Persistent organic pollutants (POPS) present in the living environment are thought to have detrimental health effects on the population, with pregnant women and the developing foetus being at highest risk. We report on the levels of selected POPs in maternal blood of 155 delivering women residing in seven regions within the Sao Paulo State, Brazil. The following selected POPs were measured in the maternal whole blood: 12 polychlorinated biphenyls (PCBs) congeners (IUPAC Nos. 99, 101, 118, 138, 153, 156, 163, 170, 180, 183, 187, 194); dichlordiphenyltrichloroethane p,p'-DDT, diphenyldichloroethylene p,p'-DDE and other pesticides such as hexachlorocyclohexanes (alpha-HCH, beta-HCH, gamma-HCH), hexachlorobenzene (HCB), chlordane derivatives cis-chlordane, trans-chlordane. oxy-chlordane, cis-nonachlor and trans-nonachlor. Statistical comparisons between regions were performed only on compounds having concentrations above LOD in 70% of the samples. PCB118 congener was found to be highest in the industrial site (mean 4.97 ng/g lipids); PCB138 congener concentration was highest in the Urban 3 site (mean 4.27 ng/g lipids) and congener PCB153 was highest in the industrial and Urban 3 sites with mean concentration of 7.2 ng/g lipids and 5.89 ng/g lipids respectively. Large differences in levels of p,p'-DDE between regions were observed with the Urban 3 and industrial sites having the highest concentrations of 645 ng/g lipids and 417 ng/g lipids, respectively; beta-HCH was found to be highest in the Rural 1 site; the gamma-HCH in Rural 1 and industrial; the HCB in the Rural 1 and industrial sites and oxy-chlordane and t-NC in the Rural 2 sites. As land is developed, the impervious surfaces that are created increase the amount of runoff during rainfall events, disrupting the natural hydrologic cycle, with an increment in volume of runoff and in pollutant loadings. Pollutants deposited or derived from an activity on the land surface will likely end up in stormwater runoff in some concentration, such as nutrients, sediment, heavy metals, hydrocarbons, gasoline additives, pathogens, deicers, herbicides and pesticides. Several of these pollutants are particulate-bound, so it appears clear that sediment removal can provide significant water-quality improvements and it appears to be important the knowledge of the ability of stromwater treatment devices to retain particulate matter. For this reason three different units which remove sediments have been tested through laboratory. In particular a roadside gully pot has been tested under steady hydraulic conditions, varying the characteristics of the influent solids (diameter, particle size distribution and specific gravity). The efficiency in terms of particles retained has been evaluated as a function of influent flow rate and particles characteristics; results have been compared to efficiency evaluated applying an overflow rate model. Furthermore the role of particles settling velocity in efficiency determination has been investigated. After the experimental runs on the gully pot, a standard full-scale model of an hydrodynamic separator (HS) has been tested under unsteady influent flow rate condition, and constant solid concentration at the input. The results presented in this study illustrate that particle separation efficiency of the unit is predominately influenced by operating flow rate, which strongly affects the particles and hydraulic residence time of the system. The efficiency data have been compared to results obtained from a modified overflow rate model; moreover the residence time distribution has been experimentally determined through tracer analyses for several steady flow rates. Finally three testing experiments have been performed for two different configurations of a full-scale model of a clarifier (linear and crenulated) under unsteady influent flow rate condition, and constant solid concentration at the input. The results illustrate that particle separation efficiency of the unit is predominately influenced by the configuration of the unit itself. Turbidity measures have been used to compare turbidity with the suspended sediments concentration, in order to find a correlation between these two values, which can allow to have a measure of the sediments concentration simply installing a turbidity probe.

La inundación en áreas urbanas por sobrecarga de las redes de drenaje es un problema recurrente de importancia creciente. Las cubiertas vegetadas (naturadas) retienen parte de la lámina de agua precipitada, reduciendo el escurrimiento superficial y generando hidrogramas de escorrentía directa con caudales pico menores y más retardados. Estas propiedades hacen que esta tecnología pueda contribuir a reducir la sobrecarga de cauces urbanos. Los resultados obtenidos a lo largo de casi dos años de estudio permitieron estimar una capacidad de retención de las cubiertas ensayadas (en la Ciudad de Buenos Aires), que ha sido variable en función de la precipitación, del grado de cobertura y profundidad del sustrato. Es así que el porcentaje retenido ha sido alto (73% a 100%) con precipitaciones menores o iguales a los 20 mm, alrededor del 60% con lluvias de 35 a 40 mm, y con precipitaciones cercanas a 100 mm los porcentajes de retención se redujeron notablemente, alcanzando valores cercanos al 30%. Estos resultados posicionan las cubiertas vegetadas, para las condiciones y sitio del ensayo, como una alternativa dentro del manejo hídrico integrado en cuencas urbanas.

