The ability of blocking ELISAs and haemagglutination-inhibition (HI) tests to detect antibodies in sera from chickens challenged with either Avibacterium (Haemophilus) paragallinarum isolate Hp8 (serovar A) or H668 (serovar C) was compared. Serum samples were examined weekly over the 9 weeks following infection. The results showed that the positive rate of serovar A specific antibody in the B-ELISA remained at 100% from the second week to the ninth week. In chickens given the serovar C challenge, the highest positive rate of serovar C specific antibody in the B-ELISA appeared at the seventh week (60% positive) and was then followed by a rapid decrease. The B-ELISA gave significantly more positives at weeks 2, 3, 7, 8 and 9 post-infection for serovar A and at week 7 post-infection for serovar C. In qualitative terms, for both serovar A and serovar C infections, the HI tests gave a lower percentage of positive sera at all time points except at 9 weeks post-infection with serovar C. The highest positive rate for serovar A HI antibodies was 70% of sera at the fourth and fifth weeks post-infection. The highest rate of serovar C HI antibodies was 20% at the fifth and sixth weeks post-infection. The results have provided further evidence of the suitability of the serovar A and C B-ELISAs for the diagnosis of infectious coryza.

The bacterial genus Helicobacter is a member of the Campylobacteriales in the Epsilonproteobacteria subphylum, and is comprised of organisms that are morphologically similar to Campylobacter. An outbreak of acute respiratory disease in layers was diagnosed as being of dual nature due to fowlpox and infectious laryngotracheitis using a multidisciplinary approach including virus isolation, histopathology, electron microscopy and polymerase chain reaction (PCR). The diagnosis was based on virus isolation of gallid herpesvirus 1 (GaHV-1) in chicken kidney cells and fowlpox virus (FWPV) in 9-day-old chicken embryonated eggs inoculated via the chorioallantoic membrane. The histopathology of tracheas from dead birds revealed intra-cytoplasmic and intra-nuclear inclusions suggestive of poxvirus and herpesvirus involvement. The presence of FWPV was further confirmed by electron microscopy, PCR and histology. All FWPV isolates contained the long terminal repeats of reticuloendotheliosis virus as demonstrated by PCR. GaHV-1 isolates were detected by PCR and were shown to have a different restriction fragment length polymorphism pattern when compared with the chicken embryo origin SA2 vaccine strain; however, they shared the same pattern with the Intervet chicken embryo origin vaccine strain. This is a first report of dual infection of chickens with GaHV-1 and naturally occurring FWPV with reticuloendotheliosis virus insertions. Further characterization of the viruses was carried out and the results are reported here.

Coccidiosis is a costly enteric disease of chickens caused by protozoan parasites of the genus Eimeria. Disease diagnosis and management is complicated since there are multiple Eimeria species infecting chickens and mixed species infections are common. Current control measures are only partially effective and this, combined with concerns over vaccine efficacy and increasing drug resistance, demonstrates a need for improved coccidiosis diagnosis and control. Before improvements can be made, it is important to understand the species commonly infecting poultry flocks in both backyard and commercial enterprises. The aim of this project was to conduct a survey and assessment of poultry Eimeria across Australia using genetic markers, and create a collection of isolates for each Eimeria species. A total of 260 samples (faecal or caecal) was obtained, and survey results showed that Eimeria taxa were present in 98% of commercial and 81% of backyard flocks. The distribution of each Eimeria species was widespread across Australia, with representatives of all species being found in every state and territory, and the Eimeria species predominating in commercial flocks differed from those in backyard flocks. Three operational taxonomic units also occurred frequently in commercial flocks highlighting the need to understand the impact of these uncharacterised species on poultry production. As Eimeria infections were also frequent in backyard flocks, there is a potential for backyard flocks to act as reservoirs for disease, especially as the industry moves towards free range production systems. This Eimeria collection will be an important genetic resource which is the crucial first step in the development of more sophisticated diagnostic tools and the development of new live vaccines which ultimately will provide savings to the industry in terms of more efficient coccidiosis management.

