Biofilms represent the predominant mode of microbial growth in the natural environment. Bacillus subtilis is a ubiquitous Gram-positive soil bacterium that functions as an effective plant growth-promoting agent. The biofilm matrix is composed of an exopolysaccharide and an amyloid fiber-forming protein, TasA, and assembles with the aid of a small secreted protein, BslA. Here we show that natively synthesized and secreted BslA forms surface layers around the biofilm. Biophysical analysis demonstrates that BslA can self-assemble at interfaces, forming an elastic film. Molecular function is revealed from analysis of the crystal structure of BslA, which consists of an Ig-type fold with the addition of an unusual, extremely hydrophobic "cap" region. A combination of in vivo biofilm formation and in vitro biophysical analysis demonstrates that the central hydrophobic residues of the cap are essential to allow a hydrophobic, nonwetting biofilm to form as they control the surface activity of the BslA protein. The hydrophobic cap exhibits physiochemical properties remarkably similar to the hydrophobic surface found in fungal hydrophobins; thus, BslA is a structurally defined bacterial hydrophobin. We suggest that biofilms formed by other species of bacteria may have evolved similar mechanisms to provide protection to the resident bacterial community.

Clostridium perfringens est ubiquitaire dans l’environnement. Ce microorganisme peut être retrouvé dans la flore normale du tractus gastro-intestinal des mammifères et peut également causer une variété d’infections intestinales. Le phénotype de résistance à la bacitracine a déjà été rapporté chez C. perfringens mais les gènes associés n’ont pas été caractérisés. Dans cette étude, 24 des 99 isolats de C. perfringens aviaires testés ont démontré une résistance à la bacitracine. Les analyses ont révélé la présence d’un transporteur ABC ainsi que d’une undécaprénol kinase surproduite. Ces deux mécanismes semblent être codés par l’opéron bcrABDR. En amont et en aval des gènes bcr, un élément IS1216-like a été identifié, celui-ci pouvant jouer un rôle dans la dissémination de la résistance à la bacitracine. Des analyses d’hybridation sur ADN ont révélé que les gènes bcrABDR étaient localisés sur le chromosome. De plus, il a été démontré que les gènes bcr étaient exprimés en présence de bacitracine. Plusieurs études ont associé la tolérance aux antibiotiques et aux désinfectants à la formation de biofilm. Dans la littérature, peu d’informations sont disponibles sur le biofilm de C. perfringens. La majorité des isolats testés dans cette étude ont démontré la formation d’un biofilm. L’analyse de la matrice a démontré que celle-ci contenait des protéines, de l’ADN extracellulaire ainsi que des polysaccharides liés en bêta-1,4. Une meilleure survie des cellules en biofilm a été observée suite à une exposition à de fortes concentrations d’antibiotiques. Une exposition à de faibles doses de certains antibiotiques semblait diminuer le biofilm formé alors que pour d’autres, le biofilm semblait augmenter. Dans la présente étude, la susceptibilité des biofilms de C. perfringens à la désinfection a été également analysée. Les résultats ont démontré que la formation de biofilm protégeait les cellules de l’action du monopersulfate de potassium, des ammoniums quaternaires, du peroxyde d’hydrogène et du glutéraldéhyde. Toutefois, l’hypochlorite de sodium a été démontré comme étant efficace contre le biofilm de C. perfringens. Il a été démontré que les biofilms mixtes de C. perfringens cultivés en présence de Staphylococcus aureus ou d’Escherichia coli étaient plus résistants à la désinfection en comparaison aux biofilms simples de S. aureus ou d’E. coli. Toutefois, le biofilm simple de C. perfringens était plus résistant à la désinfection que les biofilms mixtes. Finalement, les profils de transcription entre les populations planctoniques et en biofilm ont été analysés par séquençage d’ARN. L’analyse transcriptomique du biofilm a identifié 238 gènes différentiellement exprimés entre les deux conditions. Les gènes négativement régulés sont impliqués dans la virulence, la production d’énergie, le métabolisme des sucres ainsi que dans la biosynthèse des acides gras et des acides aminés alors que les gènes induits sont impliqués dans la réponse au stress et au stress oxydatif, dans la biosynthèse d’acides gras et de phospholipides ainsi que dans la virulence. Cette étude décrit pour la première fois la découverte des gènes associés à la résistance à la bacitracine chez C. perfringens. Elle rapporte également de nouvelles données sur la matrice du biofilm, la tolérance aux antibiotiques et aux désinfectants ainsi que sur le transcriptome du biofilm de C. perfringens.

