岷江柏(Cupressus chenggiana S. Y. Hu)是我国川甘地区特有的珍稀濒危乔木,一般生长在干旱的河谷区,在涵养水源和保护水 土等方面起着重要的作用。本文选择4个岷江柏种群,采用了野外调查和室内实验相结合的研究方法,调查岷江柏种群结实状况, 分析种子和球果形态特征,阐明种子发芽的基本特征,研究岷江柏种子贮藏过程中几个生理指标的动态变化特点,目的是为岷江柏 种苗繁育、自然更新能力评估以及珍稀濒危机制分析提供理论依据。研究得出如下结论:1.岷江柏球果呈椭球形,长为1.5~ 2.2cm,宽为1.5~1.9cm,质量为1.7~4.2g,球果鳞片数量为8~11片,球果内种子数量一般在40~70粒。岷江柏种子为椭圆形,长 为3.58~4.02mm,宽为3.10~3.15mm,厚为0.96~1.11mm,千粒重为3.1~3.5g。岷江柏的结实率很低,并且有显著的地理差异和 大小年差异。2. 岷江柏种子发芽温度范围是5℃~30℃,其中种子的适宜发芽温度范围是10℃~25℃。种子最适发芽温度随着贮藏 时间的增加而变化。在适宜温度范围内,种子发芽周期为20d。温度对种子的发芽势和T50有显著影响,对种子发芽率没有显著影响 ;光照有利于种子发芽;岷江柏种子的发芽特征是岷江柏保护种子资源、防止物种濒危的一种环境适应,有助于岷江柏种子提高发 芽率和幼苗的存活率。岷江柏种子是一种耐贮藏的正常性种子,在短期贮藏过程中,贮藏温度和种子含水量对于种子生理指标和种 子发芽没有显著影响。3. 岷江柏种子在短期贮藏过程中,千粒重没有显著变化;含水量都经历了先下降,再稳定的过程;粗脂肪 含量和可溶性糖含量逐渐降低;可溶性蛋白含量和丙二醛含量逐渐增加;脯氨酸含量在贮藏1~7个月时变化差异不明显,但是贮藏 7~10个月后显著增加。岷江柏种子的各个生理指标之间的相关性差异不显著。4. 岷江柏球果和种子的形态特征存在显著的地理差 异。岷江柏种子的发芽能力的地理性差异不大,种群间差异不大。岷江柏种群的地理差异由种群特征、生境特征和气候特征共同决 定。5. 在岷江柏的人工繁育中,对于刚刚采集的种子,发芽温度在15℃~25℃比较适合,其中以25℃最佳;而对于短期贮藏(4~ 10个月)后的种子,发芽温度在10℃~25℃均可,以15℃~20℃为最佳。野外播种的最适时间为4~6月,6~9月的间歇性干旱和降 水波动可能是限制岷江柏自然更新的因素之一。在短期贮藏过程中,种子可以采用常规室温贮藏,可以节约成本。Cupressus chenggiana is a specific and endangered plant in Sichuan and Gansu provinces of China, and it usually grows in dry valley and plays an important role in water supply and soil and water conservation in the dry valley of alpine and canyon region of southwest China. The research selected four Cupressus chenggiana populations and used the methods of the field investigation and the lab experiments. The fruiting characters of Cupressus chenggiana populations, the morphological characters of seeds and cones, the germination characters of seeds and the store physiological dymatics of several factors of seeds have been studied in order to give some theoretical advices on the artificial propagation and the ability of natural regeneration and the endangered principle of Cupressus chenggiana in the paper. The main results may be clarified as follows: 1. The cones of Cupressus chenggiana are ellipsoidal, length ranged from 1.5 to 2.2cm, with ranged from 1.5 to 1.9 cm, weight ranged from 1.7 to 4.2g, the number of cone squama ranged from 8 to 11, and the seed number of per cone ranged from 40 to 70. The seeds of Cupressus chenggiana are elliptical, length ranged from 3.58 to 4.02 mm, width ranged from 3.10 to 3.15 mm, thickness ranged from 0.96 to 1.11 mm, and the weight of 1000 seeds ranged from 3.1 to 3.5g. The fruiting rate of Cupressus chenggiana is very low, and the fruiting period of Cupressus chenggiana has the geographical differences and the big or small year differences. 2. Seed germination temperature is between 5℃ and 30℃, while the suited temperature is between 10℃ and 25℃. The optimum temperature of seed germination will change as the store time of seeds changes logner. The cycle of seed germination can persist 20 days in the range of the suited temperature. The germination temperatures have significant influences on the germination potential and T50, but have no significant infuluences on the germination rate. The photoperiod is in favor of seed germination. The characters of Cupressus chenggiana seed germination represent a kind of environmental adaptability to protect the seed sources and endangered species, and it can give help to increase the germination rate of seeds and the livability of seedings. The seeds of Cupressus chenggiana are a kind of orthodox seeds that can endure the long time storage. In the short time storage, the store temperatures and the moisture contents of seeds have no significant infuluences on the physiological factors and the germination of seeds, but the store time has significant influences on the physiological factors of seeds. 3. In the short store course of Cupressus chenggianna seeds, the 1000 seed weight has no significant variation; The moisture content descends at the beginning of the storage, but has no significant variation later; The crude fat content and the soluble sugar content descend gradually; The soluble protein content and MDA content increase gradually; The praline content has no significant variation after 1~7 months storage, but increase significantly after 7~10 months storage. The correlations of different physiological factors are not significant. 4. The morphological characters of cones and seeds of four populations exist significant differences. The germination of Cupressus chenggiana seeds has no significant geographical variation. The geographical variation of Cupressus chenggiana populations can be ascribed to the population characters, climate and environment. 5. In the course of artificial propagation of Cupressus chenggiana, it is favored that the germination temperature of newly collected seeds is between 15℃ and 25℃, while the optimum temperature is 25℃. After the short storage ranged from 4 months to 10 months, it is favored that the germination temperature is between 10℃ and 25℃, while the optimum temperature is ranged from 15℃ to 20℃. The field sowing optimum time is between April and June, and the interval drought and fallrain fluctuation between July and September may be one of the reasons that restrict natural regeneration of Cupressus chenggiana. In the short storage, seeds can be stored in the condition of room temperature.

