In the current study we investigated whether ego depletion negatively affects attention regulation under pressure in sports by assessing participants' dart throwing performance and accompanying gaze behavior. According to the strength model of self-control, the most important aspect of self-control is attention regulation. Because higher levels of state anxiety are associated with impaired attention regulation, we chose a mixed design with ego depletion (yes vs. no) as between-subjects and anxiety level (high vs. low) as within-subjects factor. Participants performed a perceptual-motor task requiring selective attention, namely, dart throwing. In line with our expectations, depleted participants in the high-anxiety condition performed worse and displayed a shorter final fixation on bull's eye, demonstrating that when one's self-control strength is depleted, attention regulation under pressure cannot be maintained. This is the first study that directly supports the general assumption that ego depletion is a major factor in influencing attention regulation under pressure.

As higher levels of state anxiety are associated with impaired attention regulation (Nieuwenhuys & Oudejans, 2012) we chose a mixed design with ego depletion (yes vs. no) as between-subjects and anxiety level (high vs. low) as within-subjects factor. A total of 28 right-handed students participated in our study (Mage = 23.4, SDage = 2.5; 10 female; no professional dart experience). Participants performed a perceptual-motor task requiring selective attention, namely, dart throwing. The task was performed while participants were positioned high and low on a climbing wall (i.e., with high and low levels of anxiety). In line with our expectations, a mixed-design ANOVA revealed that depleted participants in the high anxiety condition performed worse (p < .001) and displayed a shorter final fixation on bull’s eye (p < .01) than in the low anxiety condition, demonstrating that when one is depleted attention regulation under pressure cannot be maintained. This is the first study that directly supports the general assumption that ego depletion is a major factor in influencing attention regulation under pressure.

We present the crystal structures of the SEC14-like domain of supernatant protein factor (SPF) in complex with squalene and 2,3-oxidosqualene. The structures were resolved at 1.75 Å (complex with squalene) and 1.6 Å resolution (complex with 2,3-oxidosqualene), leading in both cases to clear images of the protein/ substrate interactions. Ligand binding is facilitated by removal of the Golgi-dynamics (GOLD) C-terminal domain of SPF, which, as shown in previous structures of the apo-protein, blocked the opening of the binding pocket to the exterior. Both substrates bind into a large hydrophobic cavity, typical of such lipid-transporter family. Our structures report no specific recognition mode for the epoxide group. In fact, for both molecules, ligand affinity is dominated by hydrophobic interactions, and independent investigations by computational models or differential scanning micro-calorimetry reveal similar binding affinities for both ligands. Our findings elucidate the molecular bases of the role of SPF in sterol endo-synthesis, supporting the original hypothesis that SPF is a facilitator of substrate flow within the sterol synthetic pathway. Moreover, our results suggest that the GOLD domain acts as a regulator, as its conformational displacement must occur to favor ligand binding and release during the different synthetic steps.

