Le CD40 ligand (CD40L) est un régulateur important de la réponse immunitaire et un contributeur clé dans les maladies auto-immunes. Nous avons rapporté précédemment que le CD40L se liait à l’intégrine α5β1, toutefois, les conséquences fonctionnelles de cette interaction demeurent inconnues. Les lymphocytes T sont au centre de la pathogénèse des maladies auto-immunes. Ils expriment, lors de celles-ci, des quantités aberrantes d’intégrines β1 faisant en sorte que la liaison CD40L/α5β1 pourrait être d’une haute importance dans les réponses inflammatoires. Dans cette étude, nous avons démontré que la forme soluble du CD40L (sCD40L) se liait aux lymphocytes T primaires ainsi qu’aux cellules Jurkat E6.1 et ce, dépendamment de l’intégrine α5β1. L’interaction du CD40L avec l’α5β1 lymphocytaire a induit l’activation des voies anti-apoptotiques dont les MAPKs (les protéines kinases mitogène activée) et les PI3 kinases (PI3K). La liaison du sCD40L à l’α5β1 n’a pas induit son changement structural ni son adhésion à la FN (fibronectine). Ceci pourrait avoir des conséquences directes sur la survie des cellules T lors de la progression des maladies inflammatoires. Ces résultats soulignent l’impact de l’interaction CD40L/α5β1 sur la fonction biologique des lymphocytes T et ils pourraient expliquer leur survie et leur persistance au niveau des sites d’inflammations durant les maladies auto- immunes.

TGF-beta1 levels increase after vascular injury and promote vascular smooth muscle cell (VSMC) proliferation. We define a nonviral gene delivery system that targets alphavbeta3 and alpha5beta1 integrins that are expressed on proliferating VSMCs and strongly induced by TGF-beta1. A 15-amino acid RGDNP-containing peptide from American Pit Viper venom was linked to a Lys(16) peptide as vector (molossin vector) and complexed with Lipofectamine or fusogenic peptide for delivery of luciferase or beta-galactosidase reporter genes to primary cultures of human, rabbit, and rat VSMCs. Preincubation of VSMCs with TGF-beta1 for 24 h, but not with PDGF-BB, interferon-gamma, TNF-alpha, nor PMA, increased alphavbeta3 and alpha5beta1 expressions on VSMCs and enhanced gene delivery of molossin vector. Thus beta-galactosidase activity increased from 35 +/- 5% (controls) to 75 +/- 5% after TGF-beta1 treatment, and luciferase activity increased fourfold over control values. Potential use of this system in vessel bypass surgery was examined in an ex vivo rat aortic organ culture model after endothelial damage. Molossin vector system delivered beta-galactosidase to VSMCs in the vessel wall that remained for up to 12 days posttransfection. The molossin vector system, when combined with TGF-beta1, enhances gene delivery to proliferating VSMCs and might have clinical applications for certain vasculoproliferative diseases.

Uppsatsen undersöker hur de olika panoreringsteknikerna, direkt/ambient och direkt-överallt, för 5.1 surround uppfattas utav lyssnare. Enligt direkt/ambient ska alla direkta källor panoreras till frontkanalerna (L, C, R) och endast ambiens finnas i surroundkanalerna (Ls, Rs). Enligt direkt-överallt kan direkta källor panoreras till alla kanaler. Ljudstimuli i tre genrer har utav undertecknad framställts, rock/hårdrock, jazz/fusion och hip hop. Ett exempel enligt direkt/ambient och ett enligt direkt-överallt framställdes i varje genre. Vidare har två lyssningsundersökningar ägt rum, en med ”vana” lyssnare i ett kontrollrum samt en med ”vanliga” lyssnare i hemmiljö. Undersökningarna visade att de flesta föredrog direkt-överallt-produktionerna och dessa uppfattades även som mest omslutande vilket har visat sig vara en viktig egenskap för en musikproduktion i surround.

The enzyme 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) catalyzes the reaction between shikimate 3-phosphate and phosphoenolpyruvate to form 5-enolpyruvylshikimate 3-phosphate, an intermediate in the shikimate pathway, which leads to the biosynthesis of aromatic amino acids. EPSPS exists in an open conformation in the absence of substrates and/or inhibitors and in a closed conformation when bound to the substrate and/or inhibitor. In the present report, the H/D exchange properties of EPSPS from Mycobacterium tuberculosis (Mt) were investigated for both enzyme conformations using ESI mass spectrometry and circular dichroism (CD). When the conformational changes identified by H/D exchanges were mapped on the 3-D structure, it was observed that the apoenzyme underwent extensive conformational changes due to glyphosate complexation, characterized by an increase in the content of alpha-helices from 40% to 57%, while the beta-sheet content decreased from 30% to 23%. These results indicate that the enzyme underwent a series of rearrangements of its secondary structure that were accompanied by a large decrease in solvent access to many different regions of the protein. This was attributed to the compaction of 71% of alpha-helices and 57% of beta-sheets as a consequence of glyphosate binding to the enzyme. Apparently, MtEPSPS undergoes a series of inhibitor-induced conformational changes, which seem to have caused synergistic effects in preventing solvent access to the core of molecule, especially in the cleft region. This may be part of the mechanism of inhibition of the enzyme, which is required to prevent the hydration of the substrate binding site and also to induce the cleft closure to avoid entrance of the substrates.
