Infectious diseases often hamper the production of aquatic organisms in aquaculture systems, causing economical losses, environmental problems and consumer safety issues. The conventional way aquaculture producers had to control pathogens was by means of synthetic antibiotics and chemicals. This procedure had consequences in the emergence of more resilient pathogens, drug contamination of seafood products and local ecosystems. To avoid the repercussions of antibiotic use, vaccination has greatly replaced human drugs in western fish farms. However there is still massive unregulated antibiotic use in third world fish farms, so less expensive therapeutic alternatives for drugs are desperately needed. An alternative way to achieve disease control in aquaculture is by using natural bioactive organic compounds with antibiotic, antioxidant and/or immunostimulant properties. Such diverse biomolecules occur in bacteria, algae, fungi, higher plants and other organisms. Fatty acids, nucleotides, monosaccharides, polysaccharides, peptides, polyphenols and terpenoids, are examples of these substances. One promising source of bioactive compounds are salt tolerant plants. Halophytes have more molecular resources and defence mechanisms, when compared with other tracheophytes, to deal with the oxidative stresses of their habitat. Many halophytes have been used as a traditional food and medical supply, especially by African and Asian cultures. This scientific work evaluated the antibiotic, antioxidant, immunostimulant and metal chelating properties of Atriplex halimus L., Arthrocnemum macrostachyum Moric., Carpobrotus edulis L., Juncus acutus L. and Plantago coronopus L., from the Algarve coast. The antibiotic properties were tested against Listonella anguillarum, Photobacterium damselae piscicida and Vibrio fischeri. The immunostimulant properties were tested with cytochrome c and Griess assays on Sparus aurata head-kidney phagocytes. J. acutus ether extract inhibited the growth of P. damselae piscicida. A. macrostachyum, A. halimus, C. edulis, Juncus acutus and P. coronopus displayed antioxidant, copper chelating and iron chelating properties. These plants show potential as sources of bioactive compounds with application in aquaculture and in other fields.

O Amyloodinium ocellatum é um parasita dinoflagelado que aparece em aquaculturas por todo o mundo, infectando as brânquias e o tegumento dos peixes. Esta parasita é particularmente relevante na aquacultura mediterrânica onde os surtos causam perdas consideráveis. Apesar de já terem sido experimentados vários tratamentos, nenhum deles se revelou efectivo na erradicação do parasita e ainda alguns dos químicos utilizados podem ter importantes efeitos nocivos sobre o ambiente. Deste modo, o objectivo deste estudo foi testar novos fármacos para impedir o desenvolvimento e/ou eliminar o parasita em peixes já infectados. Os fármacos utilizados neste estudo foram endoperóxidos, uma recente classe de antimaláricos. Na primeira parte deste estudo realizaram-se ensaios in vitro onde se testaram os efeitos de diferentes concentrações (0,1mM; 1 mM; 2mM e 2,5mM) dos fármacos NAD17, NAD19, LCD93 e LCD67A sobre a divisão dos tomontes. Às 24 e 48 horas de exposição a 0,1mM NAD19 e 1mM LCD93, os tomontes observados não tinham entrado em divisão. Na segunda parte realizou-se um ensaio in vivo no qual os fármacos foram administrados a juvenis de dourada (Sparus aurata) a uma concentração de 10μmol/kg através de tratamento oral. No ensaio in vivo avaliou-se o possível efeito profiláctico dos fármacos na infecção através da análise da abundância e prevalência do parasita, da eficácia dos fármacos e dos efeitos do parasita e do tratamento na fisiologia e no eixo do stress dos animais infectados através de análises enzimáticas e de parâmetros bioquímicos do sangue. As medições realizadas ao plasma sanguíneo e às brânquias de douradas com diferentes níveis de infecção de A. ocellatum revelaram que não existe diferença significativa entre os controlos e os endoperóxidos para a dose testada. Será necessário realizar mais ensaios para obter conclusões sobre o efeito dos endoperóxidos no controlo do parasita de peixes A. ocellatum.

