Contrails and especially their evolution into cirrus-like clouds are thought to have very important effects on local and global radiation budgets, though are generally not well represented in global climate models. Lack of contrail parameterisations is due to the limited availability of in situ contrail measurements which are difficult to obtain. Here we present a methodology for successful sampling and interpretation of contrail microphysical and radiative data using both in situ and remote sensing instrumentation on board the FAAM BAe146 UK research aircraft as part of the COntrails Spreading Into Cirrus (COSIC) study.

Recent laboratory measurements show that absorption by the water vapour continuum in near-infrared windows may be about an order of magnitude higher than assumed in many radiation codes. The radiative impact of the continuum at visible and near-infrared wavelengths is examined for the present day and for a possible future warmer climate (with a global-mean total column water increase of 33%). The calculations use a continuum model frequently used in climate models (‘CKD’) and a continuum model where absorption is enhanced at wavelengths greater than 1 µm based on recent measurements (‘CAVIAR’). The continuum predominantly changes the partitioning between solar radiation absorbed by the surface and the atmosphere; changes in top-of-atmosphere net irradiances are smaller. The global-mean clear-sky atmospheric absorption is enhanced by 1.5 W m−2 (about 2%) and 2.8 W m−2 (about 3.5%) for CKD and CAVIAR respectively, relative to a hypothetical no-continuum case, with all-sky enhancements about 80% of these values. The continuum is, in relative terms, more important for radiation budget changes between the present day and a possible future climate. Relative to the no-continuum case, the increase in global-mean clear-sky absorption is 8% higher using CKD and almost 20% higher using CAVIAR; all-sky enhancements are about half these values. The effect of the continuum is estimated for the solar component of the water vapour feedback, the reduction in downward surface irradiance and precipitation change in a warmer world. For CKD and CAVIAR respectively, and relative to the no-continuum case, the solar component of the water vapour feedback is enhanced by about 4 and 9%, the change in clear-sky downward surface irradiance is 7 and 18% more negative, and the global-mean precipitation response decreases by 1 and 4%. There is a continued need for improved continuum measurements, especially at atmospheric temperatures and at wavelengths below 2 µm.

We report on the assembly of tumor necrosis factor receptor 1 (TNF-R1) prior to ligand activation and its ligand-induced reorganization at the cell membrane. We apply single-molecule localization microscopy to obtain quantitative information on receptor cluster sizes and copy numbers. Our data suggest a dimeric pre-assembly of TNF-R1, as well as receptor reorganization toward higher oligomeric states with stable populations comprising three to six TNF-R1. Our experimental results directly serve as input parameters for computational modeling of the ligand-receptor interaction. Simulations corroborate the experimental finding of higher-order oligomeric states. This work is a first demonstration how quantitative, super-resolution and advanced microscopy can be used for systems biology approaches at the single-molecule and single-cell level.

Transforming growth factor beta (TGF-beta) plays a role both in the induction of Treg and in the differentiation of the IL-17-secreting T cells (Th17) which drive inflammation in experimental autoimmune encephalomyelitis (EAE). We investigated the role that thrombospondin-1 (TSP-1) dependent activation of TGF-beta played in the generation of an encephalitic Th17 response in EAE. Upon immunization with myelin oligodendrocyte glycoprotein peptide (MOG(35-55)), TSP-1 deficient (TSP-1(null)) mice and MOG(35-55) TCR transgenic mice that lack of TSP-1 (2D2.TSP-1(null)) exhibited an attenuated form of EAE, and secreted lower levels of IL-17. Adoptive transfer of in vitro-activated 2D2.TSP-1(null) T cells induced a milder form of EAE, independent of TSP-1 expression in the recipient mice. Furthermore, in vitro studies demonstrated that anti-CD3/anti-CD28 pre-activated CD4+ T cells transiently upregulated latent TGF-beta in a TSP-1 dependent way, and such activation of latent TGF-beta was required for the differentiation of Th17 cells. These results demonstrate that TSP-1 participates in the differentiation of Th17 cells through its ability to activate latent TGF-beta, and enhances the inflammatory response in EAE. We study the loading of sodium atoms into a magneto-optical trap from current-controlled sodium metal dispensers. Contrary to what was previously reported [V. Wippel, C. Binder, W. Huber, L Windholz, M. Allegrini, F. Fuso, E. Arimondo, Eur. Phys. J. D 17 (2001) 2851 we demonstrate a significantly higher number of trapped atoms that make Na dispensers a feasible source of atoms for cold-atom studies. The inherent rise in pressure that naturally arises from metal dispensers as they are heated to release atoms is partially controlled by placing the metal dispensers near the pumping port where an ion pump is connected. We also study the effects of placing the sodium dispensers at different distances from the main vacuum chamber where the atoms are trapped and the effectiveness of using a Zeeman slower to cool the atoms as they emerge from the dispensers. We observe trapping of up to 1.9 x 10(8) atoms, which is significantly higher by almost three orders of magnitude than previously reported experiments. (C) 2008 Elsevier B.V. All rights reserved.

