Através deste estudo, avaliou-se o nível de conhecimento e conduta dos farmacêuticos, responsáveis técnicos (RT) em drogarias, quanto a alguns aspectos da legislação farmacêutica e sanitária. Foram entrevistados 45 RT em 2007, sendo que 19 desconheciam a definição dos medicamentos de referência, 21 do genérico e 25 do similar. Todos sabiam da permissão de aplicação de medicamentos injetáveis. Porém, apenas dois sabiam da permissão da aplicação de penicilínicos, metade desconhecia que é permitido fracionar medicamentos fracionáveis em drogaria, 18 não sabiam dispensar corretamente uma prescrição pelo princípio ativo e 37 acreditavam que o farmacêutico pode intercambiar qualquer medicamento de marca pelo genérico. A maioria dos RT apresentou nível regular e insatisfatório de conduta e de conhecimento sobre a legislação profissional e sanitária, o que sinaliza um problema na formação acadêmica e de atualização permanente, podendo comprometer as diretrizes da política nacional de medicamentos. ABSTRACT Assessment of knowledge and behavior of pharmacists with technical responsibility for drugstores This paper is a study of the level of knowledge and conduct of the legally responsible pharmacist-in-charge (PIC) at drugstores, regarding certain aspects of pharmaceutical and health legislation. In 2007, 45 PICs were interviewed, of whom 19 did not know the definition of original (innovative) branded drugs, 21 of generic drugs and 25 of similar branded drugs. All PICs knew that it is permitted to administer injectable drugs in the drugstore. However, only two knew that penicillins can be injected, half of them did not know that certain medicines can be fractionated in drugstores, 18 did not know how to dispense correctly a prescription for an active ingredient and 37 believed that the pharmacist can swap any brand name drug with its generic counterpart. Most PICs showed an unsatisfactory level of conduct and knowledge of the professional and health legislation, which points to problems in the undergraduate and in-service training of pharmacists, that could jeopardize the national drug policy guidelines. Among the three forms of relativistic Hamiltonian dynamics proposed by Dirac in 1949, the front form has the largest number of kinematic generators. This distinction provides useful consequences in the analysis of physical observables in hadron physics. Using the method of interpolation between the instant form and the front form, we introduce the interpolating scattering amplitude that links the corresponding time-ordered amplitudes between the two forms of dynamics and provide the physical meaning of the kinematic transformations as they allow the invariance of each individual time-ordered amplitude for an arbitrary interpolation angle. We discuss the rationale for using front form dynamics, nowadays known as light-front dynamics (LFD), and present a few explicit examples of hadron phenomenology that LFD uniquely can offer from first-principles quantum chromodynamics. In particular, model-independent constraints are provided for the analyses of deuteron form factors and the N Delta transition form factors at large momentum transfer squared Q(2). The swap of helicity amplitudes between the collinear and non-collinear kinematics is also discussed in deeply virtual Compton scattering.

This work has as objective to show the vibrational analysis as a method of predictive maintenance as an auxiliar procedure in the fail detection in equipments, most specifically in the rotative ones, and with that help the maintenance team to have conditions to foresee the right time to do the swap of the components of the systems, what would lead to failures. Knowing the exact moment the resources of people and money can be focused in the critic operations to the plant. This technic has been already studied for more then 30 years and was widely used in this work, not only as an equipment condition verification method, but also, after the equipment replacement, was used to prove that the new fan was operating under the best work conditions and with that maintenance could return to contol the vibration level of the equipment, not being necessary any kind of intervention

Using the method of Social Networks Analysis, we’ll try to see if politicians swap votes between them seeking support for their projects, to this end, we’ll first map the behavior, activities and contacts of the Senators of the Brazilian Federative Republic within all the fixed committees which had some vote during the 52nd Legislature, then we can see the most popular and central Senators to each committee. After this analysis we wish to answer the following question: did logrolling exist in the Brazilian Senate? Previously the empirical analysis and response to the problems posed, we’ll present the necessary theoretical introduction understood by reviewing the literature on relevant subjects, whether they are purely theoretical with respect to the exchange of support, or analytical about the Brazilian political system.

