During initial development, both X chromosomes are active in females, and one of them must be silenced at the appropriate time in order to dosage compensate their gene expression levels to male counterparts. Silencing involves epigenetic mechanisms, including histone deacetylation. Major X chromosome inactivation (XCI) in bovine occurs between hatching and implantation, although in vitro culture conditions might disrupt the silencing process, increasing or decreasing X-linked gene expression. In this study, we aimed to address the roles of histone deacetylase inhibition by trichostatin A (TSA) on female preimplantation development.We tested the hypothesis that by enhancing histone acetylation, TSA would increase the percentage of embryos achieving 16-cell stage, reducing percentage of embryos blocked at 8-cell stage, and interfere with XCI in IVF embryos. We noticed that after TSA treatment, acetylation levels in individual blastomeres of 8-16 cell embryos were increased twofold on treated embryos, and the samewas detected for blastocysts. Changes among blastomere levels within the same embryo were diminished on TSA group, as low-acetylated blastomeres were no longer detected. The percentage of embryos that reached the 5th cleavage cycle 118 h after IVF, analyzed by Hoechst staining, remained unaltered after TSA treatment. Then, we assessed XIST and G6PD expression in individual female bovine blastocysts by quantitative real-time PCR. Even though G6PD expression remained unaltered after TSA exposure, XIST expression was eightfold decreased, and we also detected a major decrease in the percentage of blastocysts expressing detectable XIST levels after TSA treatment. Based on these results, we conclude that HDAC is involved on XCI process in bovine embryos, and its inhibition might delay X chromosome silencing and attenuate aberrant XIST expression described for IVF embryos. © 2013 Society for Reproduction and Fertility.

A Clibadium sylvestre é largamente distribuída na região amazônica, onde é conhecida como cunambi ou cunhambi, e sua ingestão causa embriaguez, ou mesmo morte dos peixes, demonstrando propriedade ictiotóxica. Os compostos existentes nas folhas da Clibadium sylvestre são poderosos estimulantes do sistema nervoso central, suas folhas contêm substâncias com potencial convulsivante. As alterações eletroencefalográficas, crise convulsiva e os efeitos de drogas no controle do comportamento convulsivo foram estudados bem como a via metabólica dos componentes acetato de cunaniol e cunaniol. O trabalho foi realizado em ratos wistar machos adultos, tratados com DE50 de 2,92 mg/kg ou DL50 de 3,64 mg/kg de cunaniol a via de administração utilizada foi a intraperitoneal. Após a administração do cunaniol, a evolução das crises convulsivas foram observadas, permitindo classificá-las de acordo com a intensidade de apresentação e relacionar com a concentração plasmática do cunaniol. Os parâmetros eletroencefalográficos, da atuação das drogas no controle das convulsões e a característica cíclica foram determinadas e avaliadas. A análise de plasma obtido por cromatografia líquida após a aplicação das substâncias convulsivantes indicam que o acetato de cunaniol sofre desacetilação dando origem ao cunaniol, droga responsável pelo quadro convulsivo. Dados eletrocorticográficos demonstraram cinco padrões de traçados diferentes durante registro de 4 horas permanecendo com alterações de traçado por 12 horas após aplicação. As drogas utilizadas para prevenir o desencadeamento das convulsões, as mais efetivas foram o Diazepam, o Fenobarbital e a Quetamina. O comportamento convulsivo foi classificado em cinco estágios. Para a ocorrência dos estágios 4 e 5 não houve diferenças estatísticas quanto à concentração plasmática de cunaniol.

The aim of this study was to evaluate citric acid application with or without chitosan on the maintenance of the quality of litchi 'Bengal'. It was used litchi on mature maturation stage. After selection, fruits were immersed for one minute on the solution of citric acid and chitosan: [1]Control - without immersion; [2] citric acid 300g L-1, [3] citric acid 300 g L-1 + chitosan 0,3%, [4]citric acid 300 g L-1 + chitosan 0,6%, [5] citric acid 600 g L-1, [6] citric acid 600 g L-1 + chitosan 0,3%, [7] citric acid 600 g L-1 + chitosan 0,6%. Chitosan of low molecular weight and deacetylation degree of 75,58% (Sigma - Aldrich®) were used. After immersion, fruits were put to drain the excess of solution. Then, they were stored in cool chamber at 5ºC, previously sanitized, for 20 days. The experiment was conducted following a completely randomized design, with a factorial scheme composed by seven coating solutions and five sampling dates. Every five days were evaluated: loss of fresh mass of fruits; color; content of anthocyanins and the activity of peel enzymes polyphenoloxidase and peroxidase. It was verified that the solution of citric acid 600 g L-1, associated or not to chitosan, and the combination of chitosan 0,3% to 300 g L-1 of citric acid were the most efficient to maintain the quality of litchi 'Bengal' for 20 days at 5ºC.

