducted in the city of Recife. 21/36 of the Special Zones of Social Interest (ZEIS), were randomly selected for the present study. From 10,664 persons screened, 683 were positive and the prevalence rate for microfilaraemia (mf) varied from 0.6% to 14.9%. A mean mf prevalence of 6.5%, showed that the infection occurs in a wide geographic distribution in Greater Recife and that the intensity of transmission is a real and potential threat to public health in affected communities. Mf rate among males and females differed significantly. Due to the rapid increase in population, unplanned urban settlements, poor sanitary facilities and the favorable geographical conditions to the development of the vector, filariasis may actually be increasing in Recife.

Primary powders of Bacillus sphaericus strain S2 isolated from soil samples in Brazil, and strain 2362 were produced in a 14 liter fermentor. Growth patterns and sporulation observed in three trials with strains S2 and 2362 in the fermentor were similar. Second-instar larvae of Culex quinquefasciatus, Anopheles albimanus, Anopheles quadrimaculatus, and Aedes aegypti exposed for 48 hr to strain S2 responded with LC50 values of 0.25, 5.95, 12.28 and 140.0 ppb of lyophilized primary powder, respectively. Under the same conditions, strain 2362 resulted in LC50 values of 0.39, 7.16, 16.93 and 307.0 ppb of lyophilized primary powder, respectively, in those mosquito larvae. Statistical analysis of the bioassay data did not show significant differences among LC50 values observed in B. sphaericus strains S2 and 2362, at the 0.05 level. Toxins of strains S2 and 2362 were extracted at pH 12 with NaOH. Electrophoresis of the extracts in polyacrylamide gel under denaturing conditions revealed the 51 and 42 kDa toxins in both S2 and 2362 B. sphaericus strains. The presence of the 42 kDa peptide in the extracts was confirmed by Western blot and Elisa, with anti-42 kDa IgG previously prepared from strain 2362.

Bacillus thuringiensis (Bt) subsp. medellin (Btmed) produces parasporal crystalline inclusions which are toxic to mosquito larvae. It has been shown that the inclusions of this bacterium contain mainly proteins of 94, 68 and 28-30 kDa. EcoRI partially digested total DNA of Btmed was cloned by using the Lambda Zap II cloning kit. Recombinant plaques were screened with a mouse policlonal antibody raised against the 94 kDa crystal protein of Btmed. One of the positive plaques was selected, and by in vivo excision, a recombinant pBluescript SK(-) was obtained. The gene encoding the 94 kDa toxin of Btmed DNA was cloned in a 4.4 kb DNA fragment. Btmed DNA was then subcloned as a EcoRI/EcoRI fragment into the shuttle vector pBU4 producing the recombinant plasmid pBTM3 and used to transform by electroporation Bt subsp. israelensis (Bti) crystal negative strain 4Q2-81. Toxicity to mosquito larvae was estimated by using first instar laboratory reared Aedes aegypti, and Culex quinquefasciatus larvae challenged with whole crystals. Toxicity results indicate that the purified inclusions from the recombinant Bti strain were toxic to all mosquito species tested, although the toxicity was not as high as the one produced by the crystal of the Btmed wild type strain. Poliacrylamide gel electrophoresis indicate that the inclusions produced by the recombinant strain Bti (pBTM3) were mainly composed of the 94 kDa protein of Btmed, as it was determined by Western blot

Heartworm, a chronic fatal mosquito-borne canine disease, is frequently diagnosed in dogs from the State of Rio de Janeiro, where its prevalence is 29.7% in the city of Niterói. Nevertheless it is rarely detected in cats (0.8%) from the same state. Dogs are the primary source of infection to mosquitoes, because cats either do not demonstrate microfilaremia or it is too low and transient for transmission. A mosquito survey was conducted in Itacoatiara, Niterói, from March 1995 to February 1996, using canine, feline and human baits. A total of 21 mosquito species (3,888 females) was collected and biting frequency was highest at dusk. The four species collected most frequently (88.9%) were: Aedes taeniorhynchus (30% of the total catch; with the peak in May/June); Culex quinquefasciatus (22.5%; August/October); Aedes scapularis (19.4%; August, October/November and January) and Culex declarator (17%; November/January). Human baits were attractive to these species and dogs were significantly more attractive to them than cats. Ae. taeniorhynchus, Cx. quinquefasciatus, Ae. scapularis, Cx. declarator and Cx. nigripalpus are the most likely mosquito species to transmit Dirofilaria immitis parasites to dogs and may transmit the parasite to humans. It is also suggested that the vector to cats belongs to the genus Culex

Bacillus thuringiensis produces d-endotoxins that require proteolytic processing to become active. The activation of the B. thuringiensis subsp. medellin 28 kDa (Cyt1Ab1) cytolytic toxin by trypsin, chymotrypsin and gut extract from Culex quinquefasciatus larvae was analyzed. The Cyt1Ab1 toxin of B. thuringiensis subsp. medellin was processed by all proteases tested to fragments between 23 and 25 kDa, while processing of the Cyt1Aa1 toxin produce fragments between 22.5 and 24.5 kDa. The Cyt1Ab1 toxin was preferentially processed at the alkaline pH of 12. The in vitro proteolytic processing of the Cyt1Ab1 toxin by C. quinquefasciatus larvae midgut extract showed a 25 kDa fragment; a similar result was observed when the activation was performed in the in vivo experiments. The solubilized Cyt1Ab1 toxin and the protease resistant cores generated by in vitro processing showed hemolytic activity but not mosquitocidal activity. Amino terminal sequence of the C. quinquefasciatus gut extract resistant fragment indicated that the cutting site was located between Lys31 and Asp32, with a sequence DDPNEKNNHNS; while for the trypsin-resistant fragment the cutting site was determined between Leu29 and Arg30, and for the chymotrypsin-resistant fragment between Arg30 and Lys31.

Mosquito control with biological insecticides, such as Bacillus sp. toxins, has been used widely in many countries. However, rapid sedimentation away from the mosquito larvae feeding zone causes a low residual effect. In order to overcome this problem, it has been proposed to clone the Bacillus toxin genes in aquatic bacteria which are able to live in the upper part of the water column. Two strains of Asticcacaulis excentricus were chosen to introduce the B. sphaericus binary toxin gene and B. thuringiensis subsp. medellin cry11Bb gene cloned in suitable vectors. In feeding experiments with these aquatic bacteria, it was shown that Culex quinquefasciatus, Aedes aegypti, and Anopheles albimanus larvae were able to survive on a diet based on this wild bacterium. A. excentricus recombinant strains were able to express both genes, but the recombinant strain expressing the B. sphaericus binary toxin was toxic to mosquito larvae. Crude protease A. excentricus extracts did not degrade the Cry11Bb toxin. The flotability studies indicated that the recombinant A. excentricus strains remained in the upper part of the water column longer than the wild type Bacillus strains.

We report here the reappearence of Aedes aegypti in the Rimac district, and summarize the history of this mosquito species in Peru since its first detection in 1852. On March 17 2000 were found Ae. aegypti and Culex quinquefasciatus in Mariscal Castilla town, Flor de Amancaes, San Juan de Amancaes, El Altillo and Santa Rosa in the Rimac district, Lima Province.