Records of total organic carbon (TOC) and C37 alkenones were used as indicators for past primary productivity in the western and eastern Arabian Sea. Data from GeoB 3005, an open ocean site in the western Arabian Sea upwelling area, are compared with similar records of GeoB 3007 from the Owen Ridge, Ocean Drilling Program (ODP) Site 723 from the continental margin off Oman and MD 900963 from the eastern Arabian Sea. TOC/C37 alkenone records together with other proxies used to reconstruct upwelling intensity, indicate periods of high productivity in tune with precessional forcing all over the Arabian Sea. Based on their phase-relationship to variations in boreal summer insolation they can be divided into three groups. In the western Arabian Sea the precession-related phasing is different between productivity proxies and those for summer monsoon wind strength and upwelling intensity. TOC and C37 alkenone records from the western Arabian Sea lag the other monsoonal indicators by about 5 kyr, but lead productivity indicators from the eastern Arabian Sea by 3 kyr. Based on the differences in phase relationships associated with the precessional cycling between productivity and monsoonal proxies in the western Arabian Sea it is proposed that the TOC/C37 alkenone signal in the western Arabian Sea document a combined signal of moderate SW monsoon winds and of strengthened and prolonged NE monsoon winds. In the eastern Arabian Sea the phasing hints to coincidence between maximum productivity and stronger NE monsoon winds associated with precession-related maxima in ice volume. In contrast, variations in paleoproductivity at site GeoB 3007 from the Owen Ridge indicate productivity maxima during glacial substages 8.2, 6.2 and 2.2, whereas precessionrelated changes are of only minor importance at this location. The results of frequency analyses confirm that productivity at site GeoB 3007 responds predominantly to glacialinterglacial climate changes, while site GeoB 3005 from the open ocean upwelling region near the Gulf of Aden is dominated by precessional insolation. A possible explanation for the pattern revealed at the Owen Ridge is the periodic NW-SE displacement of the Findlater Jet axis, which separates the region of open ocean upwelling to the northwest from downwelling to the southeast ofthe jet. The carbon isotopes of planktic foraminifera reflect nutrient related d13C variations of dissolved inorganic carbon. The difference between the planktic foraminifera Globigerinoides ruber (w), living in the upper 50 m of the water column, and the deeper Iiving Neogloboquadrina dutertrei (Delta d13Cr-d) of core GeoB 3005 displays nutrient variations in the upwelling area near the Gulf of Aden. The results of cross-spectral analyses between Deltad13Cr-d of GeoB 3005 and proxies for SW monsoon intensity indicate, too, a dissociation of productivity from monsoonal upwelling intensity. Instead, productivity depends mainly on the availability of nutrients, while upwelling intensity of sub-surface water masses seems to be of only secondary importance. Additionally, sea surface temperatures (SSTs) were reconstructed using the unsaturation ratio of C37 alkenones. Alkenone SSTs reflect annual mean temperatures rather than explicitly the season of upwelling. This is evident from alkenone SSTs in a transect of surface sediments extending from the inner Gulf of Aden into the western Arabian Sea. The alkenone-derived SST records of GeoB 3005 from the open ocean upwelling region near the Gulf of Aden and GeoB 3007 from the Owen Ridge reveal similar variations with high SSTs during interglacial and low SSTs during glacial periods. The glacial marine oxygen isotope stage (MIS) 6 remains relatively warm and was not as cold as MIS 3 to 4 and 8 according to the alkenone SST. Similar variation-patterns were reconstructed in the coastal upwelling area off Oman for ODP Site 723 as weIl as in the eastern Arabian Sea for MD 900963, where upwelling is not as pronounced as in the western Arabian Sea. Spectral-analyses indicate that SST changes are in good agreement with the modulation of low-latitude precessional insolation changes by eccentricity. However, they do not show the pronounced cydicity in the precessional frequency band, which is characteristic for variations in paleoproductivity. Although the overall variation pattern is very similar, a dose comparison between the western (GeoB 3005) and the eastern Arabian Sea (MD 900963) shows larger differences between both sites during cold intervals than during periods of warm SSTs. This is attributed to a more effective cooling of surface waters in the western Arabian Sea by prolonged NE monsoon winds during times of expanded Northern Hemisphere ice-sheets, thereby lowering the annual mean SSTs stronger than in the eastern Arabian Sea.

A three node, displacement based, acoustic element is developed. In order to avoid spurious rotational modes, a higher order stiffness is introduced. The higher order stiffness is developed from an incompatible strain field which computes element volume changes under nodal rotational displacements fields. The higher order strain satisfies the IET requirements, non affecting convergence. The higher order stiffness is modulated, element by element, with a factor. Thus, the displacement based formulation is capable of placing the spurious rotational modes over the range of physical compressional modes that can be accurately captured by the mesh.