Visual field losses associated with mercury (Hg) exposure have only been assessed in patients exposed to methylmercury. Here we evaluate the automated visual field in 35 ex-workers (30 males; 44.20+/-5.92 years) occupationaly exposed to mercury vapor and 34 controls (21 males; 43.29+/-8.33 years). Visual fields were analyzed with the Humphrey Field Analyzer II (model 750i) using two tests: the standard automated perimetry (SAP, white-on-white) and the short wavelength automated perimetry (SWAP, blue-on-yellow) at 76 locations within a 27 degrees central visual field. Results were analyzed as the mean of the sensitivities measured at the fovea, and at five successive concentric rings, of increasing eccentricity, within the central field. Compared to controls, visual field sensitivities of the experimental group measured using SAP were lower for the fovea as well as for all five eccentricity rings (p<0.05). Sensitivities were significantly lower in the SWAP test (p<0.05) for four of the five extra-foveal eccentricity rings; they were not significant for the fovea (p = 0.584) or for the 15 degrees eccentricity ring (p = 0.965). These results suggest a widespread reduction of sensitivity in both visual field tests. Previous reports in the literature describe moderate to severe concentric constriction of the visual field in subjects with methylmercury intoxication measured manually with the Goldman perimeter. The present results amplify concerns regarding potential medical risks of exposure to environmental mercury sources by demonstrating significant and widespread reductions of visual sensitivity using the more reliable automated perimetry. Red cell haemoglobin is the fundamental oxygen-transporting molecule in blood, but also a potentially tissue-damaging compound owing to its highly reactive haem groups. During intravascular haemolysis, such as in malaria and haemoglobinopathies(1), haemoglobin is released into the plasma, where it is captured by the protective acute-phase protein haptoglobin. This leads to formation of the haptoglobin-haemoglobin complex, which represents a virtually irreversible non-covalent protein-protein interaction(2). Here we present the crystal structure of the dimeric porcine haptoglobin-haemoglobin complex determined at 2.9 angstrom resolution. This structure reveals that haptoglobin molecules dimerize through an unexpected beta-strand swap between two complement control protein (CCP) domains, defining a new fusion CCP domain structure. The haptoglobin serine protease domain forms extensive interactions with both the alpha- and beta-subunits of haemoglobin, explaining the tight binding between haptoglobin and haemoglobin. The haemoglobin-interacting region in the alpha beta dimer is highly overlapping with the interface between the two alpha beta dimers that constitute the native haemoglobin tetramer. Several haemoglobin residues prone to oxidative modification after exposure to haem-induced reactive oxygen species are buried in the haptoglobin-haemoglobin interface, thus showing a direct protective role of haptoglobin. The haptoglobin loop previously shown to be essential for binding of haptoglobin-haemoglobin to the macrophage scavenger receptor CD163 (ref. 3) protrudes from the surface of the distal end of the complex, adjacent to the associated haemoglobin alpha-subunit. Small-angle X-ray scattering measurements of human haptoglobin-haemoglobin bound to the ligand-binding fragment of CD163 confirm receptor binding in this area, and show that the rigid dimeric complex can bind two receptors. Such receptor cross-linkage may facilitate scavenging and explain the increased functional affinity of multimeric haptoglobin-haemoglobin for CD163 (ref. 4).

This thesis is focused on the financial model for interest rates called the LIBOR Market Model. In the appendixes, we provide the necessary mathematical theory. In the inner chapters, firstly, we define the main interest rates and financial instruments concerning with the interest rate models, then, we set the LIBOR market model, demonstrate its existence, derive the dynamics of forward LIBOR rates and justify the pricing of caps according to the Black’s formula. Then, we also present the Swap Market Model, which models the forward swap rates instead of the LIBOR ones. Even this model is justified by a theoretical demonstration and the resulting formula to price the swaptions coincides with the Black’s one. However, the two models are not compatible from a theoretical point. Therefore, we derive various analytical approximating formulae to price the swaptions in the LIBOR market model and we explain how to perform a Monte Carlo simulation. Finally, we present the calibration of the LIBOR market model to the markets of both caps and swaptions, together with various examples of application to the historical correlation matrix and the cascade calibration of the forward volatilities to the matrix of implied swaption volatilities provided by the market.

