The goal of this trial was to estimate the total dry matter (TDMI) and daily pasture dry matter intakes (PDMI) by lactating crossbred Holstein - Zebu cows grazing elephant grass (Pennisetum purpureum Schum.) paddocks submitted to different rest periods. Three groups of 24 cows were used during two years. The paddocks were grazed during three days at the stocking rate of 4.5 cows/ha. Treatments consisted of resting periods of 30 days without concentrate and resting periods of 30, 37.5 and 45 days with 2 kg/cow/day of 20.6% crude protein concentrate. From July to October, pasture was supplemented with chopped sugarcane plus 1% urea. Total daily dry matter intake was estimated using the extrusa in vitro dry matter digestibility and the fecal output with chromium oxide. Regardless of the treatment the estimated average TDMI was 2.7, 2.9 and 2.9±0.03% and the mean PDMI was 1.9, 2.1 and 2.1±0.03% of body weight in the first, second and third grazing day, respectively (P<0.05). Only during the summer pasture quality was the same whichever the grazing day. Sugarcane effectively replaced grazing pasture, mainly in the first day when pasture dry matter intake was lowest.

Community-level patterns of functional traits relate to community assembly and ecosystem functioning. By modelling the changes of different indices describing such patterns - trait means, extremes and diversity in communities - as a function of abiotic gradients, we could understand their drivers and build projections of the impact of global change on the functional components of biodiversity. We used five plant functional traits (vegetative height, specific leaf area, leaf dry matter content, leaf nitrogen content and seed mass) and non-woody vegetation plots to model several indices depicting community-level patterns of functional traits from a set of abiotic environmental variables (topographic, climatic and edaphic) over contrasting environmental conditions in a mountainous landscape. We performed a variation partitioning analysis to assess the relative importance of these variables for predicting patterns of functional traits in communities, and projected the best models under several climate change scenarios to examine future potential changes in vegetation functional properties. Not all indices of trait patterns within communities could be modelled with the same level of accuracy: the models for mean and extreme values of functional traits provided substantially better predictive accuracy than the models calibrated for diversity indices. Topographic and climatic factors were more important predictors of functional trait patterns within communities than edaphic predictors. Overall, model projections forecast an increase in mean vegetation height and in mean specific leaf area following climate warming. This trend was important at mid elevation particularly between 1000 and 2000 m asl. With this study we showed that topographic, climatic and edaphic variables can successfully model descriptors of community-level patterns of plant functional traits such as mean and extreme trait values. However, which factors determine the diversity of functional traits in plant communities remains unclear and requires more investigations.

The objective of this work was to evaluate the effect of drought on genetic parameters and breeding values of cassava. The experiments were carried out in a completely randomized block design with three replicates, under field conditions with (WD) or without (FI) water deficit. Yield of storage roots (RoY), shoot (ShY), and starch (StY), as well as the number of roots (NR), and root dry matter content (DMC) were evaluated in 47 cassava accessions. Significant differences were observed among accessions; according to heritability, these differences had mostly a genetic nature. Heritability estimates for genotypic effects () ranged from 0.25±0.12 (NR) to 0.60±0.18 (DMC), and from 0.51±0.17 (NR) to 0.80±0.21 (RoY and StY) for WD and FI, respectively, as a consequence of greater environmental influence on WD. Selective accuracy was lower in WD, and ranged from 0.71 (NR) to 0.89 (RoY, DMC, and StY). However, genetic gains were quite high and ranged from 24.43% (DMC) to 113.41% (StY), in WD, and from 8.5% (DMC) to 75.70% (StY) in FI. These genetic parameters may be useful for defining which selection strategies, breeding methods, and experimental designs are more suitable to obtain cassava genetic gains for tolerance to drought.

The effects of 0, 30 and 60 mM NaCl and substrates (red peat, sand or 3:1:1 [w/w] mixture of peat, sand, or soil) on vegetative growth of lulo, an Andean fruit species, during 12 weeks were studied. The experiment was carried out by using 2000 cm³ of polypropylene plastic pots under greenhouse conditions. Plant height, number of leaves and nodes, leaf area, total plant dry matter (DM), and shoot/root ratio were evaluated. With the increase of salt concentration, the plant height, the number of leaves and nodes, the leaf areas and plant dry mass DM decreased, whereas shoot/root ratio increased. Sand grown lulo plants were most affected by salinity and presented total mortality at 60 mM NaCl. On the other hand, plants held either in peat or in substrate mixture developed larger height, greater leaf and node numbers, higher leaf area and dry matter content. Shoot/root ratio in control (soil) and sand-grown plants (30 mM NaCl) was lower.

