Cromoglycate is accumulated on a poly-L-lysine (PLL) modified carbon electrode best from pH 4 solution, where it is anionic and the PLL is cationic, and at which pH the cromoglycate gives a good reduction peak at -0.82 V. The PLL film can be regenerated readily by washing the electrode with 3 M sodium hydroxide solution, in which the PLL is deprotonated. Regeneration of the film is not required as frequently when larger amounts of PLL are incorporated into it. This allows standard addition procedures to be carried out without regenerating the electrode. Linear calibration graphs have been obtained typically in the range 0.1 - 1.5 mug ml(-1). Detection limits have been calculated to be 10 ng ml(-1). The standard addition method has been applied satisfactorily to diluted urine solutions. (C) 2003 Elsevier B.V. B.V. All rights reserved.

A film of poly-L-lysine (PLL) adheres better to the surface of a glassy carbon electrode when the PLL is partially cross-linked by means of glutaraldehyde. A film composition of 97.5% PLL/2.5% glutaraldehyde gives good adhesion and retains the anionic exchange capability of the PLL. The performance of the film was tested with hexacyanoferrate(III) using electrochemical and nonelectrochemical accumulation.

The detection limit (about 0.017 mu g mL(-1)) for voltammetric determination of iodide (peak at +0.87 V vs. Ag/AgCl at pH 2) at a glutaraldehyde-cross-linked poly-L-lysine modified glassy carbon electrode involving oxidation to iodine was found to be several orders of magnitude lower than that for the voltammetric determination on a bare glassy carbon electrode. This method was applied successfully to the determination of iodide in two medicinal formulations. Idoxuridine was determined indirectly at the same electrode by accumulating it first at -0.8 V vs. Ag/AgCl. At this potential the C-I bond in the adsorbed idoxuridine is reduced giving iodide, which is then determined at the modified electrode. The method was successfully applied to the determination of idoxuridine in a urine sample.

Lysine-ketoglutaratc reductase catalyzes the first step of lysine catabolism in maize (Zea mays L.) endosperm. The enzyme condenses L-lysine and α-ketoglutarate into saccharopine using NADPH as cofactor. It is endosperm-specific and has a temporal pattern of activity, increasing with the onset of kernel development, reaching a peak 20 to 25 days after pollination, and thereafter decreasing as the kernel approaches maturity. The enzyme was extracted from the developing maize endosperm and partially purified by ammonium-sulfate precipitation, anion-exchange chromatography on DEAE-cellulose, and affinity chromatography on Blue-Sepharose CL-6B. The preparation obtained from affinity chromatography was enriched 275-fold and had a specific activity of 411 nanomoles per minute per milligram protein. The native and denaturated enzyme is a 140 kilodalton protein as determined by polyacrylamide gel electrophoresis. The enzyme showed specificity for its substrates and was not inhibited by either aminoethyl-cysteine or glutamate. Steady-state product-inhibition studies revealed that saccharopine was a noncompetitive inhibitor with respect to α-ketoglutarate and a competitive inhibitor with respect to lysine. This is suggestive of a rapid equilibriumordered binding mechanism with a binding order of lysine, α-ketoglutarate, NADPH. The enzyme activity was investigated in two maize inbred lines with homozygous normal and opaque-2 endosperms. The pattern of lysine-ketoglutarate reductase activity is coordinated with the rate of zein accumulation during endosperm development. A coordinated regulation of enzyme activity and zein accumulation was observed in the opaque-2 endosperm as the activity and zein levels were two to three times lower than in the normal endosperm. Enzyme extracted from L1038 normal and opaque-2 20 days after pollination was partially purified by DEAE-cellulose chromatography. Both genotypes showed a similar elution pattern with a single activity peak eluted at approximately 0.2 molar KCL. The molecular weight and physical properties of the normal and opaque-2 enzymes were essentially the same. We suggest that the Opaque-2 gene, which is a transactivator of the 22 kilodalton zein genes, may be involved in the regulation of the lysine-ketoglutarate reductase gene in maize endosperm. In addition, the decreased reductase activity caused by the opaque-2 mutation may explain, at least in part, the elevated concentration of lysine found in the opaque-2 endosperm.