Preeclampsia (PE) is a pregnancy specific syndrome characterized by a systemic inflammatory response, with higher intensity than that observed in normal pregnancy. Cells of the immune system, such as monocytes and granulocytes are endogenously activated and secrete high levels of free radicals and inflammatory cytokines. The objective of this study was to assess the activation state of monocytes from pregnant women with preeclampsia by endogenous expression of TLR2 e TLR4 receptors and to correlate the expression of TLR2 and TLR4 on monocytes surface of pregnant women with PE with the production of tumor necrosis factor-alpha (TNF- and interleukin-10 (IL-10) by these cells stimulated or not with peptidoglycan (PG) and lipopolysaccharide (LPS), as agonists agents of TLR2 and TLR4, respectively. We evaluated 15 pregnant women with PE, 15 normotensive pregnant women (NT) and 15 non-pregnant (NP). Peripheral blood monocytes were incubates in the presence or absence of LPS or PG. The supernatant obtained after 18h of culture was aspirated and used for TNF- and IL-10 determination by enzyme immunoassay (ELISA). The endogenous expression of TLR2 and TLR4 receptors was evaluated by flow cytometry. Our results showed significant highly concentrations of TNF- and TLR4 expression in monocytes of preeclamptic women when compared with NT and NP. Normal pregnant women presented higher levels of IL-10 in comparison with PE and NP groups. TLR2 expression was similar in the three groups studied. Therefore, our study highlights the important role of TLR4 in PE and the consequent high production of TNF- by monocytes of these patients, as well as the potential mechanism involving low levels of IL-10 in the pathophysiology of the disease. These observations demonstrate the strong link between the pathology of PE and the immune system of these patients

J Appl Physiol 113: 1456-1465, 2012. First published August 30, 2012; doi:10.1152/japplphysiol.00936.2011.-This study investigated the involvement of prostaglandins and regulated on activation, normal T cell expressed and secreted (RANTES), in fever induced by live Staphylococcus aureus (no. 25923, American Type Culture Collection) injection in rats. S. aureus was injected intraperitoneally at 10(9), 10(10), and 2 x 10(10) colony-forming units (CFU)/cavity, and body temperature (T-b) was measured by radiotelemetry. The lowest dose of S. aureus induced a modest transient increase in T-b, whereas the two higher doses promoted similar long-lasting and sustained T-b increases. Thus, the 10(10) CFU/cavity dose was chosen for the remaining experiments. The T-b increase induced by S. aureus was accompanied by significant decreases in tail skin temperature and increases in PGE(2) levels in the cerebrospinal fluid (CSF) and hypothalamus but not in the venous plasma. Celecoxib (selective cyclooxygenase-2 inhibitor, 2.5 mg/kg po) inhibited the fever and the increases in PGE(2) concentration in the CSF and hypothalamus induced by S. aureus. Dipyrone (120 mg/kg ip) reduced the fever from 2.5 to 4 h and the PGE(2) increase in the CSF but not in the hypothalamus. S. aureus increased RANTES in the peritoneal exudate but not in the CSF or hypothalamus. Met-RANTES (100 mu g/kg iv), a chemokine (C-C motif) receptor (CCR)1/CCR5 antagonist, reduced the first 6 h of fever induced by S. aureus. This study suggests that peripheral (local) RANTES and central PGE(2) production are key events in the febrile response to live S. aureus injection. As dipyrone does not reduce PGE(2) synthesis in the hypothalamus, it is plausible that S. aureus induces fever, in part, via a dipyrone-sensitive PGE(2)-independent pathway.

Etablierung von Expressionsystemen für Gene der Indolalkaloid-Biosynthese unter besonderer Berücksichtigung von Cytochrom P450-Enzymen In der vorliegenden Arbeit wurden Enzyme aus der Arzneipflanze Rauvolfia serpentina bearbeitet. Es wurde versucht, das an der Biosynthese des Alkaloids Ajmalin beteiligte Cytochrom P450-Enzym Vinorin-Hydroxylase heterolog und funktionell zu exprimieren. Ein zunächst unvollständiger, unbekannter Cytochrom P450-Klon konnte komplettiert und eindeutig mittels heterologer Expression in sf9-Insektenzellen als Cinnamoyl-Hydroxylase identifiziert werden. Die Tauglichkeit des Insektenzellsystems für die Untersuchung der Vinorin-Hydroxylase ist auf Grund der deacetylierenden Wirkung der Insektenzellen auf das Substrat Vinorin nicht gegeben. Im Rahmen des Homology Cloning Projektes konnten mehrere Volllängenklone und diverse Teilsequenzen von neuen Cytochrom P450-Klonen ermittelt werden. Ausserdem wurde durch das unspezifische Binden eines degenerierten Primers ein zusätzlicher Klon gefunden, der der Gruppe der löslichen Reduktasen zugeordnet werden konnte. Diese putative Reduktase wurde auf die Aktivität von mehreren Schlüsselenzymen der Ajmalin-Biosynthese durch heterologe Expression in E.coli und anschliessende HPLC-gestützte Aktivitätstests ohne Erfolg geprüft. Bedingt durch die Untauglichkeit des Insektenzellsystems für die Identifizierung der Vinorin-Hydroxylase, wurde ein neuartiges Modul-gestütztes, pflanzliches Expressionsystem etabliert, um vorhandene P450-Volllängenklone auf Vinorin- Hydroxylaseaktivität testen zu können. Die Funktionalität des Systems konnte durch die heterologe Expression der Polyneuridinaldehyd Esterase bestätigt werden. Trotzdem war es bis jetzt nicht möglich, die Cinnamoyl-Hydroxylase als Kontrollenzym für das pflanzliche System oder aber die gesuchte Vinorin- Hydroxylase in aktiver Form zu exprimieren.
