The aim of this study was to adapt the methodology of electrical conductivity (EC) test and determine the physiological quality of seeds of B. virgilioides. Seeds were separated into three batches according to tegument color (1-yellow, 2-orange, and 3-red/black). The EC test analyzed three quantities of seeds (25, 50 and 100), three volumes of water (25, 50 and 100 mL), and six times of seed imbibition (12, 24, 36, 48, 60 and 72), at 25 °C with 6 replications. Germination test was employed to compare the results. The EC test allowed the differentiation of batches in three levels of vigor - the same was observed in germination test: batch 1 presented the best seed vigor, followed by batches 2 and 3. The use of 25, 50 or 100 seeds imbibed in 50 mL of water for 24 hours is recommended.

Due to the expansion of the wild crop to regions with higher temperatures is important to develop cultivars adapted to high heat. The aim was to select tests for evaluating seed physiological quality of wild radish to estimating genetic characters in order to select cultivars adapted to high temperature conditions. Hundred of half-brothers of wild radish were subjected to germination test and vigor (first count of germination, classification of seedling vigor, accelerated aging test, germination and testing of the first count at high temperature as well as seedling emergence in field. The germination test, first count test, accelerated aging and high germination test (20-35°C) can be used for the selection of wild radish crop populations adapted to germination and field emergence under high temperatures.

The efficiency of vigor tests in assessing the physiological potential of seeds depends on their standardization for different species. In this context, the research aimed to study specific methodologies to evaluate the vigor of spinach seeds (Tetragonia tetragonoides). For this purpose, five lots of spinach seed cv. New Zealand were submitted to germination, first count of germination, seedling emergence in the field, accelerated aging (with and without use of saturated solution of NaCl at 41and 45 ºC for 24,48 and 72 hours), controlled deterioration (18,21 and 24% seed water content adjustments for 24 hours at 45 ºC) and electrical conductivity tests with variations inwater volume (25, 50 and 75 mL), seed quantity (25 and 50) and soaking period (1, 2, 4, 8,12, 16 and 24 hours) at 25 ºC. The first count test allowed obtaining preliminary information about seed vigor, and other tests in general showed similarity with seedling emergence in the field. Thus, it was concluded that traditional accelerated aging test 24 h /41 ºC and accelerated aging test with saturated salt solution for 24 h at 41 ºC and 45 ºC, and controlled deterioration test with 21% seed water content / 24 h / 45 ºC were all efficient for evaluating the physiological potential of spinach seeds.

Two experiments were conducted in order to study the effects of seed number, temperature and time of soaking sesame seeds on the results of the electrical conductivity test for physiological quality of seeds. In the experiment I evaluated the electrical conductivity varying the number of seeds (25, 50 and 100) and temperature (20, 25 and 30° C) in experiment II is different soaking periods (2, 4, 6, 8, 12, 16, 20 and 24 h). The tests were performed with three cultivars (cv. Trebol, cv. Cnpa G4 e cv. Comum). The treatments in factorial 3x3x3 and 3x8 in experiments I and II, respectively. The experimental design was completely randomized, with four replications. Means were compared by Tukey test at 5% probability. The characterization of cultivars held through the germination and vigor tests (physical, physiological and biochemical), which rated the Trebol cultivar seeds with better physiological then the Cnpa G4 and Comum. The electrical conductivity test was conducted with four sub-samples of 25 pure seeds soaked in 75 mL of deionized water at 25° C, to separate the lots from 2 hours of soaking, being feasible to evaluate the quality sesame seeds.

Electrical conductivity has been proposed as a rapid test to evaluate seed vigor; however, few researches have emphasized methodologies to its use in seeds of medicinal plants, such as chamomile. The objective of the research was to evaluate the electrical conductivity of chamomile seeds affected by different imbibition times and sample size. The evaluations consisted of moisture content, germination and vigor (first count of germination) to seed initial characterization. Then, it was evaluated the electrical conductivity, affected by imbibition time (6, 12, 24 e 48 hours) and seed amount per sample (25, 50, 75, 100). The completely randomized design was used with four replications, arranged as a 4 x 4 factorial. Means were compared by the Tukey test at 5% of probability. It was concluded that the electrical conductivity of chamomile seeds is affected by the number of seeds per sample and imbibition time isolately.