We show how a circuit analysis, used widely in electrical engineering, finds application to problems of light wave injection and transport in subwavelength structures in the optical frequency range. Lumped circuit and transmission-line analysis may prove helpful in the design of plasmonic devices with standard, functional properties.

Sob o acrônimo BOP (Bottom of Pyramid), muito se tem discutido sobre as vantagens empresariais de atender a uma classe emergente de aproximadamente 4 bilhões de pessoas ao redor do mundo (Prahalad 2005), como uma das formas de se defender da estagnação do consumo nos países mais desenvolvidos. Se hoje a discussão já está mais bem delineada, é muito em função de artigos de estudiosos como Prahalad, Hammond e Yunus que colocaram a base da pirâmide no centro do debate. Há empresas referência mundial nessa transição de atendimento para o BOP, como a GE na China ou a Unilever na Índia. Mas para o caso brasileiro, parece ainda não haver igual expressão, sendo a emergência do BOP mais recente. Algumas dessas empresas orientadas para as classes emergentes têm mudado seu modelo de negócios. Outras criam submarcas voltadas às classes emergentes e sofrem os impactos dessas decisões sobre a arquitetura de marcas. E, finalmente, há empresas que utilizam um conjunto de signos e códigos de comunicação na oferta de seus produtos e serviços que acreditam ser bem aceitos transversalmente nas classes sociais. O objetivo geral da dissertação foi avaliar quais códigos e signos de comunicação as empresas com posicionamento premium devem utilizar numa comunicação com essa classe emergente, sem correr o risco de afastar seu público-alvo original (das classes AB). O método inclui a ferramenta denominada Value Reframing (Reenquadramento de Valor), para a busca de novos valores para um produto ou serviço em um novo contexto de mercado. Tal ferramenta não prevê teste de rejeição dentro do público original. Então foram coletados: 1) os valores mais bem aceitos pela nova classe média; 2) os valores avaliados e não rejeitados por consumidores da classe AB. Em conclusão, é possível utilizar signos e códigos com aceitação nas duas classes (como uso do humor nas mensagens ou a sensação de que a mensagem foi direcionada para a pessoa). A adoção desses elementos podem ser mais bem sucedidos se acompanhados de uma discussão sobre o modelo de negócios vigente, para atender melhor a nova classe média.

