A menopausa e a hipertensão podem alterar o remodelamento cardiovascular, porém pouco se sabe sobre sua associação no remodelamento ventricular esquerdo e na aorta. As ratas foram separadas em quatro grupos com seis animais cada: grupo Sham, OVX (ratas ooforectomizadas), 2K1C (ratas com dois rins, um clipe), e grupo 2K1C+OVX com período experimental de 11 semanas. O ventrículo esquerdo (VE) e a aorta torácica foram removidos e analisados (microscopia de luz, imuno-histoquímica e estereologia). A citologia vaginal mostrou que os animais dos grupos Sham e 2K1C ciclaram normalmente, entretanto, os animais dos grupos OVX e OVK+2K1C permaneceram na fase do diestro ou proestro. Comparado ao grupo Sham, a pressão arterial aumentou 12% no grupo OVX e 35% maior nos grupos 2K1C e OVX+2K1C. A relação massa do VE/comprimento da tíbia e a área seccional média de cardiomiócitos aumentaram em todos os grupos com exceção do grupo Sham. A vascularização intramiocárdica foi reduzida cerca de 30% em relação ao grupo Sham, não havendo diferença significativa entre os grupos OVX, 2K1C e OVX+2K1C. O tecido conjuntivo cardíaco teve um aumento superior a 45% nos grupos 2K1C e OVX+2K1C comparados ao grupo Sham, sem diferença entre o os animais do grupo Sham e OVX. O número de núcleos de cardiomiócitos do VE foi gradualmente menor nos grupos OVX, 2K1C e OVX+2K1C, sem diferença entre os dois últimos grupos. Imuno-histoquímica positiva para receptor AT1 da Ang II nas células musculares lisas da túnica média da aorta foi observado em todos os grupos. Estes resultados indicam que a ooforectomia e a hipertensão renovascular agem aumentando a pressão arterial independentemente, com conseqüente remodelamento cardíaco adverso, com estímulo maior da hipertensão renovascular que da menopausa induzida cirurgicamente.

Esteróides Anabolizantes Androgênicos (EAA) têm sido usados por atletas com o objetivo de melhorar a massa muscular. O abuso de EAA está associado com distúrbios urogenitais, inclusive com disfunção erétil. Contudo as alterações morfológicas penianas decorrentes do uso de EAA não foram descritas. O objetivo é avaliar as alterações morfológicas do pênis de ratos púberes e adultos tratados cronicamente com doses supra-fisiológicas de EAA. No trabalho foram usados quarenta e oito ratos machos Wistar, divididos em quatro grupos: ratos controle, com 105 dias (C105) e com 65 dias de idade (C65) submetidos a injeção de veículo e ratos tratados, com 105 dias (T105) e com 65 dias de idade (T65), submetidos à injeção de decanoato de nandrolona na dose de 10 mg/Kg, uma vez por semana, durante oito semanas. Os ratos foram mortos, seus pênis foram coletados, fixados e processados de maneira rotineira para histologia. Cortes de 5m de espessura foram corados com Tricrômico de Masson, Vermelho de Picrosirius e analisados em microscopia de luz. As densidades de superfície do espaço sinusoidal, músculo liso e tecido conjuntivo do corpo cavernoso foram calculadas pelo método de contagem de pontos. A área do pênis, do corpo cavernoso (com e sem túnica albugínea) e da túnica albugínea foram medidos em cortes transversais. As médias dos grupos foram comparadas pelo teste t de Student. Em todos os casos, a significância foi fixada em um valor de probabilidade de 0,05. Nos resultados entre outras diferenças, destaca-se uma diminuição do corpo cavernoso sem túnica albugínea de 12,5% no grupo T105 e de 10,9% no grupo T65, em comparação com os seus controles. A densidade de superfície de músculo liso cavernoso apresentou uma diminuição de 5,6% e 12,9% nos grupos T65 e T105, comparando-se com os seus controles. O espaço sinusoidal aumentou em 17% no grupo T105 e diminuiu em 9,6% no grupo T65. Concluimos que o uso de altas doses de EAA promoveu mudanças estruturais no pênis dos ratos adultos, e estas podem estar envolvidas na disfunção erétil.