Prevalence of vitamin B12 deficiency is very common in elderly people and can reach values as high as 40.5% of the population. It can be the result of the interaction among several factors. Vitamin B12 deficiencies have been associated with neurological, cognitive deterioration, haematological abnormalities and cardiovascular diseases that have an important influence on the health of the elderly and their quality of life. It is necessary to approach the problems arisen from the lack of data relative to them. The main objective of this thesis was to analyse the evolution of vitamin B12 status and related parameters, lipid and haematological profiles and their relationship to health risk factors, and to functional and cognitive status over one year and to determine the effect of an oral supplementation of 500 μg of cyanocobalamin for a short period of 28 days. An additional objective was to analyze the possible effects of medicine intakes on vitamin B status. Three studies were performed: a) a one year longitudinal follow-up with four measure points; b) an intervention study providing an oral liquid supplement of 500 μg of cyanocobalamin for a 28 days period; and c) analysis of the possible effect of medication intake on vitamin B status using the ATC classification of medicines. The participants for these studies were recruited from nursing homes for the elderly in the Region of Madrid. Sixty elders (mean age 84 _ 7y, 19 men and 41 women) were recruited for Study I and 64 elders (mean age 82 _ 7y, 24 men and 40 women) for Study II. For Study III, baseline data from the initially recruited participants of the first two studies were used. An informed consent was obtained from all participants or their mentors. The studies were approved by the Ethical Committee of the University of Granada. Blood samples were obtained at each examination date and were analyzed for serum cobalamin, holoTC, serum and RBC folate and total homocysteine according to laboratory standard procedures. The haematological parameters analyzed were haematocrit, haemoglobin and MCV. For the lipid profile TG, total cholesterol, LDL- and HDLcholesterol were analyzed. Anthropometric measures (BMI, skinfolds [triceps and subscapular], waist girth and waist to hip ratio), functional tests (hand grip, arm and leg strength tests, static balance) and MMSE were obtained or administered by trained personal. The vitamin B12 supplement of Study II was administered with breakfast and the medication intake was taken from the residents’ anamnesis. Data were analyzed by parametric and non-parametric statistics depending on the obtained data. Comparisons were done using the appropriate ANOVAs or non-parametric tests. Pearsons’ partial correlations with the variable “time” as control were used to define the association of the analyzed parameters. XIII The results showed that: A) Over one year, in relationship to vitamin B status, serum cobalamin decreased, serum folate and mean corpuscular volumen increased significantly and total homocysteine concentrations were stable. Regarding blood lipid profile, triglycerides increased and HDL-cholesterol decreased significantly. Regarding selected anthropometric measurements, waist circumference increased significantly. No significant changes were observed for the rest of parameters. B) Prevalence of hyperhomocysteinemia was high in the elderly studied, ranging from 60% to 90 % over the year depending on the cut-off used for the classification. LDL-cholesterol values were high, especially among women, and showed a tendency to increase over the year. Results of the balance test showed a deficiency and a tendency to decrease; this indicates that the population studied is at high risk for falls. Lower extremity muscular function was deficient and showed a tendency to decrease. A highly significant relationship was observed between the skinfold of the triceps and blood lipid profile. C) Low cobalamin concentrations correlated significantly with low MMSE scores in the elderly studied. No correlations were observed between vitamin B12 status and functional parameters. D) Regarding vitamin B12 status, holo-transcobalamin seems to be more sensitive for diagnosis; 5-10% of the elderly had a deficiency using serum cobalamin as a criterion, and 45-52% had a deficiency when using serum holotranscobalamin as a criterion. E) 500 μg of cyanocobalamin administered orally during 28 days significantly improved vitamin B12 status and significantly decreased total homocysteine concentrations in institutionalized elderly. No effect of the intervention was observed on functional and cognitive parameters. F) The relative change (%) of improvement of vitamin B12 status was higher when using serum holo-transcobalamin as a criterion than serum cobalamin. G) Antiaenemic drug intake normalized cobalamin, urologic drugs and corticosteroids serum folate, and psychoanaleptics holo-transcobalamin levels. Drugs treating pulmonary obstruction increased total homocysteine concentration significantly. H) The daily mean drug intake was 5.1. Fiftynine percent of the elderly took medication belonging to 5 or more different ATC groups. The most prevalent were psycholeptic (53%), antiacid (53%) and antithrombotic (47%) drugs.

