This thesis reports on the experimental investigation of controlled spin dependent interactions in a sample of ultracold Rubidium atoms trapped in a periodic optical potential. In such a situation, the most basic interaction between only two atoms at one common potential well, forming a micro laboratory for this atom pair, can be investigated. Spin dependent interactions between the atoms can lead to an intriguing time evolution of the system. In this work, we present two examples of such spin interaction induced dynamics. First, we have been able to observe and control a coherent spin changing interaction. Second, we have achieved to examine and manipulate an interaction induced time evolution of the relative phase of a spin 1/2-system, both in the case of particle pairs and in the more general case of N interacting particles. The first part of this thesis elucidates the spin-changing interaction mechanism underlying many fascinating effects resulting from interacting spins at ultracold temperatures. This process changes the spin states of two colliding particles, while preserving total magnetization. If initial and final states have almost equal energy, this process is resonant and leads to large amplitude oscillations between different spin states. The measured coupling parameters of such a process allow to precisely infer atomic scattering length differences, that e.g. determine the nature of the magnetic ground state of the hyperfine states in Rubidium. Moreover, a method to tune the spin oscillations at will based on the AC-Zeeman effect has been implemented. This allowed us to use resonant spin changing collisions as a quantitative and non-destructive particle pair probe in the optical lattice. This led to a series of experiments shedding light on the Bosonic superfluid to Mott insulator transition. In a second series of experiments we have been able to coherently manipulate the interaction induced time evolution of the relative phase in an ensemble of spin 1/2-systems. For two particles, interactions can lead to an entanglement oscillation of the particle pair. For the general case of N interacting particles, the ideal time evolution leads to the creation of spin squeezed states and even Schrödinger cat states. In the experiment we have been able to control the underlying interactions by a Feshbach resonance. For particle pairs we could directly observe the entanglement oscillations. For the many particle case we have been able to observe and reverse the interaction induced dispersion of the relative phase. The presented results demonstrate how correlated spin states can be engineered through control of atomic interactions. Moreover, the results point towards the possibility to simulate quantum magnetism phenomena with ultracold atoms in optical traps, and to realize and analyze many novel quantum spin states which have not been experimentally realized so far.

The subject of the presented thesis is the accurate measurement of time dilation, aiming at a quantitative test of special relativity. By means of laser spectroscopy, the relativistic Doppler shifts of a clock transition in the metastable triplet spectrum of ^7Li^+ are simultaneously measured with and against the direction of motion of the ions. By employing saturation or optical double resonance spectroscopy, the Doppler broadening as caused by the ions' velocity distribution is eliminated. From these shifts both time dilation as well as the ion velocity can be extracted with high accuracy allowing for a test of the predictions of special relativity. A diode laser and a frequency-doubled titanium sapphire laser were set up for antiparallel and parallel excitation of the ions, respectively. To achieve a robust control of the laser frequencies required for the beam times, a redundant system of frequency standards consisting of a rubidium spectrometer, an iodine spectrometer, and a frequency comb was developed. At the experimental section of the ESR, an automated laser beam guiding system for exact control of polarisation, beam profile, and overlap with the ion beam, as well as a fluorescence detection system were built up. During the first experiments, the production, acceleration and lifetime of the metastable ions at the GSI heavy ion facility were investigated for the first time. The characterisation of the ion beam allowed for the first time to measure its velocity directly via the Doppler effect, which resulted in a new improved calibration of the electron cooler. In the following step the first sub-Doppler spectroscopy signals from an ion beam at 33.8 %c could be recorded. The unprecedented accuracy in such experiments allowed to derive a new upper bound for possible higher-order deviations from special relativity. Moreover future measurements with the experimental setup developed in this thesis have the potential to improve the sensitivity to low-order deviations by at least one order of magnitude compared to previous experiments; and will thus lead to a further contribution to the test of the standard model.

