[ES]Este trabajo tiene como objetivo analizar las soluciones existentes para el diseño de las líneas mixtas. Se trata de un tipo de líneas bastante complejas de diseñar y que por lo general han sido poco estudiadas por la sociedad académica, a pesar de las numerosas ventajas que ofrece en comparación a las líneas simples, mucho más investigadas. Para facilitar la comprensión de su diseño, y antes de su estudio, se realiza un análisis de las características de las líneas mixtas. Posteriormente se introduce una metodología desarrollada por Lluis Cuatrecasas (Presidente del Instituto Lean Management de España), que permite salvar la brecha que existe a su juicio entre los estudios sobre los diseños de este tipo de líneas y su aplicación a la empresa. Finalmente, se ilustra dicha metodología mediante un caso práctico y se evalúan sus fortalezas y debilidades.

I. The binding of the intercalating dye ethidium bromide to closed circular SV 40 DNA causes an unwinding of the duplex structure and a simultaneous and quantitatively equivalent unwinding of the superhelices. The buoyant densities and sedimentation velocities of both intact (I) and singly nicked (II) SV 40 DNAs were measured as a function of free dye concentration. The buoyant density data were used to determine the binding isotherms over a dye concentration range extending from 0 to 600 µg/m1 in 5.8 M CsCl. At high dye concentrations all of the binding sites in II, but not in I, are saturated. At free dye concentrations less than 5.4 µg/ml, I has a greater affinity for dye than II. At a critical amount of dye bound I and II have equal affinities, and at higher dye concentration I has a lower affinity than II. The number of superhelical turns, τ, present in I is calculated at each dye concentration using Fuller and Waring's (1964) estimate of the angle of duplex unwinding per intercalation. The results reveal that SV 40 DNA I contains about -13 superhelical turns in concentrated salt solutions.

The free energy of superhelix formation is calculated as a function of τ from a consideration of the effect of the superhelical turns upon the binding isotherm of ethidium bromide to SV 40 DNA I. The value of the free energy is about 100 kcal/mole DNA in the native molecule. The free energy estimates are used to calculate the pitch and radius of the superhelix as a function of the number of superhelical turns. The pitch and radius of the native I superhelix are 430 Å and 135 Å, respectively.

A buoyant density method for the isolation and detection of closed circular DNA is described. The method is based upon the reduced binding of the intercalating dye, ethidium bromide, by closed circular DNA. In an application of this method it is found that HeLa cells contain in addition to closed circular mitochondrial DNA of mean length 4.81 microns, a heterogeneous group of smaller DNA molecules which vary in size from 0.2 to 3.5 microns and a paucidisperse group of multiples of the mitochondrial length.

II. The general theory is presented for the sedimentation equilibrium of a macromolecule in a concentrated binary solvent in the presence of an additional reacting small molecule. Equations are derived for the calculation of the buoyant density of the complex and for the determination of the binding isotherm of the reagent to the macrospecies. The standard buoyant density, a thermodynamic function, is defined and the density gradients which characterize the four component system are derived. The theory is applied to the specific cases of the binding of ethidium bromide to SV 40 DNA and of the binding of mercury and silver to DNA.

Com o objetivo de investigar a influência que o molibdênio exerce nas propriedades das camadas passivas das armaduras de aços austeníticos, barras redondas dos aços UNS S30400 e 31600 foram submetidas à ação de soluções que simulam as que são encontradas nos poros de concreto. Para caracterizar o efeito do molibdênio na resistência daqueles aços à corrosão por pites, foram conduzidos ensaios de circuito aberto, polarização anódica, espectroscopia de impedância eletroquímica, análise de Mott-Schottky, tração em baixa taxa de deformação e análise das superfícies por microscopias óptica e eletrônica de varredura. Os resultados obtidos demonstram que, de uma forma geral, o aço UNS S30400 (sem molibdênio) apresentou maior resistência à corrosão localizada que o UNS S31600 (2% de Mo) nas soluções alcalinas cloretadas naturalmente aeradas, comportamento este inverso ao que se observa nos mesmos materiais quando submetidos a soluções cloretadas neutras ou ácidas.

A generalized acoustic equation is used to identify the mechanisms driving combustion instability. The relationship between the unsteady rate of heat release and the flow is found to influence significantly the frequency of oscillation. A kinematic flame model is reviewed and used to describe the unsteady combustion in a premixed ducted flame and in a typical lean premixed industrial gas turbine. Waxed duplex cartons collected from different prawn freezing factories were evaluated for their physico-chemical properties such as bursting strength, puncture resistance, water proofness, tearing, strength, tensile strength, elongation, moisture content, thickness, weight of the carton, dimension, wax content and saponifiable matter. The results are discussed from the point of view of formulation of standards for this most widely employed packaging material for frozen fishery products in the country.

Deteriorative changes in physical properties of corrugated fibre-board master cartons and waxed duplex cartons during frozen storage under commercial conditions were studied. Such changes due to prolonged exposure of these boards to moisture in the laboratory, effect of repeated wax-coating on the water resisting capacity of the boards and protection provided by increasing wax contents in the boards against water absorption and consequent deterioration in physical properties are reported.

This paper analyzes the forced response of swirl-stabilized lean-premixed flames to high-amplitude acoustic forcing in a laboratory-scale stratified burner operated with CH4 and air at atmospheric pressure. The double-swirler, double-channel annular burner was specially designed to generate high-amplitude acoustic velocity oscillations and a radial equivalence ratio gradient at the inlet of the combustion chamber. Temporal oscillations of equivalence ratio along the axial direction are dissipated over a long distance, and therefore the effects of time-varying fuel/air ratio on the response are not considered in the present investigation. Simultaneous measurements of inlet velocity and heat release rate oscillations were made using a constant temperature anemometer and photomultiplier tubes with narrow-band OH*/CH* interference filters. Time-averaged and phase-synchronized CH* chemiluminescence intensities were measured using an intensified CCD camera. The measurements show that flame stabilization mechanisms vary depending on equivalence ratio gradients for a constant global equivalence ratio (φg=0.60). Under uniformly premixed conditions, an enveloped M-shaped flame is observed. In contrast, under stratified conditions, a dihedral V-flame and a toroidal detached flame develop in the outer stream and inner stream fuel enrichment cases, respectively. The modification of the stabilization mechanism has a significant impact on the nonlinear response of stratified flames to high-amplitude acoustic forcing (u'/U∼0.45 and f=60, 160Hz). Outer stream enrichment tends to improve the flame's stiffness with respect to incident acoustic/vortical disturbances, whereas inner stream stratification tends to enhance the nonlinear flame dynamics, as manifested by the complex interaction between the swirl flame and large-scale coherent vortices with different length scales and shedding points. It was found that the behavior of the measured flame describing functions (FDF), which depend on radial fuel stratification, are well correlated with previous measurements of the intensity of self-excited combustion instabilities in the stratified swirl burner. The results presented in this paper provide insight into the impact of nonuniform reactant stoichiometry on combustion instabilities, its effect on flame location and the interaction with unsteady flow structures. © 2011 The Combustion Institute.