O presente trabalho teve como objetivo avaliar a produção de peixes em dois diferentes sistemas de cultivo, policultivo e aquacultura multi-trófica integrada, tomando como referência o crescimento das espécies, o estudo patológico das brânquias e análise de conteúdo estomacal das espécies envolvidas no ensaio (Sparus aurata, Diplodus cervinus, D. puntazzo, D. sargus e D. vulgaris), e por fim foi feita uma abordagem teórica para identificar os custos envolvidos no sistema. Para realização do ensaio foram usados seis tanques, sendo dois tratamentos, em regime triplicado. Os resultados indicam uma boa adaptação dos peixes em ambos os sistemas. A biomassa inicial foi de 0,12 Kg.mˉ3, sendo formada por: 12% Sparus aurata (119,5g), 33% Diplodus puntazzo (134,7g), 45% D. sargus (90,3g), 3% D. cervinus (37,6g) e 7% D. vulgaris (93,1g) e em três tanques também com 42 Kg de Holoturia tubulosa, pepinos do mar, e 75 Kg de Crassostrea angulata, ostras. D. cervinus, D. vulgaris e D. sargus apresentaram baixa taxa de crescimento, ≤ 0,7g/dia, ambas as espécies, e Diplodus cervinus apresentou baixa taxa de sobrevivência em ambos os sistemas (57,8% poli cultivo e 73,3% IMTA). Diplodus puntazzo foi a espécie que melhor se adaptou ao sistema de policultivo simples. Sparus aurata foi a espécie que apresentou melhor taxa de crescimento e melhor taxa de sobrevivência nos dois sistemas. Os conteúdos alimentares apresentaram semelhança entre Diplodus cervinus, D. vulgaris e Sparus aurata, semelhantes também foram os itens alimentares identificados em D. sargus e D. puntazzo, entretanto a preferência alimentar das últimas espécies citadas são opostas as das três primeiras. S. aurata e D. puntazzo que apresentaram maior peso médio foram preferencialmente infestadas por Lamellodiscus spp.. Nas espécies com pesos médios inferiores, como D. cervinus, D. sargus e D. vulgaris foram maiores as infestações por Microcotyle spp.

Although overfishing is a concern for many fish stocks, it was for a long time only associated with commercial fishing exploitation, with less or no attention being given to the recreational fisheries. Recent research has shown however that the impact of recreational fishing on particular species can be considerable, and that the recreational harvest needs to be taken into account if fisheries are to be accurately assessed and effectively managed. In Portugal, the first recreational fishing regulations were only recently implemented. However, mirroring other European countries, regulations lacked scientific support, and specific knowledge of the activity was limited to a few studies with limited coverage. This thesis aimed to characterize the biological and socioeconomic aspects of the recreational shore angling activity in southern Portugal, to investigate whether the regulations in place were adequate and effective, and to provide recommendations for improved management and conservation of the inshore fisheries resources. A combined aerial-roving survey was conducted to gather data on fishing effort, catch, fishing trips and socioeconomic aspects (including anglers’ perceptions of regulations) of the recreational angling activity. The analysis of anglers’ catches suggested that compliance with daily bag limits was high, with less than 0.5% of creels exceeding the 10 kg angler-1 day-1 bag limit. Overall, 11.5% of the retained fishes were undersized, but non-compliance with minimum size limits was found to be high for some species (e.g. seabass, 73% undersized). In terms of the impact of recreational shore angling, the total estimated catches corresponded to less than 1% of the commercial landings for the same period (shared species). However, shore angling catches for white sea bream (Diplodus sargus) were found to be considerable, corresponding to 65% of the commercial landings (39.4% of total catch). In terms of anglers’ perceptions about the recreational fishing regulations in Portugal, the present study has shown that the majority of anglers accepted the existence of some kind of SRF regulations, but in general there was a partial or total disagreement with the recreational fishing restrictions recently put in place. Most anglers perceived themselves as not being involved in the decision-making process and claimed that some restrictions lacked a meaningful rationale (e.g. prohibition of fishing from piers/jetties). Fishers’ awareness with regard to specific aspects of the restrictions (such as