This thesis deals with inflation theory, focussing on the model of Jarrow & Yildirim, which is nowadays used when pricing inflation derivatives. After recalling main results about short and forward interest rate models, the dynamics of the main components of the market are derived. Then the most important inflation-indexed derivatives are explained (zero coupon swap, year-on-year, cap and floor), and their pricing proceeding is shown step by step. Calibration is explained and performed with a common method and an heuristic and non standard one. The model is enriched with credit risk, too, which allows to take into account the possibility of bankrupt of the counterparty of a contract. In this context, the general method of pricing is derived, with the introduction of defaultable zero-coupon bonds, and the Monte Carlo method is treated in detailed and used to price a concrete example of contract. Appendixes: A: martingale measures, Girsanov's theorem and the change of numeraire. B: some aspects of the theory of Stochastic Differential Equations; in particular, the solution for linear EDSs, and the Feynman-Kac Theorem, which shows the connection between EDSs and Partial Differential Equations. C: some useful results about normal distribution.

The ferric uptake regulator protein Fur regulates iron-dependent gene expression in bacteria. In the human pathogen Helicobacter pylori, Fur has been shown to regulate iron-induced and iron-repressed genes. Herein we investigate the molecular mechanisms that control this differential iron-responsive Fur regulation. Hydroxyl radical footprinting showed that Fur has different binding architectures, which characterize distinct operator typologies. On operators recognized with higher affinity by holo-Fur, the protein binds to a continuous AT-rich stretch of about 20 bp, displaying an extended protection pattern. This is indicative of protein wrapping around the DNA helix. DNA binding interference assays with the minor groove binding drug distamycin A, point out that the recognition of the holo-operators occurs through the minor groove of the DNA. By contrast, on the apo-operators, Fur binds primarily to thymine dimers within a newly identified TCATTn10TT consensus element, indicative of Fur binding to one side of the DNA, in the major groove of the double helix. Reconstitution of the TCATTn10TT motif within a holo-operator results in a feature binding swap from an holo-Fur- to an apo-Fur-recognized operator, affecting both affinity and binding architecture of Fur, and conferring apo-Fur repression features in vivo. Size exclusion chromatography indicated that Fur is a dimer in solution. However, in the presence of divalent metal ions the protein is able to multimerize. Accordingly, apo-Fur binds DNA as a dimer in gel shift assays, while in presence of iron, higher order complexes are formed. Stoichiometric Ferguson analysis indicates that these complexes correspond to one or two Fur tetramers, each bound to an operator element. Together these data suggest that the apo- and holo-Fur repression mechanisms apparently rely on two distinctive modes of operator-recognition, involving respectively the readout of a specific nucleotide consensus motif in the major groove for apo-operators, and the recognition of AT-rich stretches in the minor groove for holo-operators, whereas the iron-responsive binding affinity is controlled through metal-dependent shaping of the protein structure in order to match preferentially the major or the minor groove.

