This paper reviews the literature on fluoride-releasing composite resins. These materials have been available for several years, with fluoride release being achieved by adding soluble or sparingly soluble fluoride salts to the formulation. However, this has been shown to lead to a gradual reduction in the mechanical properties. These materials are also unable to undergo "fluoride recharge". Experimental fluoride-releasing composites have been prepared which supply fluoride by alternative mechanisms that do have the potential for fluoride recharge but, so far, these materials have not been made available for use in patients. Fluoride-releasing composite resins have been shown to be effective in preventing secondary caries in vitro. They have also been shown to reduce the size and depth of carious lesions. However, information on their clinical effectiveness is limited and the paper concludes that there is an urgent need for research on this topic.

It is shown experimentally that subinhibitory concentrations of a number of toxic, or other agents that are typically inhibitory (copper, cadmium, tributyl tin fluoride, reduced salinity), may stimulate the growth of colonies of the hydroid Campanularia flexuosa, exhibiting a phenomenon known as hormesis. It is suggested that the stimulation of growth is not due to the specific properties of the different toxicants, but to an adaptive response of the hydroid to the inhibitory effect that they have in common. Growth is regulated by a control mechanism and it is proposed that the increased growth is a consequence of overcorrections to low levels of an inhibitory challenge. Examination of the toxicological literature shows that hormesis is a more common occurrence that is generally supposed, and it is suggested that the explanation given here might apply in other cases of hormesis.

Cereal grains are the dominant source of cadmium in the human diet, with rice being to the fore. Here we explore the effect of geographic, genetic, and processing (milling) factors on rice grain cadmium and rice consumption rates that lead to dietary variance in cadmium intake. From a survey of 12 countries on four continents, cadmium levels in rice grain were the highest in Bangladesh and Sri Lanka, with both these countries also having high per capita rice intakes. For Bangladesh and Sri Lanka, there was high weekly intake of cadmium from rice, leading to intakes deemed unsafe by international and national regulators. While genetic variance, and to a lesser extent milling, provide strategies for reducing cadmium in rice, caution has to be used, as there is environmental regulation as well as genetic regulation of cadmium accumulation within rice grains. For countries that import rice, grain cadmium can be controlled by where that rice is sourced, but for countries with subsistence rice economies that have high levels of cadmium in rice grain, agronomic and breeding strategies are required to lower grain cadmium.

In a large scale survey of rice grains from markets (13 countries) and fields (6 countries), a total of 1578 rice grain samples were analysed for lead. From the market collected samples, only 0.6% of the samples exceeded the Chinese and EU limit of 0.2 μg g− 1 lead in rice (when excluding samples collected from known contaminated/mine impacted regions). When evaluating the rice grain samples against the Food and Drug Administration's (FDA) provisional total tolerable intake (PTTI) values for children and pregnant women, it was found that only people consuming large quantities of rice were at risk of exceeding the PTTI from rice alone. Furthermore, 6 field experiments were conducted to evaluate the proportion of the variation in lead concentration in rice grains due to genetics. A total of 4 of the 6 field experiments had significant differences between genotypes, but when the genotypes common across all six field sites were assessed, only 4% of the variation was explained by genotype, with 9.5% and 11% of the variation explained by the environment and genotype by environment interaction respectively. Further work is needed to identify the sources of lead contamination in rice, with detailed information obtained on the locations and environments where the rice is sampled, so that specific risk assessments can be performed.

Research into the composition of cereal grains is motivated by increased interest in food quality. Here multi-element analysis is conducted on leaves and grain of the Bala x Azucena rice mapping population grown in the field. Quantitative trait loci (QTLs) for the concentration of 17 elements were detected, revealing 36 QTLs for leaves and 41 for grains. Epistasis was detected for most elements. There was very little correlation between leaf and grain element concentrations. For selenium, lead, phosphorus and magnesium QTLs were detected in the same location for both tissues. In general, there were no major QTL clusters, suggesting separate regulation of each element. QTLs for grain iron, zinc, molybdenum and selenium are potential targets for marker assisted selection to improve seed nutritional quality. An epistatic interaction for grain arsenic also looks promising to decrease the concentration of this carcinogenic element. © Springer Science+Business Media B.V. 2009.