BACKGROUND: Previously, tachyplesin gene (tac) has been successfully transferred into Undaria pinnatifida gametophytes using the method of microprojectile bombardment transformation. The objectives of this study were to compare and evaluate the performance of bubble-column and airlift bioreactors to determine a preferred configuration of bioreactor for vegetative propagation of transgenic U. pinnatifida gametophytes, and to then investigate the influence of light on vegetative propagation of these gametophytes, including incident light intensity, photoperiod and light quality to resolve the problems of rapid vegetative propagation within the selected bioreactor. RESULTS: Experimental results showed that final dry cell density in the airlift bioreactor was 12.7% higher than that in the bubble-column bioreactor under the optimal aeration rate of 1.2 L air min(-1) L-1 culture. And a maximum final dry cell density of 2830 mg L-1 was obtained within the airlift bioreactor using blue light at 40 mu mol m(-2) s(-1) with a light/dark cycle of 14/10 (h). Polymerase chain reaction (PCR) analysis indicated that genes (bar and tac) were not lost during rapid vegetative propagation within the airlift bioreactor. CONCLUSION: The airlift bioreactor was shown to be much more suitable for rapid vegetative propagation of transgenic U. pinnatifida gametophytes than the bubble-column bioreactor in the laboratory. The use of blue light allows improvement of vegetative propagation of transgenic U. pinnatifida gametophytes. (C) 2009 Society of Chemical Industry
Formation and early development of tetraspores of Polysiphonia urceolata (Rhodomelaceae, Rhodophyta)

Polysiphonia urceolata is one type of potential commercial red seaweeds used for breeding and cultivation, because of its significant biochemical and biomedical application. However, the information of breeding and seedling incubation for cultivation is limited, especially the early development. In this study, tetrasporohyte and gametophyte of P. urceolata were taken as the study materials in Huiquan Bay, Qingdao, China. The cleaned and sterilized tetrasporophytes and gametophytes were pre-cultured in sterilized seawater, then nurtured at 18A degrees C, 25 mu mol photons m(-2) s(-1) in 12:12 h (light:dark) photoperiod. Continuous observation under microscope showed that the early development consists of bipolar division stage and seedling stage. In the division stage, tetraspores germinate into bipolar sporelings that further differentiate into a colorless rhizoidal portion and a lightly pigmented upright shoot. The lightly pigmented rhizoidal cell develops to a rhizoid and the larger pigmented cell transforms to an erect axis. In the seedling stage, several quasi-protuberances appear on the erect axis and form juvenile seedlings. The results demonstrate the culture of P. urceolata from tetraspores under laboratory conditions.

