The study objective was to investigate an acute case of Chagas disease in the San Pedro de Shishita community, Pebas District, in the Peruvian Amazon basin, a non-endemic area. Both parents of the index case (acute case) were thoroughly interviewed, a seroepidemiological survey was carried out in the community, parasitological exams were carried out only in relatives of the index case, and triatomine bugs were searched for inside houses, peridomiciliary, and in wild environments. Seroprevalence for IgG anti-T. cruzi antibodies was 1/104 (0.96%), using an ELISA test and an indirect immunofluorescence assay. Panstrongylus geniculatus and Rhodnius pictipes adults were found. The index case is autochthonous from San Pedro de Shishita, but the source of transmission is unknown.

Tyrosine hydroxylase (TH) deficiency is an inborn error of dopamine biosynthesis and a cause of early parkinsonism. Two clinical phenotypes have been described. Type “B”: early onset severe encephalopathy; type “A”: later onset, less severe and better response to L-dopa. We aimed to study the expression of several key dopaminergic and gabaergic synaptic proteins in the cerebrospinal fluid (CSF) of a series of patients with TH deficiency and their possible relation with the clinical phenotype and response to L-DOPA. Dopamine transporter (DAT), D2-receptor and vesicularmonoamine transporter (VMAT2)weremeasured in the CSF of 10 subjectswith THdeficiency byWestern blot analysis. In 3 patients, data of pre- and post-treatmentwith L-DOPA were available, and in one of them, GABA vesicular transporter was determined. Results were compared to an age-matched control population. The concentration of D2-receptors in CSFwas significantly higher in patients with TH deficiency than in controls. Similarly, DAT and vesicular monoamine transporter type 2 were up-regulated. Studies performed before LDOPA, and on L-DOPA therapy showed a paradoxical response with D2 receptor expression increase as L-Dopa doses and homovanillic concentration gradually raised in a B phenotype patient. The opposite results were found in two patients with A phenotype. However, this is a very small sample, and further studies are needed to conclude robust differences between phenotypes. Synaptic proteins are detectable in the CSF and their quantification can be useful for understanding the pathophysiology of neurotransmitter defects and potentially to adjust and personalize treatments in the future.

INTRODUÇÃO: É frequente a associação da malária com complicações como prematuridade, retardo no crescimento intrauterino, baixo peso ao nascer e mortalidade infantil, efeitos pouco estudados em áreas hipoendêmicas para malaria. O objetivo deste estudo foi analisar a relação da malária gestacional com estes efeitos em recém-nascidosnuma região endêmica para malária na Colômbia, entre 1993 e 2007. MÉTODOS: Foram estudadas as características em 1.716 recém-nascidos num estudo de coorte. Fez-se seguimento em 394 gestantes com malária (27% por Plasmodium falciparum e 73% por P. vivax) e 1.322 sem malária. RESULTADOS: Foi encontrada uma relação entre a exposição à malária na gestação e o risco maior de baixo peso ao nascer (RR = 1,37; 1,03-1,83), assim como estatura baixa (RR = 1,52; 1,25-1,85), retardo no crescimento intrauterino (RR = 1,29; 1,0-1,66) e prematuridade (RR = 1,68; 1,3-2,17). A frequência de nascimentos prematuros foi maior nas mães com malária por P. falciparum (77%) que aquelas com P. vivax (RR = 1,77; IC 95%: 1,2-2,6). CONCLUSÕES: O baixo peso ao nascer e o retardo no crescimento foi associado com malária na gestação na Colômbia. A infecção por P. vivax foi relacionada com efeitos adversos sobre o recém-nascido, de modo semelhante em relação ao P. falciparum.

Maturity models are adopted to minimise our complexity perception over a truly complex phenomenon. In this sense, maturity models are tools that enable the assessment of the most relevant variables that impact on the outputs of a specific system. Ideally a maturity model should provide information concerning the qualitative and quantitative relationships between variables and how they affect the latent variable, that is, the maturity level. Management systems (MSs) are implemented worldwide and by an increasing number of companies. Integrated management systems (IMSs) consider the implementation of one or several MSs usually coexisting with the quality management subsystem (QMS). It is intended in this chapter to report a model based on two components that enables the assessment of the IMS maturity, considering the key process agents (KPAs) identified through a systematic literature review and the results collected from two surveys.