Työn tavoitteena oli selvittää koivun kasteluvarastoinnin kannattavuus selluteollisuudessa. Lisäksi tutkittiin, kuinka kastelu vaikuttaa puuaineeseen varastoinnin aikana ja kuinka koivun kasteluvarastointi vaikuttaa puun kuorintaan ja haketukseen, keitettävyyteen, vaalenevuuteen sekä sellun laatuun. Enocellin puukentälle rakennettiin kasteluvarasto, jossa varastoitiin 40,000 m3sob koivua. Kastelu oli päällä huhtikuusta lokakuuhun asti. Kastelun vaikutusta puuaineen muutoksiin arvioitiin lahotutkimusten avulla. Tehdaskoeajoissa verrattiin tuoretta, kasteluvarastoitua ja kuivavarastoitua koivua. Puuaines säilyi lähes muuttumattomana yhden kesän kasteluvarastoinnissa. Kastellulla koivulla terveen puun osuus oli yli 85 % kesän lopussa, kun se oli alle 20 % kuivavarastoidulla koivulla. Kuorinnan puuhäviö laskee selvästi kastelukoivulla ja myös hakkeen laatu oli parempaa kuin kuivavarastoidulla koivulla. Kastelukoivulla hakkeen kuoripitoisuus oli vain 0.13 %. Kuoren kuiva-aine oli 12 prosenttiyksikköä alhaisempi kuin kuivalla koivulla, mutta kuoren lämpöarvossa ero oli vain 1 €/ADt. Varastointimenetelmällä ei ollut vaikutusta hakkeen keitettävyyteen, mutta tuoreella puulla keitettävyys oli parempi kuin varastoidulla puulla. Sellun asetoniuutepitoisuus oli samalla tasolla tuoreella ja kastellulla puulla. Kuivalla syyspuulla uutetaso oli korkeampi, vaikka hartsisaippuan annostusta nostettiin 10 kg/ADt. Betulinolitaso oli kastellulla puulla erittäin alhainen puun hyvän kuoriutuvuuden vuoksi. Kastellun ja tuoreen puun vaalenevuus oli parempi kuin kuivalla puulla. Aktiivikloorin kulutus oli 3 – 4 kg/ADt alhaisempi kuin kuivalla syyspuulla. Puun varastoinnilla ei ollut vaikutusta sellun laatuun. Koivun kasteluvarastoinnin kannattavuus on erittäin hyvä. Tuotantokustannukset määritettiin tuoreelle, kastellulle, kierrätetylle sekä kuivalle koivulle. Kasteluvarastointi laskee tuotantokustannuksia noin 10 €/ADt verrattuna kierrätettyyn koivuun. Kuivavarastoidun puun käyttö nostaa tuotantokustannuksia noin 5 €/ADt verrattuna kastelukoivuun. Kierrätetyn ja kuivavarastoidun puun kustannusero johtuu kierrätyskustannuksista. Kasteluvarastolle, jota käytettiin kesällä 2004, takaisinmaksuaika on vain 0.4 vuotta. Jos tavoiteltu takaisinmaksuaika olisi kaksi vuotta, niin perusinvestointi 80,000 m3sob varastolle voisi maksaa noin 370 k€.

Various strength properties of paper are measured to tell how well it resists breaks in a paper machine or in printing presses. The most often measured properties are dry tensile strength and dry tear strength. However, in many situations where paper breaks, it is not dry. For example, in web breaks after the wet pressing the dry matter content can be around 45%. Thus, wet-web strength is often a more critical paper property than dry strength. Both wet and dry strength properties of the samples were measured with a L&W tensile tester. Originally this device was not designed for the measurement of the wet web tensile strength, thus a new procedure to handle the wet samples was developed. The method was tested with Pine Kraft (never dried). The effect of different strength additives on the wet-web and dry paper tensile strength was studied. The polymers used in this experiment were aqueous solution of a cationic polyamidoamine-epichlorohydrin resin (PAE), cationic hydrophilised polyisocyanate and cationic polyvinylamine (PVAm). From all three used chemicals only Cationic PAE considerably increased the wet web strength. However it was noticed that at constant solids content all chemicals decreased the wet web tensile strength. So, since all chemicals enhanced solid content it can be concluded that they work as drainage aids, not as wet web strength additives. From all chemicals only PVAm increased the dry strength and two other chemicals even decreased the strength. As chemicals were used in strong diluted forms and were injected into the pulp slurry, not on the surface of the papersheets, changes in samples densities did not happen. Also it has to be noted that all these chemicals are mainly used to improve the wet strength after the drying of the web.