O objetivo deste estudo é avaliar a propagação de choques econômicos de alguns países sobre o crescimento econômico brasileiro, com principal destaque para China, Estados Unidos da América (EUA) e Argentina, que são os principais parceiros comerciais do Brasil. O aumento do comércio com a China tornou o Brasil muito mais vulnerável a choques no PIB chinês e menos vulnerável, do que no passado recente, a choques no PIB americano, enquanto que a influência da Argentina manteve-se estável. Foi aplicada a metodologia Vetor Autorregressivo Global (Global Var – GVAR), introduzida por Pesaran, Schuermann e Weiner (2004), Garratt, Lee, Pesaran e Shin (2006) e Dées, Di Mauro, Pesaran e Smith (2007), para analisar os canais de comércio e a transmissão de choques entre o resto do mundo e o Brasil. Usando dados trimestrais a partir de 1990 até o final de 2013, foi possível constatar que o aumento da relevância da economia Chinesa na balança comercial Brasileira exerce pressão sobre o crescimento econômico do Brasil. Em suma, a China tornou-se mais relevante para o crescimento econômico do Brasil do que os EUA e a Argentina.

Background: Biopharmaceutical drugs are mainly recombinant proteins produced by biotechnological tools. The patents of many biopharmaceuticals have expired, and biosimilars are thus currently being developed. Human granulocyte colony stimulating factor (hG-CSF) is a hematopoietic cytokine that acts on cells of the neutrophil lineage causing proliferation and differentiation of committed precursor cells and activation of mature neutrophils. Recombinant hG-CSF has been produced in genetically engineered Escherichia coli ( Filgrastim) and successfully used to treat cancer patients suffering from chemotherapy-induced neutropenia. Filgrastim is a 175 amino acid protein, containing an extra N-terminal methionine, which is needed for expression in E. coli. Here we describe a simple and low-cost process that is amenable to scaling-up for the production and purification of homogeneous and active recombinant hG-CSF expressed in E. coli cells.Results: Here we describe cloning of the human granulocyte colony-stimulating factor coding DNA sequence, protein expression in E. coli BL21(DE3) host cells in the absence of isopropyl-beta-D-thiogalactopyranoside ( IPTG) induction, efficient isolation and solubilization of inclusion bodies by a multi-step washing procedure, and a purification protocol using a single cationic exchange column. Characterization of homogeneous rhG-CSF by size exclusion and reverse phase chromatography showed similar yields to the standard. The immunoassay and N-terminal sequencing confirmed the identity of rhG-CSF. The biological activity assay, in vivo, showed an equivalent biological effect (109.4%) to the standard reference rhG-CSF. The homogeneous rhG-CSF protein yield was 3.2 mg of bioactive protein per liter of cell culture.Conclusion: The recombinant protein expression in the absence of IPTG induction is advantageous since cost is reduced, and the protein purification protocol using a single chromatographic step should reduce cost even further for large scale production. The physicochemical, immunological and biological analyses showed that this protocol can be useful to develop therapeutic bioproducts. In summary, the combination of different experimental strategies presented here allowed an efficient and cost-effective protocol for rhG-CSF production. These data may be of interest to biopharmaceutical companies interested in developing biosimilars and healthcare community.