Klinische Manifestationen einer Cytomegalovirus (CMV)-Infektion gefährden den therapeutischen Erfolg der hämatopoetischen Stammzelltransplantation (HSCT). Dabei stellt insbesondere die Reaktivierung von latentem CMV im HSCT-Rezipienten das häufigste Infektionsrisiko dar. Die Inzidenz der CMV-Erkrankung kann durch Rekonstitution adoptiv transferierter CMV-spezifischer CD8 T-Zellen im HSCT-Rezipienten reduziert werden. Das Modell der sogenannten adoptiven Immuntherapie wurde zunächst im murinen Modell entwickelt und bereits in klinischen Studien bestätigt. Jedoch ist der adoptive Transfer (AT) aufgrund der nur limitiert zur Verfügung stehenden therapeutisch effektiven Zellzahlen zurzeit in der klinischen Routine nicht einsetzbar.rnZiel dieser Arbeit war daher die präklinische Evaluierung einer Kombinationstherapie aus AT einer limitierten Anzahl CMV-spezifischer T-Zellen und deren in vivo Expansion durch therapeutische Vakzinierung nach HSCT. Zur Testung dieser Therapie wurde ein murines Modell auf der Grundlage von rekombinanten murinen CMV (mCMV) und rekombinanten HCMV Dense Bodies (DB) etabliert. Beide exprimieren das gut charakterisierte MHC-Klasse-I Kb-restringierte SIINFEKL-Peptid (OVA257-264) des Ovalbumins (OVA) bzw. die Funktions-verlustmutante SIINFEKA als Modellantigen. In den rekombinanten mCMV, mCMV-Δm157Luc/m164-SIINFEKL/-A (mCMV-SIINFEKL/-A), wurde mittels orthotopen Peptid-austauschs das m164257-265 Peptid des gp36,5/m164 Proteins deletiert und durch das SIINFEKL- bzw. SIINFEKA-Peptid ersetzt. Anhand von Priming-Analysen konnte gezeigt werden, dass nach Infektion von C57BL/6 Mäusen mit mCMV-SIINFEKL SIINFEKL-spezifische T-Zellen nachweisbar sind und das im CMV-Genom integrierte SIINFEKL funktional prozessiert und präsentiert wird. Parallel hierzu konnte nach Immunisierung mit DB-SIINFEKL in vivo ein SIINFEKL-spezifisches CD8 T-Zell-Priming induziert werden. In weiteren Experimenten konnte nach DB-SIINFEKL-Immunisierung im poplitealen Lymphknoten sowie in der Milz eine Proliferation von adoptiv transferierten CD8 T-Zellen beobachtet werden. Die anschließenden Challenge-Versuche zeigten, dass eine DB-SIINFEKL-Immunisierung epitopspezifisch vor einer hoch dosierten Challenge-Infektion mit mCMV-SIINFEKL schützt. Im AT-Modell konnte gezeigt werden, dass adoptiv transferierte OT-I Zellen hämatoablativ behandelte Rezipienten epitopspezifisch vor einer mCMV-SIINFEKL-Infektion schützen können, wobei der erzielte Schutz durch zusätzliche Vakzinierung mit DB-SIINFEKL deutlich verbessert werden konnte. Im Anschluss konnte im HSCT-Rezipienten erstmals eine durch zusätzliche Vakzinierung signifikante Verbesserung des protektiven Potenzials adoptiv transferierter OT-I Zellen bestätigt werden. Diese Verstärkung der Protektion ermöglicht die Reduktion der Anzahl der für den Schutz benötigten Zellzahl und erhöht damit die Effizienz der adoptiven Immuntherapie.

After the 2008 financial crisis, the financial innovation product Credit-Default-Swap (CDS) was widely blamed as the main cause of this crisis. CDS is one type of over-the-counter (OTC) traded derivatives. Before the crisis, the trading of CDS was very popular among the financial institutions. But meanwhile, excessive speculative CDSs transactions in a legal environment of scant regulation accumulated huge risks in the financial system. This dissertation is divided into three parts. In Part I, we discussed the primers of the CDSs and its market development, then we analyzed in detail the roles CDSs had played in this crisis based on economic studies. It is advanced that CDSs not just promoted the eruption of the crisis in 2007 but also exacerbated it in 2008. In part II, we asked ourselves what are the legal origins of this crisis in relation with CDSs, as we believe that financial instruments could only function, positive or negative, under certain legal institutional environment. After an in-depth inquiry, we observed that at least three traditional legal doctrines were eroded or circumvented by OTC derivatives. It is argued that the malfunction of these doctrines, on the one hand, facilitated the proliferation of speculative CDSs transactions; on the other hand, eroded the original risk-control legal mechanism. Therefore, the 2008 crisis could escalate rapidly into a global financial tsunami, which was out of control of the regulators. In Part III, we focused on the European Union’s regulatory reform towards the OTC derivatives market. In specific, EU introduced mandatory central counterparty clearing obligation for qualified OTC derivatives, and requires that all OTC derivatives shall be reported to a trade repository. It is observable that EU’s approach in re-regulating the derivatives market is different with the traditional administrative regulation, but aiming at constructing a new market infrastructure for OTC derivatives.