Tutkimuksen tavoitteena oli kartoittaa alueellisen jäteyhtiön Kymenlaakson Jäte Oy:n mahdollisuuksia rakeistaa ja termisesti kuivata mekaanisesti kuivattua mädätysjäännöstä sekä mahdollisuuksia toimittaa termisesti kuivattua materiaalia energiahyötykäyttöön. Tutkimuksessa selvitettiin myös kokemuksia lattialämmityksen käyttämisestä mädätysjäännöksen kuivaukseen. Tutkimuksessa perehdyttiin erilaisiin rakeistus- ja kuivausmenetelmiin sekä termisen kuivurin valintaan vaikuttaviin asioihin. Kuvaukset perustuvat kirjallisuudesta ja internetistä saatuihin tietoihin. Tekniikkakuvausten pohjalta lähdettiin kyselemään tarjouksia termisiä kuivauslaitteistoja myyviltä yrityksiltä. Tarjoukset pyydettiin kuiva-ainepitoisuuden muutokselle 30 %:sta 90 %:iin ja oletettiin, että kuivaukseen on käytettävissä lämpöä viideltä kaatopaikkakaasua käyttävältä mikroturbiinilta. Tutkimuksen aikana saatiin tarjous kuudelta yritykseltä. Saadut tarjoukset esiteltiin tiivistetysti raportissa ja kokonaisuudessaan ne sisällytettiin Kymenlaakson Jäte Oy:n laajempaan raporttiin, joka ei ole julkinen. Yritykset antoivat hyvin erilaisia tietoja siitä, mitä tarjoukseen sisältyy, joten tarjoukset eivät olleet suoraan vertailukelpoisia. Tarjouksista myös havaittiin, että jos Mäkikylän biokaasulaitokselta vastaanotettaisiin enimmäismäärä (19 500 t/a) mädätysjäännöstä, mikroturbiineilta saatava lämpömäärä ei riittäisi kuivaamaan kaikkea mädätysjäännöstä 90 % kuiva-ainepitoisuuteen. Tutkimuksen aikana huomattiin myös, että sitovan tarjouksen saamiseksi mädätysjäännös tulee toimittaa testattavaksi, jolloin saadaan vahvistus kuivausmenetelmän soveltuvuudesta kyseiselle materiaalille. Tutkimuksessa selvitettiin myös, minkälaisia kokemuksia löytyy lattialämmityksen käyttämisestä kuivaukseen niin Suomesta kuin maailmalta ja voiko menetelmää käyttää mädätysjäännöksen kuivaukseen. Kyseistä menetelmää on käytetty tehostamaan aurinkokuivausta, joten tutkimuksen aikana perehdyttiin erityisesti aurinkokuivaukseen liittyviin tieteellisiin artikkeleihin. Lattialämmityksen käytöstä löytyi niin heikkouksia kuin vahvuuksia. Suomessa aurinkokuivauksen ja lattialämmityksen yhdistelmä ei ole kuitenkaan päätynyt laajaan käyttöön ja syynä voidaan nähdä muun muassa kylmät ja pimeät vuodenajat sekä suuri pinta-alan tarve. Tutkimusraportissa selvitettiin lisäksi polttolaitosten edustajien kiinnostusta ja rajoituksia ottaa vastaan termisesti kuivattua mädätysjäännöstä. Tutkimuksen aikana otettiin yhteyttä alle 100 km etäisyydellä Kymenlaakson Jäte Oy:stä sijaitsevien jätteenpolttoluvan omaavien yritysten edustajiin. Saatuja vastauksia käsiteltiin tiivistetysti raportissa ja vastaukset sisällytettiin kokonaisuudessaan Kymenlaakson Jäte Oy:n laajempaan raporttiin, joka ei ole julkinen. Puhelinhaastattelujen pohjalta nähtiin, että yrityksillä on kiinnostusta materiaalia kohtaan, mutta samalla vastauksiin vaikuttavat mädätysjäännöksen analyysitulokset. Poltto-ominaisuuksiin liittyvät analyysit tullaan toteuttamaan vuoden 2012 aikana. Laitoksilla oli myös vaihtelevia rajoituksia materiaalia kohtaan, mutta analyysituloksista riippuen materiaalia voidaan hyödyntää energiana tuhansia tai jopa kymmeniä tuhansia tonneja vuodessa alle 100 km etäisyydellä Kymenlaakson Jäte Oy:stä.

Competition between maize and signalgrass can economically cripple the intercropping by the reduced yield of maize and dry matter content of the forage. In seeking to define plant arrangements which make this system more efficient, this research was held with the objective of assessing the effects of interference of densities of signalgrass (Urochloa Brizantha) on nutrition and on maize grain yield. Two field experiments were conducted in a randomized block design with four replications. Treatments were arranged similarly in both experiments, in a 2 x 4 factorial design, the first factor being the dose of Nicosulfuron herbicide applied (0 and 8 g ha-1) and the second factor being the forage seeding rates (0, 2, 4 and 6 kg of seeds per hectare). The interference of signalgrass reduced foliar nitrogen, potassium and phosphorus content in maize plants intercropped with the forage. Higher values of grain yield were observed with the reduction of the spacing and the application of the recommended herbicide underdose (8 g ha-1). It was concluded that, regardless of the seeding density of U. Brizantha, reducing the maize seeding inter-rows spacing, combined with the application of an underdose of Nicosulfuron, caused a positive effect by reducing the initial forage growth, resulting in less interference of Urochloa brizantha on nutrient uptake by the maize plants and grain yield of the crop.

