Das Zytoskelett eukaryotischer Zellen besteht aus drei verschiedenen Protein-Netzwerken: den Aktinfilamenten, Mikrotubuli und Intermediärfilamenten. Intermediärfilamente wurden ursprünglich als statische Strukturen angesehen, die die mechanische Stabilisierung der Zellen übernehmen. In den letzten Jahren hat sich dieses Bild jedoch geändert: Intermediärfilament-Netzwerke sind hochdynamisch und unterliegen kontinuierlichen Veränderungen, welche durch Phosphorylierungen reguliert werden. Sie interagieren mit anderen Zytoskelett-Proteinen und greifen in die Regulation von Schlüsselsignalwegen, die Zellwachstum und Zellteilung sowie Apoptose und Stressantwort bestimmen, ein. Die Mechanismen der Filamentplastizität konnten bisher jedoch nicht vollständig aufgeklärt werden. So ist beispielsweise unklar, wo Auf- und Abbau der Filamente stattfindet und welche Faktoren an der Netzwerkmodulation beteiligt sind. Ziel meiner Arbeit war es, einen Beitrag zur Aufklärung dieser Mechanismen am Beispiel der epithelialen Keratin-Intermediärfilamente zu leisten. Mit Hilfe von mikroskopischen Zeitrafferaufnahmen von fluoreszenzmarkierten Zellklonen wurden Nukleationszentren in der Zellperipherie identifiziert, in denen Keratinfilamentvorläufer gebildet werden. Es handelt sich dabei um fokale Adhäsionskomplexe, die als Anheftungsstellen zwischen der extrazellulären Matrix und dem intrazellulären Aktinfilament-System dienen. Es konnte gezeigt werden, dass diese Filamentvorläufer-Entstehung für alle untersuchten Keratinisoformen gültig ist und in epitelialen als auch nicht-epithelialen Zelltypen abläuft. Knock-Down der Adhäsionskomponente Talin verhinderte die Keratinfilamentbildung. Modulation der fokalen Adhäsionskinase, die den Auf- und Abbau der Adhäsionskomplexe koordiniert, beeinflusste ebenso die Bildung der Keratinfilamentnetzwerke. Es konnte weiterhin beobachtet werden, dass die N-terminalen Isoformen IE und IF des Zytolinkers Plectin in fokalen Adhäsionen lokalisieren und damit möglicherweise an der Vernetzung von Keratinfilamentvorläufern, Zelladhäsionen und Aktinfilamenten beteiligt sind. Letztlich stellte sich heraus, dass die Bildung der Keratinfilamentvorläufer unabhängig von Proteintranslation ist. In den mikroskopischen Zeitrafferaufnahmen wurde im Anschluss an die Keratinfilamentbildung ein kontinuierlicher zentripetaler Transport der wachsenden Vorläuferpartikel beobachtet. An Hand von pharmakologischen Experimenten konnte gezeigt werden, dass dieser Transport Aktinfilament-abhängig ist. Zeitgleich kommt es zu Partikelfusion und Integration in das periphere Netzwerk, das sich weiterhin in Richtung auf das Zellzentrum bewegt. Mit Hilfe von Photoaktivierungsversuchen und Zellfusionsexperimenten konnte die Hypothese bestätigt werden, dass der Abbau der einwandernden Keratinfilamente in lösliche, rasch diffusible Zwischenstufen den kontinuierlichen peripheren Neuaufbau ermöglicht. Aus den Beobachtungen und bereits bekannten Ergebnissen wurde ein Modell des Keratin-Zyklus entwickelt, das die folgenden Stadien umfasst: Nukleation von Keratinfilamentvorläufern an fokalen Adhäsionen in der Zellperipherie, Elongation und Fusion der Keratinfilamentvorläufer bei zeitgleichem Aktinfilament-abhängigem zentripetalen Transport, Integration der Keratinfilamentvorläufer in das periphere Netzwerk, Bündelung der Filamente, Filamentabbau in lösliche Untereinheiten und Neubeginn des Zyklus in der Zellperipherie. Eine Störung dieses Zyklus liegt bei mutierten Keratinen vor, welche die Ursache von Blasen-bildenden Hauterkrankungen sind. In der vorliegenden Arbeit wurde am Beispiel von Keratin 6a-Mutanten, welche die Hauterkrankung Pachyonychia congenita verursachen, gezeigt, dass bei diesen Keratinen die Nukleation zwar im Bereich der Adhäsionskomplexe regelrecht abläuft, die anschließende Elongation und Netzwerkbildung aber gestört ist, so dass statt dessen kurzlebige, hyperphosphorylierte Granula entstehen. Der resultierende frustrane Keratin-Zyklus in der Zellperipherie ist stark beschleunigt und kann durch p38-Inhibierung gestoppt werden. Bei Proteasomeninhibierung wird der Zyklus in Richtung der Granulabildung verschoben. In dieser Arbeit wird erstmals das Keratin-Tretmühlen-Modell vorgestellt, das den regulierbaren Auf- und Abbau-Zyklus des Keratinnetzwerks beschreibt. Damit liegen testbare Hypothesen für die Aufklärung der Keratinfilament-Plastizität in physiologischen und pathologischen Situationen vor, die nach unseren ersten Ergebnissen auch von Relevanz für andere Intermediärfilamenttypen sind.

