Tomato plants inoculated with Meloidogyne javanica juveniles infected with Pasteuria penetrans were grown in a glasshouse (20-32degreesC) for 36, 53, 71 and 88 days and in a growth room (26-29degreesC) for 36, 53, 71 and 80 days. Over these periods the numbers of P penetrans endospores in infected M. javanica females and the weights of individual infected females increased. In the growth room, most spores (2.03 x 10(6)) were found after 71 days. However, in the glasshouse the rate of increase was slower and spore numbers were still increasing at the final sampling at 88 days (2.04 x 10(6)), as was the weight of the nematodes (72 mug). Weights of uninfected females reached a maximum of 36.2 and 43.1 mug after 71 days in the growth room and glasshouse, respectively.

The aim of this study was to analyze LEP and DGAT1 gene polymorphisms in 3 Nelore lines selected for growth and to evaluate their effects on growth and carcass traits. Traits analyzed were birth, weaning, and yearling weight, rump height, LM area, backfat thickness, and rump fat thickness obtained by ultrasound. Two SNP in the LEP gene [LEP 1620(A/G) and LEP 305(T/C)] and the K232A mutation in the DGAT1 gene were analyzed. The sample consisted of 357 Nelore heifers from 2 lines selected for yearling weight and a control line, established in 1980, at the Estacao Experimental de Zootecnia de Sertaozinho (Sertaozinho, Brazil). Three genotypes were obtained for each marker. Differences in allele frequencies among the 3 lines were only observed for the DGAT1 K232A polymorphism, with the frequency of the A allele being greater in the control line than in the selected lines. The DGAT1 K232A mutation was associated only with rump height, whereas LEP 1620(A/G) was associated with weaning weight and LEP 305(T/C) with birth weight and backfat thickness. However, more studies, with larger data sets, are necessary before these makers can be used for marker-assisted selection.

Alzheimer`s disease is an ultimately fatal neurodegenerative disease, and BACE-1 has become an attractive validated target for its therapy, with more than a hundred crystal structures deposited in the PDB. In the present study, we present a new methodology that integrates ligand-based methods with structural information derived from the receptor. 128 BACE-1 inhibitors recently disclosed by GlaxoSmithKline R&D were selected specifically because the crystal structures of 9 of these compounds complexed to BACE-1, as well as five closely related analogs, have been made available. A new fragment-guided approach was designed to incorporate this wealth of structural information into a CoMFA study, and the methodology was systematically compared to other popular approaches, such as docking, for generating a molecular alignment. The influence of the partial charges calculation method was also analyzed. Several consistent and predictive models are reported, including one with r (2) = 0.88, q (2) = 0.69 and r (pred) (2) = 0.72. The models obtained with the new methodology performed consistently better than those obtained by other methodologies, particularly in terms of external predictive power. The visual analyses of the contour maps in the context of the enzyme drew attention to a number of possible opportunities for the development of analogs with improved potency. These results suggest that 3D-QSAR studies may benefit from the additional structural information added by the presented methodology.