In recent years, there have been studies that show a correlation between the hyperactivity of children and use of artificial food additives, including colorants. This has, in part, led to preference of natural products over products with artificial additives. Consumers have also become more aware of health issues. Natural food colorants have many bioactive functions, mainly vitamin A activity of carotenoids and antioxidativity, and therefore they could be more easily accepted by the consumers. However, natural colorant compounds are usually unstable, which restricts their usage. Microencapsulation could be one way to enhance the stability of natural colorant compounds and thus enable better usage for them as food colorants. Microencapsulation is a term used for processes in which the active material is totally enveloped in a coating or capsule, and thus it is separated and protected from the surrounding environment. In addition to protection by the capsule, microencapsulation can also be used to modify solubility and other properties of the encapsulated material, for example, to incorporate fat-soluble compounds into aqueous matrices. The aim of this thesis work was to study the stability of two natural pigments, lutein (carotenoid) and betanin (betalain), and to determine possible ways to enhance their stability with different microencapsulation techniques. Another aim was the extraction of pigments without the use of organic solvents and the development of previously used extraction methods. Stability of pigments in microencapsulated pigment preparations and model foods containing these were studied by measuring the pigment content after storage in different conditions. Preliminary studies on the bioavailability of microencapsulated pigments and sensory evaluation for consumer acceptance of model foods containing microencapsulated pigments were also carried out. Enzyme-assisted oil extraction was used to extract lutein from marigold (Tagetes erecta) flower without organic solvents, and the yield was comparable to solvent extraction of lutein from the same flowers. The effects of temperature, extraction time, and beet:water ratio on extraction efficiency of betanin from red beet (Beta vulgaris) were studied and the optimal conditions for maximum yield and maximum betanin concentration were determined. In both cases, extraction at 40 °C was better than extraction at 80 °C and the extraction for five minutes was as efficient as 15 or 30 minutes. For maximum betanin yield, the beet:water ratio of 1:2 was better, with possibly repeated extraction, but for maximum betanin concentration, a ratio of 1:1 was better. Lutein was incorporated into oil-in-water (o/w) emulsions with a polar oil fraction from oat (Avena sativa) as an emulsifier and mixtures of guar gum and xanthan gum or locust bean gum and xanthan gum as stabilizers to retard creaming. The stability of lutein in these emulsions was quite good, with 77 to 91 percent of lutein being left after storage in the dark at 20 to 22°C for 10 weeks whereas in spray dried emulsions the retention of lutein was 67 to 75 percent. The retention of lutein in oil was also good at 85 percent. Betanin was incorporated into the inner w1 water phase of a water1-in-oil-inwater2 (w1/o/w2) double emulsion with primary w1/o emulsion droplet size of 0.34 μm and secondary w1/o/w2 emulsion droplet size of 5.5 μm and encapsulation efficiency of betanin of 89 percent. In vitro intestinal lipid digestion was performed on the double emulsion, and during the first two hours, coalescence of the inner water phase droplets was observed, and the sizes of the double emulsion droplets increased quickly because of aggregation. This period also corresponded to gradual release of betanin, with a final release of 35 percent. The double emulsion structure was retained throughout the three-hour experiment. Betanin was also spray dried and incorporated into model juices with different pH and dry matter content. Model juices were stored in the dark at -20, 4, 20–24 or 60 °C (accelerated test) for several months. Betanin degraded quite rapidly in all of the samples and higher temperature and a lower pH accelerated degradation. Stability of betanin was much better in the spray dried powder, with practically no degradation during six months of storage in the dark at 20 to 24 °C and good stability also for six months in the dark at 60 °C with 60 percent retention. Consumer acceptance of model juices colored with spray dried betanin was compared with similar model juices colored with anthocyanins or beet extract. Consumers preferred beet extract and anthocyanin colored model juices over juices colored with spray dried betanin. However, spray dried betanin did not impart any off-odors or off-flavors into the model juices contrary to the beet extract. In conclusion, this thesis describes novel solvent-free extraction and encapsulation processes for lutein and betanin from plant sources. Lutein showed good stability in oil and in o/w emulsions, but slightly inferior in spray dried emulsions. In vitro intestinal lipid digestion showed a good stability of w1/o/w2 double emulsion and quite high retention of betanin during digestion. Consumer acceptance of model juices colored with spray dried betanin was not as good as model juices colored with anthocyanins, but addition of betanin to real berry juice could produce better results with mixture of added betanin and natural berry anthocyanins could produce a more acceptable color. Overall, further studies are needed to obtain natural colorants with good stability for the use in food products.