Accuracy in the liquid hydrocarbons custody transfer is mandatory because it has a great economic impact. By far the most accurate meter is the positive displacement (PD) meter. Increasing such an accuracy may adversely affect the cost of the custody transfer, unless simple models are developed in order to lower the cost, which is the purpose of this work. PD meter consists of a fixed volume rotating chamber. For each turn a pulse is counted, hence, the measured volume is the number of pulses times the volume of the chamber. It does not coincide with the real volume, so corrections have to be made. All the corrections are grouped by a meter factor. Among corrections highlights the slippage flow. By solving the Navier-Stokes equations one can find an analytical expression for this flow. It is neither easy nor cheap to apply straightforward the slippage correction; therefore we have made a simple model where slippage is regarded as a single parameter with dimension of time. The model has been tested for several PD meters. In our careful experiments, the meter factor grows with temperature at a constant pace of 8?10?5?ºC?1. Be warned

La condición física, o como mejor se la conoce hoy en día el “fitness”, es una variable que está cobrando gran protagonismo, especialmente desde la perspectiva de la salud. La mejora de la calidad de vida que se ha experimentado en los últimos años en las sociedades desarrolladas, conlleva un aumento de la esperanza de vida, lo que hace que cada vez más personas vivan más años. Este rápido crecimiento de la población mayor de 60 años hace que, un grupo poblacional prácticamente olvidado desde el punto de vista de la investigación científica en el campo de la actividad física y del deporte, cobre gran relevancia, con el fin de poder ayudar a alcanzar el dicho “no se trata de aportar años a la vida sino vida a lo años”. La presente memoria de Tesis Doctoral tiene como principal objetivo valorar los niveles de fitness en población mayor española, además de analizar la relación existente entre el fitness, sus condicionantes y otros aspectos de la salud, tales como la composición corporal y el estado cognitivo. Entendemos que para poder establecer futuras políticas de salud pública en relación a la actividad física y el envejecimiento activo es necesario conocer cuáles son los niveles de partida de la población mayor en España y sus condicionantes. El trabajo está basado en los datos del estudio multicéntrico EXERNET (Estudio Multi-céntrico para la Evaluación de los Niveles de Condición Física y su relación con Estilos de Vida Saludables en población mayor española no institucionalizada), así como en los datos de dos estudios, llevados a cabo en población mayor institucionalizada. Se han analizado un total de 3136 mayores de vida independiente, procedentes de 6 comunidades autónomas, y 153 mayores institucionalizados en residencias de la Comunidad de Madrid. Los principales resultados de esta tesis son los siguientes: a) Fueron establecidos los valores de referencia, así como las curvas de percentiles, para cada uno de los test de fitness, de acuerdo a la edad y al sexo, en población mayor española de vida independiente y no institucionalizada. b) Los varones obtuvieron mejores niveles de fitness que las mujeres, excepto en los test de flexibilidad; existe una tendencia a disminuir la condición física en ambos sexos a medida que la edad aumenta. c) Niveles bajos de fitness funcional fueron asociados con un aumento en la percepción de problemas. d) El nivel mínimo de fitness funcional a partir del cual los mayores perciben problemas en sus actividades de la vida diaria (AVD) es similar en ambos sexos. e) Niveles elevados de fitness fueron asociados con un menor riesgo de sufrir obesidad sarcopénica y con una mejor salud percibida en los mayores. f) Las personas mayores con obesidad sarcopénica tienen menor capacidad funcional que las personas mayores sanas. g) Niveles elevados de fuerza fueron asociados con un mejor estado cognitivo siendo el estado cognitivo la variable que más influye en el deterioro de la fuerza, incluso más que el sexo y la edad. ABSTRACT Fitness is a variable that is gaining in prominence, especially from the health perspective. Improvement of life quality that has been experienced in the last few years in developed countries, leads to an expanded life expectancy, increasing the numbers of people living longer. This population consisting of people of over 60 years, an almost forgotten population group from the point of view of scientific research in the field of physical activity and sport, is becoming increasingly important, with the main aim of helping to achieve the saying “do not only add years to life, but also add life to