Limitations in quality bedding material have resulted in the growing need to re-use litter during broiler farming in some countries, which can be of concern from a food-safety perspective. The aim of this study was to compare the Campylobacter levels in ceca and litter across three litter treatments under commercial farming conditions. The litter treatments were (a) the use of new litter after each farming cycle; (b) an Australian partial litter re-use practice; and (c) a full litter re-use practice. The study was carried out on two farms over two years (Farm 1, from 2009–2010 and Farm 2, from 2010–2011), across three sheds (35,000 to 40,000 chickens/shed) on each farm, adopting three different litter treatments across six commercial cycles. A random sampling design was adopted to test litter and ceca for Campylobacter and Escherichia coli, prior to commercial first thin-out and final pick-up. Campylobacter levels varied little across litter practices and farming cycles on each farm and were in the range of log 8.0–9.0 CFU/g in ceca and log 4.0–6.0 MPN/g for litter. Similarly the E. coli in ceca were ∼log 7.0 CFU/g. At first thin-out and final pick-up, the statistical analysis for both litter and ceca showed that the three-way interaction (treatments by farms by times) was highly significant (P < 0.01), indicating that the patterns of Campylobacter emergence/presence across time vary between the farms, cycles and pickups. The emergence and levels of both organisms were not influenced by litter treatments across the six farming cycles on both farms. Either C. jejuni or C. coli could be the dominant species across litter and ceca, and this phenomenon could not be attributed to specific litter treatments. Irrespective of the litter treatments in place, cycle 2 on Farm 2 remained campylobacter-free. These outcomes suggest that litter treatments did not directly influence the time of emergence and levels of Campylobacter and E. coli during commercial farming.

The problem of ‘wet litter’, which occurs primarily in grow-out sheds for meat chickens (broilers), has been recognised for nearly a century. Nevertheless, it is an increasingly important problem in contemporary chicken-meat production as wet litter and associated conditions, especially footpad dermatitis, have developed into tangible welfare issues. This is only compounded by the market demand for chicken paws and compromised bird performance. This review considers the multidimensional causal factors of wet litter. While many causal factors can be listed it is evident that the critical ones could be described as micro-environmental factors and chief amongst them is proper management of drinking systems and adequate shed ventilation. Thus, this review focuses on these environmental factors and pays less attention to issues stemming from health and nutrition. Clearly, there are times when related avian health issues of coccidiosis and necrotic enteritis cannot be overlooked and the development of efficacious vaccines for the latter disease would be advantageous. Presently, the inclusion of phytate-degrading enzymes in meat chicken diets is routine and, therefore, the implication that exogenous phytases may contribute to wet litter is given consideration. Opinion is somewhat divided as how best to counter the problem of wet litter as some see education and extension as being more beneficial than furthering research efforts. However, it may prove instructive to assess the practice of whole grain feeding in relation to litter quality and the incidence of footpad dermatitis. Additional research could investigate the relationships between dietary concentrations of key minerals and the application of exogenous enzymes with litter quality.

1. The biopotencies relative to beta-carotene of several apocarotenoids, such as 8'-, 10'- and 12'-apo-beta-carotenal and methyl 8'-apo-beta-carotenoate, were investigated in rats, on a molar basis, by both curative-growth assay and liver-storage tests. 2. In the curative-growth assays, on a molar basis the biopotencies of 8'-, 10'- and 12'-apo-beta-carotenal and methyl 8'-apo-beta-carotenoate were 72, 78, 72 and 53% respectively, whereas on a weight basis the corresponding values were 93, 111, 111 and 63%, with respect to beta-carotene taken as 100%. In terms of yield of vitamin A, these values were much lower in the liver-storage tests. 3. When 8'-apo-beta-carotenal was fed, the unchanged aldehyde together with small amounts of the corresponding alcohol and larger proportions of the acid rapidly appeared in the tissues of both rats and chickens. The 8'-apocarotenol, 8'-apocarotenoic acid and its methyl ester were absorbed unchanged. The free acid disappeared most rapidly from the tissues, but its methyl ester persisted in the tissues longest. 4. On the basis of these observations it is suggested that most of an apocarotenal is oxidized to the corresponding acid, which, in turn, is mostly degraded to retinoic acid, with small proportions of it being attacked by the dioxygenase system giving retinal.