Laser cutting implementation possibilities into paper making machine was studied as the main objective of the work. Laser cutting technology application was considered as a replacement tool for conventional cutting methods used in paper making machines for longitudinal cutting such as edge trimming at different paper making process and tambour roll slitting. Laser cutting of paper was tested in 70’s for the first time. Since then, laser cutting and processing has been applied for paper materials with different level of success in industry. Laser cutting can be employed for longitudinal cutting of paper web in machine direction. The most common conventional cutting methods include water jet cutting and rotating slitting blades applied in paper making machines. Cutting with CO2 laser fulfils basic requirements for cutting quality, applicability to material and cutting speeds in all locations where longitudinal cutting is needed. Literature review provided description of advantages, disadvantages and challenges of laser technology when it was applied for cutting of paper material with particular attention to cutting of moving paper web. Based on studied laser cutting capabilities and problem definition of conventional cutting technologies, preliminary selection of the most promising application area was carried out. Laser cutting (trimming) of paper web edges in wet end was estimated to be the most promising area where it can be implemented. This assumption was made on the basis of rate of web breaks occurrence. It was found that up to 64 % of total number of web breaks occurred in wet end, particularly in location of so called open draws where paper web was transferred unsupported by wire or felt. Distribution of web breaks in machine cross direction revealed that defects of paper web edge was the main reason of tearing initiation and consequent web break. The assumption was made that laser cutting was capable of improvement of laser cut edge tensile strength due to high cutting quality and sealing effect of the edge after laser cutting. Studies of laser ablation of cellulose supported this claim. Linear energy needed for cutting was calculated with regard to paper web properties in intended laser cutting location. Calculated linear cutting energy was verified with series of laser cutting. Practically obtained laser energy needed for cutting deviated from calculated values. This could be explained by difference in heat transfer via radiation in laser cutting and different absorption characteristics of dry and moist paper material. Laser cut samples (both dry and moist (dry matter content about 25-40%)) were tested for strength properties. It was shown that tensile strength and strain break of laser cut samples are similar to corresponding values of non-laser cut samples. Chosen method, however, did not address tensile strength of laser cut edge in particular. Thus, the assumption of improving strength properties with laser cutting was not fully proved. Laser cutting effect on possible pollution of mill broke (recycling of trimmed edge) was carried out. Laser cut samples (both dry and moist) were tested on the content of dirt particles. The tests revealed that accumulation of dust particles on the surface of moist samples can take place. This has to be taken into account to prevent contamination of pulp suspension when trim waste is recycled. Material loss due to evaporation during laser cutting and amount of solid residues after cutting were evaluated. Edge trimming with laser would result in 0.25 kg/h of solid residues and 2.5 kg/h of lost material due to evaporation. Schemes of laser cutting implementation and needed laser equipment were discussed. Generally, laser cutting system would require two laser sources (one laser source for each cutting zone), set of beam transfer and focusing optics and cutting heads. In order to increase reliability of system, it was suggested that each laser source would have double capacity. That would allow to perform cutting employing one laser source working at full capacity for both cutting zones. Laser technology is in required level at the moment and do not require additional development. Moreover, capacity of speed increase is high due to availability high power laser sources what can support the tendency of speed increase of paper making machines. Laser cutting system would require special roll to maintain cutting. The scheme of such roll was proposed as well as roll integration into paper making machine. Laser cutting can be done in location of central roll in press section, before so-called open draw where many web breaks occur, where it has potential to improve runability of a paper making machine. Economic performance of laser cutting was done as comparison of laser cutting system and water jet cutting working in the same conditions. It was revealed that laser cutting would still be about two times more expensive compared to water jet cutting. This is mainly due to high investment cost of laser equipment and poor energy efficiency of CO2 lasers. Another factor is that laser cutting causes material loss due to evaporation whereas water jet cutting almost does not cause material loss. Despite difficulties of laser cutting implementation in paper making machine, its implementation can be beneficial. The crucial role in that is possibility to improve cut edge strength properties and consequently reduce number of web breaks. Capacity of laser cutting to maintain cutting speeds which exceed current speeds of paper making machines what is another argument to consider laser cutting technology in design of new high speed paper making machines.