years”. The principal aim of the current thesis was to assess physical fitness levels in Spanish elderly people, of over 65 years, analyzing relationship between physical fitness, its determinants, and other aspects of health such as body composition and cognitive status. In order to establish further public health policies in relation to physical activity and active ageing it is necessary to identify the starting physical fitness levels of the Spanish population and their determinants. The work is based on data from the EXERNET multi-center study ("Multi-center Study for the Evaluation of Fitness levels and their relationship to Healthy Lifestyles in noninstitutionalized Spanish elderly"), and on data from two studies conducted in institutionalized elderly people: a total of 3136 non-institutionalized elderly, from 6 Regions of Spain, and 153 institutionalized elderly in nursing homes of Madrid. The main outcomes of this thesis are: a) sex- and age-specific physical fitness normative values and percentile curves for independent and non-institutionalized Spanish elderly were established. b) Greater physical fitness was present in the elderly men than in women, except for the flexibility test, and a trend toward decreased physical fitness in both sexes as their age increased. c) Lower levels of functional fitness were associated with increased perceived problems. d) The minimum functional fitness level at which older adults perceive problems in their ADLs, is similar for both sexes e) Higher levels of physical fitness were associated with a reduced risk of suffering sarcopenic obesity and better perceived health among the elderly. f) The elderly with sarcopenic obesity have lower physical functioning than healthy counterparts. g) Higher strength values were associated with better cognitive status with cognitive status being the most influencing variable in strength deterioration even more than sex and age.

The monocarboxylate transporter (MCT) family member MCT1 transports lactate into and out of myocytes. Oxidative cells import lactate through MCT1 as a substrate, being the role of MCT1 in glycolysis-derived lactate efflux less clear. MCT1 T1470A polymorphism (rs1049434), which has been related with lactate metabolism and sports specialty 1, 2, could be an influencing factor for exercise adherence. Therefore the aim of this study was to relate the adherence to different training modalities with the T1470A MCT1 polymorphism in overweight and obese men following a weight loss program (WLP).

En este trabajo se aborda una cuestión central en el diseño en carga última de estructuras de hormigón armado y de fábrica: la posibilidad efectiva de que las deformaciones plásticas necesarias para verificar un estado de rotura puedan ser alcanzadas por las regiones de la estructura que deban desarrollar su capacidad última para verificar tal estado. Así, se parte de las decisiones de diseño que mediante mera estática aseguran un equilibrio de la estructura para las cargas últimas que deba resistir, pero determinando directamente el valor de las deformaciones necesarias para llegar a tal estado. Por tanto, no se acude a los teoremas de rotura sin más, sino que se formula el problema desde un punto de vista elastoplástico. Es decir, no se obvia el recorrido que la estructura deba realizar en un proceso de carga incremental monótono, de modo que las regiones no plastificadas contribuyen a coaccionar las libres deformaciones plásticas que, en la teoría de rotura, se suponen. En términos de trabajo y energía, se introduce en el balance del trabajo de las fuerzas externas y en el de la energía de deformación, aquella parte del sistema que no ha plastificado. Establecido así el balance energético como potencial del sistema es cuando la condición de estacionariedad del mismo hace determinados los campos de desplazamientos y, por tanto, el de las deformaciones plásticas también. En definitiva, se trata de un modo de verificar si la ductilidad de los diseños previstos es suficiente, y en qué medida, para verificar el estado de rotura previsto, para unas determinadas cargas impuestas. Dentro del desarrollo teórico del problema, se encuentran ciertas precisiones importantes. Entre ellas, la verificación de que el estado de rotura a que se llega de manera determinada mediante el balance energético elasto-plástico satisface las condiciones de la solución de rotura que los teoremas de carga última predicen, asegurando, por tanto, que la solución determinada -unicidad del problema elásticocoincide con el teorema de unicidad de la carga de rotura, acotando además cuál es el sistema de equilibrio y cuál es la deformada de colapso, aspectos que los teoremas de rotura no pueden asegurar, sino sólo el valor