1. (1) The relative abilities of the various cell fractions of rat and chicken liver to oxidize and reduce retinal and 8'- and 12'-apo-β-carotenal were investigated and it has been shown that, while retinal is exclusively oxidized by the soluble fraction, the apocarotenals are mostly oxidized by the particulate fractions of the homogenate. 2. (2) Addition of NAD+ or NADP+ markedly activated the oxidation of the apocarotenals, but not of retinal by the particulate fractions. 3. (3) Considerable amounts of retinal and 8'-, 10'- and 12'-apo-β-carotenal were isolated from the intestine of chickens fed β-carotene and these apocarotenoids were conclusively identified. 4. (4) Significant amounts of 8'-, 10'- and 12'-apo-β-carotenoic acids were isolated from the intestine of rats given 8'-apo-β-carotenal and these apocarotenoic acids were also conclusively identified. 5. (5) In the light of these observations it is suggested that during conversion to vitamin A, the β-carotene molecule is simultaneously attacked by the dioxygenase at several double bonds, the primary attack being at the central double bond and a tentative scheme for the mechanism of conversion is proposed.

En el presente estudio se evaluaron tres periodos de alimento de retiro o acabado en broilers (o, 3, 6 y 9 días antes de la matanza) el cual contiene 3,225 kcal EM/kg de alimento y 19% de proteína, ajustándose a los requerimientos nutricionales recomendados por Arbor Acres Farm Inc., (1992), en especial a la proporción energia-proteina. La evaluación tuvo lugar en la Granja Buenos Aires propiedad de la Empresa Tip-Top Industrial, S.A., con una duración de 42 días, en donde se utilizaron 800 pollos de engorde sin sexar (mixtos) de la línea Peterson-Arbor Acres de un día de edad, dichos pollos fueron distribuidos aleatoriamente en cuatro tratamientos: T1 suministro de alimento de retiro por nueve días), T2 suministro de alimento de retiro por seis días), T3, (Suministro de alimento de retiro por tres días) y T4 (testigo, con cero días de alimento de retiro), con cuatro repeticiones cada uno, distribuidos en un Diseño completamente al Azar sometidos a la prueba de rangos múltiples de Duncan. Las variables estudiadas fueron: consumo de alimento, peso vivo, ganancia de peso, conversión alimenticia, mortalidad vs viabilidad, rendimiento en la canal, análisis económico. No se encontraron diferencias significativas (P<0.05) entre los tratamientos al final del ensayo para el consumo de alimento, conversión alimenticia y costo de alimento, pero no así ara peso vivo y ganancia de peso. El porcentaje de mortalidad acumulada total fue de 2.5% y los rendimientos de la canal fueron: (90.74)T1 (88.98)T2, (85.86)T3,y (90.31)T4, Se corolo que el periodo de suministro de alimento de retiro que permitió los mejores pesos vivos, ganancias de pesos y conversiones alimenticias fue el periodo de tres días (T3), sin embargo, obtuvo el mayor consumo total para generar un peso promedio en la canal de 3.34 lbs, además, presentó el mayor costo alimenticio entre los tratamientos experimentales (T1, T2, T3) y bajo rendimiento en la canal, contrario al T1 que aunque los parámetros productivos fueron menores que el T3 obtuvo el mayor rendimiento en la canal con un menor costo alimenticio. El alimento de retiro no ejerció efecto sobre las variables estudiadas lo que demuestra que dicho alimento pueda ser una alternativa viable para disminuir costo alimenticio.