Nowadays, image analysis is one of the most modern tools in evaluating physiological potential of seeds. This study aimed at verifying the efficiency of the seedling imaging analysis to assess physiological potential of wheat seeds. The seeds of wheat, cultivars IAC 370 and IAC 380, each of which represented by five different lots, were stored during four months under natural environmental conditions of temperature (T) and relative humidity (RH), in municipality of Piracicaba, Stated of São Paulo, Brazil. For this, bimonthly assessments were performed to quantify moisture content and physiological potential of seeds by means of tests of: germination, first count, accelerated aging, electrical conductivity, seedling emergence, and computerized analysis of seedlings, using the Seed Vigor Imaging System (SVIS®). It has been concluded that the computerized analyses of seedling through growth indexes and vigor, using the SVIS®, is efficient to assess physiological potential of wheat seeds.

Os efeitos do vigor de lotes de sementes sobre a emergência das plântulas e estabelecimento do estande, principalmente sob condições menos favoráveis de ambiente, estão bem documentados na literatura. Porém, há necessidade de intensificar a pesquisa para esclarecer as relações entre o potencial fisiológico das sementes e o desempenho das plantas em campo; este foi o principal objetivo deste estudo.Utilizaram-se duas cultivares de ervilha, 'Telefone Alta' (crescimento indeterminado) e 'Itapuã' (determinado), cada uma representada por quatro lotes armazenados, durante oito meses, em três ambientes: laboratório, câmara fria e seca (10 ºC e 30% de umidade relativa do ar) e ambiente controlado (20 ºC e 70% de umidade relativa do ar); este procedimento permitiu criar diferenças entre o potencial fisiológico dos lotes de cada cultivar. Após a determinação da germinação e do vigor (condutividade elétrica, envelhecimento acelerado, comprimento e emergência de plântulas), foi conduzido ensaio de campo, realizando-se avaliações do estande inicial e final, altura e área foliar de plantas, número de vagens e produção de grãos verdes e secos. O vigor das sementes de ervilha influencia a emergência de plântulas e o estabelecimento do estande em campo, especialmente em lotes pouco vigorosos. O vigor de sementes de ervilha afeta negativamente o desenvolvimento das plantas e a produção final, quando há redução acentuada do estande; a extensão desses efeitos é proporcional à intensidade dessa redução.

The evaluation of seed vigor is an important factor for detection of lots of high quality seeds, so that development of procedures to evaluate the physiological potential has been an important tool in quality control programs seeds. In this sense the study aimed to adapt the methodologies of accelerated aging, electrical conductivity and potassium leaching to evaluate Moringa oleifera seed vigor LAM.. Therefore, four lots of moringa seeds were subjected to the germination tests, seedling emergence, speed of emergence index, emergence first count, length and dry mass of seedlings and cold test for their physiological characterization, in addition to accelerated aging, electrical conductivity and potassium leaching. The experimental design was completely randomized with four replications of 50 seeds and the means compared by Tukey test at 5% probability. For accelerated aging the periods were studied aging 12, 24 and 72 hours at 40, 42 and 45°C. For the electrical conductivity test was used to a temperature of 25°C for periods of 4, 8, 12, 16 and 24 hours of immersion in 75 to 125 mL of distilled water, using 25 to 50 seeds, and for potassium leaching test samples were used 25 to 50 seeds, placed in plastic cups containing 70 and 100 mL of distilled water at 25°C for periods of 1, 2, 3, 4, 5 and 6 hours. From the results obtained, it can be inferred that the methods best fit for the accelerated aging test Moringa seeds were a temperature of 40°C for 12 to 72 hours, 42°C 72 hours 45°C 24 hours . In the electrical conductivity test Moringa seeds, the combination of 50 seeds in 75 mL distilled water for a period of immersion of 4 hours and 50 seeds in 125 mL of 4 hours were efficient for the differentiation of lots of Moringa seeds as to vigor and for potassium leaching test moringa seeds, the combination of 50 seeds in 100mL of distilled water allowed the separation of lots of four levels of vigor, at 2 hours of immersion, showing promise in evaluate the quality of moringa seeds.