Biofilm forming multidrug resistant Staphylococcus spp. are major reservoirs for transmission of ophthalmic infections. They were isolated from ocular patients suffering from conjunctivitis. In this study we analyzed biofilm forming ability, antibiotic resistance profile of the Staphylococcus spp. isolated from clinical ocular patients, and their phylogenetic relationship with other community MRSA. Sixty Staphylococcus spp. strains isolated from clinical subjects were evaluated for their ability to form biofilm and express biofilm encoding ica gene. Among them 93% were slime producers and 87% were slime positive. Staphylococcus aureus and S. epidermidis were dominant strains among the isolates obtained from ocular patients. The strains also exhibited a differential biofilm formation quantitatively. Antibiotic susceptibility of the strains tested with Penicillin G, Ciprofloxacin, Ofloxacin, Methicillin, Amikacin, and Gentamicin indicated that they were resistant to more than one antibiotic. The amplicon of ica gene of strong biofilm producing S. aureus strains, obtained by polymerase chain reaction, was sequenced and their close genetic relationship with community acquired MRSA was analyzed based on phylogenetic tree. Our results indicate that they are genetically close to other community acquired MRSA

Im Rahmen der vorliegenden Arbeit wurde eine größere Anzahl an E. faecalis Isolaten aus Vaginalabstrichen, erstmals insbesondere von Patientinnen, die an Bakterieller Vaginose litten, untersucht und mit E. faecalis Stämme aus verschiedenen anderen klinischen Bereichen auf das Vorkommen von Virulenzfaktoren verglichen. Weiterhin wurden Korrelationen zwischen bestimmten Faktoren und der Menge an produziertem Biofilm erstellt, um mögliche Zusammenhänge zum Mechanismus der Biofilm-Bildung zu erfassen. Mittels statistischer Analysen konnte hinsichtlich der 150 untersuchten E. faecalis Isolate nachgewiesen werden, dass keine signifikanten Unterschiede der Inzidenzen von Virulenzfaktoren (esp, asa1, gelE, GelE, cylA, β-Hämolyse) zwischen den Stämmen der verschiedenen Herkunftsbereiche bestanden. In Bezug auf das Auftreten von Biofilm-Bildung zeigte sich ein erhöhtes Vorkommen bei Stämmen aus Urin sowie invasiver Herkunft (insgesamt jeweils ca. 70 % mäßige und starke Biofilm-Bildner) im Vergleich zu E. faecalis Isolaten aus Wunden oder Faeces (je ca. 40 %). Statistische Auswertungen bzgl. des Zusammenhangs eines oder einer Kombination von Virulenzfaktoren mit der Menge an gebildetem Biofilm wiesen darauf hin, dass Isolate, die das esp Gen besaßen, in erhöhtem Maße zur Biofilm-Bildung befähigt waren. Dies zeigte einen gewissen Einfluss des Zellwandproteins auf die Fähigkeit zur Biofilm-Bildung bei E. faecalis. Allerdings wurden stets auch Stämme identifiziert, die die Fähigkeit zur Biofilm-Bildung trotz des Fehlens der jeweils untersuchten genetischen Determinante bzw. der Determinanten aufwiesen, so dass auf das Vorhandensein weiterer, unbekannter Einflussfaktoren auf den Mechanismus der Biofilm-Bildung bei E. faecalis geschlossen werden konnte. Unter 78 untersuchten E. faecium Isolaten aus verschiedenen klinischen Bereichen konnte lediglich ein Stamm (1,3 %) als mäßiger Biofilm-Bildner charakterisiert werden, so dass die Fähigkeit bei dieser Spezies in der hier untersuchten Region unter diesen Bedingungen kaum nachgewiesen werden konnte. Einen Schwerpunkt dieser Arbeit bildete die Untersuchung zum Vorkommen von Virulenzfaktoren und Biofilm-Bildung bei E. faecalis Isolaten aus Vaginalabstrichen. Bzgl. des Auftretens von Virulenzfaktoren und Biofilm-Produktion konnte kein Unterschied zwischen Stämmen assoziiert mit Bakterieller Vaginose und Isolaten einer Vergleichsgruppe festgestellt werden. Allerdings zeigte eine Gegenüberstellung mit den untersuchten E. faecalis Stämmen aus anderen klinischen Bereichen, dass die 80 Isolate aus Vaginalabstrichen eine ähnlich hohe Inzidenz bestimmter Virulenzfaktoren wie Stämme aus Faeces, Urin, Wunden oder invasiver Herkunft sowie eine mit den Isolaten aus Urin und invasiver Herkunft vergleichbar hohe Fähigkeit zur Biofilm-Bildung aufwiesen (ca. 75 % mäßige und starke Biofilm-Bildner). Dies deutete auf eine Verbreitung der Biofilm-Bildungsfähigkeit bei E. faecalis Stämmen der Vaginalflora und somit auf eine große Bedeutung der Eigenschaft für Isolate dieser Herkunft hin. Die statistische Auswertung der Korrelationen von Virulenzfaktoren mit der Menge an gebildetem Biofilm lieferte ähnliche Ergebnisse wie die Analysen bzgl. der 150 E. faecalis Isolate aus anderen klinischen Bereichen und untermauerte die Annahme, dass zusätzliche Faktoren zu den hier untersuchten Determinanten bei E. faecalis vorhanden sein müssen, die Einfluss auf den Mechanismus der Biofilm-Bildung nehmen. Deshalb konzentrierte sich ein weiterer Teil der vorliegenden Arbeit auf die Herstellung und Charakterisierung von E. faecalis Biofilm-Spontanmutanten, um bisher noch ungeklärte Mechanismen oder neue Faktoren zu erkennen, die Einfluss auf die Biofilm-Bildung bei E. faecalis nehmen. Die Untersuchung einer Mutante (1.10.16) und ihres Wildtypstamms lieferte erstmals den phänotypischen Nachweis des HMW-Komplexes der drei Bee-Proteine sowie die Identifizierung konservierter Pili-Motive dieser Proteine. Des Weiteren schien die in diesem Cluster ebenfalls codierte Sortase-1 dasjenige Enzym zu sein, das höchstwahrscheinlich die Bindung des Proteins Bee-2 innerhalb dieses HMW-Komplexes katalysiert. Insofern lieferten diese Untersuchungen neue, konkrete Hinweise zur Rolle des bee Genclusters bei der Pili-Biogenese und Biofilm-Bildung von E. faecalis. Darüber hinaus stellen Erkenntnisse aus der Charakterisierung von zwei weiteren hergestellten Biofilm-Spontanmutanten viel versprechende Ausgangspunkte für zukünftige Untersuchungen dar, die ein weitergehendes Verständnis der molekularen Mechanismen der Biofilm-Bildung bei E. faecalis erzielen könnten.