The inactivation kinetics of enzymes polyphenol oxidase (PPO) and peroxidase (POD) was studied for the batch (discontinuous) microwave treatment of green coconut water. Inactivation of commercial PPO and POD added to sterile coconut water was also investigated. The complete time-temperature profiles of the experimental runs were used for determination of the kinetic parameters D-value and z-value: PPO (D(92.20 degrees C) = 52 s and z = 17.6 degrees C); POD (D(92.92 degrees C) = 16 s and z = 11.5 degrees C); PPO/sterile coconut water: (D(84.45 degrees C) = 43 s and z = 39.5 degrees C) and POD/sterile coconut water: (D(86.54 degrees C) = 20 s and z = 19.3 degrees C). All data were well fitted by a first order kinetic model. The enzymes naturally present in coconut water showed a higher resistance when compared to those added to the sterilized medium or other simulated solutions reported in the literature. The thermal inactivation of PPO and POD during microwave processing of green coconut water was significantly faster in comparison with conventional processes reported in the literature. Os recursos hídricos superficiais e subterrâneos tem-se mostrado contaminados, cada vez mais freqüentemente, por substâncias orgânicas e inorgânicas, em nível de traços, prejudicando os seus usos mais nobres, por exemplo, abastecimento público. Este estudo teve por finalidade principal verificar a eficiência de rejeição por membranas poliméricas, de alguns compostos orgânicos tipo, naftaleno, carbofurano, tricloroetileno (TCE) e metil paration, em baixas concentrações. Empregou-se testes hidrodinâmicos de filtração, tipo "dead-end", com taxa de aplicação média de 13,16 m3.m-2.h-1 através das membranas comerciais de características distintas, uma de ultra (UE-50) e duas de nanofiltração (XN-40 e TS-80). Avaliou-se, também, a preparação de água contaminada para a filtração por membranas, através do prétratamento por oxidação do carbofurano em meio líquido, usando um desinfetante padrão, o hipoclorito de sódio. Após a oxidação desde composto, testes hidrodinâmicos de filtração foram realizados para verificar, também, a eficiência de rejeição dos sub-produtos. Por fim, foram realizados testes de envelhecimento das membranas que poderiam ser atacadas pelos compostos orgânicos em solução aquosa. Todos os ensaios foram realizados à temperatura de 25 oC, empregando pressão de filtração de 4 atm. Os compostos orgânicos foram detectados por várias técnicas, principalmente cromatografia gasosa e líquida, sequenciada por espectrometria de massas. Para acompanhar o envelhecimento das membranas, em seis meses de uso, foi empregada microscopia de força atômica, através da medição de porosidade e de rugosidade superficiais. Em termos médios, nas três faixas de concentrações testadas, o metil paration foi o composto mais eficientemente removido pelas membranas em todos os testes realizados, com 59,82 % de rejeição pela membrana UE-50, 43,97 % pela membrana XN-40 e 56,12 % pela TS-80. O TCE foi rejeitado 28,42 %, 23,87 % e 21,42 % pelas membranas UE-50, XN-40 e TS-80, respectivamente. O naftaleno, foi rejeitado 20,61 %, 14,85 % e 12,63 % pelas membranas UE-50, XN-40 e TS-80, respectivamente. Para o carbofurano, a percentagem de rejeição pelas membranas UE-50, XN-40 e TS-80 foi, respectivamente, de 4,51 %, 4,88 % e 2,92 %. Dentre todas as membranas testadas, a membrana de polisulfona UE-50, de ultrafiltração, produziu a melhor eficiência na rejeição de metil paration, tricloroetileno e naftaleno, com 59,82 %, 28,43 % e 20,61 % de rejeição, na ordem. A membrana de poliamida-uréia TS-80, de nanofiltração, proporcionou maior rejeição (56,12 %) que a membrana de poliamida XN-40, de nanofiltração (43,97 %), para o metil paration. Ao contrário, a membrana XN-40 mostrou uma rejeição maior (23,87 %) que a TS-80 para o tricloroetileno (21,42 %). Para o naftaleno, a membrana XN-40 também mostrou uma rejeição (14,85 %) maior que a TS-80 (12,63 %). A eficiência de rejeição do carbofurano foi a menor de todos os compostos ensaiados, independente da membrana. Se for assumido que a membrana de ultra deverá ser seguida pela de nanofiltração, a eficiência conjunta UE-50 + XN-40 será, em termos médios, para o naftaleno, carbofurano, tricloroetileno e metil paration, respectivamente, 32,24%, 9,19%, 45,60% e 77,44%. Para o conjunto UE-50 + TS-80 será de: 30,64%, 7,29%, 43,92% e 83,51%, não havendo diferenças estatisticamente significantes entre os dois conjuntos. Nos testes de pré-tratamento por oxidação do carbofurano, observou-se a formação do carbofurano hidrolisado e mais dois sub-produtos principais, SP-1 e SP-2, que foram rejeitados, 47,85 % e 92,80 %, respectivamente, pela membrana XN-40. Na verificação das perdas das características morfológicas pelo uso prolongado das membranas, sob ataque de compostos orgânicos em baixa concentração ("envelhecimento") verificou-se que a rugosidade (0, 3 e 6 meses) e a porosidade (0, 3 e 6 meses) foram consideravelmente alteradas. A porosidade da membrana XN-40 aumentou 42,86% e a da membrana TS-80 aumentou 34,57%, quando imersas por 3 meses. A rugosidade das membranas XN-40 e TS-80 nos testes de imersão por 3 meses, aumentou 5,37% e 291% respectivamente. Nos testes de imersão em 6 meses, o aumento da porosidade das membranas XN-40 e TS-80 foi de 29,67% e 18,52% respectivamente, enquanto que a rugosidade aumentou 25,27% e 155% para as mesmas. Conclui-se que membranas de nanofiltração poliméricas necessitam de prétratamentos para rejeitar, com segurança, e colocar dentro dos padrões de potabilidade, águas contaminadas com os compostos orgânicos tipo testados e que seu uso prolongado irá afetar as suas características de rejeição dos contaminates.

