The study of ligand binding specificities of the lipid binding proteins : recombinant human a-tocopherol transport protein (a-ttp), supernatant protein factor (spf) and S. cerevisiae Sec 14p for vitamin e (rrr-a-tocopherol) and other hydrophobic ligands.

One of the various functions of proteins in biological systems is the transport of small molecules, for this purpose proteins have naturally evolved special mechanisms to allow both ligand binding and its subsequent release to a target site; a process fundamental to many biological processes. Transport of Vitamin E (a-tocopherol), a lipid soluble antioxidant, to membranes helps in the protection of polyunsaturated fatty acids against peroxidative damage. In this research, the ligand binding characteristics of several members of the CRALTRIO family of lipid binding proteins was examined; the recombinant human a-Tocopherol Transfer Protein (a-TIP), Supernatant Protein Factor (SPF)ffocopherol Associated Protein (TAP), Cellular Retinaldehyde Binding Protein (CRALBP) and the phosphatidylinositol transfer protein from S. cerevisiae Sec 14p. Recombinant Sec 14p was expressed and purified from E. coli for comparison of tocopherol binding to the two other recombinant proteins postulated to traffic a-tocopherol. Competitive binding assays using [3H]-a-tocopherol and Lipidex-l000 resin allowed determination of the dissociation constants ~) of the CRAL-TRIO proteins for a-tocopherol and - 20 hydrophobic ligands for evaluation of the possible biological relevance of the binding interactions observed. The KIs (nM) for RRR-a-tocopherol are: a-TIP: 25.0, Sec 14p: 373, CRALBP: 528 and SPFffAP: 615. This indicates that all proteins recognize tocopherol but not with the same affinity. Sec 14p bound its native ligand PI with a KI of381 whereas SPFffAP bound PI (216) and y-tocopherol (268) similarly in contrast to the preferential binding ofRRR-a-tocopherol by a-TIP. Efforts to adequately represent biologically active SPFff AP involved investigation of tocopherol binding for several different recombinant proteins derived from different constructs and in the presence of different potential modulators (Ca+2, Mg+2, GTP and GDP); none of these conditions enhanced or inhibited a-tocopherol binding to SPF. This work suggests that only aTTP serves as the physiological mediator of a-tocopherol, yet structural homology between proteins allows common recognition of similar ligand features. In addition, several photo-affmity analogs of a-tocopherol were evaluated for their potential utility in further elucidation of a-TTP function or identification of novel tocopherol binding proteins.

Waltmar Theatre steps, Moulton Hall, Chapman College, Orange, California, ca. 1975. Designed by Leason Pomeroy III & Associates of Orange, using a tilt-up concrete construction method. Completed in 1975, this 44,592 sq.ft. building is named in memory of an artist and patroness of the arts, Nellie Gail Moulton. Within this structure are the departments of Art, Communications, and Theatre/Dance as well as the Guggenheim Gallery and Waltmar Theatre. Waltmar Theatre was a gift from the late Walter and Margaret Schmid. The Guggenheim Gallery is used for the art exhibits presented by the art department and other departments on campus.

Nucleation in supersaturated solutions 3He in 4He at negative pressure.

Depending on the 3He concentration, thermal nucleation in 3-4He supersaturated liquid mixtures at negative pressures may be originated either by bubble or by 3rich drop formation. We have investigated this phenomenon within a density-functional approach, determining the regions in the pressure¿3He-concentration plane where bubbles or drops likely drive the nucleation process. As an illustrative example, we also give the homogeneous nucleation pressure corresponding to 50 and 100 mK temperature.

La vivenda rural asturiana.

Este tutorial dirigido a alumnos de Enseñanza Secundaria pretende analizar la evolución histórica de la vivienda, centrándose en las características de la vivienda rural en Asturias. Los contenidos se estructuran en tres apartados: 1) Vivienda y vivienda rural: La evolución histórica de la vivienda; Los tipos de vivienda; La vivienda rural y el paisaje. 2) Vivienda rural en Asturias: Materiales y construcción; Vivienda campesina y tipo terrena; Vivienda campesina de planta elevada; Vivienda campesina de dos plantas; Otras viviendas; Construcciones complementarias. 3) La vivienda rural hoy: Viejas y nuevas construcciones; Diversidad de usos; Aspectos culturales, paisajísticos y medioambientales. Los objetivos que se pretenden son: Aproximar al reconocimiento de la vivienda humana como lugar que evidencia y determina una serie de aspectos directamente relacionados con las estructuras económicas, sociales y culturales de cada pueblo. Ayudar a conocer la historia de la vivienda humana y su vinculación en cada momento y lugar no sólo con las características geográficas del entorno y los rasgos económicos y socio-culturales de cada pueblo, sino además y de forma inexcusable con los recursos disponibles y los conocimientos técnicos-soluciones correspondientes. Facilitar la comprensión de la vinculación existente entre esos distintos tipos de vivienda y las condiciones vitales de cada pueblo-cultura, en particular con sus formas de vida y las características de su medio natural. Profundizar en particular en las características de las viviendas rurales en los medios geográficos atlántico, mediterráneo y de montaña. Exponer y enseñar a reconocer los diferentes tipos de materiales y técnicas de construcción utilizadas en la vivienda rural tradicional en Asturias. Facilitar la comprensión y correcta utilización del vocabulario vinculado a los elementos constructivos más característicos en la vivienda rural asturiana. Mostrar las tipologías constructivas rurales más significativas en diversos espacios geográficos del Principado, su vinculación específica con distintos modelos económicos y sus elementos más significativos. Mostrar asimismo las construcciones complementarias más habituales en el medio rural asturiano, sus funciones y tipologías, su sentido dentro de las unidades económicas y sociales que caracterizan cada modelo de vivienda y que caracterizaron en particular formas de vida con frecuencia hoy desaparecidas. Mostrar las modificaciones experimentadas por la vivienda rural en Asturias como consecuencia de las profundas transformaciones económicas y sociales, derivando en nuevas funciones de las mismas, como residencia principal o secundaria y aún con frecuencia como vivienda de finalidad turística o con usos relacionados con el sector terciario. Analizar algunas de estas transformaciones desde perspectivas ecológicas y estéticas, enseñando a adoptar ante las nuevas tendencias y soluciones un planteamiento crítico, que no eluda la necesidad de conciliar las exigencias de la modernidad con el respeto a la tradición cultural y de patrimonio que en buena medida representa la vivienda rural.