The receiver-operating characteristic (ROC) curve is the most widely used measure for evaluating the performance of a diagnostic biomarker when predicting a binary disease outcome. The ROC curve displays the true positive rate (or sensitivity) and the false positive rate (or 1-specificity) for different cut-off values used to classify an individual as healthy or diseased. In time-to-event studies, however, the disease status (e.g. death or alive) of an individual is not a fixed characteristic, and it varies along the study. In such cases, when evaluating the performance of the biomarker, several issues should be taken into account: first, the time-dependent nature of the disease status; and second, the presence of incomplete data (e.g. censored data typically present in survival studies). Accordingly, to assess the discrimination power of continuous biomarkers for time-dependent disease outcomes, time-dependent extensions of true positive rate, false positive rate, and ROC curve have been recently proposed. In this work, we present new nonparametric estimators of the cumulative/dynamic time-dependent ROC curve that allow accounting for the possible modifying effect of current or past covariate measures on the discriminatory power of the biomarker. The proposed estimators can accommodate right-censored data, as well as covariate-dependent censoring. The behavior of the estimators proposed in this study will be explored through simulations and illustrated using data from a cohort of patients who suffered from acute coronary syndrome.

The Great Lakes lie within a region of East Africa with very high human genetic diversity, home of many ethno-linguistic groups usually assumed to be the product of a small number of major dispersals. However, our knowledge of these dispersals relies primarily on the inferences of historical, linguistics and oral traditions, with attempts to match up the archaeological evidence where possible. This is an obvious area to which archaeogenetics can contribute, yet Uganda, at the heart of these developments, has not been studied for mitochondrial DNA (mtDNA) variation. Here, we compare mtDNA lineages at this putative genetic crossroads across 409 representatives of the major language groups: Bantu speakers and Eastern and Western Nilotic speakers. We show that Uganda harbours one of the highest mtDNA diversities within and between linguistic groups, with the various groups significantly differentiated from each other. Despite an inferred linguistic origin in South Sudan, the data from the two Nilotic-speaking groups point to a much more complex history, involving not only possible dispersals from Sudan and the Horn but also large-scale assimilation of autochthonous lineages within East Africa and even Uganda itself. The Eastern Nilotic group also carries signals characteristic of West-Central Africa, primarily due to Bantu influence, whereas a much stronger signal in the Western Nilotic group suggests direct West-Central African ancestry. Bantu speakers share lineages with both Nilotic groups, and also harbour East African lineages not found in Western Nilotic speakers, likely due to assimilating indigenous populations since arriving in the region ~3000 years ago.

En el actual marco de creciente innovación pedagógica, debido entre otros factores, a la irrupción de nuevas herramientas informáticas, la enseñanza a distancia (e-learning y/o b-learning) va ocupando cada vez más espacio en la oferta educativa de diversas instituciones. En esta dirección, en la Universidade do Minho, y concretamente en el Área de Estudos Espanhóis e Hispano-Americanos,[1] hemos dedicado considerables esfuerzos a la ampliación de nuestra oferta desde 2010: primero en la elaboración e implementación del Curso de Formación Especializada en Español Lengua Extranjera, modalidad b-learning (3 ediciones; 2010-2013), y, actualmente, con el Máster Universitario en Español Lengua Segunda / Lengua Extranjera (vid. www.melsle.ilch.uminho.pt), también b-learning. En las siguientes páginas, nos proponemos compartir una serie de experiencias y reflexiones que han ido surgiendo durante estos años acerca de la formación universitária de profesores de Español Lengua Extranjera, en general, con recurso a la modalidade b-learning; para ello, nos centraremos en los siguientes aspectos: (i) caracterización general y problematización de la enseñanza a distancia en la Universidade do Minho; (ii) descripción del Máster Universitario en Español Lengua Segunda / Lengua Extranjera, acerca del cual detallaremos algunas prácticas adoptadas, relacionadas com la enseñanza e-learning como, por ejemplo, (iii) la coordinación pedagógica o (iv) los enfoques metodológicos adoptados a partir de la experiencia de una Unidad Curricular concreta.

