During the construction of five residential buildings in the city of Taubate, State of São Paulo, it was possible to carry out one comprehensive investigation of the behavior of precast concrete piles in clay shales. This paper describes the results of Dynamic Load Tests (DLT's) executed in three piles with different diameters and with the same embedded length. The tests were monitored using the PDA(R) (Pile Driving Analyzer) and the pile top displacement was measured by pencil and paper procedure. From the curves of RMX versus DMX resulted from CASE(R) method, CAPWAPC(R) analyses were made for signals where the maximum mobilized soil resistance was verified. The results were compared with the predicted bearing capacity using the semi-empirical method of Decourt & Quaresma (1978) and Decourt (1982) based on SPT values and the description of the soil profile. Some comments related to the values of quake and damping used for clay shales in the analyses are also presented.

Currently, the major drawback of gene therapy is the gene transfection rate. The two main types of vectors that. are used in gene therapy are based on viral or non-viral gene delivery systems. There are several non-viral systems that can be used to transfer foreign genetic material into the human body. In order to do so, the DNA to be transferred must escape the processes that affect the disposition of macromolecules. These processes include the interaction with blood components, vascular endothelial cells and uptake by the reticuloendothelial system. Furthermore, the degradation of therapeutic DNA by serum nucleases is also a potential obstacle for functional delivery to the target cell. Cationic polymers have a great potential for DNA complexation and may be useful as non-viral vectors for gene therapy applications. The objective of this review was to address the state of the art in gene therapy using synthetic and natural polycations and the latest strategies to improve the efficiency of gene transfer into the cell.

Many studies have assessed the process of forest degradation in the Brazilian Amazon using remote sensing approaches to estimate the extent and impact by selective logging and forest fires on tropical rain forest. However, only a few have estimated the combined impacts of those anthropogenic activities. We conducted a detailed analysis of selective logging and forest fire impacts on natural forests in the southern Brazilian Amazon state of Mato Grosso, one of the key logging centers in the country. To achieve this goal a 13-year series of annual Landsat images (1992-2004) was used to test different remote sensing techniques for measuring the extent of selective logging and forest fires, and to estimate their impact and interaction with other land use types occurring in the study region. Forest canopy regeneration following these disturbances was also assessed. Field measurements and visual observations were conducted to validate remote sensing techniques. Our results indicated that the Modified Soil Adjusted Vegetation Index aerosol free (MSAVI(af)) is a reliable estimator of fractional coverage under both clear sky and under smoky conditions in this study region. During the period of analysis, selective logging was responsible for disturbing the largest proportion (31%) of natural forest in the study area, immediately followed by deforestation (29%). Altogether, forest disturbances by selective logging and forest fires affected approximately 40% of the study site area. Once disturbed by selective logging activities, forests became more susceptible to fire in the study site. However, our results showed that fires may also occur in undisturbed forests. This indicates that there are further factors that may increase forest fire susceptibility in the study area. Those factors need to be better understood. Although selective logging affected the largest amount of natural forest in the study period, 35% and 28% of the observed losses of forest canopy cover were due to forest fire and selective logging combined and to forest fire only, respectively. Moreover, forest areas degraded by selective logging and forest fire is an addition to outright deforestation estimates and has yet to be accounted for by land use and land cover change assessments in tropical regions. Assuming that this observed trend of land use and land cover conversion continues, we predict that there will be no undisturbed forests remaining by 2011 in this study site. Finally, we estimated that 70% of the total forest area disturbed by logging and fire had sufficiently recovered to become undetectable using satellite data in 2004. (C) 2010 Elsevier B.V. In the Cuiabá region-State of Mato Grosso, Central Brazil-primary gold mineralization is hosted by two generations of quartz veins in Precambrian metamorphic terrains of the Cuiabá Group. Gold is mined from the veins and mainly from the eluvial horizons that cover the deeply altered basement. In the lodes gold occurs as small particles (less than 1 mm) associated with pyrite and contains up to 5% Ag. Larger particles and nuggets of almost pure gold are found in the iron duricrust which caps the upper levels of the weathering profile. It is difficult to determine the average grade of this kind of deposit but some prospects in the Cuiabá region produce up to 2 g gold per ton of ore. Lateritization is responsible for both the formation of the iron crust and the concentration of gold within the regolith. Under a tropical climate, the supergene alteration of phyllites of the Cuiabá Group has led to the formation of a weathering profile consisting typically of saprolite, mottled clay zone and duricrust, from bottom to top. The duricrust is directly derived form the in situ weathering of phyllites. Geochemical balance calculations indicate that in the transition from the saprolite to the duricrust lateritization has promoted a progressive loss of Si, Al and K, and more than 500% of absolute Fe enrichment. Gold underwent a supergene evolution related to the development of the weathering profile. In the saprolite and mottled clay zone, associated with quartz and oxidized sulfides, gold dissolves as demonstrated by corrosion features at the surface of the particles. The formation of secondary gold in the duricrust is indicated by the larger size of the nuggets, their higher fineness and the close relationship between gold and the neoformed iron oxy-hydroxides. © 1991.