Nd ISOTOPES IN THE PROVENANCE OF TERRIGENOUS AND CARBONATE ROCKS AND SEDIMENTS OF THE POTIGUAR BASIN, NORTHEASTERN BRAZIL. Mesozoic and Cenozoic rocks from the Potiguar Basin, including terrigenous and carbonate sediments have been investigated to identify their isotopic signature and source areas. Additionally, this study aims to determine the provenance of terrigenous and carbonate sediments on the Brazilian Continental shelf adjacent to Potiguar Basin. The Sm-Nd isotopic signatures of the rocks yielded model ages (TDM) in the range of 2,19- 2,88 Ga, indicating archean to paleoproterozoic sources from the basement. The terrigenous sediments yielded model ages (TDM) in the range of 2,31-2,26 Ga, from 17,5 to 0 cm depth. Despite the small number of samples, limited variations of provenance ages indicates the homogenization of the sediments, probably due to the strong influence of the basement, as the main source of sediments to the shelf. The Sm-Nd isotopic signatures of the carbonate sediments yielded model ages (TDM) in the range of 2,09-2,61 Ga, indicating archean to paleoproterozoic sources from the basement. The results also indicate that the shelf sediments are mainly derived from the Açu River or other small rivers from the Setentrional Sector of Rio Grande do Norte State. The littoral drift doesn´t seem to contribute with sediments from the Oriental Sector since isotopic signatures from this sector were not detected.

Prototheca zopfii has been considered one of the most important causes of environmental mastitis in Brazil. These algae are refractory to conventional therapy and cause great damage to the mammary gland. The present study evaluated the in vitro algaecide effect of sodium hypochlorite and iodine based antiseptics on 27 P. zopfii strains isolated from the milk of cattle. Low concentrations of sodium hypochlorite (0.0390625-0.15625%) and iodine (0.15625-0.625%) were effective against the isolates. These antiseptics may be recommended for hygiene routines, pre and postdipping and cauterization of bovine mammary glands infected by P. zopfii. (C) 2009 Elsevier Ltd. All rights reserved.

The effects of grain-based diets from C3 or C4-cycle plants on muscle delta C-13 change process in Nile tilapia (Oreochromis niloticus) fingerlings were investigated. Two groups of sex reversal males Nile tilapia fingerlings were fed with isoproteic (32.0% DP) and isocaloric (3200 kcal DE/kg) diets, differing from each other by their delta C-13. Muscle samples were collected and the carbon isotopic composition was measured. For C4 diet, the formula for the muscle delta C-13 change related to the intake time of a new diet was delta C-13=- 14.88 - 9.2 1 e(-0.0209t) and the half-life (T) of the muscle carbon was 33.2 days. For C3 diet, the formula was delta C-13 = - 25.43 + 8.59e(-0.0533), with T = 13 days. The C3 diet was considered more appropriate based on its palatability and consequent larger food intake than the C4 diet, resulting in an increased muscle delta C-13 change rate. However, for future studies, would be necessary to mix both the C3 and C4 feedstuffs to formulate diets nutritionally appropriated, with contrasting stable isotopes signatures. Tissue delta C-13 change rate is therefore indicated as a promising tool to better understand the biotic and abiotic factors that influence nutrients utilization from the diet and animal growth. Mitotic and meiotic chromosomes of several populations of Eurysternus caribaeus (Coleoptera: Scarabaeidae) were analysed through conventional staining, C-banding, base-specific fluorochromes, silver nitrate staining and fluorescent in situ hybridization (FISH). All specimens showed 2n = 8 in their karyotypes, with a neo-XY sex system (Y is a submetacentric and X a metacentric) and three pairs of submetacentric autosomes. The analysis of constitutive heterochromatin (CH) revealed small blocks located in the centromeric region of all chromosomes which do not present positive staining under the fluorochromes CMA3 and DAPI. Silver nitrate staining revealed that the nucleolar organizer region (NORs) is associated with the sex chromosomes. The FISH technique revealed that rDNA sites in the X and Y are different in size. Data from different populations indicate that the diploid number reduction (2n = 8) observed in E. caribaeus is established and presumably has preceded the dispersion of this species. Moreover, this reduction occasioned the translocation of rDNA sites to the sex chromosomes, X and Y, an uncommon pattern in Scarabaeidae that was observed for the first time by the FISH in this work.
