The Indian edible oyster Crassostrea madrasensis (Preston) is known to be a highly suitable candidate species for culture. Though Q, madrasensis has been subjected to intensive research, there has been no significant attempt to culture this oyster commercially. One major reason for the lack of interest in oyster culture could be the disparity in growth, survival and production reported by earlier workersf from different regions along the Indian coast. Greater predictability of production can create confidence and encourage entrepreneurs interested in oyster culture. The present study, which is a detailed investigation on the influence of various environmental variables on growth and reproduction of Q, madrasensis, is not confined to the impact of only hydrological parameters but is also extended to study the effect of different degrees of aerial exposure on growth and survival. The main objective of the study is to develop a background for subsequent development of a site suitability index for culture of Q, madrasensis along the Indian coast. Two sets of experiments were conducted during the present study. Details of the experiments are presented in the thesis under two major chapters comprising four sections each. Each chapter has a separate introduction, materials and methods, results and discussion. .

A detailed study of the hydrography of the Cochin Backwaters, the habitat off crassostrea madrasensis has been carried out. Data pertaining to air temperature, water temperature, salinity, dissolved oxygen, turbidity and rainfall have been collected and presented. The temperature fluctuation was in the range of 5°C only and that of salinity between 1.1%o and 32.9%o. Fairly steady salinity has been recorded during the pre-monsoon period (February to May) and drastic declension during the monsoon period (June-September).Dissolved oxygen varied between 2.5 ml/l and 6.5 ml/l. Turbidity was highest in June (27.9 p.p.m.) and minimum (10.2 p.p.m.) in February. A detailed study on marine biofouling in the Cochin Backwaters has been made with special reference to primary film, settlement and growth of the fouling organisms such as hydroids, bryozoans, tube-dwelling polychaetes, barnacles and modiolus

Little is known about the microbial diversity associated with marine macroorganisms, despite the vital role microorganisms may play in marine ecosystems. The aim of the present study was to investigate the diversity of bacteria and fungi isolated from eight marine invertebrate and one algae samples. Data derived from ARDRA and sequencing analyses allowed the identification of marine-derived microorganisms isolated from those samples. Microbial strains identified up to the genus level revealed 144 distinct ribotypes out of 256 fungal strains and 158 distinct ribotypes out of 181 bacterial strains. Filamentous fungi were distributed among 24 different genera belonging to Ascomycota, Zygomycota and Basidiomycota, some of which had never been reported in the literature as marine invertebrate-inhabiting fungi (Pestalotiopsis, Xylaria, Botrysphaeria and Cunnninghamella). Bacterial isolates were affiliated to 41 different genera, being Bacillus, Ruegeria, Micrococcus, Pseudovibrio and Staphylococcus the most abundant ones. Results revealed an unexpected high microbial diversity associated to the macroorganisms which have been collected and suggested the selection of certain microbial taxonomic groups according to the host. The combined data gathered from this investigation contribute to broaden the knowledge of microbial diversity associated to marine macroorganisms, including as a promising source for the discovery of new natural products. The Curimataú estuary is located in the oriental coast of Rio Grande do Norte State in Brazil. Its importance resides in the fact that this region possesses one of the last portions of preserved mangrove in the Rio Grande do Norte State. Nevertheless, it has been severely affected by many anthropogenic activities, as sugarcane monoculture and shrimp farming. Former works demonstrated that an accumulation of heavy metals is occurring in oysters in this estuary, and perhaps it could be explained by the input of metals in this ecosystem deriving from the shrimp farming. To better understanding the origin of these metals, bottom sediment samples, cores and suspended particulate matter were collected for a characterization of metal concentrations (Al, Ba, Cd, Cu, Cr, Fe, Mn, Ni, Pb, Zn) and to determine the potentially bioavailable metals. Additionally, the enrichment ratio for each element analyzed was calculated. The mineralogical composition of sediment samples and cores were obtained by X-ray diffraction. Moreover, data of orbital remote sensing were used in order to detect and quantify suspended matter by applying a logarithmic algorithm. Geochemical data of bottom sediments and cores revealed that, excepting Ba and Pb, the elements analyzed presented concentrations characteristic of an unpolluted ecosystem (Al: 0,25 - 8,76 %; Ba: 3,03 - 870 µg.g-1; Cd: < 0,25 µg.g-1; Cr: 1,72 - 82,4 µg.g-1; Cu: 0,12 -25,3 µg.g-1; Pb: 0,38 - 23,7 µg.g-1; Fe: 0,10 - 5,82 %; Mn: 15,1 - 815 µg.g-1; Ni: 0,14 - 36,1 µg.g-1; Zn: 1,37 - 113 µg.g-1). During the dry season a distribution pattern was observed, with higher metal concentrations in the margins, decreasing toward the central portion of the channel. These metal concentrations were well correlated with mineralogical compositions, with clay minerals prevailing at the margins, and quartz and feldspar in the center. However, this pattern was not observed during the wet season, probably because of the high water flux that disturbed bottom sediments. But, as observed for the dry season, a good correlation between metal concentrations and mineralogical composition was also observed for the wet season, with high metal concentrations where there were high quantities of clay minerals. Low enrichment ratios were obtained for the majority of elements analyzed, excepting for Mn, Ba and Pb. Manganese presented the higher ratios downstream for both seasons, and it can be an evidence of anthropogenic impact by shrimp farming. As barium and lead concentrations in sediment samples presented analytical problems during the total sample digestion, one cannot be sure that the ratios obtained correspond to the reality. The highest metal concentrations in particulate matter were obtained in the portion dominated by fluvial transport for all metals analyzed, excepting for copper. Barium and zinc were the only elements that presented elevated concentrations that are not common of unpolluted ecosystems (Ba: 5730 - 8355 µg.g-1; Zn: 3899 - 4348 µg.g-1). However, these high concentrations could not be related to the shrimp farming and waste waters from the town of Canguaretama, once they were obtained from the fluvial particulate matter, that is upstream from the activities above mentioned. The application of the logarithmic algorithm to the processed LANDSAT image was well succeeded, although the acquired image does not correspond exactly to the field campaigns. The IKONOS image provided very detailed views of the suspended sediment concentration at the estuary, as the mixture of distinct water flows at the confluence of Cunhaú and Curimataú rivers, with more turbid waters from Cunhaú river, that is directly affected by effluents from shrimp farming and urban waste waters deriving from the town of Canguaretama