We have constructed plasmids to be used for in vitro signature-tagged mutagenesis (STM) of Campylobacter jejuni and used these to generate STM libraries in three different strains. Statistical analysis of the transposon insertion sites in the C. jejuni NCTC 11168 chromosome and the plasmids of strain 81-176 indicated that their distribution was not uniform. Visual inspection of the distribution suggested that deviation from uniformity was not due to preferential integration of the transposon into a limited number of hot spots but rather that there was a bias towards insertions around the origin. We screened pools of mutants from the STM libraries for their ability to colonize the ceca of 2-week-old chickens harboring a standardized gut flora. We observed high-frequency random loss of colonization proficient mutants. When cohoused birds were individually inoculated with different tagged mutants, random loss of colonization-proficient mutants was similarly observed, as was extensive bird-to-bird transmission of mutants. This indicates that the nature of campylobacter colonization in chickens is complex and dynamic, and we hypothesize that bottlenecks in the colonization process and between-bird transmission account for these observations.

Con el objetivo de reducir los costos de alimentación en broilers se le retiró al alimento finalizador los aditivos más caros y se evaluaron los principales índices productivos. Durante un período de 42 días los pollos de engorde fueron evaluados en la granja avícola "Buenos Aires" propiedad de la empresa Tip-Top industrial S.A., se utilizaron 800 pollos de engorde de un día de edad de la estirpe Peterson x Arbor Acres (sin sexar), se realizó un DCA con cuatro tratamientos y cuatro réplicas cada uno y fueron sometidos a la prueba de Duncan para determinar la superioridad por tratamiento y a la vez se le efectúo Análisis de Costo por alimentación. Los pollos del TI (12 días), T2 (nueve días) y T3 (seis días) de retiro, consumieron al final del período de crianza 3.44, 3.44 y 3.47 kg, mientras que para T4 (tres días) fue de 3.52 kg. El peso vivo final hasta las seis semanas fue de 1.87, 1.87, 1.89 y 1.90 kg. Para los TI, T2, T3 y T4 respectivamente. La ganancia media de peso a los 28-42 días fue de 0.81, 0.79, 0.84 y 0.80 kg para los tratamientos TI, T2, T3 y T4 respectivamente. La conversión alimenticia por tratamiento fue de 1.84 (TI, T2 y T3) y 1.85 (T4). El rendimiento en la canal fue de 82.00, 83.00, 85.00, y 85.00 para los tratamientos 1, 2, 3 y 4 respectivamente. No se encontró diferencia significativa (P<0.05) para las variables mencionadas en estudio sometidas al diseño estadístico, la mortalidad total acumulada fue de 2.37%. Usando el alimento de retiro seis días antes de la matanza se disminuyeron los costos alimenticios en $ 0.016 por kg de carne procesada.

El presente trabajo fue realizado en la granja avícola "La Trinidad", propiedad de la Empresa perteneciente a la TIP - TOP Industrial S.A. ubicado en el Km. 26 Carretera a Masaya, entrada principal a Nindirí 5 Km al Norte de la comunidad San Francisco, el experimento se realizó entre el 28 de Febrero y 10 de Abril de 1996, se utilizaro n 85.680 pollos de engorde (machos y hembras), de un día de nacido incluyéndose para dichos experimentos cuatro tratamientos con dos repeticiones cada uno, T1 en el cual se aplicó un programa de 23 hrs/luz durante el intervalo de edad de los 12 a los 42 días, T2 a partir de los 15 días a 42 días con un suministro de 23 H/Luz; T3 a partir de 18dlas a 42 días con 23 h/Luz (Tratamiento testigo), T4 entre los 21 días a los 42 días con 23 h/Luz. Las variables estudiadas para el experimento fueron las siguientes: Consumo de Alimento, Peso Vivo, Conversión de Alimento, Ganancia Media Diaria y Mortalidad.