La tesi s'ha estructurat en tres apartats que, en conjunt, han de permetre determinar les possibilitats d'aprofitament dins la mateixa indústria alimentària de la fracció plasmàtica de la sang de porc generada per escorxadors que utilitzen sistemes oberts de recollida higiènica. 1. En la primera part s'analitza la composició de la sang higiènica que s'està recollint actualment i s'estudien les característiques tant físico-químiques com microbiològiques que determinen la seva qualitat. La caracterització s'ha realitzat amb sang recollida en diferents escorxadors industrials de les comarques de Girona i s'ha centrat principalment en l'estudi de la contaminació microbiològica i el nivell d'hemòlisi de la sang. S'ha fet un disseny experimental que ha permès alhora valorar l'efecte d'alguns factors sobre la qualitat de la sang: possibles diferències relacionades amb (1) la climatologia del període de l'any en el qual es fa la recollida, (2) particularitats dels escorxadors (grandària, sistemes de dessagnat, tipus, dosi i sistema de dosificació de l'anticoagulant, condicions de processament, maneig i emmagatzematge després de la recollida, etc.). Els resultats obtinguts ens permeten constatar que, en les condicions actuals, la sang que s'està recollint en els escorxadors estudiats no es pot considerar adequada per a una matèria primera de productes destinats a alimentació humana. La major part de la microbiota contaminant s'adquireix en el propi sagnador. S'ha constatat que el sistema de dessagnat en posició horitzontal podria ser una mesura útil per minimitzar la contaminació d'origen fecal o provinent de la pell de l'animal sacrificat i que la separació immediata de les fraccions en el propi escorxador també pot contribuir a reduir la contaminació. Així doncs, en el benentès que l'efectivitat pot obtenir-se del conjunt de mesures preses, més que de l'aplicació d'una sola d'elles, es suggereix la introducció d'una sèrie d'actuacions que potser permetrien reduir els nivells de contaminació que s'obtenen actualment. El tractament mecànic de la sang, el sistema d'addició d'anticoagulant, el volum i concentració de la solució anticoagulant afegida i el període d'emmagatzematge són els factors responsables de l'hemòlisi; mentre que nivells elevats de contaminació microbiològica i el tipus d'anticoagulant utilitzat deterrminen la velocitat d'increment de l'hemòlisi de sang refrigerada. S'ha constatat que quan la sang no pot ser processada immediatament i s'ha d'emmagatzemar en refrigeració és millor utilitzar citrat sòdic enlloc de polifosfat com a anticoagulant ja que l'increment d'hemòlisi es dóna més lentament. 2. El segon apartat s'ha centrat en la fracció plasmàtica de la sang. S'ha utilitzat la deshidratació per atomització com a tecnologia de conservació del plasma i s'ha fet una caracterització del producte en pols resultant des del punt de vista de composició i qualitat. A més de la contaminació microbiològica, que determina la qualitat higiènico-sanitària del producte, s'ha realitzat un estudi de les propietats funcionals que podrien fer del plasma un producte útil en la formulació d'aliments (capacitat escumant, emulsionant, gelificant). S'ha fet especial incidència en (1) determinar l'efecte del procés tecnològic de deshidratació sobre la funcionalitat del producte i (2) estudiar l'estabilitat del plasma deshidratat durant el període d'emmagatzematge. En les condicions de deshidratació per atomització aplicades no es provoca desnaturalització de la fracció proteica i s'obté un producte suficientment deshidratat, amb una aw<0,4 per permetre suposar una bona estabilitat. Algunes mostres de plasma deshidratat analitzades presenten nivells detectables de determinats residus (sulfonamides i corticosteroides). La qualitat microbiològica del producte en pols reflecteix l'elevada contaminació que contenia la matèria primera utilitzada, tot i que la deshidratació per atomització ha comportat la reducció en una unitat logarítmica de la càrrega contaminant. Els recomptes generals de microorganismes són encara preocupants i més tenint en compte que s'ha evidenciat la presència de toxines estafilocòciques en algunes mostres. L'avaluació de les propietats funcionals del producte deshidratat en relació a les que presentava el plasma líquid ens ha permès comprovar que: (1) El procés de deshidratació no ha afectat la solubilitat de les proteïnes. Això, junt amb el fet que no s'obtinguin diferències significatives en l'anàlisi calorimètrica de mostres líquides o deshidratades, permet concloure que el procés no provoca desnaturalització proteica. (2) No s'observen efectes negatius del procés tecnològic sobre la capacitat escumant ni en l'activitat emulsionant de les proteïnes plasmàtiques, dues propietats funcionals que possibiliten l'aplicació del plasma amb aquestes finalitats en l'elaboració d'alguns aliments. (3) La deshidratació tampoc perjudica de manera important les característiques dels gels que s'obtenen per escalfament, ja que els gels obtinguts a partir del plasma líquid i del plasma deshidratat presenten la mateixa capacitat de retenció d'aigua i no s'observen diferències en la microestructura de la xarxa proteica d'ambdós tipus de gel. Tanmateix, els que s'obtenen a partir del producte en pols mostren una menor resistència a la penetració. L'estudi d'estabilitat ens ha permès comprovar que la mostra de plasma deshidratat per atomització perd algunes de les seves propietats funcionals (facilitat de rehidratació, capacitat de retenció d'aigua i fermesa dels gels) si s'emmagatzema a temperatura ambient, mentre que aquestes característiques es mantenen un mínim de sis mesos quan el producte en pols es conserva a temperatura de refrigeració. 