The morphology of 20 species of weeds that occur in the most common crops in Maringá, Paraná State, Brazil and adjacent regions were described at early developmental stages, in order to identify the species in the field and provide information for taxonomic and ecological works. Sixteen species featured epigeal-foliaceous seedlings. The seedling in Cardiospenuum halicacabum is epigeal-fleshy; in Cenchrus echinatus and Digitaria horizontalis it is hypogeal and in Commelina virginica is epigeal-cryptocotyledonar. The differentiation of the first eophyll occurred within ten (Digitaria horizontalis) to 35 days (Bidens pilosa) and the first metaphyll appeared within 22 (Commelina virginica) to 49 days (Acanthospenuum australe). The number of eophylls varied from one to four, and the phyllotaxis varied from alternate to opposite.

The aim of this study was to investigate whether handling and acclimatization could affect the biomarker responses in oysters Crassostrea gigas. Adult oysters were sampled in a farming area, subjected to handling stress during two hours (shell cleaning and transport), and then acclimatized in laboratory for 2, 3 and 4 weeks. Groups of five oysters were sampled before and after the handling (T0 and T1, respectively), and after 2, 3 and 4 weeks acclimatization. During the acclimatization, water was renewed daily, food given twice a day and temperature and salinity maintained at 22 °C and 25 ppt, respectively. One group, in another tank, was kept in similar conditions and was exposed for 1 week to 0.1 % diesel after the 2-weeks acclimatization period. After exposure, gills were immediately frozen in liquid N 2 for biochemical analyses. Higher expression of heat-shock proteins (HSP70) was observed after handling, and after acclimatization periods of 3-week and 4-week, compared to the T0 group. The diesel exposed group did not show elevated levels of HSP70, when compared to the 3-week acclimatized group. The activity of glutathione S-transferase (GST) was unchanged after handling, but was lower after all acclimatization periods, compared to the T0 group. Exposure to diesel caused an increase in GST activity compared to the 3-week acclimatized group, but not compared to T0. The activity of catalase (CAT), acetylcholinesterase (AChE), and the MDA levels remained unchanged during the whole experiment. These results point to the need of a special care in laboratory and field experiments employing HSP70 and GST as biomarkers. (Supported by CNPq-CTPetro to ACDB.). © 2008 Published by Elsevier Ltd.