La realizzazione di stati non classici del campo elettromagnetico e in sistemi di spin è uno stimolo alla ricerca, teorica e sperimentale, da almeno trent'anni. Lo studio di atomi freddi in trappole di dipolo permette di avvicinare questo obbiettivo oltre a offrire la possibilità di effettuare esperimenti su condesati di Bose Einstein di interesse nel campo dell'interferometria atomica. La protezione della coerenza di un sistema macroscopico di spin tramite sistemi di feedback è a sua volta un obbiettivo che potrebbe portare a grandi sviluppi nel campo della metrologia e dell'informazione quantistica. Viene fornita un'introduzione a due tipologie di misura non considerate nei programmi standard di livello universitario: la misura non distruttiva (Quantum Non Demolition-QND) e la misura debole. Entrambe sono sfruttate nell'ambito dell'interazione radiazione materia a pochi fotoni o a pochi atomi (cavity QED e Atom boxes). Una trattazione delle trappole di dipolo per atomi neutri e ai comuni metodi di raffreddamento è necessaria all'introduzione all'esperimento BIARO (acronimo francese Bose Einstein condensate for Atomic Interferometry in a high finesse Optical Resonator), che si occupa di metrologia tramite l'utilizzo di condensati di Bose Einstein e di sistemi di feedback. Viene descritta la progettazione, realizzazione e caratterizzazione di un servo controller per la stabilizzazione della potenza ottica di un laser. Il dispositivo è necessario per la compensazione del ligh shift differenziale indotto da un fascio laser a 1550nm utilizzato per creare una trappola di dipolo su atomi di rubidio. La compensazione gioca un ruolo essenziale nel miglioramento di misure QND necessarie, in uno schema di feedback, per mantenere la coerenza in sistemi collettivi di spin, recentemente realizzato.

Drought strongly influences root activities in crop plants and weeds. This paper is focused on the performance of the heavy metal accumulator Solanum nigrum, a plant which might be helpful for phytoremediation. The water potential in a split root system was decreased by the addition of polyethylene glycol (PEG 6000). Rubidium, strontium and radionuclides of heavy metals were used as markers to investigate the uptake into roots, the release to the shoot via the xylem, and finally the basipetal transport via the phloem to unlabeled roots. The uptake into the roots (total contents in the plant) was for most makers more severely decreased than the transport to the shoot or the export from the shoot to the unlabeled roots via the phloem. Regardless of the water potential in the labeling solution, 63Ni and 65Zn were selectively redistributed within the plant. From autoradiographs, it became evident that 65Zn accumulated in root tips, in the apical shoot meristem and in axillary buds, while 63Ni accumulated in young expanded leaves and roots but not in the meristems. Since both radionuclides are mobile in the phloem and are, therefore, well redistributed within the plant, the unequal transfer to shoot and root apical meristems is most likely caused by differences in the cell-to-cell transport in differentiation zones without functional phloem (immature sieve tubes).

The alkali metals cesium, rubidium, lithium and sodium were introduced together with strontium via flaps into leaf laminas or into the stem of maturing, intact winter wheat (Triticum aestivum L. cv. Arina) grown in a field. Long-distance transport of these elements and the influence of the application date and of different application positions were investigated. The phloem-immobile Sr served as a marker for the distribution of the xylem sap in the plants. Dry matter accumulation in the grains and the transpiration per shoot were not markedly affected by the treatments as compared to control plants. The phloem mobility was rather high for Cs and Rb. Li was almost immobile in the phloem (similarly to Sr). An application into the cut stem xylem below the second leaf node contributed more to the contents in the grains than an application into the flag leaf. An earlier feeding date led to a higher accumulation in the grains. The marked losses of the elements applied during maturation (most pronounced for Li) can be explained by leakage in the rain.

