The aim of this study was to evaluate the quality of spray droplet spectrum on coffee tree, by using different equipments and syrup volumes. The experiment was set up in a coffee farm, Acaiá variety, Campos Gerais city, MG, Brazil. The treatments were three syrup volumes (150, 300 and 600 L/ha). The lower volume was sprayed with the pneumatic sprayer Martignani, Whirlwind B612 Autonom-Trac mode, and the other volume rates using the conventional hydraulic sprayer to coffee tree. It was possible concludes that the spray volume can be reduced in applications on coffee tree using the pneumatic sprayer.

Essential steps to providing reliable results using the Analytical Quality Assurance Cycle

Considering how demand for quality assurance (QA) has grown in analytical laboratories, we show the trends in analytical science, illustrated through international standard ISO/IEC 17025, validation, measurements of uncertainty, and quality-control (QC) measures. A detailed review of the history of analytical chemistry indicates that these concepts are consistently used in laboratories to demonstrate their traceabilities and competences to provide reliable results. We propose a new approach for laboratory QA, which also develops a diagram to support routine laboratories (which generally apply a quality system, such as ISO/IEC 17025) or research laboratories (that have some difficult applying this international standard). This approach, called the Analytical Quality Assurance Cycle (AQAC), presents the major QA concepts and the relationships between these concepts in order to provide traceability and reliable results. The AQAC is a practical tool to support the trend towards QA in analytical laboratories. (C) 2012 Elsevier Ltd. All rights reserved.

Tre guide francesi sull'Italia del Sud: analisi argomentativa del discorso turistico

Le but de ce mémoire est d'identifier les caractéristiques typiques du discours touristique à travers une analyse argumentative de trois guides touristiques d'éditeurs et de langue français sur l'Italie du Sud, respectivement le guide «Routard», «Petit futé» et «Le guide bleu». Dans le premier chapitre, j'ai tracé l'histoire du développement et de la diffusion de ce genre textuel à travers les siècles. Ensuite, à travers une analyse non seulement linguistique mais qui prend en compte aussi les éléments et les stéréotypes culturels, j'ai mis en exergue les différences de style des trois guides et la manière dont ils présentent une destination peu connue au public français telle que l'Italie du Sud. En particulier, il a été possible, à travers les textes des guides, de retrouver une représentation pas toujours stéréotypée des Italiens et des Français aussi. Le dernier chapitre est consacré à une étude purement linguistique des textes touristiques, dont j'ai pris en considération des éléments récurrents comme la formule, le lexique évaluatif, la technique du balancement axiologique et la poéticité du langage. En conclusion, avec les outils fournis par l'analyse argumentative et culturelle, il a été possible de répertorier les éléments du style et du langage que l'on pourrait considérer comme typiques du discours de promotion touristique et, en même temps, d'identifier les différences entre les trois guides qui montrent comment un même genre peut se décliner de multiples façons différentes.

Erkennung der alpha-Kette des GM-CSF-Rezeptors (CSF2RA) durch einen HLA-unabhängigen alpha:beta-T-Zellrezeptor