Xylella fastidiosa is a phytopathogen that causes diseases in different plant species. The development of disease symptoms is associated to the blockage of the xylem vessels caused by biofilm formation. In this study, we evaluated the sensitivity of biofilm and planktonic cells to copper, one of the most important antimicrobial agents used in agriculture. We measured the exopolysaccharides (EPS) content in biofilm and planktonic cells and used real-time reverse transcription polymerase chain reaction to evaluate the expression of the genes encoding proteins involved in cation/multidrug extrusion (acrA/B, mexE/czcA, and metI) and others associated with different copper resistance mechanisms (copB, cutA1, cutA2, and cutC) in the X. fastidiosa biofilm formed in two different media. We confirmed that biofilms are less susceptible to copper than planktonic cells. The amount of EPS seems to be directly related to the resistance and it varies according to the media where the cells are grown. The same was observed for gene expression. Nevertheless, some genes seem to have a greater importance in biofilm cells resistance to copper. Our results suggest a synergistic effect between diffusion barriers and other mechanisms associated with bacterial resistance in this phytopathogen. These mechanisms are important for a bacterium that is constantly under stress conditions in the host.

Background: The ability of biofilm formation seems to play an essential role in the virulence of coagulase-negative staphylococci (CNS). The most clearly characterized component of staphylococcal biofilms is the polysaccharide intercellular adhesin (PIA) encoded by the icaADBC operon. Biofilm production was studied in 80 coagulase-negative staphylococci (CNS) strains isolated from clinical specimens of newborns with infection hospitalized at the Neonatal Unit of the University Hospital, Faculty of Medicine of Botucatu, and in 20 isolates obtained from the nares of healthy individuals without signs of infection. The objective was to compare three phenotypic methods with the detection of the icaA, icaD and icaC genes by PCR. Findings: Among the 100 CNS isolates studied, 82% tested positive by PCR, 82% by the tube test, 81% by the TCP assay, and 73% by the CRA method. Using PCR as a reference, the tube test showed the best correlation with detection of the ica genes, presenting high sensitivity and specificity. Conclusions: The tube adherence test can be indicated for the routine detection of biofilm production in CNS because of its easy application and low cost and because it guarantees reliable results with excellent sensitivity and specificity. © 2010 Cunha et al; licensee BioMed Central Ltd.