The ontogeny of IgM-producing cells was studied in juvenile mandarin fish Simperca chuatsi, an important fish in China's aquaculture sector. The IgM-producing cells were localised through in situ hybridisation with a probe complementary to the Ig mu-chain in lymphoid-related tissues, including head kidney, spleen, thymus, intestine and gills. In head kidney, transcripts of Ig mu were first detected at 20 days post-hatching (dph) with a few positive signals. and the number of IgM-producing cells increased obviously from 39 dph onwards. At 136 dph, a large amount of positive cells were observed in the entire organ with clusters of these cells located around the blood vessels. In spleen, IgM-producing cells were found from 26 dph onwards, followed by an increase until 67 dph: clusters of positive cells were also detected around blood vessels at 102 dph. In thymus, IgM-producing cells were first observed at 39 dph; thereafter, no obvious increase was detected until 78 dph. The positive cells in thymus were distributed mainly in the outer zone of thymus. A few IgM-producing cells were still observed in thymus of 1-year-old mandarin fish. IgM-producing cells were not detected in the intestine until 87 dph, with several discrete positively stained cells distributed in the lamina propria. IgM-producing cells, scattered mainly in primary gill filaments around blood vessels, were detected in gills from 90 dph. As in other teleosts, these results indicated that the head kidney appears to be the primary organ for IgM production in mandarin fish, and IgM-producing cells exist in all organs examined in the present study, implying their lymphoid role in fish. The substitution of dietary docosahexaenoic acid (DHA) with eicosapentaenoic acid (EPA) reduces larval growth in gilthead sea bream. However, the value of EPA when dietary DHA is able to meet the requirements of the larvae has not been sufficiently studied. Dietary phosphoacylgliceride levels also affect fish growth and it has been suggested that they enhance lipid transport in developing larvae. The present experiment was carried out to further study the effect of dietary lecithin and eicosapentaenoic acid on growth, survival, stress resistance,. larval fatty acid composition and lipid transport, when DHA is present in the microdiets of gilthead:sea bream. Eighteen thousand gilt-head sea bream larvae of 4.99+/-0.53 mm total length were fed three microdiets tested by triplicate: a control diet [2% soybean lecithin (SBL) and 2.89% EPA], a low EPA diet,(2% SBL and 1.63% EPA) and a no SBL diet (0% SBL and 2.71% EPA). Handling, temperature and salinity tests determined larval resistance to stress. The results show that when dietary DHA levels are high, but dietary arachidonic acid (ARA) levels are about 0.2%, EPA is necessary to improve larval growth, and survival. Larval EPA content, but not DHA or ARA, was affected by dietary EPA levels. Increased dietary EPA improved larval stress resistance to handling and temperature tests, which could be related to its possible role as a regulator of cortisol production whereas it did not affect stress resistance after salinity shock. Larvae fed the no SBL diet showed a lower lipid content characterized by a low proportion of saturated and monounsaturated fatty acids, together with a significant reduction in the appearance of lipoprotein particles in the lamina propria and in the size of such particles, denoting a critical reduction in dietary lipid transport and utilization, and lower larval growth and survival rates.

Estudo realizado sobre a cadeia produtiva de mandioca em alguns município do Amazonas. A metodologia utilizada envolveu levantamento bibliográfico, aplicação de questionários e consultas pessoais a diversos seguimentos do ambiente organizacional e institucional e da própria cadeia produtiva. Com base no estudo, foi constatado que: a) o mandiocultor pratica uma agricultura itinerante, de baixo nivel tecnologico, e utilizacao da mesma area por, no maximo, dois anos; b) a farinha produzida no Estado nao e' suficiente para atendimento do mercado interno, sendo necessaria sua importacao (31%) de outros Estados; c) o principal produto da mandioca e' a raiz, tanto para a producao de farinha (mandioca brava ou mansa) como para o consumo direto (mandioca mansa ou macaxeira); d) o tucupi, a goma (fecula) e a farinha de mandioca sao os subprodutos mais utilizados; e) os canais de comercializacao dos produtos e subprodutos estao a mercer de sete intermediacoes, o que provoca desvios dos recursos que deveriam ser destinados ao mandiocultor; f) limitacoes estruturais e tecnologicas quanto a agroindustria (casa de farinha), estradas, eletrificacao etc.; g) carencia de credito rural, assitencia tecnica e pesquisa; e h) nao adocao das novas tecnologias disponiveis. Existe, pois, a necessidade de incrementacao de uma politica agricola regional para que o mandiocultor possa aumentar a producao, a produtividade, a qualidade do produto e subprodutos e da propria renda, o que resultaria, inevitavelmente, na fixacao do homem no campo