Fluctuating light intensity had a more significant impact on growth of gametophytes of transgenic Laminaria japonica in a 2500 ml bubble-column bioreactor than constant light intensity. A fluctuating light intensity between 10 and 110 mu E m(-2) s(-1), with a photoperiod of 14 h:10 h light:dark, was the best regime for growth giving 1430 mg biomass l(-1).

A sporeling culture method using gametophyte clones developed in the early 1990s led to egg discharge occurring in the dark 5 min after the start of the dark period under growth under a 11:13 L-D photoperiod. The course of egg discharge could be disturbed by light, with irradiance as low as 5-6 mu mol photon m(-2) s(-1) causing 75-80% of the discharged eggs to detach from the oogonia and consequently to die within several hours. In order to enhance outgrowth rate of young sporophytes, a study was conducted to test the effect of controlling darkness in the period 2-3 h after dusk. When the slides were transferred from the standard 11:13 L-D regime to continuous light, eggs were discharged 5 min after the end of the light phase and peaked 5-l5 min later on first day after transfer, indicating that the female gametes "remember" the light-dark regime. This suggests the existence of an endogenous circadian rhythm. During the second and third days, very few eggs were discharged throughout the 11 h of the normal light phase of the L-D regime, indicating the inhibitory effect of continuous light and that the rhtyhm is easily damped by light.

Two photoperiodic responses, the development of sporophylls and hairs, have been quantified in sporophytes of the brown alga Undaria pinnatifida. In a final experiment, the algae were cultivated in outdoor, 2000-L seawater tanks in a greenhouse for up to 12 weeks, and daylength was regulated by automatic blinds mounted on top of the tanks. Vegetative young sporophytes were treated under short-day (SD; 8 h light per day) or long-day conditions (LD; 16 h light per day), at 12 h light per day or in a night-break regime (NB; 8 h light per day, 7.5 h dark, 1 h light, 7.5 h dark). The earliest sporophyll development was observed 6, 7 or 9 weeks under LD, NB or SD conditions, respectively. After 12 weeks the sporophylls were significantly longer and wider under LD or NB conditions than in the SD regime, and only half of the experimental algae had formed sporophylls under SD conditions, but all algae under LD or NB conditions. In a foregoing 7-week culture experiment performed in 300-L indoor tanks, enhanced sporophyll formation had also been observed under LD and not under SD conditions (NB omitted). In both experiments, blade elongation rates remained high until the end of the experiments in SD, but declined during sporophyll initiation in LD, NB or at 12 h light per day. Another difference caused by photoperiod was observed in regard to the development of surface hair spots which occurred in both experiments on the blades in LD, NB or at 12 h light per day with identical densities, but were completely lacking under SD conditions. It is concluded that U. pinnatifida is a facultatative long-day plant in regard to reproduction forming vigorously sporophylls in long days, and an obligate long-day plant in regard to hair formation.

Morphology and culture studies on germlings of Sargassum thunbergii (Mertens et Roth) Kuntze were carried out under controlled laboratory conditions. Growth characteristics of these germlings grown under different temperatures (from 10 to 25A degrees C), irradiances (from 9 to 88 mu mol photons m(-2) s(-1)), and under blue and white light conditions are described. The development of embryonic germlings follows the classic "8 nuclei 1 egg" type described for Sargassaceae. Fertilized eggs spent 5-6 h developing into multicellular germlings with abundant rhizoids after fertilization. Under conditions of 20A degrees C, 44 mu mol photons m(-2) s(-1) and photoperiod of 12 h, young germlings with one or two leaflets reached 2-3 mm in length after 8 weeks. Temperature variations (10, 15, 20, 25A degrees C) under 88 mu mol photons m(-2) s(-1) significantly influenced the growth rate within the first week, although this effect became less obvious after 8 weeks, especially at 15 and 20A degrees C. Variation in germling growth was highly significant under different irradiances (9, 18, 44, 88 mu mol photons m(-2) s(-1)) at 25A degrees C. Low temperature (10A degrees C) reduced germling growth. Growth of germlings cultured under blue light was lower than in white light. Optimal growth of these germlings occurred at 25A degrees C and 44 mu mol photons m(-2) s(-1).