The adhesion of Candida albicans to surfaces is the prerequisite for occurrence of denture stomatitis, a common disease diagnosed among denture wearers. A routine of denture cleansing is essential to prevent biofilm formation and the onset of this infection. The aim of this study was to investigate the effectiveness of combining brushing and cleansing agents in killing C. albicans biofilm. Disks of acrylic resin were made, sterilized, and inoculated with C. albicans (107 cfu/mL). After incubation (37°C/48 h), specimens were randomly assigned to 10 experimental groups (n=9): 5 subjected to brushing with distilled water or cleansing agents - dentifrice slurry, 2% chlorhexidine gluconate (CHX), 1% sodium hypochlorite (NaOCl), and Polident fresh cleanse® (combined method) - and 4 exposed to the cleansing agents without brushing (immersion). Non-cleansed specimens were used as positive controls. The viability of cells was evaluated by XTT reduction method. Results were analyzed by Mann-Whitney and Kruskal-Wallis tests (α=0.05). The combined method was significantly more effective (p<0.0001) in reducing biofilm viability than the immersion. Brushing with CHX and NaOCl resulted in 100% removal of the biofilm. Immersion in the agents reduced significantly (p<0.0001) the biofilm viability, with CHX being the most effective (p<0.0001). The use of the combined method of brushing with cleansing agents is an effective method to reduce C. albicans biofilm, being CHX and NaOCl the most effective solutions.

This in vitro study evaluated the effect of photodynamic therapy (PDT) on the multispecies biofilm of Candida albicans, Candida glabrata, and Streptococcus mutans. Standardized fungal and bacterial suspensions were cultivated appropriately for each species and inoculated in 96-well microtiter plates for mix-biofilm formation. After 48 h of incubation, the biofilms were submitted to PDT (P + L+) using Photodithazine® (PDZ) at 100, 150, 175, 200, or 250 mg/mL for 20 min and 37.5 J/cm2 of light-emitting diode (LED) (660 nm). Additional samples were treated only with PDZ (P + L-) or LED (P-L+), or neither (control, P-L-). Afterwards, the biofilms were evaluated by quantification of colonies (CFU/mL), metabolic activity (XTT reduction assay), total biomass (crystal violet staining), and confocal scanning laser microscopy (CSLM). Data were analyzed by one-way ANOVA and Tukey tests (p < 0.05). Compared with the control, PDT promoted a significant reduction in colonies viability of the three species evaluated with 175 and 200 mg/mL of PDZ. PDT also significantly reduced the metabolic activity of the biofilms compared with the control, despite the PDZ concentration. However, no significant difference was found in the total biomass of samples submitted or not to PDT. For all analysis, no significant difference was verified among P-L-, P + L-, and P-L+. CSLM showed a visual increase of dead cells after PDT. PDT-mediated PDZ was effective in reducing the cell viability of multispecies biofilm. © 2013 Springer-Verlag London.

Listeria monocytogenes, considered as one of the most important foodborne pathogens, is easily found on surfaces, particularly in the form of a biofilm. Biofilms are aggregates of cells that facilitate the persistence of these pathogens in food processing environments conferring resistance to the processes of cleaning and may cause contamination of food during processing, thus, representing a danger to public health. Little is known about the dynamics of the formation and regulation of biofilm production in L.monocytogenes, but several authors reported that the luxS gene may be a precursor in this process. In addition, the product of the inlA gene is responsible for facilitating the entry of the microorganism into epithelial cells that express the receptor E-cadherin, also participates in surface attachment. Thus, 32 strains of L.monocytogenes isolated from different foods (milk and vegetables) and from food processing environments were analyzed for the presence of these genes and their ability to form biofilms on three different surfaces often used in the food industry and retail (polystyrene, glass and stainless steel) at different temperatures (4, 20 and 30°C). All strains had the ilnA gene and 25 out of 32 strains (78.1%) were positive for the presence of the luxS gene, but all strains produced biofilm in at least one of the temperatures and materials tested. This suggests that genes in addition to luxS may participate in this process, but were not the decisive factors for biofilm formation. The bacteria adhered better to hydrophilic surfaces (stainless steel and glass) than to hydrophobic ones (polystyrene), since at 20°C for 24h, 30 (93.8%) and 26 (81.3%) produced biofilm in stainless steel and glass, respectively, and just 2 (6.2%) in polystyrene. The incubation time seemed to be an important factor in the process of biofilm formation, mainly at 35°C for 48h, because the results showed a decrease from 30 (93.8%) to 20 (62.5%) and from 27 (84.4%) to 12 (37.5%), on stainless steel and glass, respectively, although this was not significant (. p=0.3847). We conclude that L.monocytogenes is capable of forming biofilm on different surfaces independent of temperature, but the surface composition may be important factor for a faster development of biofilm. © 2013 Elsevier Ltd.