In the developing chicken embryo yolk sac vasculature, the expression of arterial identity genes requires arterial hemodynamic conditions. We hypothesize that arterial flow must provide a unique signal that is relevant for supporting arterial identity gene expression and is absent in veins. We analyzed factors related to flow, pressure and oxygenation in the chicken embryo vitelline vasculature in vivo. The best discrimination between arteries and veins was obtained by calculating the maximal pulsatile increase in shear rate relative to the time-averaged shear rate in the same vessel: the relative pulse slope index (RPSI). RPSI was significantly higher in arteries than veins. Arterial endothelial cells exposed to pulsatile shear in vitro augmented arterial marker expression as compared with exposure to constant shear. The expression of Gja5 correlated with arterial flow patterns: the redistribution of arterial flow provoked by vitelline artery ligation resulted in flow-driven collateral arterial network formation and was associated with increased expression of Gja5. In situ hybridization in normal and ligation embryos confirmed that Gja5 expression is confined to arteries and regulated by flow. In mice, Gja5 (connexin 40) was also expressed in arteries. In the adult, increased flow drives arteriogenesis and the formation of collateral arterial networks in peripheral occlusive diseases. Genetic ablation of Gja5 function in mice resulted in reduced arteriogenesis in two occlusion models. We conclude that pulsatile shear patterns may be central for supporting arterial identity, and that arterial Gja5 expression plays a functional role in flow-driven arteriogenesis.

TIE2 is a vascular endothelial-specific receptor tyrosine kinase essential for the regulation of vascular network formation and remodeling. Previously, we have shown that the 1.2-kb 5' flanking region of the TIE2 promoter is capable of directing beta-galactosidase reporter gene expression specifically into a subset of endothelial cells (ECs) of transgenic mouse embryos. However, transgene activity was restricted to early embryonic stages and not detectable in adult mice. Herein we describe the identification and characterization of an autonomous endothelial-specific enhancer in the first intron of the mouse TIE2 gene. Furthermore, combination of the TIE2 promoter with an intron fragment containing this enhancer allows it to target reporter gene expression specifically and uniformly to virtually all vascular ECs throughout embryogenesis and adulthood. To our knowledge, this is the first time that an in vivo expression system has been assembled by which heterologous genes can be targeted exclusively to the ECs of the entire vasculature. This should be a valuable tool to address the function of genes during physiological and pathological processes of vascular ECs in vivo. Furthermore, we were able to identify a short region critical for enhancer function in vivo that contains putative binding sites for Ets-like transcription factors. This should, therefore, allow us to determine the molecular mechanisms underlying the vascular-EC-specific expression of the TIE2 gene.