the rationale for minimum size limits) was found to be very limited. During the same period, catches from sport fishing competitions were examined to test for differences with the recreational activity in terms of catches, and evaluate long term trends in catch and mean size of fish. Catches of the sport fishing competitions were found to be different from those observed for recreational fishing, being dominated by different species (e.g. garfish, mullets), and suggesting different fishing strategies of the the two types of anglers. High percentages of undersized fish were observed to be captured (and retained) during the competitions (in particular seabass, with 100% undersized), probably as a result of a single allowable minimum size (AMS) of 15 cm for all species in use in competitions. Lastly, catch and release fishing experiments were carried out to assess post-release mortality of three recreationally important species: two banded sea bream Diplodus vulgaris; black sea bream Spondyliosoma cantharus; and gilthead sea bream Sparus aurata. Post-release mortalities were found to be low (0-12%). The main predictor of mortality for Sparus aurata was anatomical hooking location, with 63% of the fishes that died being deeply hooked. The results support the release of fish, either from mandatory (e.g. minimum landing sizes) or voluntary practices. In summary, this thesis has demonstrated that the impact of recreational fishing for particular species is significant and needs to be taken into account for more effective management and stock assessment purposes. It has also highlighted several management issues that should be addressed in order to promote more adequate regulations in the future and prevent noncompliance issues. A periodic monitoring of the recreational fishing activity including all fishing modes (i.e. spear fishing, boat, and shore angling) would also be beneficial to ensure a timely knowledge on the global recreational fishing activity and support future management actions.

The identification of genes involved in signaling and regulatory pathways, and matrix formation is paramount to the better understanding of the complex mechanisms of bone formation and mineralization, and critical to the successful development of therapies for human skeletal disorders. To achieve this objective, in vitro cell systems derived from skeletal tissues and able to mineralize their extracellular matrix have been used to identify genes differentially expressed during mineralization and possibly new markers of bone and cartilage homeostasis. Using cell systems of fish origin and techniques such as suppression subtractive hybridization and microarray hybridization, three genes never associated with mechanisms of calcification were identified: the calcium binding protein S100-like, the short-chain dehydrogenase/reductase sdr-like and the betaine homocysteine S-methyltransferase bhmt3. Analysis of the spatial-temporal expression of these 3 genes by qPCR and in situ hybridization revealed: (1) the up-regulation of sdr-like transcript during in vitro mineralization of gilthead seabream cell lines and its specificity for calcified tissues and differentiating osteoblasts; (2) the up-regulation of S100-like and the down-regulation of bhmt3 during in vitro mineralization and the central role of both genes in cartilaginous tissues undergoing endo/perichondral mineralization in juvenile fish. While expression of S100-like and bhmt3 was restricted to calcified tissues, sdr-like transcript was also detected in soft tissues, in particular in tissues of the gastrointestinal tract. Functional analysis of gene promoters revealed the transcriptional regulation of the 3 genes by known regulators of osteoblast and chondrocyte differentiation/mineralization: RUNX2 and RAR (sdr-like), ETS1 (s100-like; bhmt3), SP1 and MEF2c (bhmt3). The evolutionary relationship of the different orthologs and paralogs identified within the scope of this work was also inferred from taxonomic and phylogenetic analyses and revealed novel protein subfamilies (S100-like and Sdr-like) and the explosive diversity of Bhmt family in particular fish groups (Neoteleostei). Altogether our results contribute with new data on SDR, S100 and BHMT proteins, evidencing for the first time the role for these three proteins in mechanisms of mineralization in fish and emphasized their potential as markers of mineralizing cartilage and bone in developing fish.