Por ser especie heliofila, a seringueira e' plantada em pleno sol. Por esse motivo, foi testado o comportamento de plantio, em trilhas abertas na capoeira, de toco alto que, por medir de 2,10m a 2,40m de altura e possuir maior quantidade de reservas do que as mudas tradicionais, traria as vantagens de reducao do custo de preparo da area, evitando a derruba e queima, com preservacao da materia organica do solo, alem da incorporacao gradual da biomassa da capoeira. As copas de Hevea pauciflora apresentam derrama natural tardia, sugerindo que seu ponto de compensacao luminosa e' mais baixo do que o de H. brasiliensis. Desse modo, copas enxertadas com H. pauciflora poderiam tolerar melhor a fase de crescimento a sombra parcial da capoeira. O tratamento com copa propria apresentou o menor perimetro de caule, com valores tambem baixos nos blocos em pleno sol. Os incrementos anuais em perimetro de caule, apos a decapitacao dos enxerto de copa, permaneceram muito baixos na capoeira, nos tres anos consecutivos, e atingiram valores sucessivamente mais altos em pleno sol, exceto com a copa propria. Os resultados com as copas enxertadas de H. pauciflora demostram, portanto, ser inviavel o plantio em capoeira, nas condicoes descritas neste trabalho.

Inflammatory bowel diseases (IBD), encompasses a range of chronic, immune-mediated inflammatory disorders that are usually classified under two major relapsing conditions, Crohn’s Disease (CD) and ulcerative colitis (UC). Extensive studies in the last decades have suggested that the etiology of IBD involves environmental and genetic factors that lead to dysfunction of epithelial barrier with consequent deregulation of the mucosal immune system and inadequate responses to gut microbiota.Over the last decade, the microbial species that has attracted the most attention, with respect to CD etiology, is Eschericia coli. In CD tissue, E. coli antigens have also been identified in macrophages within the lamina propria, granulomas, and in the germinal centres of mesenteric lymph nodes of patients. They have been shown to adhere to and invade intestinal epithelial cells whilst also being able to extensively replicate within macrophages. Through the work of genome-wide association studies (GWAS), there is growing evidence to suggest that the microbial imbalance between commensal and pathogenic bacteria in the gut is aided by a defect in the innate immune system. Autophagy represents a recently investigated pathway that is believed to contribute to the pathogenesis of CD, with studies identified a variant of the autophagy gene, ATG16L1, as a susceptibility gene. The aim of my thesis was to study the cellular and molecular mechanism promoted by E.coli strains in epithelial cells and to assess their contribution to IBD pathology. To achieve this we focused on developing both an in vitro and in vivo model of AIEC infection. This allowed us to further our knowledge on possible mechanisms utilised by AIEC that promoted their survival, as well as developing a better understanding of host reactions. We demonstrate a new survival mechanism promoted by E.coli HM605, whereby it induces the expression of the anti-apoptotic proteins Bcl-XL and BCL2, all of which is exacerbated in an autophagy deficient system. We have also demonstrated the presence of AIEC-induced inflammasome responses in epithelial cells which are exacerbated in an autophagy deficient system and expression of NOD-like receptors (NLRs) which might mediate inflammasome responses in vivo. Finally, we used the Citrobacter rodentium model of infectious colitis to identify Pellino3 as an important mediator in the NOD2 pathway and regulator of intestinal inflammation. In summary, we have developed robust and versatile models of AIEC infection as well as provide new insights into AIEC mediated survival pathways. The collected data provides a new perception into why AIEC bacteria are able to prosper in conditions associated with Crohn’s disease patients with a defect in autophagy.

BACKGROUND: Interleukin-10 (IL-10) is currently being extensively studied in clinical trials for the treatment of Crohn's disease (CD). Only marginal effects have, however, been reported, and the dose-response curve was bell-shaped contrasting with the reported data from in vitro experiments. AIM: To use another in vitro model to analyze the effect of rhIL-10 and rhIL-4 on the spontaneous mucosal TNF-alpha secretion in patients with CD, and to characterize the phenotype of the cells targeted by rhIL-10. METHODS: Non-inflamed colon biopsies from CD patients were cultured for 16 hours in presence of different concentrations of rhIL-10 or rhIL-4. The numbers of TNF-alpha-secreting cells among isolated lamina propria mononuclear cells (LPMNC) were estimated by Elispot. RESULTS: Both rhIL-10 and rhIL-4 down-regulate TNF-alpha secretion by LPMNC from CD patients, with a more pronounced effect with rhIL-10. These effects were closely linked to the cytokine concentrations used, with a bell-shaped dose-response curve. Residual TNF-alpha secretion, in the presence of optimal rhIL-10 concentration was mainly attributable to CD3+ T cells. In contrast, at higher rhIL-10 concentrations, CD3- cells contributed significantly to the TNF-alpha secretion. CONCLUSIONS: The in vitro model we used, demonstrates that IL-4, but mostly IL-10, efficiently suppresses TNF-alpha secretion in LPMNC from CD patients, with a dose-response curve similar to results obtained in vivo. Resistance at high rhIL-10 concentrations was associated with a change in the phenotype of TNF-alpha-secreting cells.

