Computer simulation is a powerful tool to predict microstructure and its evolution in dynamic and post-dynamic recrystallization. CAFE proposed as an appropriate approach by combining finite element (FE) method and cellular automata (CA) for recrystallization simulation. In the current study, a random grid cellular automaton (CA), as micro-scale model, based on finite element (FE), as macro-scale method, has been used to study initial and evolving microstructural features; including nuclei densities, dislocation densities, grain size and grain boundary movement during dynamic recrystallization in a C-Mn steel. An optimized relation has been established between mechanical variables and evolving microstructure features during recrystallization and grain growth. In this model, the microstructure is defined as cells located within grains and grain boundaries while dislocations are randomly dispersed throughout microstructure. Changes of dislocation density during deformation are described considering hardening, recovery and recrystallization. Recrystallization is assumed to initiate near grain boundaries and nucleation rate was considered constant (site-saturated condition). The model produced a mathematical formulation which captured the initial and evolving microstructural entities and linked their effects to measurable macroscopic variables (e.g. stress).

Simulation of materials processing has to face new difficulties regarding proper description of various discontinuous and stochastic phenomena occurring in materials. Commonly used rheological models based on differential equations treat material as continuum and are unable to describe properly several important phenomena. That is the reason for ongoing search for alternative models, which can account for non-continuous structure of the materials and for the fact, that various phenomena in the materials occur in different scales from nano to mezo. Accounting for the stochastic character of some phenomena is an additional challenge. One of the solutions may be the coupled Cellular Automata (CA) – Finite Element (FE) multi scale model. A detailed discussion about the advantages given by the developed multi scale CAFE model for strain localization phenomena in contrast to capabilities provided by the conventional FE approaches is a subject of this work. Results obtained from the CAFE model are supported by the experimental observations showing influence of many discontinuities existing in the real material on macroscopic response. An immense capabilities of the CAFE approach in comparison to limitations of the FE method for modeling of real material behavior is are shown this work as well.

An investigation of the application of a multi scale CAFE model to prediction of the strain localization phenomena in industrial processes, such as extrusion, is presented in this work. Extrusion involves the formation of a strong strain localization zone, which influences the final product microstructure and may lead to a coarse grain layer close to the surface. Modelling of the shape of this zone and prediction of the strain magnitude will allow computer aided design of the extrusion process and optimisation of the technological parameters with respect to the microstructure and properties of the products. Thus, the particular objective of this work is comparison of the FE and CAFE predictions of strain localization in the shear zone area in extrusion. Advantages and disadvantages of the developed CAFE model are also discussed on the basis of the simulation results.

Abstract A detailed description of possibilities given by the developed Cellular Automata—Finite Element (CAFE) multi scale model for prediction of the initiation and propagation of micro shear bands and shear bands in metallic materials subjected to plastic deformation is presented in the work. Particular emphasis in defining the criterion for initiation of micro shear and shear bands, as well as in defining the transition rules for the cellular automata, is put on accounting for the physical aspects of these phenomena occurring in two different scales in the material. The proposed approach led to the creation of the real multi scale model of strain localization phenomena. This model predicts material behavior in various thermo-mechanical processes. Selected examples of applications of the developed model to simulations of metal forming processes, which involve strain localization, are presented in the work. An approach based on the Smoothed Particle Hydrodynamic, which allows to overcome difficulties with remeshing in the traditional CAFE method, is a subject of this work as well. In the developed model remeshing becomes possible and difficulties limiting application of the CAFE method to simple deformation processes are solved. Scanning tunneling microscopy (STM) images of 1,10-phenanthroline (PHEN) and dipyrido[3,2-a:2‘,3‘-c]phenazine (DPPZ) on Au(111) are recorded using both in situ and ex situ techniques. The images of PHEN depict regimes of physisorption and chemisorption, whereas DPPZ is only physisorbed. All physisorbed structures are not pitted and fluctuate dynamically, involving aligned (4 × 4) surface domains with short-range (ca. 20 molecules) order for PHEN but unaligned chains with medium-range (ca. 100 molecules) order for DPPZ. In contrast, the chemisorbed PHEN monolayers remain stable for days, are associated with surface pitting, and form a (4 × √13)R46° lattice with long-range order. The density of pitted atoms on large gold terraces is shown to match the density of chemisorbed molecules, suggesting that gold adatoms link PHEN to the surface. For PHEN, chemisorbed and physisorbed adsorbate structures are optimized using plane-wave density-functional theory (DFT) calculations for the surface structure. Realistic binding energies are then obtained adding dispersive corrections determined using complete-active-space self-consistent field calculations using second-order perturbation theory (CASPT2) applied to cluster-interaction models. A fine balance between the large adsorbate−adsorbate dispersive forces, adsorbate−surface dispersive forces, gold ligation energy, and surface mining energy is shown to dictate the observed phenomena, leading to high surface mobility and substrate/surface lattice incommensurability. Increasing the magnitude of the dispersive forces through use of DPPZ, rather than PHEN, to disturb this balance produced physisorbed monolayers without pits and/or surface registration but with much longer-range order. Analogies are drawn with similar but poorly understood processes involved in the binding of thiols to Au(111).

Esta dissertação propõe analisar o programa das Bibliotecas Parque do Governo do Estado do Rio de Janeiro por meio do estudo de caso da biblioteca da Rocinha, terceiro equipamento cultural deste gênero inaugurado no segundo mandato da gestão do governador Sérgio Cabral (2011-2014) e o primeiro a ser instalado dentro de uma favela carioca. Entre os objetivos específicos deste trabalho estão entender como se deu o processo de negociação do poder público com lideranças da comunidade para a implementação da biblioteca na Rocinha; identificar as práticas dos usuários do equipamento público e as percepções destes usuários em relação ao espaço cultural bem como compreender a dinâmica de funcionamento da unidade, levando em consideração aspectos como planejamento versus demanda, usos não programados, conflitos e as alternativas criadas a partir do entendimento da realidade dos usuários e de suas necessidades, já que a biblioteca é essencialmente um equipamento cultural em construção permanente.