O cádmio (Cd) é um metal não essencial e é considerado um poluente prioritário pela comunidade europeia. Este metal atinge o ambiente no decurso de várias actividades antropogénicas e tende a concentrar-se nos solos e sedimentos, onde está potencialmente disponível para as plantas, sendo posteriormente transferido através da cadeia trófica. Neste contexto, o principal objectivo da presente dissertação foi o estudo dos efeitos da assimilação e da acumulação de Cd em plantas e as suas consequências para animais consumidores. Numa primeira fase, foram estudados os principais efeitos fisiológicos e genotóxicos do Cd em plantas. As plantas de alface (Lactuca sativa L.) expostas a Cd apresentaram um decréscimo na eficiência fotossintética, aumento de peroxidação lipídica e alterações significativas na actividade de enzimas de stress oxidativo. Estas alterações culminaram num decréscimo do crescimento da parte aérea no final da exposição. As respostas obtidas pelos parâmetros bioquímicos sugerem que estes poderão ser utilizados como eventuais biomarcadores em testes ecotoxicológicos com Cd em abordagens integrantes em conjunto com parâmetros clássicos. Os efeitos mutagénicos de Cd foram avaliados através da determinação da instabilidade de microsatélites (IM). Não foi observada IM, nem nas folhas nem nas raízes de plantas de alface com 5 semanas de idade expostas a 100 μM Cd durante 14 dias, no entanto observou-se IM em raízes de alface exposta a 10 μM Cd durante 28 dias desde a germinação. A idade da planta e a maior acumulação de Cd nas raízes poderão explicar os resultados obtidos. A clastogenicidade de Cd foi analisada em três espécies vegetais com diferentes capacidades de destoxificação e acumulação de metais através de citometria de fluxo. Foram detectadas alterações significativas nos parâmetros analisados em raízes alface, mas não nas espécies Thlaspi caerulescens J & C Presl e Thlaspi arvense L. Estes resultados sugerem que o stress provocado pelo Cd originou clastogenicidade como consequência da perda de porções de cromossomas, uma vez que o conteúdo de ADN nuclear diminuiu. A transferência trófica através da cadeia alimentar permanece muito pouco estudada em termos ecotoxicológicos. A distribuição subcellular de metais num organismo pode ser utilizada para compreender a transferência trófica de um metal na cadeia alimentar. Como tal, numa última parte é estudado de que modo a distribuição subcellular do Cd em plantas com perfis de acumulação de Cd distintos afecta a biodisponibilidade e transferência trófica de Cd para isópodes. A distribuição de Cd entre as 4 fracções subcelulares obtidas através de centrifugação diferencial revelou a existência de diferenças significativas entre as espécies de plantas. Estes resultados em conjunto com a avaliação directa da eficiência de assimilação (EA) de Cd individual de cada uma das quatro fracções subcelulares das plantas em estudo, resultou em informação de grande relevância para a explicação das diferenças observadas na EA de Cd por parte de isópodes alimentados com folhas de diferentes espécies de plantas. Com base nos resultados obtidos, o Cd ligado a proteínas estáveis à temperatura (e.g. metaloteoninas e fitoquelatinas) é o menos biodisponível, sendo assim o que menos contribuiu para a transferência trófica, enquanto que o Cd ligado a proteínas desnaturadas pela temperatura foi a fracção mais disponível para transferência trófica de Cd ao isópode. Estes resultados realçam a relevância ecológica da distribuição subcelular de Cd em plantas que tem influência directa na tranferência trófica deste metal para os consumidores e ainda o facto de que alterações na distribuição subcelular de Cd em plantas devido a diferentes mecanismos de destoxificação poderá ter um impacto directo na transferência trófica de Cd para o animal consumidor.