BACKGROUND/AIMS: The intestinal immune system faces large amounts of antigens, and its regulation is tightly balanced by cytokines. In this study, the effect of intestinal flow diversion on spontaneous secretion of interleukin (IL)-4 and interferon (IFN)- gamma was analysed. METHODS: Eight patients (two with Crohn's disease, four with ulcerative colitis, and two with previous colon cancer) carrying a double lumen small bowel stoma after a total colectomy procedure were included in the study. For each patient, eight biopsy samples were taken endoscopically from both the diverted and non-diverted part of the small bowel. Intraepithelial lymphocytes (IELs) and lamina propria lymphocytes (LPLs) were isolated separately and assayed for numbers of cells spontaneously secreting IL-4 and/or IFN-gamma by an ELISPOT technique. RESULTS: Compared with the non-diverted mucosa, a significant decrease in the number of spontaneously IFN-gamma secreting CD3 lymphocytes was observed in the diverted small bowel mucosa among both IELs (p = 0.008) and LPLs (p = 0.007). The same results, although less significant, were obtained for IL-4, especially in LPLs (p = 0.01). CONCLUSION: The intestinal content influences the spontaneous secretion of IFN-gamma and IL-4 by intestinal lymphocytes. These results could help to elucidate the anti-inflammatory role of split ileostomy in patients suffering from inflammatory bowel diseases.

Vein grafting results in the development of intimal hyperplasia with accompanying changes in guanine nucleotide-binding (G) protein expression and function. Several serum mitogens that act through G protein-coupled receptors, such as lysophosphatidic acid, stimulate proliferative pathways that are dependent on the G protein betagamma subunit (Gbetagamma)-mediated activation of p21ras. This study examines the role of Gbetagamma signaling in intimal hyperplasia by targeting a gene encoding a specific Gbetagamma inhibitor in an experimental rabbit vein graft model. This inhibitor, the carboxyl terminus of the beta-adrenergic receptor kinase (betaARK(CT)), contains a Gbetagamma-binding domain. Vein graft intimal hyperplasia was significantly reduced by 37% (P<0.01), and physiological studies demonstrated that the normal alterations in G protein coupling phenotypically seen in this model were blocked by betaARK(CT) treatment. Thus, it appears that Gbetagamma-mediated pathways play a major role in intimal hyperplasia and that targeting inhibitors of Gbetagamma signaling offers novel intraoperative therapeutic modalities to inhibit the development of vein graft intimal hyperplasia and subsequent vein graft failure.

The peptide tyrosine tyrosine (PYY) is produced and secreted from L cells of the gastrointestinal mucosa. To study the anatomy and function of PYY-secreting L cells, we developed a transgenic PYY-green fluorescent protein mouse model. PYY-containing cells exhibited green fluorescence under UV light and were immunoreactive to antibodies against PYY and GLP-1 (glucagon-like peptide-1, an incretin hormone also secreted by L cells). PYY-GFP cells from 15 μm thick sections were imaged using confocal laser scanning microscopy and three-dimensionally (3D) reconstructed. Results revealed unique details of the anatomical differences between ileal and colonic PYY-GFP cells. In ileal villi, the apical portion of PYY cells makes minimal contact with the lumen of the gut. Long pseudopod-like basal processes extend from these cells and form an interface between the mucosal epithelium and the lamina propria. Some basal processes are up to 50 μm in length. Multiple processes can be seen protruding from one cell and these often have a terminus resembling a synapse that appears to interact with neighboring cells. In colonic crypts, PYY-GFP cells adopt a spindle-like shape and weave in between epithelial cells, while maintaining contact with the lumen and lamina propria. In both tissues, cytoplasmic granules containing the hormones PYY and GLP-1 are confined to the base of the cell, often filling the basal process. The anatomical arrangement of these structures suggests a dual function as a dock for receptors to survey absorbed nutrients and as a launching platform for hormone secretion in a paracrine fashion.