This study investigated the effects of stocking density on the growth and fatty acid (FA) of Brycon insignis metabolism. Fingerlings (360) were distributed into eight ponds at two stocking densities (105 and 210 g/m(3)). The analysis of growth showed that the condition factor (K) and the coefficient of variation (CV) for body mass were not affected by stocking density. However, final body mass and length, specific growth rate (SGR), and weight gain (WG) were higher in the low stocking density group, which also presented a higher feed efficiency (FE) and survival (S). By contrast, muscle protein levels were higher in the high stocking density group. The plasma and muscle lipid content were not affected by stocking density, but fish reared at lower stocking density presented higher lipid concentration in the liver, with no differences in hepatosomatic index values. Even with the differences observed in metabolic and growth parameters, plasma cortisol was not affected by stocking density. The FA profile in the muscle and liver neutral fraction were not affected by stocking density, but the FA in the polar fractions differed between the two stocking densities. In the liver, total polyunsaturated fatty acids (PUFA) and PUFA n - 3 increased in higher stocking density, mainly due to an increase in docosahexaenoic acid (DHA). In addition, PUFA n - 6 were also increased in the higher stocking density group, mainly due to an increase in arachidonic acid (AA) and docosadienoic acid (22:2n - 6). In the muscle polar fraction, the saturated fatty acids (SFA) and monounsaturated fatty acids (MUFA) decreased in the animals from the higher stocking density group, and this reduction was compensated by an increase in PUFA n - 3 and PUFA n - 6, mainly the FA with 20-22 carbons (20:4n - 6: 22:4:n - 6; 22:5n - 6, 22:5n - 3, and 22:6n - 3). A different profile was observed for the C18 PUFAs, mainly 18:2n - 6 and 18:4n - 6, which were higher in the lower density stocking group. The data suggest that when living in high stocking density, B. insignis differentially utilizes the hepatic lipids as energy source and remodels the membrane fatty acids, with higher amounts of DHA in the polar muscle fraction compensated for by a decrease in MUFA. The adenohypophysis (AH) of juvenile pirarucu (Arapaima gigas), a representative species of the Osteoglossomorpha (bonytongue fishes, one of the oldest living groups of the teleosts), was studied using histochemical and immunocytochemical methods. The AH is comprised of the pars distalis (PD), without a clear distinction between rostral pars distalis (RPD) and proximal pars distalis (PPD), and the pars intermedia (PI). The neurohypophysis (NH) is positioned on top of the PD and penetrates and branches into the PI. In the most rostral dorsal portion of the PD, adrenocorticotropic cells and fusiform gonadotropic cells were found. In the central PD, scarce prolactin-producing cells and growth-hormone-producing cells were located mainly in the dorsal part, whereas round gonadotropic cells were abundant in the ventral portion of this region. Human thyrotropin immunoreactive cells were not found in the entire AH. In the PI, melanotropic, some adrenocorticotropic, and somatolactin-producing cells were located intermingled surrounding the neurohypophyseal branches. Our results showed that the A. gigas pituitary has some basal characteristics between the ancient Actinopterygii and the more derived teleosts.

An experiment was conducted with barramundi (Lates calcarifer) juveniles to examine the marginal efficiency of utilisation of long chain-polyunsaturated fatty acids (LC-PUFA). A series of five diets with blends of fish (anchovy) oil and poultry fat (F100:P0, F60:P40, F30:P70, F15:P85, F0:P100) were fed to 208. ±. 4.1. g fish over a 12-week period. The replacement of fish oil with poultry fat had no impact on growth performance (average final weight of 548.3. ±. 10.2. g) or feed conversion (mean = 1.14. ±. 0.02). Analysis of the whole body composition showed that the fatty acid profile reflected that of the fed diet. However it was also shown that there was a disproportional retention of some fatty acids relative to others (notably LOA, 18:2n-6 and LNA, 18:3n-3). By examining the body mass independent retention of different fatty acids with differential levels of intake of each, the marginal efficiencies of the use of these nutrients by this species were able to be determined. The differential retention of fatty acids in the meat was also examined allowing the determination of oil blending strategies to optimise meat n-3 LC-PUFA levels.