Devido às actividades antropogénicas várias substâncias químicas têm sido introduzidas no meio ambiente em concentrações que de outro modo não ocorreriam de forma tão elevada naturalmente. Assim, o conhecimento acerca das características de um químico, tais como, o potencial para se acumular em diferentes níveis tróficos, a sua mobilidade dentro do ecossistema, a toxicidade específica e a bioacumulação, é fundamental para compreender os seus efeitos nos ecossistemas. Esta tese investiga a influência de especiação, na biodisponibilidade do cádmio (Cd) para o isópode Porcellio dilatatus, incluindo os efeitos de especiação do metal: (i) na assimilação do Cd, (ii) no modo como o Cd se distribui internamente no organismo, e (iii) como a sobrevivência e a reprodução são afectadas em isópodes terrestres. Num primeiro ensaio laboratorial avaliou-se a importância da transferência trófica na assimilação do Cd em P. dilatatus. Para tal analisou-se a eficiência de assimilação (EA) do Cd em isópodes, adicionado superficialmente ao alimento (alface) na forma de Cd(NO3)2 e contaminando o meio de crescimento da alface. A hipótese era de que a alface contaminada biologicamente através do cultivo em meio hidropónico contaminado teria uma maior proporção de complexos com proteína ou conjugado na forma de Cd (ex. Cd cisteína). A EA de Cd foi maior entre os isópodes que foram alimentados com o sal (71%, SE = 7%), do que entre os isópodes que se alimentaram de alface contaminada biologicamente (52%, SE = 5%), demonstrando-se assim num teste laboratorial que é provável que a especiação do Cd influencie a taxa de assimilação e acumulação do Cd. Na experiência alimentar que se seguiu, estudou-se em detalhe a especiação do metal comparando as EA do Cd conjugado com cisteína (Cd(Cys)2) e na forma de Cd(NO3)2, com os quais se contaminou gelatina com alface. A utilização de Cd-cisteína, proporcionou uma forma experimental para explorar a biodisponibilidade do Cd complexado dentro do tecido biológico. Como esperado, a EA de Cd em isópodes alimentados com nitrato de Cd (64%, SE = 5%) foi maior do que no caso de isópodes alimentados com o conjugado de cisteína (20%, SE = 3%). De seguida estudou-se a distribuição subcelular das espécies de Cd assimilado através de um processo de fraccionamento. Supunha-se que as diferenças de especiação de Cd reflectiria diferentes estratégias de compartimentalização, com consequências ao nível da detoxificação, armazenamento celular e distribuição subcelular do metal. O “sequestro” na forma de metal biologicamente detoxificado (BDM = proteínas estáveis ao calor - HSP e grânulos ricos em metal - RMG) foi maior nos isópodes alimentados com Cd(NO3)2, sugerindo que são mais eficientes na detoxificação de Cd (22%) do que quando alimentados com Cd(Cys)2 (15%). Foi também demonstrado que os isópodes alimentados com Cd(Cys)2 possuíam níveis de armazenamento de Cd superior nas fracções sensíveis ao metal (MSF = organelos e proteínas desnaturadas pelo calor - HDP) consideradas fracções potencialmente vulneráveis e afectando os isópodes em termos de toxicidade. As diferentes distribuições internas que se seguiram à assimilação e detoxificação das diferentes espécies de Cd foram finalmente avaliadas em termos da sobrevivência e reprodução dos isópodes. O tratamento com Cd(Cys)2 teve maior mortalidade, provavelmente devido à maior disponibilidade de Cd ingerido com implicações ao nível dos processos fisiológicos. Os isópodes alimentados com Cd(NO3)2 armazenaram o Cd nos MRG, como estratégia de detoxificação, sendo mais eficientes a detoxificar o Cd ainda que aumentando a concentração total do metal que se tornou menos tóxico para o isópode. Desta forma, o Cd nos grânulos não estava disponível para os processos fisiológicos e deixou de ser tóxico. Isso poderia estar relacionado com a resistência e tolerância aos metais devido à capacidade dos isópodes compartimentalizarem o Cd no hepatopâncreas, que actua como um mecanismo de detoxificação e contribui para a tolerância a altos níveis de cádmio. Em termos de parâmetros reprodutivos, observou-se uma redução de gestações e duração da gestação na presença de ambas as espécies de metal, mas no caso do Cd(Cys)2 as gravidezes não se concluíram. O número de jovens produzido por fêmeas alimentadas com Cd(NO3)2 foi menor do que no controlo, mas os pesos dos juvenis foram superiores. Finalmente sugere-se assim que esta abordagem seja considerada em estudos do movimento trófico de metais nas cadeias alimentares dado que se espera que a especiação de metais implique diferentes fluxos, dentro de uma dada cadeia trófica.