Con el objetivo de evaluar la aplicación de alimento de retiro en broilers en 5 periodos diferentes de (0,4,6,8 y 10 días) antes del sacrificio se realizó el ensayo compuesto de 4 repeticiones cada uno, distribuidas en un diseño completamente al azar, con el uso de raciones isocalóricas e isoproteicas. Se estudiaron los siguientes variables: consumo de alimento, peso vivo, ganancia de peso, conversión alimenticia, mortalidad, rendimiento en canal y análisis financiero. Los resultados, no arrojaron diferencias significativas entre los tratamientos para: consumo de alimento, conversión alimenticia, ganancia de peso y peso vivo; pero se presentaron diferencias entre los tratamientos en cuanto a los costos de producción unitaria. El porcentaje de mortalidad acumulada alcanzó el 3% y los rendimientos de la canal fueron: para el t1 (70.2%), t2 (69.7%), t3 (70.0%), t4 (10.1%) y t5 (75.2%). El t5, fue el tratamiento que obtuvo la conversión alimenticia más elevada en comparación con el testigo y los otros tratamientos experimentales. Sin embargo, el que presentó los mejores resultados en cuanto a consumo de alimentos, rendimiento en canal, ganancia de peso, y peso vivo, fue el tratamiento (t5), con un rendimiento en canal caliente de 75.2%. Este tratamiento, presentó los mayores parámetros productivos y mayor ahorro de (29.8702 U.S $) por camada de 140 pollos, utilizando la ración de retiro 10 días antes de la matanza. Esto justifica el uso de este tipo de alimento durante este período, con lo cual queda demostrado que pueden obtenerse resultados satisfactorios y que es una alternativa para disminuir los costos de alimentación en Broilers.

Con el propósito de comparar el valor nutritivo del maíz amarillo y el sorgo rojo en la alimentación de pollos asaderos, se hizo un experimento en la (ENAG) Escuela Nacional de Agricultura y Ganadería. Se utilizaron dos raciones: 1) Maíz amarillo + base concentrada comercial. 2) Sorgo rojo + base concentrada comercial. Se usaron 80 pollos Vantress Cross de un día de nacidos, sin sexar que fueron colocados en un compartimiento por 2 semanas, donde se les suministro un mismo régimen alimenticio. Fueron vacunados, despicados, pesados e identificados para luego ser trasladados a las creadoras para dar inicio al experimento. El diseño estadístico usado fue completamente al azar, con dos tratamientos y dentro de cada tratamiento 4 lotes. La cantidad de pollos por tratamiento fue de cuarenta, con dos lotes de once pollos cada uno y los dos restantes con nueve pollos cada uno. Se tomaron datos de la ganancia de peso semanal, consumo semanal de alimento y factor de conversión semanal. Al finalizar el experimento se observo que los pollos alimentados con la ración que llevaba sorgo rojo, alcanzaron pesos mayores que los alimentados con la ración que contenía maíz amarillo, sin embargo al efectuar el análisis estadístico no fue significativo dicho aumento. Se noto que hubo un consumo mas elevado en los pollos alimentados con la ración que contenía sorgo rojo, y sin embargo el factor de conversión fue un poco mas elevado que el de los pollos alimentados con la ración que contenía maíz amarillo. Al realizar la evaluación económica se noto que el costo por pollo del tratamiento que contenía maíz amarillo fue C 0.85 mas caro que el costo por pollo del tratamiento que contenía sorgo rojo. Para efectuar los análisis estadísticos se hizo uso de las ganancias de peso de cada pollo, dicho análisis no mostró diferencias estadisticamente significativas entre los distintos lotes, por lo tanto se procedió a realizar otro análisis en donde se unió los grados de libertad de los lotes/tratamientos con los de entre individuos dentro de lotes para obtener mayor precisión y nuevamente no se encontró diferencias estadisticamente significativas entre la ración que contenía sorgo rojo y la que contenía maíz amarillo. Es de hace notar que al final del experimento, el 75 por ciento de los pollos eran hembras y se encontraban distribuidos en igual numero en ambos tratamientos. Los pollos del tratamiento de maíz amarillo presentaron coloración amarilla intenso en las patas, pico y piel, en cambio los del tratamiento con sorgo rojo fueron de un color amarillo pálido.