Chemical analyses for biotites and their host rocks from the Cabreúva (three facies) and Salto (five facies) intrusions from the multiple-centered rapakivi Itu Complex, State of São Paulo, Brazil, are presented and compared. The Cabreúva intrusion comprises different kinds of mainly even-grained biotite and hornblende-bearing syenogranites, monzogranites and quartz syenites and the Salto intrusion several types of mainly porphyritic biotite syenogranites, some of them hornblende-bearing. The biotites from the Salto intrusion (S-micas) show a more restricted composition than those from the Cabreúva intrusion (C-micas). This reflects the chemical variability of the two bodies which is smaller in the Salto intrusion and larger in the Cabreúva pluton. In the AlIV x Fet/(Fet+Mg) diagram the S- and C-micas show similar AlIV contents, around 2.2-2.3, but C-micas have higher Fet/(Fet+Mg) ratios (0.7-0.9) compared to those of S-micas (0.5-0.6). In the Mg:(Al+Fe+3+Ti):(Fe+2+Mn) diagram the S-micas are defined as Fe+2-biotites and the C-micas occupy the area between the Fe+2-biotites and the siderophyllite/lepidomelane fields, slightly overlapping the latter. In the Al2O3 × FeOt, MgO × FeOt, Al2O3 x MgO and Alt x Mg diagrams, the S-micas always lie on the calc-alkaline/alkaline boundary (or in the subalkaline field) whereas the C-micas systematically plot in the alkaline field, reflecting the higher alkalis content of the Cabreúva intrusion. In the Fet/(Fet+Mg) x SiO2 diagram, the S-micas lie on a smooth line whereas the C-micas from the different facies are separated by distinct chemical gaps reflecting the major or minor chemical overlapping of the facies from the Salto and Cabreúva intrusions.
Pollution by hexachlorobenzene and pentachlorophenol in the coastal plain of São Paulo state, Brazil

Organochlorine compounds were dumped by chemical industries during the 1970s in many areas of the coastal plain of São Paulo state in Brazil. These dumps, located on hillsides and in valleys, in both rural and urban environments, are responsible for soil and water pollution. The objective of this work was to determine how the pollutants have spread in an area occupied by a spodosol-type soil mantle. The study combines soil morphological observations with soil and water analysis of hexachlorobenzene (HCB) and pentachlorophenol (PCP) in soil toposequences. The results indicate that the highest pollutant concentrations are observed near the dump site and that the compounds contamination is increasing. A map integrating topography and chemical concentrations was created to visualize the spatial distribution of HCB levels in the landscape. Physical and chemical analyses were performed to measure HCB and PCP levels in the soil. Soil water appears to act as a vector of HCB, probably through complexation with and dispersal of dissolved organic matter. The persistence of HCB at the studied site is most likely due to the low pH values in combination with a high content of organic matter. HCB was consistently found in higher concentrations than PCP. It is plausible that the cause of this difference is that PCP is degraded more easily under sunlight than HCB and that degradation of PCP under acid conditions leads to the formation of HCB. © 2003 Published by Elsevier B.V.