Aus dem tumorreaktiven T-Zellrepertoire der Melanompatientin Ma-Mel-86/INTH, bei der im Verlauf Lymphknotenmetastasen HLA-Klasse I-negativer Tumorzellen auftraten, wurden durch Stimulation mit autologen Tumorzellen CD8+ T-Zellklone isoliert und expandiert, die auf Melanomzellen der Patientin CSF2RA (engl. GM-CSF receptor alpha chain) in HLA-unabhängiger Weise erkannten. Aus einem der T-Zellklone wurde ein CSF2RA-reaktiver α:β-T-Zellrezeptor (TCR, engl. T-cell receptor) kloniert (Bezeichnung: TCR-1A.3/46). Die α-Kette des TCR enthielt die Domänen TRAV14/DV4*01, TRAJ48*01 und TRAC*01, die β-Kette die Domänen TRBV10-3*01, TRBD2*01, TRBJ2-7*01 und TRBC2*01. Durch Austausch der humanen konstanten gegen die homologen murinen Domänen wurde der TCR optimiert (Bezeichnung: cTCR-1A.3/46) und hinsichtlich seiner Expression und Funktionalität nach retroviralem Transfer in humane PBMC (engl. peripheral blood mononuclear cells) im 51Chromfreisetzungstest, im IFN-γ-ELISpot-Assay und in einem Degranulations-Assay validiert. TCR-transgene T-Zellen lysierten nicht nur spezifisch die HLA-defizienten, CSF2RA+ Melanomlinien des Modells Ma-Mel-86, sondern erkannten auch Zelllinien verschiedener Spezies nach Transfektion von CSF2RA sowie Monozyten, Granulozyten, dendritische Zellen und ein breites Spektrum hämatologischer Malignome myeloiden Ursprungs ungeachtet deren HLA-Phänotypen. Lymphatische Zellen sowie CD34+ Blutstammzellen wurden in In vitro-Untersuchungen nicht erkannt. Der Zusatz von GM-CSF zu Zellen, die CSF2RA und CSF2RB exprimierten, inhibierte die Erkennung durch TCR-transgene PBMC, während die Koexpression der α- und der ß-Kette des GM-CSF-Rezeptors alleine keinen negativen Effekt auf die Erkennung hatte. Daraus war zu schließen, dass CSF2RA präferentiell freistehend und weniger nach Integration in den heteromultimerischen GM-CSF-Rezeptor-Komplex erkannt wurde. In der zweidimensionalen Collier-de-Perles-Visualisierung der IMGT-Datenbank (engl. International immunogenetics information system) wies der CSF2RA-reaktive TCR-1A.3/46 im Vergleich zu TCR von konventionellen, HLA-restringierten T-Zellen keine Besonderheiten auf. Darüber hinaus waren auch die von den HLA-unabhängigen T-Zellen exprimierten CD8-Moleküle identisch zu den CD8-Molekülen HLA-abhängiger CTL (engl. cytotoxic T lymphocytes). Die Präsenz von CD8-Molekülen förderte die HLA-unabhängige Erkennung von CSF2RA, schien aber dafür nicht zwingend erforderlich zu sein, da Antikörper gegen CD8 die Erkennung zu ca. 65 % blockierten und TCR-transgene CD4+ T-Zellen im Vergleich zu TCR-transduzierten CD8+ T-Zellen eine deutlich verringerte, aber noch erhaltene Funktionalität aufwiesen. Es ist derzeit nicht klar, ob HLA-unabhängige T-Zellen gegen CSF2RA im peripheren Blut der Patientin vorkamen, weil sie der im Tiermodell postulierten Thymusselektion MHC-unabhängiger TCR (Tikhonova et al., Immunity 36:79, 2012) entkommen waren, oder weil ein ursprünglich gegen einen HLA-Peptid-Komplex gerichteter TCR eine HLA-unabhängige Kreuzreaktivität aufwies. CSF2RA verbessert die Glucoseutilisation in malignen Zellen, und es wurden ihm embryotrophe Eigenschaften zugeschrieben (Spielholz et al., Blood 85:973, 1995; Sjöblom et al., Biol. Reprod. 67:1817, 2002). Damit kann CSF2RA malignes Wachstum fördern und ist somit ein potentielles Zielmolekül für die Immuntherapie. Seine HLA-unabhängige Erkennung würde sowohl die HLA-Vielfalt als auch den HLA-Verlust als typische Limitationen der T-Zellimmuntherapie umgehen. Zur Überprüfung der In vivo-Spezifität des HLA-unabhängigen TCR gegen CSF2RA und damit zum Ausschluss relevanter off-tumor-/on-target- bzw. off-tumor-/off-target-Effekte ist jedoch eine Testung in einem präklinischen Tiermodell erforderlich.

First intercomparison study on the analysis of oxygenated polycyclic aromatic hydrocarbons (oxy-PAHs) and nitrogen heterocyclic polycyclic aromatic compounds (N-PACs) in contaminated soil

Oxygenated polycyclic aromatic hydrocarbons (oxy-PAHs) and nitrogen heterocyclic polycyclic aromatic compounds (N-PACs) are toxic, highly leachable and often abundant at sites that are also contaminated with PAHs. However, due to lack of regulations and standardized methods for their analysis, they are seldom included in monitoring and risk-assessment programs. This intercomparison study constitutes an important step in the harmonization of the analytical methods currently used, and may also be considered a first step towards the certification of reference materials for these compounds. The results showed that the participants were able to determine oxy-PAHs with accuracy similar to PAHs, with average determined mass fractions agreeing well with the known levels in a spiked soil and acceptable inter- and intra-laboratory precisions for all soils analyzed. For the N-PACs, the results were less satisfactory, and have to be improved by using analytical methods more specifically optimized for these compounds.