En el levante español se producen periódicamente intensos aguaceros (gota fría), que generan grandes riadas, desbordándose los cauces y asolando los campos y las ciudades, ocasionando graves daños en la economía local y con frecuencia, por desgracia, víctimas personales. La Confederación Hidrográfica del Segura ha puesto en marcha un vasto sistema de protección, conocido como Plan de Defensa contra Avenidas, del que ya se encuentran en explotación un conjunto de presas cuyo objetivo prioritario es la laminación de las avenidas. Las presas de laminación ubicadas en esta área mediterránea son estructuras muy singulares, al estar ubicadas en cuencas extremadamente secas, con alta pendiente, rápida respuesta, picos altos de caudal, y gran poder erosivo del flujo. Las aguas arrastran en suspensión un alto porcentaje de aporte sólido y flotantes, que quedan retenidos en el embalse, aterrándolo e inutilizando los desagües. En esta tesis doctoral se analizan los criterios de actuación para la gestión de estas presas de laminación, con objeto de que se puedan introducir en las Normas de Explotación innovaciones destinadas a mejorar la eficiencia del conjunto, de manera que se consiga la máxima protección posible y al mismo tiempo se puedan aprovechar las aguas, sin menoscabo de su objetivo prioritario, para otros fines complementarios. Como consecuencia del trabajo realizado se indica la conveniencia de establecer un conjunto de normas de actuación comunes a todas las presas del sistema, como es la estrategia de recibir siempre a los hidrogramas de la riada con las compuertas de los desagües de fondo cerrados, o la conveniencia de que en todas las presas se dispongan compuertas en estos órganos. También se dictan recomendaciones en la gestión del vaciado, para conseguir el máximo aprovechamiento del agua, introduciendo el concepto innovador de los trasvases virtuales. Asimismo, se analiza la conveniencia de fijar normas diferenciales específicas atendiendo a las peculiaridades de cada presa, en cuanto se refiere al nivel máximo de explotación, al cumplimiento de los condicionantes ambientales, al mantenimiento de la calidad del agua, y a otros aspectos singulares que caracterizan a cada uno de estos embalses. The Spanish Mediterranean region hydrology is characterised by low rainfall, but concentrated in extreme convective events. Those heavy rains and storms lead to large, flash-floods which cause devastation of fields, villages and, unfortunately, loss of lives. Segura River Basin Authority has developed an ambitious plan for flood protection and control that is constituted by a system of flood protection dams, river channelling, river diversions diversions and levees. Flood protection dams placed on this area, are distinct structures, as they are located on extremely dry basins, with high slopes and limited vegetal covering. These facts, along with the very high rainfall intensity, are determinants of a rapid response and a very erosive flow. The flow transports a large rate of solids in suspension, and also debris, which many times block and clog the outlets intakes, disabling them. In such a case, the dam would be inoperative, in an emergency state and remedial measures should be taken. This PhD thesis analyses the flood protection dams operation criteria, for providing managers a methodology to analyse each individual case and also the flood protection system as a whole. Additionally practical rules and recommendations, to cope with the objectives of dam safety, flood protection and water resources use, are given. The rules consider the specific differences and risks that may be faced by the dams. To that end, they were grouped, depending on their characteristics: allowance for permanent water storage, environmental restrictions, water quality, geotechnical problems, type of structure, etc. As a result of this research, it is recommended to design these dams with valves for regulating the bottom outlets. The availability of valves is linked with the routing strategy of receiving flood with the outlets closed. This has been shown as the best measure to prevent clogging and opens the possibility of storing temporarily the water, making then a scheduled emptying of the reservoir, which could be computed as a virtual transfer to the regulating dams. Emptying should be done in accordance with the situation of the other dams of the system and of the rivers downstream. This detention time would make possible also the beginning of the sedimentation process which is necessary for using the water with the modern irrigation systems that are in use in this area.

El 5º Informe del IPCC (Panel Intergubernamental de Cambio Climático, 2014) señala que el turismo será una de las actividades económicas que mayores efectos negativos experimentará en las próximas décadas debido al calentamiento térmico del planeta. En España, el turismo es una fuente principal de ingresos y de creación de puestos de trabajo en su economía. De ahí que sea necesaria la puesta en marcha de medidas de adaptación a la nueva realidad climática que, en nuestro país, va a suponer cambios en el confort climático de los destinos e incremento de extremos atmosféricos. Frente a los planes de adaptación al cambio climático en la actividad turística, elaborados por los gobiernos estatal y regional, que apenas se han desarrollado en España, la escala local muestra interesantes ejemplos de acciones de adaptación al cambio climático, desarrolladas tanto por los municipios (energía, transporte, vivienda, planificación urbanística) como por la propia empresa turística (hoteles, campings, apartamentos). Medidas de ahorro de agua y luz, fomento del transporte público y de las energías limpias, creación de zonas verdes urbanas y adaptación a los extremos atmosféricos destacan como acciones de mitigación del cambio climático en los destinos turísticos principales de nuestro país.