O cultivo de espécies de ostras do gênero Crassostrea está em expansão no nordeste do estado do Pará, Brasil. Este estudo analisa a qualidade sanitária das ostras e da água em que são cultivadas nos municípios de São Caetano de Odivelas e Curuçá. As coletas foram realizadas mensalmente entre junho de 2009 e maio de 2010. As amostras de águas foram coletadas nas marés enchente e vazante, e cerca de 15 ostras foram obtidos a cada mês durante a maré vazante. Concentrações de coliformes foram determinadas usando a Técnica de Fermentação de Tubos Múltiplos, seguida pela identificação bioquímica das bactérias e determinação do perfil de suscetibilidade de Escherichia coli isoladas a partir de amostras de águas e ostras. A média geométrica das concentrações de coliformes termotolerantes na água foi de 119 mL MPN/100 em São Caetano de Odivelas e 163,21 MPN/100 mL em Curuçá, valores bem acima do limite de 43 mL MPN/100 estabelecido pelo Conselho Nacional do Meio Ambiente Brasileiro (CONAMA). Como a legislação brasileira relacionada à qualidade sanitária dos moluscos bivalves destina-se apenas ao produto processado, foi adotada a legislação da União Europeia, que classifica as ostras para o consumo cru em três classes sanitárias. Em São Caetano de Odivelas, apenas duas das amostras coletadas durante este estudo foram atribuídas para a classe A, sete amostras para a classe B e três amostras para a classe C. Enquanto em Curuçá três amostras foram atribuídas à classe A, sete amostras para a classe C e duas amostras para a classe C. Os resultados sugerem a necessidade de medidas mitigatórias para garantir a qualidade sanitária das ostras, tais como a aplicação de métodos de depuração.

The objective of this work was to evaluate the total and thermotolerant coliform densities in the oyster culture water of Cananeia, SP, Brazil, correlating these densities with environmental variables and tidal variations. Superficial water samples were collected in two tide conditions (spring and neap) from three areas of Cananéia municipality (Mandira, Itapitangui and Cooperostra). The three studied areas showed good conditions for the culture regarding coliform densities. The two tidal conditions differed significantly as to total coliform concentration; however, the same procedure was not performed for thermotolerant coliforms. No correlation was observed between water temperature, pH, and concentrations of total and thermotolerant coliforms. Coliform density was positively correlated with rainfall and negatively correlated with salinity. Spring and neap tides differed significantly as to coliform number. Simple diagnosis of environmental conditions of the crop fields is insufficient to assess water quality of shellfish cultivation. A continuous monitoring program of planted areas is necessary both for the assessment of water quality potential for marine culture and for ensuring safe consumption of seafood, besides constituting an important tool to understand the relationships between contamination and the involved environmental variables.