Natural environments have been worldwide affected by the growing impact of anthropogenic actions that promote the reduction or the extinction of several vertebrate species. Aquatic ecosystems represent one of the most affected environments and many fish species and/or populations have been increasingly fragmented distributed due to habitat degradation, predatory fishing, introduction of exotic species, river sedimentation, deforestation, pollution, reduction of food resource, and construction of hydroelectric dams. Actually, more than 150 Brazilian fish species, including freshwater, estuary and coastal species, can be considered threatened. Information on the diversity, conservation biology and population analysis on threatened species or populations, with several DNA markers, can be extremely useful for the success of fish species-recovery and maintenance programs. Although DNA analysis in Neotropical fish species are just beginning, they tend to increase with the widespread attention to the use of molecular approaches to minimize problems related to the risk of extinction. The accumulation of information on biology and pattern of genetic variation of fish species, associated with ecological and demographic data, and also education and respect to the nature, constitutes a crucial task to develop efficient conservation strategies in order to preserve the genetic diversity in aquatic environments.

Background: Cancer-cachexia induces a variety of metabolic disorders on protein turnorver, decreasing protein synthesis and increasing protein degradation. Controversly, insulin, other hormones, and branched-chain amino acids, especially leucine, stimulate protein synthesis and modulate the activity of translation initiation factors involved in protein synthesis. Since the tumour effects are more pronounced when associated with pregnancy, ehancing muscle-wasting proteolysis, in this study, the influence of a leucine-rich diet on the protein synthesis caused by cancer were investigated. Methods: Pregnant rats with or without Walker 256 tumour were distributed into six groups. During 20 days of experiment, three groups were fed with a control diet: C - pregnant control, W - tumour-bearing, and P - pair-fed, which received the same amount of food as ingested by the W group; three other groups of pregnant rats were fed a leucine-rich diet: L - pregnant leucine, WL - tumour-bearing, and PL - pair-fed, which received the same amount of food as ingested by the WL group. Results: The gastrocnemius muscle of WL rats showed increased incorporation of leucine in protein compared to W rats; the leucine-rich diet also prevented the decrease in plasma insulin normally seen in W. The expression of translation initiation factors increased when tumour-bearing rats fed leucine-rich diet, with increase of ∼35% for eIF2α and eIF5, ∼17% for eIF4E and 20% for eIF4G; the expression of protein kinase S6K1 and protein kinase C was also highly enhanced. Conclusion: The results suggest that a leucine-rich diet increased the protein synthesis in skeletal muscle in tumour-bearing rats possibly through the activation of eIF factors and/or the S6kinase pathway. © 2007 Ventrucci et al; licensee BioMed Central Ltd.

This study aimed to evaluate the environmental quality of the marine portion of Xixová-Japuí State Park (XJSP), an urban marine protected area, which is influenced by multiple contamination sources, by using ecotoxicological and geochemical analyses. Sediments were predominantly sandy, with low CaCO3 and organic matter contents, and presented contamination by metals (Cd,Cu,Zn). Acute toxicity was detected in three tested samples, and copepod exposed to sediments from four stations exhibited lower fecundities, despite the absence of significant effects. Contamination and toxicity seemed to be associated, suggesting that the environment is degraded and presents risks to the biota. Whole sediment TIE indicated ammonia as a main responsible for toxicity, suggesting sewage is a main contributor to sediment degradation. As external contamination sources seem to be negatively influencing the sediment quality, the park conservation objectives are not being fully reached, demanding actions to mitigate impacts. © 2013 Elsevier Ltd.

Este trabalho apresenta dados geológicos, petrográficos e mineralógicos referentes ao granito que hospeda o depósito aurífero Tocantinzinho e objetivou contribuir ao entendimento dos processos hidrotermais associados à sua gênese. O depósito ocorre em biotita monzogranito tardi a pós-tectônico, do subtipo oxidado da série ilmenita, que foi alojado a profundidades de 6 - 9 km. Esse granitoide encontra-se bastante fraturado e localmente brechado, tendo experimentado processos hidrotermais de grau fraco a moderado, os quais geraram duas principais variedades (salame e smoky) sem diferenças mineralógicas ou químicas importantes, porém macroscopicamente muito distintas. Vários tipos de alteração hidrotermal foram reconhecidos nas rochas granitoides, sendo representados principalmente por vênulas e pela substituição de minerais primários. A história hidrotermal teve início com a microclinização, durante a qual o protólito granítico foi em parte transformado na variedade salame. A temperaturas em torno de 330 oC ocorreu a cloritização, que produziu chamosita com XFe na faixa de 0,55 - 0,70. Seguiu-se a sericitização, durante a qual os fluidos mineralizadores precipitaram pirita, calcopirita, esfalerita, galena e ouro. À medida que a alteração progrediu, as soluções se saturaram em sílica e precipitaram quartzo em vênulas. No estágio mais tardio (carbonatação), provavelmente houve mistura entre fluidos aquosos e aquocarbônicos, de que teria resultado a reação entre Ca2+ e CO2 e formação de calcita. A maioria dos sulfetos encontra-se em vênulas, algumas em trama stockwork. O ouro é normalmente muito fino e ocorre principalmente como inclusões submicroscópicas ou ao longo de microfraturas em pirita e quartzo. O depósito Tocantinzinho é muito similar aos depósitos Batalha, Palito e São Jorge, e aos do campo Cuiú-Cuiú. Tipologicamente poderia ser classificado como depósito relacionado a intrusões.