Late outgrowth endothelial progenitor cells (EPCs) derived from the peripheral blood of patients with significant coronary artery disease were sodded into the lumens of small diameter expanded polytetrafluoroethylene (ePTFE) vascular grafts. Grafts (1mm inner diameter) were denucleated and sodded either with native EPCs or with EPCs transfected with an adenoviral vector containing the gene for human thrombomodulin (EPC+AdTM). EPC+AdTM was shown to increase the in vitro rate of graft activated protein C (APC) production 4-fold over grafts sodded with untransfected EPCs (p<0.05). Unsodded control and EPC-sodded and EPC+AdTM-sodded grafts were implanted bilaterally into the femoral arteries of athymic rats for 7 or 28 days. Unsodded control grafts, both with and without denucleation treatment, each exhibited 7 day patency rates of 25%. Unsodded grafts showed extensive thrombosis and were not tested for patency over 28 days. In contrast, grafts sodded with untransfected EPCs or EPC+AdTM both had 7 day patency rates of 88-89% and 28 day patency rates of 75-88%. Intimal hyperplasia was observed near both the proximal and distal anastomoses in all sodded graft conditions but did not appear to be the primary occlusive failure event. This in vivo study suggests autologous EPCs derived from the peripheral blood of patients with coronary artery disease may improve the performance of synthetic vascular grafts, although no differences were observed between untransfected EPCs and TM transfected EPCs.

The vertebrate brain actively regulates incoming sensory information, effectively filtering input and focusing attention toward environmental stimuli that are most relevant to the animal's behavioral context or physiological state. Such centrifugal modulation has been shown to play an important role in processing in the retina and cochlea, but has received relatively little attention in olfaction. The terminal nerve, a cranial nerve that extends underneath the lamina propria surrounding the olfactory epithelium, displays anatomical and neurochemical characteristics that suggest that it modulates activity in the olfactory epithelium. Using immunocytochemical techniques, we demonstrate that neuropeptide Y (NPY) is abundantly present in the terminal nerve in the axolotl (Ambystoma mexicanum), an aquatic salamander. Because NPY plays an important role in regulating appetite and hunger in many vertebrates, we investigated the possibility that NPY modulates activity in the olfactory epithelium in relation to the animal's hunger level. We therefore characterized the full-length NPY gene from axolotls to enable synthesis of authentic axolotl NPY for use in electrophysiological experiments. We find that axolotl NPY modulates olfactory epithelial responses evoked by L-glutamic acid, a food-related odorant, but only in hungry animals. Similarly, whole-cell patch-clamp recordings demonstrate that bath application of axolotl NPY enhances the magnitude of a tetrodotoxin-sensitive inward current, but only in hungry animals. These results suggest that expression or activity of NPY receptors in the olfactory epithelium may change with hunger level, and that terminal nerve-derived peptides modulate activity in the olfactory epithelium in response to an animal's changing behavioral and physiological circumstances.

Scientists interested in the smooth muscles of the urinary tract, and their control, have recently been studying cells in the interstitium of tissues that express the c-kit antigen (Kit(+) cells). These cells have morphologic features that are reminiscent of the well-described pacemaker cells in the gut, the interstitial cells of Cajal (ICC). The spontaneous contractile behavior of muscles in the urinary tract varies widely, and it is clear that urinary tract Kit(+) interstitial cells cannot be playing an identical role to that played by the ICC in the gut. Nevertheless, there is increasing evidence that they do play a role in modulating the contractile behavior of adjacent smooth muscle, and might also be involved in mediating neural control. This review outlines the properties of ICC in the gut, and gives an account of the discovery of cells in the interstitium of the main components of the urinary tract. The physiologic properties of such cells and the functional implications of their presence are discussed, with particular reference to the bladder. In this organ, Kit(+) cells are found under the lamina propria, where they might interact with the urothelium and with sensory nerves, and also between and within the smooth-muscle bundles. Confocal microscopy and calcium imaging are being used to assess the physiology of ICC and their interactions with smooth muscles. Differences in the numbers of ICC are seen in smooth muscle specimens obtained from patients with various pathologies; in particular, bladder overactivity is associated with increased numbers of these cells.