3. En l'última part, tenint en compte les conclusions derivades dels resultats dels apartats anteriors, s'han assajat tres possibles sistemes de reducció de la contaminació aplicables a la fracció plasmàtica com a pas previ a la deshidratació, per tal de millorar les característiques de qualitat microbiològica i les perspectives d'estabilitat del producte durant l'emmagatzematge. S'ha determinat l'eficàcia, i l'efecte sobre les propietats del plasma deshidratat, que poden tenir tractaments d'higienització basats en la centrifugació, la microfiltració tangencial i l'aplicació d'altes pressions. Els tractaments de bactofugació aplicats permeten reduir entre el 96 i el 98% la contaminació microbiana del plasma. Aquesta reducció s'aconsegueix tant amb un sistema discontinu com amb un sistema continu treballant a una velocitat de 12 L/h, fet que permetria adaptar el tractament de bactofugació a un procés de producció industrial. Un sistema combinat de bactofugació en continu i microfiltració tangencial permet incrementar l'eficàcia fins a un 99,9 % de reducció. Cal tenir present, però, que aquest tractament provoca també una disminució de l'extracte sec que afecta negativament les propietats funcionals del plasma líquid. Malgrat suposar una pèrdua pel que fa al rendiment, aquest efecte negatiu sobre la funcionalitat no suposaria cap inconvenient si s'utilitzés la deshidratació com a tecnologia de conservació del plasma, ja que es podria corregir l'extracte sec durant la reconstitució del producte. Caldria avaluar si la millora en la qualitat higiènico-sanitària del producte compensa o no les pèrdues que suposa aquest sistema d'higienització combinat. Amb relació als tractaments d'alta pressió, de totes les condicions de tractament assajades, les pressions de fins 450 MPa permeten obtenir plasma sense modificacions importants que impedeixin la seva deshidratació per atomització. Així doncs, les condicions de procés que s'han aplicat són pressuritzacions a 450 MPa de 15 minuts de durada. La temperatura de tractament que s'ha mostrat més eficaç en la reducció dels recomptes de microorganismes ha estat de 40ºC. Els tractaments a aquesta temperatura permeten assolir reduccions del 99,97% i disminuir en un 80% la capacitat de creixement dels microorganismes supervivents a la pressurització en relació a la que presentava la població contaminant del plasma abans del tractament. L'estudi de l'efecte d'aquest tractament (450 MPa, 15 min i 40ºC) sobre les propietats funcionals del plasma ha permès observar que la pressurització comporta una disminució en la solubilitat del producte però una millora en les propietats de superfície -estabilitat de l'escuma i activitat emulsionant- i un increment de la capacitat de retenció d'aigua i de la duresa dels gels obtinguts per escalfament. Calen més estudis per confirmar i caracteritzar aquesta millora en la funcionalitat, així com per establir si el tractament de pressurització afecta també l'estabilitat del producte durant l'emmagatzematge. De totes les tecnologies d'higienització assajades, l'alta pressió és la que permet obtenir millors resultats en el sentit de poder garantir un producte de bona qualitat microbiològica i segur, des del punt de vista sanitari i tecnològic, per a la seva utilització com a ingredient alimentari.

A survey was carried out on 55 commercial dairy farms located in the South of Chile during 1995-97. A questionnaire was developed to obtain informed estimates of dairy effluent management on those farms. Information was analysed on an annual basis using a computer spreadsheet linking all the parameters surveyed. In addition, slurry samples were taken for analysis of dry matter content (DM). Herd size varied between 50 and 800 cows per farm. A large proportion of the total volume of effluents produced came from rainfall (46%), dirty water accounted for 29% with only 25% from cow's faeces and urine. The large volume of effluents produced resulted in a reduced storage capacity (on average of 2 months) or more frequent and higher application rates to the field. Only 37% of the farmers knew the application rates of manure and there was a wide range in the quantity used per year (12 m(3)/ha to 300 m(3)/ha). Dairy effluents were applied mainly on grass (71%) throughout the year but, mostly concentrated during the winter and spring time using only surface irrigation system. The total solids contents of effluents was very low, with 62% of the samples being <4% DM. This reflected the large volumes of clean water that the storage tanks received. The information collected has identified problems in effluent management in Chilean dairy farms where research and technology transfer will be necessary to avoid pollution problems.