Strong convective events can produce extreme precipitation, hail, lightning or gusts, potentially inducing severe socio-economic impacts. These events have a relatively small spatial extension and, in most cases, a short lifetime. In this study, a model is developed for estimating convective extreme events based on large scale conditions. It is shown that strong convective events can be characterized by a Weibull distribution of radar-based rainfall with a low shape and high scale parameter value. A radius of 90km around a station reporting a convective situation turned out to be suitable. A methodology is developed to estimate the Weibull parameters and thus the occurrence probability of convective events from large scale atmospheric instability and enhanced near-surface humidity, which are usually found on a larger scale than the convective event itself. Here, the probability for the occurrence of extreme convective events is estimated from the KO-index indicating the stability, and relative humidity at 1000hPa. Both variables are computed from ERA-Interim reanalysis. In a first version of the methodology, these two variables are applied to estimate the spatial rainfall distribution and to estimate the occurrence of a convective event. The developed method shows significant skill in estimating the occurrence of convective events as observed at synoptic stations, lightning measurements, and severe weather reports. In order to take frontal influences into account, a scheme for the detection of atmospheric fronts is implemented. While generally higher instability is found in the vicinity of fronts, the skill of this approach is largely unchanged. Additional improvements were achieved by a bias-correction and the use of ERA-Interim precipitation. The resulting estimation method is applied to the ERA-Interim period (1979-2014) to establish a ranking of estimated convective extreme events. Two strong estimated events that reveal a frontal influence are analysed in detail. As a second application, the method is applied to GCM-based decadal predictions in the period 1979-2014, which were initialized every year. It is shown that decadal predictive skill for convective event frequencies over Germany is found for the first 3-4 years after the initialization.

Extreme weather events related to deep convection are high-impact critical phenomena whose reliable numerical simulation is still challenging. High-resolution (convection-permitting) modeling setups allow to switch off physical parameterizations accountable for substantial errors in convection representation. A new convection-permitting reanalysis over Italy (SPHERA) has been produced at ARPAE to enhance the representation and understanding of extreme weather situations. SPHERA is obtained through a dynamical downscaling of the global reanalysis ERA5 using the non-hydrostatic model COSMO at 2.2 km grid spacing over 1995-2020. This thesis aims to verify the expectations placed on SPHERA by analyzing two weather phenomena that are particularly challenging to simulate: heavy rainfall and hail. A quantitative statistical analysis over Italy during 2003-2017 for daily and hourly precipitation is presented to compare the performance of SPHERA with its driver ERA5 considering the national network of rain gauges as reference. Furthermore, two extreme precipitation events are deeply investigated. SPHERA shows a quantitative added skill over ERA5 for moderate to severe and rapid accumulations in terms of adherence to the observations, higher detailing of the spatial fields, and more precise temporal matching. These results prompted the use of SPHERA for the investigation of hailstorms, for which the combination of multiple information is crucial to reduce the substantial uncertainties permeating their understanding. A proxy for hail is developed by combining hail-favoring environmental numerical predictors with observations of ESWD hail reports and satellite overshooting top detections. The procedure is applied to the extended summer season (April-October) of 2016-2018 over the whole SPHERA spatial domain. The results indicate maximum hail likelihood over pre-Alpine regions and the northern Adriatic sea around 15 UTC in June-July, in agreement with recent European hail climatologies. The method demonstrates enhanced performance in case of severe hail occurrences and the ability to separate between ambient signatures depending on hail severity.

Um evento extremo de precipitação ocorreu na primeira semana do ano 2000, de 1º a 5 de janeiro, no Vale do Paraíba, parte leste do Estado de São Paulo, Brasil, causando enorme impacto socioeconômico, com mortes e destruição. Este trabalho estudou este evento em 10 estações meteorológicas selecionadas que foram consideradas como aquelas tendo dados mais homogêneos do Que outras estações na região. O modelo de distribuição generalizada de Pareto (DGP) para valores extremos de precipitação de 5 dias foi desenvolvido, individualmente para cada uma dessas estações. Na modelagem da DGP, foi adotada abordagem não-estacionaria considerando o ciclo anual e tendência de longo prazo como co-variaveis. Uma conclusão desta investigação é que as quantidades de precipitação acumulada durante os 5 dias do evento estudado podem ser classificadas como extremamente raras para a região, com probabilidade de ocorrência menor do que 1% para maioria das estações, e menor do que 0,1% em três estações.