In order to evaluate factors regulating substrate metabolism in vivo positron emitting radionuclides were used for the assessment of skeletal muscle blood flow and glucose utilization. The potassium analog, Rb-82 was used to measure skeletal muscle blood flow and the glucose analog, 18-F-2-deoxy-2-fluoro-D-glucose (FDG) was used to examine the kinetics of skeletal muscle transport and phosphorylation.^ New Zealand white rabbits' blood flow ranged from 1.0-70 ml/min/100g with the lowest flows occurring under baseline conditions and the highest flows were measured immediately after exercise. Elevated plasma glucose had no effect on increasing blood flow, whereas high physiologic to pharmacologic levels of insulin doubled flow as measured by the radiolabeled microspheres, but a proportionate increase was not detected by Rb-82. The data suggest that skeletal muscle blood flow can be measured using the positron emitting K+ analog Rb-82 under low flow and high flow conditions but not when insulin levels in the plasma are elevated. This may be due to the fact that insulin induces an increase in the Na+/K+-ATPase activity of the cell indirectly through a direct increase in the Na+/H+pump activity. This suggests that the increased cation pump activity counteracts the normal decrease in extraction seen at higher flows resulting in an underestimation of flow as measured by rubidium-82.^ Glucose uptake as measured by FDG employed a three compartment mathematical model describing the rates of transport, countertransport and phosphorylation of hexose. The absolute values for the metabolic rate of FDG were found to be an order of magnitude higher than those reported by other investigators. Changes noted in the rate constant for transport (k1) were found to disagree with the a priori information on the effects of insulin on skeletal muscle hexose transport. Glucose metabolism was however, found to increase above control levels with administration of insulin and electrical stimulation. The data indicate that valid measurements of skeletal muscle glucose transport and phosphorylation using the positron emitting glucose analog FDG requires further model application and biochemical validation. Asbestos and silica are important industrial hazards. Exposure to these dusts can result in pulmonary fibrosis and, in the case of asbestos, cancer. Although the hazards of asbestos and silica exposure have long been known, the pathogenesis of dust-related disease is not well understood. Both silica and asbestos are thought to alter the function of the alveolar macrophage, but the nature of the biochemical alteration is unknown. Therefore, this study examined the effect of asbestos and silica on the activation pathway of the guinea pig alveolar macrophage. Activation of macrophages by physiological agents results in stimulation of phospholipase C causing phosphatidyl inositol turnover and intracellular calcium mobilization. Phosphatidyl inositol turnover produces diacylglycerol which activates protein kinase C causing superoxide anion production.^ Chrysotile stimulated alveolar macrophages to produce superoxide anion. This stimulation proceeded via phospholipase C, since chrysotile stimulated phosphatidyl inositol turnover and intracellular calcium mobilization. The possible involvement of a coupling protein was evaluated by pretreating cells with pertussis toxin. Pertussis toxin pretreatment partially inhibited chrysotile stimulation, suggesting that chrysotile activates a coupling protein in an non-classical manner. Potential binding sites for chrysotile stimulation were examined using a series of nine lectins. Chrysotile-stimulated superoxide anion production was blocked by pretreatment with lectins which bound to N-acetylglucosamine, but not by lectins which bound to mannose, fucose, or N-acetylgalactosamine. In addition, incubation with the N-acetylglucosamine polymer, chitin, inhibited chrysotile-stimulated superoxide anion production, suggesting that chrysotile stimulated superoxide anion production by binding to N-acetylglucosamine residues.^ On the other hand, silica did not stimulate superoxide anion production. The effect of silica on agonist stimulation of this pathway was examined using two stimulants of superoxide anion production, N-formyl-nle-leu-phe (FNLP, which stimulates through phospholipase C) and phorbol-12,13-dibutyrate (which directly activates protein kinase C). Sublethal doses of silica inhibited FNLP-stimulated superoxide anion production, but did not affect phorbol-12,13-dibutyrate-stimulated superoxide anion production, suggesting that the site of inhibition precedes protein kinase C. This inhibition was not due to cell membrane damage, since cell permeability to calcium-45 and rubidium-86 was not increased. It is concluded that chrysotile binds to N-acetylglucosamine residues on macrophage surface glycoproteins to stimulate the physiological pathway resulting in superoxide anion production. In contrast, silica does not stimulate superoxide anion production, but it did inhibit FNLP-stimulated superoxide anion production. ^

A total of 167 samples distubuted throughout the CRP-3 drillhole from 5.77 to 787.68 mbsf and representing fine to coarse sandstones have been analysed by X-ray fluorescence spectrometry (XRF) Bulk sample geochemistry (major and trace elements) indicates a dominant provenance of detritus from the Ferrar Supergroup in the uppermost 200 mbsf of the core. A markedly increased contribution from the Beacon sandstones is recognized below 200 mbsf and down to 600 mbsf. In the lower part of CRP-3, down to 787.68 mbsf, geochemical evidence for influxes of Ferrar materials is again recorded. On the basis of preliminary magnetostratigraphic data reported for the lower 447 mbsf of the drillhole, we tentatively evaluated the main periodicities modulating the geochemical records. Our results identify a possible influence of the precession, obliquity and long-eccentricity astronomical components (21, 41, and 400 ky frequency bands) on the deposition mechanisms of the studied glaciomarine sediments.