Dans les milieux contaminés par les métaux, les organismes vivants sont exposés à plusieurs d’entre eux en même temps. Les modèles courants de prédiction des effets biologiques des métaux sur les organismes (p. ex., modèle du ligand biotique, BLM ; modèle de l’ion libre, FIAM), sont des modèles d’équilibre chimique qui prévoient, en présence d'un deuxième métal, une diminution de la bioaccumulation du métal d’intérêt et par la suite une atténuation de ses effets. Les biomarqueurs de toxicité, tels que les phytochélatines (PCs), ont été utilisés comme étant un moyen alternatif pour l’évaluation des effets biologiques. Les phytochélatines sont des polypeptides riches en cystéine dont la structure générale est (γ-glu-cys)n-Gly où n varie de 2 à 11. Leur synthèse semble dépendante de la concentration des ions métalliques ainsi que de la durée de l’ exposition de l’organisme, aux métaux. L'objectif de cette étude était donc de déterminer, dans les mélanges binaires de métaux, la possibilité de prédiction de la synthèse des phytochélatines par les modèles d’équilibres chimiques, tel que le BLM. Pour cela, la quantité de phytochélatines produites en réponse d’une exposition aux mélanges binaires : Cd-Ca, Cd-Cu et Cd-Pb a été mesurée tout en surveillant l’effet direct de la compétition par le biais des concentrations de métaux internalisés. En effet, après six heures d’exposition, la bioaccumulation de Cd diminue en présence du Ca et de très fortes concentrations de Pb et de Cu (de l’ordre de 5×10-6 M). Par contre, avec des concentrations modérées de ces deux métaux, le Cd augmente en présence de Cu et ne semble pas affecté par la présence de Pb. Dans le cas de la compétition Cd-Cu, une bonne corrélation a été observée entre la production de PC2, PC3 et PC4 et la quantité des métaux bioaccumulés. Pour la synthèse des phytochélatines et la bioaccumulation, les effets étaient considérés comme synergiques. Dans le cas du Cd-Ca, les quantités de PC3 et PC4 ont diminué avec le métal internalisé (effet antagoniste), mais ce qui était remarquable était la grande quantité de cystéine (GSH) et PC2 qui ont été produites à de fortes concentrations du Ca. Le Pb seul n’a pas induit les PCs. Par conséquent, il n’y avait pas de variation de la quantité de PCs avec la concentration de Pb à laquelle les algues ont été exposées. La détection et la quantification des PCs ont été faites par chromatographie à haute performance couplée d’un détecteur de fluorescence (HPLC-FL). Tandis que les concentrations métalliques intracellulaires ont été analysées par spectroscopie d’absorption atomique (AAS) ou par spectrométrie de masse à source plasma à couplage inductif (ICP-MS).

Increasing legislation has steadily been introduced throughout the world to restrict the use of heavy metals, particularly cadmium (Cd) and lead (Pb) in high temperature pigments, ceramics, and optoelectronic material applications. Removal of cadmium from thin-film optical and semiconductor device applications has been hampered by the absence of viable alternatives that exhibit similar properties with stability and durability. We describe a range of tin-based compounds that have been deposited and characterized in terms of their optical and mechanical properties and compare them with existing cadmium-based films that currently find widespread use in the optoelectronic and semiconductor industries. Coatings and filters for spaceflight far-infrared components require a robust, non-absorptive low-index thin film material to contrast with the typically higher refractive index infrared materials. Barium fluoride is one such material for the 10 to 20µm wavelength infrared region, however its optical and mechanical properties vary depending on the process used to deposit it in thin film form. Thin films of dielectric produced by thermal evaporation are well documented as having a lower packing density and refractive index than bulk material. The porous and columnar micro structure of these films causes possible deterioration of their performance in varied environmental conditions, primarily because of the moisture absorption. Dielectric thin films produced by the more novel technique of ion-beam sputtering are denser with no columnar micro structure and have a packing density and refractive index similar to the bulk material. A comparative study of Barium Fluoride (BaF2) thin films made by conventional thermal evaporation and ion-beam sputtering is reported. Films of similar thicknesses are deposited on Cadmium Telluride and Germanium substrates. The optical and mechanical properties of these films are then examined. The refractive index n of the films is obtained from applying the modified Manifacier's evvelope method to the spectral measurements made on a Perkin Elmer 580 spectrophotometer. An estimate is also made of the value of the extinction coefficient k in the infrared wavelength transparent region of the thin film. In order to study the mechanical properties of the BaF2 films, and evaluate their usefulness in spaceflight infrared filters and coatings, the thin film samples are subjected to MIL-F-48616 environmental tests. Comparisons are made of their performance under these tests.