BACKGROUND Angiogenesis and vascular remodelling are crucial events in tissue repair mechanisms promoted by cell transplantation. Current evidence underscores the importance of the soluble factors secreted by stem cells in tissue regeneration. In the present study we investigated the effects of paracrine factors derived from cultured endothelial progenitor cells (EPC) on rat brain endothelial cell properties and addressed the signaling pathways involved. METHODS Endothelial cells derived from rat brain (rBCEC4) were incubated with EPC-derived conditioned medium (EPC-CM). The angiogenic response of rBCEC4 to EPC-CM was assessed as effect on cell number, migration and tubular network formation. In addition, we have compared the outcome of the in vitro experiments with the effects on capillary sprouting from rat aortic rings. The specific PI3K/AKT inhibitor LY294002 and the MEK/ERK inhibitor PD98059 were used to study the involvement of these two signaling pathways in the transduction of the angiogenic effects of EPC-CM. RESULTS Viable cell number, migration and tubule network formation were significantly augmented upon incubation with EPC-CM. Similar findings were observed for aortic ring outgrowth with significantly longer sprouts. The EPC-CM-induced activities were significantly reduced by the blockage of the PI3K/AKT and MEK/ERK signaling pathways. Similarly to the outcome of the rBCEC4 experiments, inhibition of the PI3K/AKT and MEK/ERK pathways significantly interfered with capillary sprouting induced by EPC-CM. CONCLUSION The present study demonstrates that EPC-derived paracrine factors substantially promote the angiogenic response of brain microvascular endothelial cells. In addition, our findings identified the PI3K/AKT and MEK/ERK pathways to play a central role in mediating these effects.

DNA-grafted supramolecular polymers (SPs) allow the programmed organization of DNA in a highly regular, one-dimensional array. Oligonucleotides are arranged along the edges of pyrene-based helical polymers. Addition of complementary oligonucleotides triggers the assembly of individual nanoribbons resulting in the development of extended supramolecular networks. Network formation is enabled by cooperative coaxial stacking interactions of terminal GC base pairs. The process is accompanied by structural changes in the pyrene polymer core that can be followed spectroscopically. Network formation is reversible, and disassembly into individual ribbons is realized either via thermal denaturation or by addition of a DNA separator strand.

This article is the introduction to a special issue of The Developing Economies which presented the results of a research project by the Institute of Developing Economies that examined the development mechanisms in Korea and Taiwan. Our conclusion in this article is that their development mechanisms, despite their similar development patterns of export-led industrialization, have been essentially different: a government-led mechanism in Korea as opposed to a market-led mechanism in Taiwan. We verified this difference through comparative studies of the two economies covering trade balances, the growth of total factor productivity, the scale of enterprises and business groups, and the development processes of individual manufacturing sectors. In our explanatory discussion we propose that the difference in the mechanisms is based on: 1) the amount of accumulation in the economy at the time postwar industrialization started, 2) the relationship between government and society, and 3) the mechanism of social network formation.

We examine network formation via bilateral trade agreement (BTA) among three symmetric countries. Each government decides whether to form a link or not via a BTA depending on the differential of ex-post and ex-ante sum of real wages in the country. We model the governmental decision in two forms, myopic and farsighted and analyze the effects on the BTA network formation. First, we find that both myopic and farsighted games never induce the formation of star networks nor empty networks. Second, the networks resulting from myopic game coincides with those resulting from farsighted games.

Cytoplasmic dynein is one of the major motor proteins involved in intracellular transport. It is a protein complex consisting of four subunit classes: heavy chains, intermediate chains (ICs), light intermediate chains, and light chains. In a previous study, we had generated new monoclonal antibodies to the ICs and mapped the ICs to the base of the motor. Because the ICs have been implicated in targeting the motor to cargo, we tested whether these new antibodies to the intermediate chain could block the function of cytoplasmic dynein. When cytoplasmic extracts of Xenopus oocytes were incubated with either one of the monoclonal antibodies (m74–1, m74–2), neither organelle movement nor network formation was observed. Network formation and membrane transport was blocked at an antibody concentration as low as 15 μg/ml. In contrast to these observations, no effect was observed on organelle movement and tubular network formation in the presence of a control antibody at concentrations as high as 0.5 mg/ml. After incubating cytoplasmic extracts or isolated membranes with the monoclonal antibodies m74–1 and m74–2, the dynein IC polypeptide was no longer detectable in the membrane fraction by SDS-PAGE immunoblot, indicating a loss of cytoplasmic dynein from the membrane. We used a panel of dynein IC truncation mutants and mapped the epitopes of both antibodies to the N-terminal coiled-coil domain, in close proximity to the p150Glued binding domain. In an IC affinity column binding assay, both antibodies inhibited the IC–p150Glued interaction. Thus these findings demonstrate that direct IC–p150Glued interaction is required for the proper attachment of cytoplasmic dynein to membranes.