OBJETIVO: Existe uma prevalência estimada de 11,2% de alcoolismo no Brasil. As tentativas de muitos alcoolistas para abandonar o consumo resultam fracassos e recaídas. A freqüência de recaída, segundo estudos, oscila entre 10% e 30%, assinalando-se diferentes causas. O objetivo da presente pesquisa é conhecer os fatores que favorecem as recaídas. MÉTODOS: Aplicaram-se entrevistas e questionários "Fatores de Recaída" e "Razões para Beber" a 105 alcoolistas primários de ambos os sexos, e com predomínio de alcoolismo puro. RESULTADOS: Os principais fatores de recaída são a pressão social, os conflitos interpessoais, os estados emocionais negativos, assim como a própria dependência fisiológica e psicológica. CONCLUSÕES: De maneira geral, os sujeitos apresentaram grande quantidade de fatores e razões que atuam simultaneamente favorecendo a recaída.

We propose a novel hanging spherical drop system for anchoring arrays of droplets of cell suspension based on the use of biomimetic superhydrophobic flat substrates, with controlled positional adhesion and minimum contact with a solid substrate. By facing down the platform, it was possible to generate independent spheroid bodies in a high throughput manner, in order to mimic in vivo tumour models on the lab-on-chip scale. To validate this system for drug screening purposes, the toxicity of the anti-cancer drug doxorubicin in cell spheroids was tested and compared to cells in 2D culture. The advantages presented by this platform, such as feasibility of the system and the ability to control the size uniformity of the spheroid, emphasize its potential to be used as a new low cost toolbox for high-throughput drug screening and in cell or tissue engineering.
Superhydrophobic surfaces as a tool for the fabrication of hierarchical spherical polymeric carriers

Hierarchical polymeric carriers with high encapsulation efficiencies are fabricated via a biocompatible strategy developed using superhydrophobic (SH) surfaces. The carries are obtained by the incorporation of cell/BSA-loaded dextran-methacrylate (DEXT-MA) microparticles into alginate (ALG) macroscopic beads. Engineered devices like these are expected to boost the development of innovative and customizable systems for biomedical and biotechnological purposes.

Nuestro proyecto de investigación se propone describir y analizar los factores que influyen en la ausencia de estrategias empresariales y gubernamentales que propicien el desarrollo de iniciativas conjuntas (interactorales de carácter públicoprivadas) en torno al desarrollo de la Responsabilidad Social Empresaria (RSE) en Córdoba capital. A estos fines, a través del contacto con referentes claves de los sectores público-gubernamental y privado-empresarial y representantes de los principales órganos institucionalizados de intermediación entre ambos sectores, se intentará analizar una problemática que, según aproximaciones anteriores realizadas en el marco de PROETICA/ICDA-UCC, estaría ligada a las lógicas de actuación predominantes en los sectores mencionados que impedirían una adecuada vinculación intersectorial. Así, puede decirse que el predominio de patrones individuales de comportamiento que se traducen en el planteo de estrategias diferenciadas no relacionadas, los prejuicios mutuos que impiden el diálogo y el acercamiento y el desconocimiento del tema por parte del Gobierno, conllevan a una ausencia de estrategias orientadas a la promoción y el desarrollo de la RSE. Asimismo, el problema estaría vinculado con una acentuada crisis de representatividad general que afecta también a los actores que operan en la intermediación gobierno- empresas. El equipo considera que la indagación acerca de los factores que, según los actores implicados, influyen en la relación in-tersectorial, es el puntapié incial de cara a resolver las cuestiones que impiden el fortalecimiento de vínculos en pos del desarrollo de la RSE y la consecución de los fines que esta promueve.