Four trials of identical experimental design were conducted to determine the effects of temperature, dietary Lys level, and dietary Arg:Lys ratios on performance and carcass yield of male broilers. Birds of a commercial strain were grown from 21 to 42 d of age in wire-floored finishing batteries placed in environmental chambers. The chambers were programmed to provide either a constant thermoneutral temperature (21.1 C), a constant cold temperature (15.5 C), or a cycling hot diurnal temperature (25.5 to 33.3 C). Within each environment there was a factorial arrangement of three Lys levels (1.0, 1.1, and 1.2%) with four Arg:Lys ratios (1.1:1, 1.2:1, 1.3:1, and 1.4:1). Environmental temperature significantly influenced virtually every characteristic examined. Hot cyclic temperatures reduced weight gain, feed intake, and breast meat yield, and increased feed conversion, dressing percentage, leg quarter yield, and abdominal fat content. The cold environment promoted increased feed intake and mortality. Ascites and cardiomyopathy were the leading causes of death under cold exposure and thermoneutral conditions, whereas complications arising from heat exposure were the main cause of death under hot cyclic conditions. Levels of Lys affected leg quarter yield and abdominal fat content over all environments but increased breast meat yield only under cold conditions. Increasing Arg: Lys ratios improved feed conversion and dressing percentage and reduced abdominal fat content; it could not be determined whether these responses were consistent with Arg per se or were due to a nonspecific N response. As increasing Lys levels or Arg:Lys ratios did not improve weight gain, increase breast meat yield, or attenuate adverse effects due to heat or cold exposure, it is concluded that the levels of Lys and Arg suggested for 21 to 42 d by the NRC are adequate for birds of this age under the environmental conditions encountered.

One experiment was conducted to evaluate the effect of using different lysine and methionine + cystine levels on body weight recovery, performance, and egg quality of laying hens during the postmolting period. In this trial, 432 Isa Brown layers, with 72 weeks of age, were distributed in 54 cages according to a completely randomized design with six treatments and nine replicates of eight birds each. During the resting period, six diets with different digestible lysine and methionine + cystine levels were used, as follows: 0.48% digestible lysine and 0.43% methionine + cystine; 0.48% digestible lysine and 0.47% methionine + cystine; 0.48% digestible lysine and 0.52% methionine + cystine; 0.56% digestible lysine and 0.50% methionine + cystine; 0.56% digestible lysine and 0.56% methionine + cystine; 0.56% digestible lysine and 0.62% methionine + cystine. Data were submitted to analysis of variance and means were compared at by Tukey's test at 5% probability level. The different lysine and methionine + cystine levels in the diets fed during the resting period significantly influenced layer performance. The diet containing 0.56% lysine and 0.56% methionine + cystine promoted higher egg weight eggs during the second production cycle.

Egg quality of semi-heavy laying hens fed on low protein diets (14.0% CP) and on different lysine levels is evaluated, while maintaining the same ratio of digestible amino acids / digestible lysine. Four hundred and twenty commercial strain Isa Brown laying hens, 28 weeks old, were divided into 42 experimental plots. A completely randomized design with six treatments and seven replicates was employed in four production cycles of 28 days each. Treatments comprised Control - 16.92% CP; 0.750% digestible lysine. Treatments 1 to 5, with CP levels 14% and digestible lysine levels 0.600, 0.675, 0.750, 0.825 and 0.900% respectively. Levels of Treatments 1 and 2 (0.546 and 0.640% digestible Met + Cys / 0.600 and 0.675% digestible lysine) provided smaller egg size. On the other hand, eggs had higher shell percentage when compared to control diet. When compared to other digestible amino acids, digestible lysine requirement may be estimated at 0.750% in a diet with 14% CP, which corresponds to the average daily intake of 876 mg dig. lysine hen-1 day-1 and 798 mg dig. Met + Cys hen-1 day-1, without jeopardizing performance and eggs' internal and external quality.