In the last decades, the increase of industrial activities and of the request for the world food requirement, the intensification of natural resources exploitation, directly connected to pollution, have aroused an increasing interest of the public opinion towards initiatives linked to the regulation of food production, as well to the institution of a modern legislation for the consumer guardianship. This work was planned taking into account some important thematics related to marine environment, collecting and showing the data obtained from the studies made on different marine species of commercial interest (Chamelea gallina, Mytilus edulis, Ostrea edulis, Crassostrea gigas, Salmo salar, Gadus morhua). These studies have evaluated the effects of important physic and chemical parameters variations (temperature, xenobiotics like drugs, hydrocarbons and pesticides) on cells involved in the immune defence (haemocytes) and on some important enzymatic systems involved in xenobiotic biotransformation processes (cytochrome P450 complex) and in the related antioxidant defence processes (Superoxide dismutase, Catalase, Heat Shock Protein), from a biochemical and bimolecular point of view. Oxygen is essential in the biological answer of a living organism. Its consume in the normal cellular breathing physiological processes and foreign substances biotransformation, leads to reactive oxygen species (ROS) formation, potentially toxic and responsible of biological macromolecules damages with consequent pathologies worsening. Such processes can bring to a qualitative alteration of the derived products, but also to a general state of suffering that in the most serious cases can provoke the death of the organism, with important repercussions in economic field, in the output of the breedings, of fishing and of aquaculture. In this study it seemed interesting to apply also alternative methodologies currently in use in the medical field (cytofluorimetry) and in proteomic studies (bidimensional electrophoresis, mass spectrometry) with the aim of identify new biomarkers to place beside the traditional methods for the control of the animal origin food quality. From the results it’s possible to point out some relevant aspects from each experiment: 1. The cytofluorimetric techniques applied to O. edulis and C. gigas could bring to important developments in the search of alternative methods that quickly allows to identify with precision the origin of a specific sample, contributing to oppose possible alimentary frauds, in this case for example related to presence of a different species, also under a qualitative profile, but morpholgically similar. A concrete perspective for the application in the inspective field of this method has to be confirmed by further laboratory tests that take also in account in vivo experiments to evaluate the effect in the whole organism of the factors evaluated only on haemocytes in vitro. These elements suggest therefore the possibility to suit the cytofluorimetric methods for the study of animal organisms of food interest, still before these enter the phase of industrial working processes, giving useful information about the possible presence of contaminants sources that can induce an increase of the immune defence and an alteration of normal cellular parameter values. 2. C. gallina immune system has shown an interesting answer to benzo[a]pyrene (B[a]P) exposure, dose and time dependent, with a significant decrease of the expression and of the activity of one of the most important enzymes involved in the antioxidant defence in haemocytes and haemolymph. The data obtained are confirmed by several measurements of physiological parameters, that together with the decrease of the activity of 7-etossi-resourifine-O-deetilase (EROD linked to xenobiotic biotransformation processes) during exposure, underline the major effects of B[a]P action. The identification of basal levels of EROD supports the possible presence of CYP1A subfamily in the invertebrates, still today controversial, never identified previously in C. gallina and never isolated in the immune cells, as confirmed instead in this study with the identification of CYP1A-immunopositive protein (CYP1A-IPP). This protein could reveal a good biomarker at the base of a simple and quick method that could give clear information about specific pollutants presence, even at low concentrations in the environment where usually these organisms are fished before being commercialized. 3. In this experiment it has been evaluated the effect of the antibiotic chloramphenicol (CA) in an important species of commercial interest, Chamelea gallina. Chloramphenicol is a drug still used in some developing countries, also in veterinary field. Controls to evaluate its presence in the alimentary products of animal origin, can reveal ineffective whereas the concentration results to be below the limit of sensitivity of the instruments usually used in this type of analysis. Negative effects of CA towards the CYP1A- IPP proteins, underlined in this work, seem to be due to the attack of free radicals resultant from the action of the antibiotic. This brings to a meaningful alteration of the biotransformation mechanisms through the free radicals. It seems particularly interesting to pay attention to the narrow relationships in C. gallina, between SOD/CAT and CYP450 system, actively involved in detoxification mechanism, especially if compared with the few similar works today present about mollusc, a group that is composed by numerous species that enter in the food field and on which constant controls are necessary to evaluate in a rapid and effective way the presence of possible contaminations. 4. The investigations on fishes (Gadus morhua, and Salmo salar) and on a bivalve mollusc (Mytilus edulis) have allowed to evaluate different aspects related to the possibility to identify a biomarker for the evaluation of the health of organisms of food interest and consequently for the quality of the final product through 2DE methodologies. In the seafood field these techniques are currently used with a discreet success only for vertebrates (fishes), while in the study of the invertebrates (molluscs) there are a lot of difficulties. The results obtained in this work have underline several problems in the correct identification of the isolated proteins in animal organisms of which doesn’t currently exist a complete genomic sequence. This brings to attribute some identities on the base of the comparison with similar proteins in other animal groups, incurring in the possibility to obtain inaccurate data and above all discordant with those obtained on the same animals by other authors. Nevertheless the data obtained in this work after MALDI-ToF analysis, result however objective and the spectra collected could be again analyzed in the future after the update of genomic database related to the species studied. 4-A. The investigation about the presence of HSP70 isoforms directly induced by different phenomena of stress like B[a]P presence, has used bidimensional electrophoresis methods in C. gallina, that have allowed to isolate numerous protein on 2DE gels, allowing the collection of several spots currently in phase of analysis with MALDI-ToF-MS. The present preliminary work has allowed therefore to acquire and to improve important methodologies in the study of cellular parameters and in the proteomic field, that is not only revealed of great potentiality in the application in medical and veterinary field, but also in the field of the inspection of the foods with connections to the toxicology and the environmental pollution. Such study contributes therefore to the search of rapid and new methodologies, that can increase the inspective strategies, integrating themselves with those existing, but improving at the same time the general background of information related to the state of health of the considered animal organism, with the possibility, still hypothetical, to replace in particular cases the employment of the traditional techniques in the alimentary field.