This study evaluated the effects of two lipids sources of fish residue (tilapia and salmon) compared with a vegetable oil source (soybean oil) on the fatty acid profiles of male and female lambari. This experiment was developed in a completely randomized experimental design in a 3 x 2 factorial arrangement, totaling 6 treatments resulting from the combination of the three experimental diets for both sexes, with four replications for each treatment. This study involved 120 male (2.58 +/- 0.13 g) and 72 female lambari (4.00 +/- 0.09 g), fed the experimental diets twice a day until apparent satiation for a period of 60 days. Oleic, linoleic, palmitic and stearic fatty acids were found at higher concentrations in all experimental oils and diets, as well in the muscle of male and female lambari. The low amounts of arachidonic, eicosapentaenoic and docosahexaenoic acids in the experimental diets and subsequent greater concentrations in muscle tissue, suggested that lambari are able to desaturate and elongate the chain of fatty acids with 18 carbons. The fish of both sexes that received the diet with soybean oil showed high levels of n-6 fatty acids, especially of C18: 2n-6 and low levels of eicosapentaenoic and docosahexaenoic acids. The diet with salmon residue oil promoted higher levels of fatty acids of the n-3 series and resulted in the best n-3/n-6 ratio in the muscle of male and female lambari. The oils from fish residues can be a substitute for traditional fish oil and its use in the lambari diets does not impair its growth.

The objective of this work was to evaluate corn gluten meal (CGM) as a substitute for fish meal in diets for striped catfish (Pseudoplatystoma fasciatum) juveniles. Eight isonitrogenous (46% crude protein) and isoenergetic (3,450 kcal kg(-1) digestible energy) diets, with increasing levels of CGM - 0, 6, 12, 18, 24, 30, 36, and 42%-, were fed to juvenile striped catfish (113.56 +/- 5.10 g) for seven weeks. Maximum values for weight gain, specific growth rate, protein efficiency ratio and feed conversion ratio, evaluated by polynomial quadratic regression, were observed with 10.4, 11.4, 15.4 and 15% of CGM inclusion, respectively. Feed intake decreased significantly from 0.8% CGM. Mesenteric fat index and body gross energy decreased linearly with increasing levels of CGM; minimum body protein contents were observed with 34.1% CGM. Yellow pigmentation of fillets significantly increased until 26.5% CGM, and decreased from this point forth. Both plasma glucose and protein concentrations decreased with increased CGM levels. The inclusion of 10-15% CGM promotes optimum of striped catfish juveniles depending on the parameter evaluated. Yellow coloration in fillets produced by CGM diets can have marketing implications.

[ES] El jurel dentón Pseudocaranx dentex (Bloch & Schneider, 1801) es una especie habitualmente asociada al entorno de instalaciones comerciales de engorde de doradas Sparus aurata L., 1758 y lubinas Dicentrarchus labrax (L., 1758) en jaulas flotantes en las islas Canarias. Con el objetivo de conocer el potencial de esta especie para la acuicultura se ha llevado a cabo un estudio de su ciclo reproductivo anual. Durante un año se han recogido muestras de gónadas, evaluado variables morfométricas a partir de especímenes salvajes y registrado los valores de temperatura y fotoperiodo en el momento de los muestreos. Los análisis macroscópicos y microscópicos realizados de las gónadas, junto con la determinación mensual del índice gonadosomático (IGS) y del factor de condición (FC), han servido para establecer el ciclo anual de maduración de esta especie. El estadio de vitelogénesis avanzada fue observado entre mayo y noviembre, sugiriendo un prolongado periodo de desove para esta especie en aguas de las islas Canarias.