Free nitric oxide (NO) reacts with sulphydryl residues to form S-nitrosothiols, which act as NO reservoirs. We sought to determine whether thiol-preserving agents and antioxidants, such as dithiothreitol (DTT) and vitamin C, induce NO release from S-nitrosylated proteins in endothelial cell cultures to promote angiogenesis. NO release was measured directly in cell supernatants using a Sievers NO Analyser, and in vitro angiogenesis was assessed by quantifying capillary-like tube network formation of porcine aortic endothelial cells (PAEC) on growth factor-reduced Matrigel. Incubation of PAEC with DTT or vitamin C significantly increased NO release in a concentration-dependent manner. However, the nitric oxide synthase (NOS) inhibitors, L-NNA and L-NIO, had no effect on DTT- or vitamin C-induced NO release, and there was no concomitant increase in the phosphorylation of endothelial NOS at serine-1177 following DTT or vitamin C treatment. DTT and vitamin C increased capillary-like tube network formation by nine- and two-fold, respectively, and the addition of copper ions doubled the effect of vitamin C. Surprisingly, DTT maintained endothelial tube networks for up to one month under serum-free conditions, and selective inhibitors of guanylyl cyclase (ODQ) and PKG (KT-5823) blocked this, demonstrating the requirement of cyclic GMP and PKG in this process. Both DTT and vitamin C are capable of releasing sufficient NO from S-nitrosothiols to induce capillary morphogenesis. This study provides the first evidence that increased denitrosylation leads to increased bioavailability of NO, independent of NOS activity, to promote sustained angiogenesis.

Glutaredoxin-1 (Glrx) is a cytosolic enzyme that regulates diverse cellular function by removal of GSH adducts from S-glutathionylated proteins including signaling molecules and transcription factors. Glrx is up-regulated during inflammation and diabetes. Glrx overexpression inhibits VEGF-induced endothelial cell (EC) migration. The aim was to investigate the role of up-regulated Glrx in EC angiogenic capacities and in vivo revascularization in the setting of hind limb ischemia. Glrx overexpressing EC from Glrx transgenic mice (TG) showed impaired migration and network formation and secreted higher level of soluble VEGF receptor 1 (sFlt), an antagonizing factor to VEGF. After hind limb ischemia surgery Glrx TG mice demonstrated impaired blood flow recovery, associated with lower capillary density and poorer limb motor function compared to wild type littermates. There were also higher levels of anti-angiogenic sFlt expression in the muscle and plasma of Glrx TG mice after surgery. Non-canonical Wnt5a is known to induce sFlt. Wnt5a was highly expressed in ischemic muscles and EC from Glrx TG mice, and exogenous Wnt5a induced sFlt expression and inhibited network formation in human microvascular EC. Adenoviral Glrx-induced sFlt in EC was inhibited by a competitive Wnt5a inhibitor. Furthermore, Glrx overexpression removed GSH adducts on p65 in ischemic muscle and EC, and enhanced nuclear factor kappa B (NF-kB) activity which was responsible for Wnt5a-sFlt induction. Taken together, up-regulated Glrx induces sFlt in EC via NF-kB -dependent Wnt5a, resulting in attenuated revascularization in hind limb ischemia. The Glrx-induced sFlt may be a part of mechanism of redox regulated VEGF signaling.

S-glutathionylation occurs when reactive oxygen or nitrogen species react with protein-cysteine thiols. Glutaredoxin-1 (Glrx) is a cytosolic enzyme which enzymatically catalyses the reduction in S-glutathionylation, conferring reversible signalling function to proteins with redox-sensitive thiols. Glrx can regulate vascular hypertrophy and inflammation by regulating the activity of nuclear factor κB (NF-κB) and actin polymerization. Vascular endothelial growth factor (VEGF)-induced endothelial cell (EC) migration is inhibited by Glrx overexpression. In mice overexpressing Glrx, blood flow recovery, exercise function and capillary density were significantly attenuated after hindlimb ischaemia (HLI). Wnt5a and soluble Fms-like tyrosine kinase-1 (sFlt-1) were enhanced in the ischaemic-limb muscle and plasma respectively from Glrx transgenic (TG) mice. A Wnt5a/sFlt-1 pathway had been described in myeloid cells controlling retinal blood vessel development. Interestingly, a Wnt5a/sFlt-1 pathway was found also to play a role in EC to inhibit network formation. S-glutathionylation of NF-κB components inhibits its activation. Up-regulated Glrx stimulated the Wnt5a/sFlt-1 pathway through enhancing NF-κB signalling. These studies show a novel role for Glrx in post-ischaemic neovascularization, which could define a potential target for therapy of impaired angiogenesis in pathological conditions including diabetes.