Chondrus is a type of commercially produced red seaweed that widely used for food and carrageen extraction. Although the natural life history of the alga had been well understood, the factors influencing development of the tetraspore and carpospore remain poorly understood. In the perspective of seedling resources, the regulation of early development is crucial for the seedling nursing; therefore, it is necessary to understand the physiological influences during its early development. In this study, we studied the effects of temperature and irradiance on the early development of Chondrus ocellatus Holm under laboratory conditions. The released tetraspores and carpospores were cultivated at different temperatures (10-28 degrees C) and irradiances ( 10, 60 mu mol photons m(-2)s(-1)) with a photoperiod of 12L:12D. The results indicate that both tetraspores and carpospores are tolerant to temperatures of 10-25 degrees C, and have the highest relative growth rate at 20 degrees C. Irradiance variances influenced the growth of the discoid crusts, and the influence was more significant with increasing temperature; 60 mu mol photons m(-2)s(-1) was more suitable than 10 mu mol photons m(-2)s(-1). The optimum temperature and irradiance for the development of seedlings was 20 degrees C and 60 mu mol photons m(-2)s(-1), respectively.

红毛菜(Bangia Lyngb.)属于红藻门,与紫菜属同属红毛菜科,其味道和营养都优于紫菜。目前红毛菜栽培产业已在我国福建莆田展开,但栽培技术还有待提高。海藻栽培技术的发展和成熟依赖于对其生长发育过程的认识。本研究针对红毛菜发育过程及相关光合生理展开,并初步探讨了一采自山西娘子关泉淡水红毛菜群体(FWB)的系统地位。 色素突变标记的壳孢子萌发特征表明最初两次分裂产生的4细胞决定了完整植株的形态建成。成熟植株,为雌雄异体。雌性生殖器果胞的标志性分化结构为原始受精丝,环境因子是促发原始受精丝发展的外部因素,其膨大程度随受精的延迟而增大。原孢子是主要的无性生殖孢子类型,在不良环境中,藻体也会形成内生孢子或休眠包囊,或者藻体断裂后重新形成完整的植株。 红毛菜的生长发育很大程度上受环境因子的控制。高温不利于配子体的发育,15-20 ºC比较适宜。红毛菜无性繁殖的最适温度-光照组合为20 ºC-8 h,有性繁殖为15 ºC-12h。 不同发育阶段,PSII实际光合效率(Y(II))与细胞的健康状况以及光合器官完整性及其在细胞内的分布有关,而与细胞的类型关系不大。健康的假根细胞、已分化未成熟的精子以及果孢子细胞均具有很高的Y(II)。色素体由中间位变为围周位,中央大液泡(营养藻丝)和大小纤维囊泡(成熟孢子与精子)的产生,使得细胞Y(II)降低。刚放散的壳孢子Y(II)很低,说明在壳孢子由贝壳基质释放到自由水体过程,光合作用受到一定程度抑制;而2h后,Y(II)开始恢复,rbcL的转录水平非常高,为孢子的萌发储备物质和能量需求。 在失水和低盐胁迫下,藻体均维持较高的Y(II)。干出处理至藻体重量不再变化,复水后Y(II)可回复初始水平。海生红毛菜在100%淡水培养基中(约20ºC)培养7天后,部分雄性藻体依然活着。从而体现了红毛菜位居高潮带的生理优势。 FWB终生行无性繁殖,藻体形态与发生以及染色体数目(4条)与海生群体没有区别。而rbcL-rbcS Spacer序列显示,红毛菜海生群体(无性和有性)具有完全相同的序列,而FWB与它们有5bp差异,但是与欧洲、北美地区的淡水群体仅1bp不同,初步说明所有淡水红毛菜群体具有共同的原始起源。