This paper summarizes floristic and phytossociology data of 11, out of 14 plots of 1 ha, allocated along an altitudinal gradient in the Serra do Mar, Silo Paulo, Brazil. The study was conducted at Serra do Mar State Park and the plots start at the sea level (10 m - plot of Restinga Forest that occurs at Praia da Fazenda, Picinguaba, municipality of Ubatuba) up to 1100 m above sea level (the Montane Ombrophilous Dense occurs alongside the Itamambuca Trail, municipality of Silo Luis do Paraitinga). The Restinga Forest occurs in Pleistocenic Coastal Plain where the soil is classified as a sandy Quartzipsamment (Quartzenic Neosol), while along the slopes of the Serra do Mar, the Ombrophylus Dense Forest grows on the top of a pre-Cambrian crystalline basement with granitic rocks, where the soil is a sandy-loam Dystrophic Inceptisol (Cambisol/Latosol). In all 14 plots soils are acidic (pH 3 - 4), chemically poor, with high dilution of nutrients and high saturation of aluminum. In the Restinga and at the foot of the slope the climate is Tropical/Subtropical Humid (Af/Cfa), with no dry season, an average annual rainfall over 2,200 mm and an average annual temperature of 22 degrees C. Towards the top of the Serra do Mar there is a gradual cooling along the slope, but there is no reduction in rainfall, so at 1,100 m above sea level the climate is classified as Humid Subtropical (Cfa/Cfb), with no dry season and an average annual temperature of 17 degrees C. It is important to remark that, almost daily, from 400 m above sea level up to the top of slopes the mountains are covered by a dense fog. In the 14 plots 21,733 individuals with DBH >= 4.8 cm, including trees, palms and ferns, were marked, measured and sampled. The average number of individuals sampled in each plot was 1264 ind.ha(-1)(+/- 218 SE 95%). Within the parameters considered trees prevailed (71% in the Montane ODF to 90% in the Restinga Forest), followed by palms (10% in the RF and 25% in the Montane Ombrophilous Dense Forest/ODF) and ferns (0% % in the RF and 4% in the Montane ODF). Regarding these proportions the Exploited Lowlands ODF differs from the others with only 1.8% of palm trees and striking 10% of ferns. The forest canopy is irregular with heights ranging from 7 to 9 m, rarely emergent trees reach 18 m, and due to this irregularity of the canopy the amount of light that gets through sets conditions for the development of hundreds of epiphytic species. Aside from Montana ODF, where the number of dead trees was more than 5% of individuals sampled, in the other phytophysiognomies this value was below 2.5%. In the 11 plots where the floristic study was conducted we found 562 species in 195 genera and 68 families. Only seven species - Euterpe edulis Mart. (Arecaceae), Calyptranthes lucida Mart. ex DC. and Marlierea tomentosa Cambess (both Myrtaceae), Guapira opposita (Veil.) Reitz (Nyctaginaceae), Cupania oblongifolia Mart. (Sapindaceae), Cecropia glaziovii Snethl. and Coussapoa microcarpa (Schott) Rizzini (both Urticaceae) - occurred from Restinga to Montane ODF, while 12 other species did not occur only in the Restinga Forest. Families with the greatest number of species are Myrtaceae (133 spp), Fabaceae (47 spp), Rubiaceae (49) and Lauraceae (49) throughout the gradient and Monimiaceae (21) specifically in portions Montane ODF. Only in the F plot, where logging has occurred between 1950 and 1985, the abundance of palm trees has been replaced by Cyatheaceae. The study shows a peak of diversity and richness, Shannon-Weiner index (H') ranging from 3.96 to 4.48 nats.ind(-1), in the intermediate altitudes (300 to 400 m) along the slope. Several explanations for this result are raised here, including the fact that these elevations are within the limits expansions and retractions of the different phytophysiognomies of the Atlantic ODF due to climate fluctuations during the Pleistocene. The results presented in this paper demonstrate the extraordinary richness of tree species of the Atlantic Rainforest from the northeastern coast of the State of São Paulo, reinforcing the importance of its conservation throughout the altitudinal gradient. The richness of this forest justifies a long term commitment to study its dynamics and functioning through permanent plots, and monitor the impacts of climate change in this vegetation.

A protocol to produce large amounts of bioactive homogeneous human interferon β1 expressed in Escherichia coli was developed. Human interferon β1 ser17 gene was constructed, cloned and subcloned, and the recombinant protein expressed in E. coli cells. Solubilization of recombinant human interferon β1 ser17 (rhIFN-β1 ser17) was accomplished by employing a brief shift to high alkaline pH in the presence of non-ionic detergent. The recombinant protein was purifi ed by three chromatographic steps. N-terminal amino acid sequencing and mass spectrometry analysis provided experimental evidence for the identity of the recombinant protein. Reverse phase liquid chromatography demonstrated that the content of deamidates and sulphoxides was similar to a commercial standard. Size exclusion chromatography demonstrated the absence of high molecular mass aggregates and dimers. The protocol represents an effi cient and high-yield method to obtain bioactive homogeneous monomeric rhIFN-β1 ser17 protein. It may thus represent an important step towards scaling up for rhIFN-β1 ser17 large-scale production. © 2010 Villela AD, et al.