Valtra Versy y Direct

Los nuevos modelos Valtra, en sus versiones Versu y Direct, se han refinado las características tradicionales ofrecidas por Valtra añadiéndose otras muchas. De su “diseño tradicional” se destaca la facilidad de manejo en posición invertida utilizando el sistema de conducción reversible Twin-Trac. De lo "nuevo" se destacan sus nuevas transmisiones: transmisión variable continua (CVT), y transmisión múltiple escalonada con cambio bajo carga (Powershift).

Estimación de biomasa forestal mediante el uso de imágenes radar.

La importancia de la Biomasa a nivel mundial, ha llevado a que más de 130 países celebren el protocolo de Kioto sobre el cambio climático dictaminando como objetivo la reducción de las emisiones de seis gases de efecto invernadero y tres gases industriales fluorados, así como la incorporación de la fijación del CO2 como un objetivo dentro de los criterios de gestión de bosques. Entre las metodologías no destructivas para estimación de biomasa, aquí desarrolladas se describen tres técnicas que varios autores han propuesto para calcular los valores de biomasa y carbono, tal como el uso de ecuaciones alométricas por medio de la medición de variables dasométricas como el DAP, la aplicación de la teoría de huecos (v.g. DHP, TRAC), y la obtención de biomasa mediante información radar. Las imágenes radar proporcionan una clara ventaja al poder ser adquiridas en cualquier momento del día e independientemente de las condiciones climatológicas. Se han adquirido dos imágenes de sensores diferentes, tal como ALOSPALSAR que trabaja en la banda L y RADARSAT-2 que trabaja en la banda C, se aplica la metodología descrita por Saatchi et al. (2007), desarrollando los algoritmos semiempíricos propuestos para la estimación de biomasa del fuste (Ws) y biomasa de la copa (Wc), obteniendo los coeficientes a partir de información adquirida en campo. ABSTRACT The importance of biomass worldwide has led to more than 130 countries to celebrate the Kyoto Protocol, aimed at reducing emissions of six greenhouse gases and three fluorinated industrial gases, and the incorporation of the fixation of CO2 as an objective within forest management criteria. Among the non-destructive methods for estimating biomass, three techniques were developed. These have been described by some authors, as the use of allometric equations by measuring forest variables such as the DAP, the application of the Gap Theory (e.g. DHP, TRAC), as well as deriving biomass by radar information. The radar images provide a clear advantage since they can be gathered at any time of the day regardless of the weather conditions. For this purpose, two radar products have acquired from different sensors, such as ALOSPALSAR operating on L frequency band and RADARSAT-2 operating on C frequency band. The methodology applied in this work is described in Saatchi et al. (2007), that develop semiempirical algorithms for estimating stem biomass (Ws) and crown biomass (Wc). The corresponding coefficients are determined by means of regression procedures using field information derived from allometric and radiation measurements.

SMRT corepressor interacts with PLZF and with the PML-retinoic acid receptor α (RARα) and PLZF-RARα oncoproteins associated with acute promyelocytic leukemia

Retinoic acid receptors (RARs) are hormone-regulated transcription factors that control key aspects of normal differentiation. Aberrant RAR activity may be a causal factor in neoplasia. Human acute promyelocytic leukemia, for example, is tightly linked to chromosomal translocations that fuse novel amino acid sequences (denoted PML, PLZF, and NPM) to the DNA-binding and hormone-binding domains of RARα. The resulting chimeric receptors have unique transcriptional properties that may contribute to leukemogenesis. Normal RARs repress gene transcription by associating with ancillary factors denoted corepressors (also referred to as SMRT, N-CoR, TRAC, or RIP13). We report here that the PML-RARα and PLZF-RARα oncoproteins retain the ability of RARα to associate with corepressors, and that this corepressor association correlates with certain aspects of the leukemic phenotype. Unexpectedly, the PLZF moiety itself can interact with SMRT corepressor. This interaction with corepressor is mediated, in part, by a POZ motif within PLZF. Given the presence of POZ motifs in a number of known transcriptional repressors, similar interactions with SMRT may play a role in transcriptional silencing by a variety of both receptor and nonreceptor transcription factors.