Liver transplantation is the unique treatment for several end-stage diseases. Familial Amiloidotic Polineuropathy (FAP) is a neurodegenerative disease related with systemic deposition of amyloidal fiber mainly on peripheral nervous system, clinically translated by an autonomous sensitive-motor neuropathy with severe functional limitations in some cases. The unique treatment for FAP disease is a liver transplant with a very aggressive medication to muscle metabolism and force production. To our knowledge there are no quantitative characterizations of body composition, strength or functional capacity in this population.

Peripheral arterial disease (PAD) as a high incidence in general population and 12% to 20% of population with more than 60 years has already clinical symptoms, such as intermittent claudication (IC), pain, loss of strength and functional incapacity. There are already some studies who refer the possible positive effects of physical exercise in functional consequences of PAD.

Formaldehyde (FA), also known as formalin, formal and methyl aldehydes, is a colorless, flammable, strong-smelling gas. It has an important application in embalming tissues and that result in exposures for workers in the pathology anatomy laboratories and mortuaries. Occupational exposure to FA has been shown to induce nasopharyngeal cancer and has been classified as carcinogenic to humans (group 1) on the basis of sufficient evidence in humans and sufficient evidence in experimental animals. Manifold in vitro studies clearly indicated that FA is genotoxic. FA induced various genotoxic effects in proliferating cultured mammalian cells. The cytokinesis-block micronucleus (CBMN) assay was originally developped as an ideal system form easuring micronucleus (MN), however it can also be used to measure nucleoplasmic bridges (NBP) and nuclear buds (NBUD). Over the past decade another unique mechanism of micronucleus formation, known as nuclear budding has emerged. NBUDS is considered as a marker of gene amplification and/or altered gene dosage because the nuclear budding process is the mechanism by which cells removed amplified and/excess DNA.

Formaldehyde (FA) the most simple and reactive of all aldehydes, is a colorless, reactive and readily polymerizing gas at normal temperature. It has a pungent, suffocating odour that is recognized by most human subjects at concentrations below 1ppm. According to the Report on Carcinogens, FA ranks 25th in the overall U.S. chemical production with more than 11 billion pounds (5 million tons) produced each year. Is an important industrial compound that is used in the manufacture of synthetic resins and chemical compounds such as lubricants and adhesives. It has also applications as a disinfectant, preservative and is used in cosmetics. Estimates of the number of persons who are occupationally exposed to FA indicate that, at least at low levels, may occur in a wide variety of industries. The occupational settings with most extensive use of formaldehyde is in the production of resins and in anatomy and pathology laboratories. Several studies reported a carcinogenic effect in humans after inhalation of FA, in particular an increased risk for nasopharyngeal cancer. Nowadays, the International Agency for Research on Cancer (IARC) classifies FA as carcinogenic to humans (group 1), on the basis of sufficient evidence in humans and sufficient evidence in experimental animals. Manifold in vitro studies clearly indicated that FA is genotoxic. FA induced various genotoxic effects in proliferatin cultured mammalian cells. A variety of evidence suggests that the primary DNA alterations after FA exposure are DNA-protein crosslinks. Incomplete repair of DPX can lead to the formation of mutations.

Genomic damage is probably the most important fundamental cause of development and degenerative disease. It is also well established that genomic damage is produced by environmental exposure to genotoxins, medical procedures (e.g. radiation and chemicals), micronutrient deficiency (e.g. folate), lifestyle factors (e.g. alcohol, smoking, drugs and stress), and genetic factors such as inherited defects in DNA metabolism and/or repair. Tobacco smoke has been associated to a higher risk of development of cancer, especially in the oral cavity, larynx and lungs, as these are places of direct contact with many carcinogenic tobacco’s compounds. Alcohol is definitely a recognized agent that influence cells in a genotoxic form, been citied as a strong agent with potential in the development of carcinogenic lesions. Epidemiological evidence points to a strong synergistic effect between cigarette smoking and alcohol consumption in the induction of cancers in the oral cavity. Approximately 90% of human cancers originate from epithelial cells. Therefore, it could be argued that oral epithelial cells represent a preferred target site for early genotoxic events induced by carcinogenic agents entering the body via inhalation and ingestion. The MN assay in buccal cells was also used to study cancerous and precancerous lesions and to monitor the effects of a number of chemopreventive agents.

Aging in humans appears to be associated with genetic instability. The cytokinesis-blocked micronucleus assay (CBMN) is a comprehensive method for measuring chromosome breakage, DNA misrepair, chromosome loss, non-disjunction, necrosis, apoptosis and cytostasis. Age and gender are the most important demographic variables affecting the micronucleus (MN) index and studies report frequencies in females being greater than those in males by a factor of 1.2 to 1.6 depending on the age group. It has been shown that a higher MN frequency directly corresponds to a decreased efficiency of DNA repair and increased genome instability.