A series of experiments were conducted to identify the factors that affected the growth and survival of the settling flounder larvae Paralichthys olivaceus. Settling larvae 24 days after hatching (DAH) were reared in 10-l experimental tanks up to 40 DAH, and two of the following factors were changed as controlled factors in each experiment: light regime (24L:0D or 12L:12D), prey density (1500, 3000, or 5000 Artemia l(-1)), shelter (sand or no sand) and stocking density (5, 10, or 15 fish l(-1)). Early settling larvae (24-35 DAH) experienced little mortality (less than 10% of the overall mortality) that was not significantly affected by above factors. In contrast, late settling larvae (36-40 DAH) suffered high cannibalistic mortality which was significantly influenced by each of the above factors. Larvae experienced significantly lower mortality at 10 fish l(-1) level than at other densities. Larvae at 15 fish l(-1) level had higher mortality than at 5 fish l(-1) when all other factors were identical. Larvae at 3000 and 5000 Artemia l(-1) treatments survived significantly better than at 1500 Artemia l(-1), but no significant differences in larval mortality were found between the two higher densities. Larvae suffered higher mortality at low prey density or at the absence of sand when they were exposed to longer photoperiod. Low stocking density significantly improved the growth of the settling larvae. The average daily instantaneous growth rate (G) at 5 and 15 fish l(-1) treatments were 0.050 and 0.034, with the coefficient of variation (CV) in final length at 16.4 and 23.5, respectively. Daily instantaneous growth rate increased significantly from 0.033 in the 1500 Artemia l(-1) to 0.041 and 0.045 in the 3000 and 5000 Artennia l(-1), respectively, but no significant difference in larval growth existed between the two higher prey densities. These findings suggested that the optimal prey density for growth and survival of the settling flounder larvae at a stocking density of 5 - 15 fish l(-1) was around 3000 Artemia l(-1) . Larvae that were exposed to 24L showed 20% increase in growth ( G = 0.046, CV = 18.7) than those exposed to 12L ( G = 0.037, CV = 20.5). Longer exposure to light significantly improved larval growth, provided sufficient food was available. Sand substrate did not show significant effects on larval growth, possibly because the larvae spent most of the time swimming or feeding in the water column during this stage. The present study was designed to examine whether photoperiod alone was effective to induce seasonal regulations in physiology in root voles (Microtus oeconomus) from the Qinghai-Tibetan plateau noted for its extreme cold environment. Root voles were randomly assigned into either long photoperiod (LD; 16L: 8D) or short photoperiod (SD; 8L: 16D) for 4 weeks at constant temperature (20 degrees C). At the end of acclimation, SD voles showed lower body mass and body fat coupled with higher energy intake than LD voles. SD greatly enhanced thermogenic capacities in root voles, as indicated by elevated basal metabolic rate (BMR), nonshivering thermogenesis (NST), mitochondrial protein content and uncoupling protein-1 (UCP1) content in brown adipose tissue (BAT). Although no variations in serum leptin levels were found between SD and LD voles, serum leptin levels were positively correlated with body mass and body fat mass, and negatively correlated with energy intake and UCP1 content in BAT, respectively. To summarize, SD alone is effective in inducing higher thermogenic capacities and energy intake coupled with lower body mass and body fat mass in root voles. Changes in photoperiod, ambient temperature and food availability trigger seasonal acclimatization in physiology and behavior of many animals. In the present study, seasonal adjustments in body mass and in several physiological, hormonal, and biochemical markers were examined in wild-captured plateau pikas (Ochotona curzoniae) from the Qinghai-Tibetan plateau. Our results showed that plateau pikas maintained a relatively constant body mass throughout the year and showed no seasonal changes in body fat mass and circulating levels of serum leptin. However, nonshivering thermogenesis, cytochrome c oxidase activity, and mitochondrial uncoupling protein 1 (UCP1) contents in brown adipose tissues were significantly enhanced in winter. Further, serum leptin levels were positively correlated with body mass and body fat mass while negatively correlated with UCP1 contents. Together, these data suggest that plateau pikas mainly depend on increasing thermogenic capacities, rather than decreasing body mass, to cope with cold, and leptin may play a potential role in their thermogenesis and body mass regulation.