Cadmium (Cd) in air, drinking water and food has the potential to affect the health of people, mainly those who live in highly industrialized regions. Cd affects placental function, can cross the placental barrier and directly modify fetal development. Once the organism is particularly susceptible to the exposition to the Cd during the perinatal period, and that this metal can be excreted in the milk, the aim of the present work was to study the effects of the constant exposition to drinkable water containing low levels of Cd during the lactation, on the salivary glands of the rat. Female rats received ad libitum drinking water containing 300mg/l of CdCl2 throughout the whole lactation. Control animals received a similar volume of water without Cd. Lactant rats (21 day old) were killed by lethal dose of anesthetic. The salivary glands were separated, fixed in ""alfac"" solution for 24 h, and serially sectioned. The 6 mu m thick sections were stained with hematoxylin and eosin. Nuclear glandular parameters were estimated, as well as cytoplasm and cell volume, nucleus/cytoplasm ratio, number and surface density, diameters and cell thickness. Mean body weight was 34.86 g for the control group and 18.56 g for the Cd-treated group. Histologically, the glandular acini were significantly smaller, the gland ducts were similar in both groups studied. The connective tissue was more abundant. In conclusion, the salivary glands (submandibular, parotid and sublingual) showed retarded growth after Cd intoxication.

The south region of Sao Paulo city hosts the Guarapiranga dam, responsible for water supply to 25% of the city population. Their surroundings have been subject to intense and irregular occupation by people from very low socioeconomics classes. Measurements undertaken on sediment and particulate materials in the dam revealed concentrations of lead. copper, zinc and cadmium above internationally accepted limits. Epidemiological and toxicological studies undertaken by the World Health Organization in individuals exhibiting lead concentrations in blood, near or below the maximum recommended (10 mu g dl(-1)), surprisingly revealed that toxic effects are more intense in individuals belonging to low socioeconomics classes. Motivated by these facts, we aimed at the investigation of chronic incorporation of lead. as well as the use of our BIOKINETICS code, which is based on an accepted ICRP biokinetics model for lead, in order to extrapolate the results from teeth to other organs. The focus of our data taking was children from poor families, living in a small, restrict and allegedly contaminated area in Sao Paulo city. Thus, a total of 74 human teeth were collected. The average concentration of lead in teeth of children 5 to 10 years old was determined by means of a high-resolution inductively coupled plasma mass spectrometer (ICP-MS). For standardization of the measurements, an animal bone certified material (H-Animal Bone), from the International Atomic Energy Agency, was analyzed. The amount of lead in children living in the surroundings of the dam, was approximately 40% higher than those from the control region, and the average lead concentration was equal to 1.3 mu g g(-1) approximately. Grouping the results in terms of gender, tooth type and condition, it was concluded that a carious molar of boys is a much more efficient contamination pathway for lead, resulting in concentrations 70% higher than in the control region. The local structure of an ion-conducting glass with nominal composition 50B(2)O(3)-10PbO-40LiF has been investigated by complementary (7)Li, (11)B, (19)F, and (207)Pb single- and double-resonance experiments. The results give insight into the structural role of the lithium fluoride additive in borate glasses: (1) LiF is seen to actively participate in the network transformation process contributing to the conversion of three- into four-coordinate boron units, as shown by (11)B single-resonance as well as by (11)B{(19)F} and (19)F{(11)B} double-resonance experiments. (2) (19)F signal quantification experiments suggest substantial fluoride loss, presumably caused by formation of volatile BF(3). A part of the fluoride remains in the dopant role, possibly in the form of small LiF-like cluster domains, which serve as a mobile ion supply. (3) The extent of lithium-fluorine and lead-fluorine interactions has been characterized by (7)Li{(19)F} and (207)Pb{(19)F} REDOR and SEDOR experiments. On the basis of these results, a quantitative structural description of this system has been developed.

Lead (Pb) poisoning is preventable but continues to be a public health problem in several countries. Measuring Pb in the surface dental enamel (SDE) using microbiopsies is a rapid, safe, and painless procedure. There are different protocols to perform these microbiopsies, but the reliability of dental enamel lead levels (DELL) determination is dependent upon biopsy depth (BD). It is established that DELL decrease from the outermost superficial layer to the inner layer of dental enamel. The aim of this study was to determine DELL obtained by two different microbiopsy techniques on SDE termed protocol I and protocol II. Two consecutive enamel layers were removed from the same subject group (n = 138) for both protocols. Protocol I consisted of a biopsied site with a diameter of 4 mm after the application of 10 l HCl for 35 s. Protocol II involved a biopsied site of 1.6 mm diameter after application of 5 l HCl for 20 s. The results demonstrated that there were no significant differences for BD and DELL between homologous teeth using protocol I. However, there was a significant difference between DELL in the first and second layers using both protocols. Further, the BD in protocol II overestimated DELL values. In conclusion, SDE analyzed by microbiopsy is a reliable biomarker in protocol I, but the chemical method to calculate BD in protocol II appeared to be inadequate for measurement of DELL. Thus, DELL could not be compared among studies that used different methodologies for SDE microbiopsies.