O café é um dos principais produtos agrícolas, sendo considerado o segundo item em importância do comércio internacional de commodities. O gênero Coffea pertence à família Rubiaceae que também inclui outras plantas importantes. Este gênero contém aproximadamente 100 espécies, mas a produção comercial é baseada somente em duas espécies, Coffea arabica e Coffea canephora, que representam aproximadamente 70 % e 30 % do mercado total de café, respectivamente. O Projeto Genoma Café Brasileiro foi desenvolvido com o objetivo de disponibilizar os modernos recursos da genômica à comunidade científica e aos diferentes segmentos da cadeia produtiva do café. Para isso, foram seqüenciados 214.964 clones escolhidos aleatoriamente de 37 bibliotecas de cDNA de C. arabica, C. canephora e C. racemosa representando estádios específicos do desenvolvimento de células e de tecidos do cafeeiro, resultando em 130.792, 12.381 e 10.566 seqüências de cada espécie, respectivamente, após processo de trimagem. Os ESTs foram agrupados em 17.982 contigs e em 32.155 singletons. A comparação destas seqüências pelo programa BLAST revelou que 22 % não tiveram nenhuma similaridade significativa às seqüências no banco de dados do National Center for Biotechnology Information (de função conhecida ou desconhecida). A base de dados de ESTs do cafeeiro resultou na identificação de cerca de 33.000 unigenes diferentes. Os resultados de anotação das seqüências foram armazenados em base de dados online em cafe>http://www.lge.ibi.unicamp.br/cafe. Os recursos desenvolvidos por este projeto disponibilizam ferramentas genéticas e genômicas que podem ser decisivas para a sustentabilidade, a competitividade e a futura viabilidade da agroindústria cafeeira nos mercados interno e externo.

Strains of Xylella fastidiosa, isolated from sweet orange trees (Citrus sinensis) and coffee trees (Coffea arabica) with symptoms of citrus variegated chlorosis and Requeima do Cafe, respectively, were indistinguish able based on repetitive extragenic palindromic polymerase chain reaction (PCR) and enterobacterial repetitive intergenic consensus PCR assays. These strains were also indistinguishable with a previously described PCR assay that distinguished the citrus strains from all other strains of Xylella fastidiosa. Because we were not able to document any genomic diversity in our collection of Xylella fastidiosa strains isolated from diseased citrus, the observed gradient of increasing disease severity from southern to northern regions of São Paulo State is unlikely due to the presence of significantly different strains of the pathogen in the different regions. When comparisons were made to reference strains of Xylella fastidiosa isolated from other hosts using these methods, four groups were consistently identified consistent with the hosts and regions from which the strains originated: citrus and coffee, grapevine and almond, mulberry, and elm, plum, and oak. Independent results from random amplified polymorphic DNA (RAPD) PCR assays were also consistent with these results; however, two of the primers tested in RAPD-PCR were able to distinguish the coffee and citrus strains. Sequence comparisons of a PCR product amplified from all strains of Xylella fastidiosa confirmed the presence of a CfoI polymorphism that can be used to distinguish the citrus strains from all others. The ability to distinguish Xylella fastidiosa strains from citrus and coffee with a PCR-based assay will be useful in epidemiological and etiological studies of this pathogen.

This study was undertaken to investigate the toxic effect of Crotalaria spectabilis seeds added to the ration for commercial broilers during the final phase of growth. Ground seeds were added at different concentrations to the ration: 0.0% (control), 0.01%, 0.1% and 0.4%. Rations containing 0.4% caused symptoms of intoxication, beginning in the second week of the study. During the third week birds showed bristling, apathy, general weakness, distended abdomen and agglomeration. In the fourth week, four animals died. Necropsy revealed prominent ascites and severe lesions of liver, kidney and lung. Microscopic examination revealed necrosis of hepatocytes, inflammatory cell infiltration, hypertrophy and hyperplasia of biliary duct cells leading to atresia and cholestasis. Prominent cartilaginous and osseous nodules in the lungs were also present as well as degenerative, changes in the kidney tubules and necrosis of cells of the bursa. All the broilers receiving a ration with 0.1% of Crotalaria seeds showed ascites and slight lesions of the liver. The remaining groups showed no reduction in weight gains, lesions or clinical symptoms. It is concluded that broilers during the final phase of growth are sensitive to administrations of seeds of C. spectabilis in their ration. The presence of ascites and cartilaginous and osseous nodules in the lungs of the affected birds was also considered important.

From February to November 2004, 670 samples of bovine mastitic milk from which 231 Escherichia coli strains were isolated, were collected from two Brazilian states. The strains were screened for the presence of Shiga toxin-producing (stx 1 and stx 2) and intimin (eae) genes. Twenty (8.6%) strains were detected by PCR to harbor the Shiga toxin genes (8 the stx 1 gene, 12 the stx 2 gene and none both of them). Two (0.8%) of the Escherichia coli strains studied were eae positive non Shiga toxin-producing. The strains were also examined for resistance to 12 antimicrobial agents. The predominantly observed resistance was to tetracycline (92.2%), streptomycin (90.4%), nalidixic acid (88.3%), amikacin (86.5%) and cephalothin (84.8%). Multidrug resistance was found among 152 isolates (65.8%).