O SW do município de Presidente Figueiredo, localizado no Estado do Amazonas, Nordeste do Cráton Amazônico Central, Brasil, hospeda granitoides do tipo I de idade entre 1890 a 1898 Ma (Terra Preta Granito, Suíte Água Branca), hornblenda-sienogranitos do tipo A (Sienogranito Canoas da Suíte Mapuera), rochas vulcânicas ácidas à intermediárias (Grupo Iricoumé) e granitos rapakivi de idades entre 1883 a 1889 Ma (Granito São Gabriel da Suíte Mapuera), e rochas afins (quartzo-gabro-anortosito e diorito), além de quartzo-monzonito Castanhal, milonitos e hornfels. A fácies quartzo-diorito do granito Terra Preta foi formada por processos de mistura entre um dique quartzo-gabro sinplutônico e um granodiorito hornblenda. Glóbulos parcialmente assimilados de sienogranitos hornblenda Canoas e seus contatos com o granodiorito hornblenda Terra Preta sugerem que o sienogranito Canoas é um pouco mais jovem do que o Granito Terra Preta. Xenólitos do sienogranito Canoas no interior do Granito São Gabriel mostram que o granito é mais jovem do que o sienogranito Canoas. Novas evidências geológicas e petrográficas avançam na compreensão petrológica destas rochas e sugerem que, além de cristalização fracionada, assimilação e mistura de magma, desempenharam um papel importante, pelo menos em escala local, na evolução e variação composicionais dos plutons. Tal evidência é encontrada no Granito Terra Preta misturado com materiais quartzo-diorito, félsico associado ao sienogranito Canoas e nos enclaves microgranulares intermediários, que apresentam biotita e hornblenda primárias, além de dissolução plagioclásio, corrosão de feldspatos, mantos feldspatos alcalinos, segunda geração de apatita, e elevados teores xenocristais em enclaves intermediários formados a partir da fragmentação de intrusões máficas. Análises petrográficas mostram que um evento deformacional registrado na parte Ocidental da área de estudo (com deformação progressiva de E para W) é estimado entre o magmatismo pós-colisional de 1,90 Ga e as invasões do Granito São Gabriel e rochas afins máficas/intermediárias (intraplaca). No entanto, torna-se extremamente necessário obter idades absolutas para este evento metamórfico.

Polygalacturonases are enzymes involved in the degradation of pectic substances, being extensively used in food industries, textile processing, degumming of plant rough fibres, and treatment of pectic wastewaters. Polygalacturonase (PG) production by thermophilic fungus Thermoascus aurantiacus on solid-state fermentation was carried out in culture media containing sugar cane bagasse and orange bagasse in proportions of 30% and 70% (w/w) at 45°C for 4 days. PG obtained was purified by gel filtration and ion-exchange chromatography. The highest activity was found between pH 4.5 and 5.5, and the enzyme preserved more than 80% of its activity at pH values between 5.0 and 6.5. At pH values between 3.0 and 4.5, PG retained about 73% of the original activity, whereas at pH 10.0 it remained around 44%. The optimum temperature was 60–65°C. The enzyme was completely stable when incubated for 1 hour at 50°C. At 55°C and 60°C, the activity decreased 55% and 90%, respectively. The apparent molecular weight was 29.3 kDa, Km of 1.58 mg/mL and Vmax of 1553.1μmol/min/mg. The presence of Zn+2, Mn+2, and Hg+2 inhibited 59%, 77%, and 100% of enzyme activity, respectively. The hydrolysis product suggests that polygalacturonase was shown to be an endo/exoenzyme.