de la carga última a verificar. Otra precisión se basa en la particularidad de los casos en que el sistema presenta una superficie de rotura plana, haciendo infinitas las posibilidades de equilibrio para una misma deformada de colapso determinada, lo que está en la base de, aparentemente, poder plastificar a antojo en vigas y arcos. Desde el planteamiento anterior, se encuentra entonces que existe una condición inherente a cualquier sistema, definidas unas leyes constitutivas internas, que permite al mismo llegar al inicio del estado de rotura sin demandar deformación plástica alguna, produciéndose la plastificación simultánea de todas las regiones que hayan llegado a su solicitación de rotura. En cierto modo, se daría un colapso de apariencia frágil. En tal caso, el sistema conserva plenamente hasta el final su capacidad dúctil y tal estado actúa como representante canónico de cualquier otra solución de equilibrio que con idéntico criterio de diseño interno se prevea para tal estructura. En la medida que el diseño se acerque o aleje de la solución canónica, la demanda de ductilidad del sistema para verificar la carga última será menor o mayor. Las soluciones que se aparten en exceso de la solución canónica, no verificarán el estado de rotura previsto por falta de ductilidad: la demanda de deformación plástica de alguna región plastificada estará más allá de la capacidad de la misma, revelándose una carga de rotura por falta de ductilidad menor que la que se preveía por mero equilibrio. Para la determinación de las deformaciones plásticas de las rótulas, se ha tomado un modelo formulado mediante el Método de los Elementos de Contorno, que proporciona un campo continuo de desplazamientos -y, por ende, de deformaciones y de tensiones- incluso en presencia de fisuras en el contorno. Importante cuestión es que se formula la diferencia, nada desdeñable, de la capacidad de rotación plástica de las secciones de hormigón armado en presencia de cortante y en su ausencia. Para las rótulas de fábrica, la diferencia se establece para las condiciones de la excentricidad -asociadas al valor relativo de la compresión-, donde las diferencias entres las regiones plastificadas con esfuerzo normal relativo alto o bajo son reseñables. Por otro lado, si bien de manera un tanto secundaria, las condiciones de servicio también imponen un límite al diseño previo en carga última deseado. La plastificación lleva asociadas deformaciones considerables, sean locales como globales. Tal cosa impone que, en estado de servicio, si la plastificación de alguna región lleva asociadas fisuraciones excesivas para el ambiente del entorno, la solución sea inviable por ello. Asimismo, las deformaciones de las estructuras suponen un límite severo a las posibilidades de su diseño. Especialmente en edificación, las deformaciones activas son un factor crítico a la hora de decidirse por una u otra solución. Por tanto, al límite que se impone por razón de ductilidad, se debe añadir el que se imponga por razón de las condiciones de servicio. Del modo anterior, considerando las condiciones de ductilidad y de servicio en cada caso, se puede tasar cada decisión de diseño con la previsión de cuáles serán las consecuencias en su estado de carga última y de servicio. Es decir, conocidos los límites, podemos acotar cuáles son los diseños a priori que podrán satisfacer seguro las condiciones de ductilidad y de servicio previstas, y en qué medida. Y, en caso de no poderse satisfacer, qué correcciones debieran realizarse sobre el diseño previo para poderlas cumplir. Por último, de las consecuencias que se extraen de lo estudiado, se proponen ciertas líneas de estudio y de experimentación para poder llegar a completar o expandir de manera práctica los resultados obtenidos. ABSTRACT This work deals with a main issue for the ultimate load design in reinforced concrete and masonry structures: the actual possibility that needed yield strains to reach a ultimate state could be reached by yielded regions on the structure that should develop their ultimate capacity to fulfill such a state. Thus, some statically determined design decisions are posed as a start for prescribed ultimate loads to be counteracted, but finding out the determined value of the strains needed to reach the ultimate load state. Therefore, ultimate load theorems are not taken as they are, but a full elasto-plastic formulation point of view is used. As a result, the path the structure must develop in a monotonus increasing loading procedure is not neglected, leading to the fact that non yielded regions will restrict the supposed totally free yield strains under a pure ultimate load theory. In work and energy terms, in the overall account of external forces work and