We assessed the potential for using optical functional types as effective markers to monitor changes in vegetation in floodplain meadows associated with changes in their local environment. Floodplain meadows are challenging ecosystems for monitoring and conservation because of their highly biodiverse nature. Our aim was to understand and explain spectral differences among key members of floodplain meadows and also characterize differences with respect to functional traits. The study was conducted on a typical floodplain meadow in UK (MG4-type, mesotrophic grassland type 4, according to British National Vegetation Classification). We compared two approaches to characterize floodplain communities using field spectroscopy. The first approach was sub-community based, in which we collected spectral signatures for species groupings indicating two distinct eco-hydrological conditions (dry and wet soil indicator species). The other approach was “species-specific”, in which we focused on the spectral reflectance of three key species found on the meadow. One herb species is a typical member of the MG4 floodplain meadow community, while the other two species, sedge and rush, represent wetland vegetation. We also monitored vegetation biophysical and functional properties as well as soil nutrients and ground water levels. We found that the vegetation classes representing meadow sub-communities could not be spectrally distinguished from each other, whereas the individual herb species was found to have a distinctly different spectral signature from the sedge and rush species. The spectral differences between these three species could be explained by their observed differences in plant biophysical parameters, as corroborated through radiative transfer model simulations. These parameters, such as leaf area index, leaf dry matter content, leaf water content, and specific leaf area, along with other functional parameters, such as maximum carboxylation capacity and leaf nitrogen content, also helped explain the species’ differences in functional dynamics. Groundwater level and soil nitrogen availability, which are important factors governing plant nutrient status, were also found to be significantly different for the herb/wetland species’ locations. The study concludes that spectrally distinguishable species, typical for a highly biodiverse site such as a floodplain meadow, could potentially be used as target species to monitor vegetation dynamics under changing environmental conditions.

Dynamic global vegetation models (DGVMs) typically rely on plant functional types (PFTs), which are assigned distinct environmental tolerances and replace one another progressively along environmental gradients. Fixed values of traits are assigned to each PFT; modelled trait variation along gradients is thus driven by PFT replacement. But empirical studies have revealed "universal" scaling relationships (quantitative trait variations with climate that are similar within and between species, PFTs and communities); and continuous, adaptive trait variation has been proposed to replace PFTs as the basis for next-generation DGVMs. Here we analyse quantitative leaf-trait variation on long temperature and moisture gradients in China with a view to understanding the relative importance of PFT replacement vs. continuous adaptive variation within PFTs. Leaf area (LA), specific leaf area (SLA), leaf dry matter content (LDMC) and nitrogen content of dry matter were measured on all species at 80 sites ranging from temperate to tropical climates and from dense forests to deserts. Chlorophyll fluorescence traits and carbon, phosphorus and potassium contents were measured at 47 sites. Generalized linear models were used to relate log-transformed trait values to growing-season temperature and moisture indices, with or without PFT identity as a predictor, and to test for differences in trait responses among PFTs. Continuous trait variation was found to be ubiquitous. Responses to moisture availability were generally similar within and between PFTs, but biophysical traits (LA, SLA and LDMC) of forbs and grasses responded differently from woody plants. SLA and LDMC responses to temperature were dominated by the prevalence of evergreen PFTs with thick, dense leaves at the warm end of the gradient. Nutrient (N, P and K) responses to climate gradients were generally similar within all PFTs. Area-based nutrients generally declined with moisture; Narea and Karea declined with temperature, but Parea increased with temperature. Although the adaptive nature of many of these trait-climate relationships is understood qualitatively, a key challenge for modelling is to predict them quantitatively. Models must take into account that community-level responses to climatic gradients can be influenced by shifts in PFT composition, such as the replacement of deciduous by evergreen trees, which may run either parallel or counter to trait variation within PFTs. The importance of PFT shifts varies among traits, being important for biophysical traits but less so for physiological and chemical traits. Finally, models should take account of the diversity of trait values that is found in all sites and PFTs, representing the "pool" of variation that is locally available for the natural adaptation of ecosystem function to environmental change.

The objective of this trial was to evaluate the nutritional value, fermentation profile and dry matter losses of Palisadegrass silages ensiled with either dried citrus pulp, soybean hulls, chemical or microbial additives. The trial was carried out in a completely randomized experimental design and in a factorial arrangement (3 x 5), with three dry matter levels (wet forage or forage ensiled with pelleted citrus pulp or pelleted soybean hulls) and five additives (without or with the presence of bacterial inoculants or the addition of: sodium benzoate, formic acid in the concentration of 62% or 44%), totalizing 15 treatments and 60 experimental silos. The variables analyzed were: nutritional value, losses due to gases and effluents, and dry matter recovery. The use of dried citrus pulp or soybean hulls at the ensiling time increased the dry matter content (29.4 and 28,9%) and decreased the effluent production (4.1 and 3.8 kg/t of fresh matter), also providing fermentable substrate to microorganisms, resulting in increased fermentation coefficient and digestibility of silages. The use of formic acid resulted in silages with higher digestibility and increased water-soluble carbohydrates and crude protein content. This additive was also effective in reducing the losses due to gases and, as a result, increased the total dry matter recovery. The treatment containing homolactic bacteria showed similar trend of increasing the digestibility and reducing the losses due to gases. The use of sodium benzoate was less effective in altering the fermentation pattern of tropical silages. The nutritional value and total dry matter losses of silages ensiled without additives can be considered satisfactory. However, wet forage ensiled with dried citrus pulp and, mainly, with soybeans hulls showed the best results. Treatments containing formic acid had a beneficial effect on the fermentation profile of tropical grass silages.