Neuropeptides affect the activity of the myriad of neuronal circuits in the brain. They are under tight spatial and chemical control and the dynamics of their release and catabolism directly modify neuronal network activity. Understanding neuropeptide functioning requires approaches to determine their chemical and spatial heterogeneity within neural tissue, but most imaging techniques do not provide the complete information desired. To provide chemical information, most imaging techniques used to study the nervous system require preselection and labeling of the peptides of interest; however, mass spectrometry imaging (MSI) detects analytes across a broad mass range without the need to target a specific analyte. When used with matrix-assisted laser desorption/ionization (MALDI), MSI detects analytes in the mass range of neuropeptides. MALDI MSI simultaneously provides spatial and chemical information resulting in images that plot the spatial distributions of neuropeptides over the surface of a thin slice of neural tissue. Here a variety of approaches for neuropeptide characterization are developed. Specifically, several computational approaches are combined with MALDI MSI to create improved approaches that provide spatial distributions and neuropeptide characterizations. After successfully validating these MALDI MSI protocols, the methods are applied to characterize both known and unidentified neuropeptides from neural tissues. The methods are further adapted from tissue analysis to be able to perform tandem MS (MS/MS) imaging on neuronal cultures to enable the study of network formation. In addition, MALDI MSI has been carried out over the timecourse of nervous system regeneration in planarian flatworms resulting in the discovery of two novel neuropeptides that may be involved in planarian regeneration. In addition, several bioinformatic tools are developed to predict final neuropeptide structures and associated masses that can be compared to experimental MSI data in order to make assignments of neuropeptide identities. The integration of computational approaches into the experimental design of MALDI MSI has allowed improved instrument automation and enhanced data acquisition and analysis. These tools also make the methods versatile and adaptable to new sample types.

Tämän tutkimuksen tavoitteena oli selvittää kuinka yrittäjän verkostot muodostuvat ja minkälainen niiden rakenne on. Tavoitteena oli myös saada tietoa yrittäjän sosiaalisten verkostojen merkityksestä yrittäjälle. Tutkimus toteutettiin laadullisena tutkimuksena. Tutkimusaineisto kerättiin strukturoidulla haastattelulla ja tutkimusaineiston rinnalla käytettiin yrittäjien havainnollistamia piirroksia sekä tieteellisiä julkaisuja ja kirjallisuutta. Haastateltavina oli sekä nais- että miesyrittäjiä eri paikkakunnilta Etelä-Suomen alueelta. Tutkimustulosten mukaan yrittäjien verkostoissa voidaan havaita eri tasoja, jotka rakentuvat eri tavoilla ja ovat yrittäjälle merkitykseltään erilaisia. Verkoston eri tasoille sijoittuu yrittäjälle tärkeitä ihmisiä sen mukaan mitä hyötyä ja arvoa heillä yrittäjälle on. Tulokset osoittavat, että verkostojen rakentamiseen vaikuttavat sekä liiketoiminnan laatu, motiivit sekä yrittäjän persoona. Tulokset ovat linjassa tämän tutkimuksen perustana olevien teorioiden kanssa. Viitteitä löytyy myös siitä, että yrittäjän sukupuolella on myös merkitystä siihen, millaiseksi sosiaaliset verkostot rakentuvat sekä henkilökohtaisessa elämässä että liike-elämän puolella. Sosiaalisista verkostoista saatavat hyödyt ovat moninaisia. Lisätutkimuksia tulisi tehdä tehokkaiden työkalujen kehittämiseksi henkilöstön ja yrittäjän omien resurssien tunnistamiseksi. Tätä kautta myös yrittäjyyskasvatusta ja yrittäjäneuvontaa voitaisiin tehostaa.