internal strain energy, those domains in the body not reaching their ultimate state are considered. Once thus established the energy balance of the system as its potential, by imposing on it the stationary condition, both displacements and yield strains appear as determined values. Consequently, what proposed is a means for verifying whether the ductility of prescribed designs is enough and the extent to which they are so, for known imposed loads. On the way for the theoretical development of the proposal, some important aspects have been found. Among these, the verification that the conditions for the ultimate state reached under the elastoplastic energy balance fulfills the conditions prescribed for the ultimate load state predicted through the ultimate load theorems, assuring, therefore, that the determinate solution -unicity of the elastic problemcoincides with the unicity ultimate load theorem, determining as well which equilibrium system and which collapse shape are linked to it, being these two last aspects unaffordable by the ultimate load theorems, that make sure only which is the value of the ultimate load leading to collapse. Another aspect is based on the particular case in which the yield surface of the system is flat -i.e. expressed under a linear expression-, turning out infinite the equilibrium possibilities for one determined collapse shape, which is the basis of, apparently, deciding at own free will the yield distribution in beams and arches. From the foresaid approach, is then found that there is an inherent condition in any system, once defined internal constitutive laws, which allows it arrive at the beginning of the ultimate state or collapse without any yield strain demand, reaching the collapse simultaneously for all regions that have come to their ultimate strength. In a certain way, it would appear to be a fragile collapse. In such a case case, the system fully keeps until the end its ductility, and such a state acts as a canonical representative of any other statically determined solution having the same internal design criteria that could be posed for the that same structure. The extent to which a design is closer to or farther from the canonical solution, the ductility demand of the system to verify the ultimate load will be higher or lower. The solutions being far in excess from the canonical solution, will not verify the ultimate state due to lack of ductility: the demand for yield strains of any yielded region will be beyond its capacity, and a shortcoming ultimate load by lack of ductility will appear, lower than the expected by mere equilibrium. For determining the yield strains of plastic hinges, a Boundary Element Method based model has been used, leading to a continuous displacement field -therefore, for strains and stresses as well- even if cracks on the boundary are present. An important aspect is that a remarkable difference is found in the rotation capacity between plastic hinges in reinforced concrete with or without shear. For masonry hinges, such difference appears when dealing with the eccentricity of axial forces -related to their relative value of compression- on the section, where differences between yield regions under high or low relative compressions are remarkable. On the other hand, although in a certain secondary manner, serviceability conditions impose limits to the previous ultimate load stated wanted too. Yield means always big strains and deformations, locally and globally. Such a thing imposes, for serviceability states, that if a yielded region is associated with too large cracking for the environmental conditions, the predicted design will be unsuitable due to this. Furthermore, displacements must be restricted under certain severe limits that restrain the possibilities for a free design. Especially in building structures, active displacements are a critical factor when chosing one or another solution. Then, to the limits due to ductility reasons, other limits dealing with serviceability conditions shoud be added. In the foresaid way, both considering ductility and serviceability conditions in every case, the results for ultimate load and serviceability to which every design decision will lead can be bounded. This means that, once the limits are known, it is possible to bound which a priori designs will fulfill for sure the prescribed ductility and serviceability conditions, and the extent to wich they will be fulfilled, And, in case they were not, which corrections must be performed in the previous design so that it will. Finally, from the consequences derived through what studied, several study and experimental fields are proposed, in order to achieve a completeness and practical expansion of the obtained results.