La acuicultura es uno de los sectores con mayor crecimiento en la producción animal mundial, con una tasa superior al 5% anual en la última década (Izquierdo et al., 2008). En la acuicultura, el éxito del cultivo de cualquier especie de peces marinos está limitado por la cantidad y la calidad de la producción en masa de sus alevines (Izquierdo y Fernández - Palacios, 1997). Por lo tanto, para cubrir las crecientes demandas de la producción de semillas, es necesario mejorar la calidad nutritiva de sus larvas, lo cual todavía constituye una de las principales limitaciones para el desarrollo del cultivo de especies de peces marinos (Watanabe et al., 1983; Yúfera y Pascual, 1984; Sargent et al. 1997; Izquierdo et al., 2000). Aunque la producción Mediterránea del cultivo de peces marinos se ha incrementado en varias especies, la dorada Sparus aurata continúa siendo la especie mas cultivada (Izquierdo, 2005), con una producción anual de alevines que supera los 120 millones/ año. La demanda de alevines de buena calidad está aumentando a un ritmo del 10% anual, pero el éxito de la producción de los juveniles se ve muy afectado por la eficacia de la primera alimentación y la calidad nutricional de la dieta de arranque (Kolkovski et al., 1993; Sargent et al., 1997; Izquierdo et al., 2000). En general, la dorada y la lubina europea (Dicentrarchus labrax) son las especies más importantes de peces marinos criados en la región mediterránea y han caracterizado el desarrollo de la acuicultura marina en esta región en las últimas tres décadas (FAO, 1999). Además, en la producción, de ambas especies, se prevee una mayor expansión (Basurco y Abellán, 1999). Sin embargo, a pesar de que el engorde de estas especies está bien controlado, el conocimiento de sus necesidades nutricionales, en comparación con otras especies como salmónidos y carpas, sigue siendo incompleto (NRC, 1993). Por lo tanto, para obtener un mejor crecimiento y una mayor tasa de supervivencia, es esencial una dieta que responda a las necesidades nutricionales de las larvas, tanto cualitativas como cuantitativas (Kolkovski et al., 1993; Sargent et al., 1997). Además, incluso antes de que comience el desarrollo larvario, los huevos de los peces deben contener todos los nutrientes que cubran los requerimientos adecuados para el desarrollo del embrión (Izquierdo y Fernández - Palacios, 1997). En los últimos años, las investigaciones han prestado gran atención a la importancia de los lípidos dietéticos para larvas de peces marinos (Izquierdo et al., 2003), puesto que son esenciales para el crecimiento y el desarrollo de los mismos (Watanabe, 1982; Sargent et al., 1999a). Como reflejo de esta importancia, la dieta para dorada y lubina se ha convertido en una dieta altamente energética (25% de lípidos) en comparación con la década anterior (12% de lípidos) (Izquierdo et al., 2003). Los lípidos dietéticos proporcionan una fuente rica de energía y fosfolípidos y son fundamentales para la estructura de las biomembranas. Los lípidos dietéticos también sirven como vehículos para la absorción de otros nutrientes, como las vitaminas liposolubles A, D, E, y K, y pigmentos naturales o sintéticos. Los lípidos son componentes de las hormonas y precursores para la síntesis de diversos metabolitos funcionales, como las prostaglandinas y otros eicosonoides. Además, los lípidos dietéticos son reconocidos como uno de los factores nutricionales más importantes que afectan el crecimiento y la supervivencia de las larvas (Watanabe et al., 1983), porque constituyen materiales esenciales para la formación normal de célula, las membranas de los tejidos y el desarrollo de órganos (Izquierdo et al., 1998, 2003; Pousaò et al., 2003). Sin embargo, la utilización de los lípidos dietéticos por las larvas puede verse afectada directa o indirectamente por varios cambios morfológicos y fisiológicos que ocurren durante el desarrollo larvario. En los últimos años ha habido más interés en todos estos aspectos nutricionales de los lípidos en las larvas de peces, debido a la importancia de utilización de los lípidos dieteticos para el óptimo crecimiento y supervivencia larvaria (Izquierdo et al., 2000).