The focus of this study was to evaluate the effect of lysine on the productive (weight and length gain, apparent feed conversion, specific growth rate and condition factor) and reproductive performance of Rhamdia voulezi males confined in net cages during the first reproductive cycle. The following parameters were assessed: seminal parameters (motility rate and duration, survival, sperm concentration, morphology, pH and osmolarity), hormonal parameters (cortisol and testosterone), testicular tissue (histomorphology), organosomatic indices (gonadosomatic, hepatosomatic and viscerosomatic indices) and composition of essential amino acids, crude protein and moisture of whole carcass. Four hundred fish were used, distributed in a random experimental design with four treatments and four replications in 16 net cages with 25 fish each. The treatments consisted of four different diets prepared so as to contain the following levels of lysine: 1.20, 1.40, 1.60 and 1.80%, with 30% crude protein and 3500 kcal kg(-1) digestible energy for 185 days (Jul./12-Jan./13). Eighteen males were selected per treatment, and they all released semen after slight abdominal pressure. The males were weighed, measured, submitted to hypophysation (2.5 mg kg(-1) carp pituitary extract), and then had their semen and blood collected. The fish were sacrificed by cervical dislocation, dissected, and the testes, liver, fat and guts were removed and weighed. The effects (p < 0.05) for the means of final weight, weight gain, apparent feed conversion and condition factor were observed for the analysis of productive performance. With regard to the reproductive parameters, only the seminal volume was affected (p < 0.05). Thus, the levels of testosterone showed quadratic effect (p < 0.05). The anatomy and the histomorphology of the testes were similar between the treatments during the spermiation period. With regard to the organosomatic indices, there was no influence (p > 0.05) between the treatments. The amino acids in the carcass were not affected (p > 0.05). The increment of lysine in the diet provided linear increase for weight gain and seminal volume and linear decrease for feed conversion in R. voulezi broodstocks confined in net cages. The aim of this study was to determine the coefficients of the Goettingen model for Redbro birds and estimate the digestible lysine requirements. To determine the model parameters, three nitrogen balance trials were performed in Periods I (14-28 days), II (42-56 days) and III (70-84 days), using 42 birds per trial. The birds were individually housed and subjected to six diets with increasing levels of nitrogen, with lysine as the limiting amino acid (deficient by 20% in relation to other amino acids). Dietary nitrogen concentrations were 8, 16, 24, 32, 40 and 48 g/kg. A control diet was added to confirm lysine as the first limiting amino acid. Nitrogen balance trials were divided into 5 days of adaptation and two periods of excreta collection, each one of 5 days. The response of the birds to a control diet confirmed that lysine was the first limiting amino acid. The adjustment of the exponential functions between nitrogen retention or excretion and nitrogen intake allowed estimation of parameters of the Goettingen model. The maximum potential for nitrogen retention was 3276, 2585 and 2603 mg/BWkg0.67.day, nitrogen maintenance requirement was 225, 135 and 122 mg/BWkg0.67.day and efficiency of nitrogen utilisation was 313 x 10(-6), 406 x 10(-6) and 415 x 10(-6) in the phases of 14-28, 42-56 and 70-84 days. The digestible lysine intake for Periods I, II and III, based on 60% of the maximum potential for nitrogen retention, was 711, 989 and 1272 mg/day (1.225%, 1.137% and 1.09% of lysine in the diet for a daily feed intake of 58, 87 and 117 g/day), respectively.

The objectives of this study were to (1) evaluate if dietary lysine (Lys) has an effect on the free amino acid (FAA) pool of Yellow Perch Perca flavescens eggs, and (2) determine how dietary Lys influences the reproductive performance and eggs fertilization rate (embryo survival) of female Yellow Perch. Two-year-old Yellow Perch of initial size of approximately 75 g were randomly distributed into six 400-L tanks at 32 +/- 1 fish per tank. This experiment included two wheat gluten-based diets in triplicate Lys-deficient ([-] Lys) and Lys-supplemented ([+]Lys; 2.23% in dry feed) diets. Females from the reference group were fed a commercial diet. Females from reference, (+) Lys, and (-) Lys groups were stripped and their eggs divided into 0.4-1.2-g portions and mixed with sperm (21.4 +/- 4.3 mu L) from either reference, (+) Lys, or (-) Lys males. The mean weight of Yellow Perch females and mean total weight of ovulated eggs were the greatest in the reference group compared with both (+) Lys and (-) Lys groups. There were no differences in the ratio of weight of eggs to female body weight as well as egg size among groups. There was no difference among treatments in the concentration of free amino acids except glutamic and aspartic acids in Yellow Perch eggs. There was significant effect of female dietary treatments on the egg fertilization rate averaged across all males. The higher fertilization rate was observed in the reference and (+) Lys groups compared with the (-) Lys group. The effect of female dietary treatment on the egg survival was also dependent on the dietary treatment of males.