To investigate the potential role of tenascin-C (TN-C) on endothelial sprouting we used bovine aortic endothelial cells (BAECs) as an in vitro model of angiogenesis. We found that TN-C is specifically expressed by sprouting and cord-forming BAECs but not by nonsprouting BAECs. To test whether TN-C alone or in combination with basic fibroblast growth factor (bFGF) can enhance endothelial sprouting or cord formation, we used BAECs that normally do not sprout and, fittingly, do not express TN-C. In the presence of bFGF, exogenous TN-C but not fibronectin induced an elongated phenotype in nonsprouting BAECs. This phenotype was due to altered actin cytoskeleton organization. The fibrinogen globe of the TN-C molecule was the active domain promoting the elongated phenotype in response to bFGF. Furthermore, we found that the fibrinogen globe was responsible for reduced cell adhesion of BAECs on TN-C substrates. We conclude that bFGF-stimulated endothelial cells can be switched to a sprouting phenotype by the decreased adhesive strength of TN-C, mediated by the fibrinogen globe.

We analyzed the distribution of the cauliflower mosaic virus (CaMV) aphid transmission factor (ATF), produced via a baculovirus recombinant, within Sf9 insect cells. Immunogold labeling revealed that the ATF colocalizes with an atypical cytoskeletal network. Detailed observation by electron microscopy demonstrated that this network was composed of microtubules decorated with paracrystalline formations, characteristic of the CaMV ATF. A derivative mutant of the ATF, unable to self-assemble into paracrystals, was also analyzed. This mutant formed a net-like structure, with a mesh of four nanometers, tightly sheathing microtubules. Both the ATF– and the derivative mutant–microtubule complexes were highly stable. They resisted dilution-, cold-, and calcium-induced microtubule disassembly as well as a combination of all three for over 6 hr. CaMV ATF cosedimented with microtubules and, surprisingly, it bound to Taxol-stabilized microtubules at high ionic strength, thus suggesting an atypical interaction when compared with that usually described for microtubule-binding proteins. Using immunofluorescence double labeling we also demonstrated that the CaMV ATF colocalizes with the microtubule network when expressed in plant cells.

During the aging process, mammals lose up to a third of their skeletal muscle mass and strength. Although the mechanisms underlying this loss are not entirely understood, we attempted to moderate the loss by increasing the regenerative capacity of muscle. This involved the injection of a recombinant adeno-associated virus directing overexpression of insulin-like growth factor I (IGF-I) in differentiated muscle fibers. We demonstrate that the IGF-I expression promotes an average increase of 15% in muscle mass and a 14% increase in strength in young adult mice, and remarkably, prevents aging-related muscle changes in old adult mice, resulting in a 27% increase in strength as compared with uninjected old muscles. Muscle mass and fiber type distributions were maintained at levels similar to those in young adults. We propose that these effects are primarily due to stimulation of muscle regeneration via the activation of satellite cells by IGF-I. This supports the hypothesis that the primary cause of aging-related impairment of muscle function is a cumulative failure to repair damage sustained during muscle utilization. Our results suggest that gene transfer of IGF-I into muscle could form the basis of a human gene therapy for preventing the loss of muscle function associated with aging and may be of benefit in diseases where the rate of damage to skeletal muscle is accelerated.

Studies were conducted to identify a 64-kD thylakoid membrane protein of unknown function. The protein was extracted from chloroplast thylakoids under low ionic strength conditions and purified to homogeneity by preparative sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Four peptides generated from the proteolytic cleavage of the wheat 64-kD protein were sequenced and found to be identical to internal sequences of the chloroplast-coupling factor (CF1) α-subunit. Antibodies for the 64-kD protein also recognized the α-subunit of CF1. Both the 64-kD protein and the 61-kD CF1 α-subunit were present in the monocots barley (Hordeum vulgare), maize (Zea mays), oat (Avena sativa), and wheat (Triticum aestivum); but the dicots pea (Pisum sativum), soybean (Glycine max Merr.), and spinach (Spinacia oleracea) contained only a single polypeptide corresponding to the CF1 α-subunit. The 64-kD protein accumulated in response to high irradiance (1000 μmol photons m−2 s−1) and declined in response to low irradiance (80 μmol photons m−2 s−1) treatments. Thus, the 64-kD protein was identified as an irradiance-dependent isoform of the CF1 α-subunit found only in monocots. Analysis of purified CF1 complexes showed that the 64-kD protein represented up to 15% of the total CF1 α-subunit.
