The process of coarsening of an ensemble of clusters is investigated for the case that elastic strains due to matrix - cluster interactions change the process qualitatively as compared with dependencies established theoretically first by Lifshitz and Slezov. Such a qualitatively different behavior occurs always when the energy of elastic deformation in cluster growth increases more rapidly than linear with the volume of a cluster. Analytic solutions, for limiting cases, as well as numerical solutions, for the general case of coarsening in an ensemble of pores with a given pore size distribution, are presented. Possible applications are discussed.

The moisture sorption isotherms of Chilean papaya were determined at 5, 20, and 45 ºC, over a relative humidity range of 10-95%. The GAB, BET, Oswin, Halsey, Henderson, Smith, Caurie and Iglesias-Chirife models were applied to the sorption experimental data. The goodness of fit of the mathematical models was statistically evaluated by means of the determination coefficient, mean relative percentage deviation, sum square error, root-mean-square error, and chi-square values. The GAB, Oswin and Halsey models were found to be the most suitable for the description of the sorption data. The sorption heats calculated using the Clausius-Clapeyron equation were 57.35 and 59.98 kJ·mol-1, for adsorption and desorption isotherms, respectively.

In the present work, a method was developed and validated for the quantification of benzyl isothiocyanate (BITC) in the fruits of Carica papaya. The quantification of this compound was carried out by gas chromatography (GC) with selective detectors - nitrogen phosphorus detector (NPD) and flame photometric detector (FPD). The performance of these detectors showed a higher sensitivity of the NPD with a broader linear range of detection. The LOD/LOQ were 0.038/0.100 µg/mL for NPD and 5.78/19.29 µg/mL for FPD. The recovery of the method for BITC was 90,64%. An average value of BITC concentration in all the analyzed samples was 16,23 µg BITC/g.

The objective of this study was to monitor 11 organophosphorus pesticides in samples of papaya, bell pepper, and banana, commercialized in the metropolitan area of Vitória (ES, Brazil). The pesticides were determined by an optimized and validated method using high performance liquid chromatography with tandem mass spectrometry (HPLC-MS/MS). All three samples exhibited a matrix effect for most of the pesticides, mainly with signal suppression, and therefore the calibration curves were produced in matrices. Linearity revealed coefficients of determination (r2) greater than 0.9895 for all pesticides and recovery results ranged from between 76% and 118% with standard deviation no greater than 16%. Precision showed relative standard deviation values lower than 19% and HorRat values lower than 0.7, considering all pesticides. Limits of quantification were less than 0.01 mg/kg for all pesticides. Regarding analysis of the samples (50 of each), none of the pesticides exceeded the maximum residue limit determined by Brazilian legislation.

Neste trabalho estudou-se a proteção de melancia (Citrullus lanatus) com estirpes fracas do Papaya ringspot virus, estirpe melancia, em condições de casa de vegetação e em campo. Os resultados mostraram que a proteção foi eficiente em ambos os testes. Nos testes em campo porém, as plantas protegidas da cultivar Crimson Sweet tiveram uma redução no peso médio de frutos por planta de, aproximadamente, 50% e 10,8%, em comparação às plantas não protegidas que estavam sadias, no primeiro e no segundo teste, respectivamente. O mesmo fato ocorreu com a cultivar Crimson Tide, na qual se registrou uma redução de 36,4%, aproximadamente, em um único teste. Além da redução na produção, outro problema encontrado na premunização da melancia foi a necessidade de duas inoculações sucessivas das mudas, em dias consecutivos, para aumentar a freqüência de plantas infetadas.

Papaya ringspot virus (PRSV) is the causal agent of the main papaya (Carica papaya) disease in the world. Brazil is currently the world's main papaya grower, responsible for about 40% of the worldwide production. Resistance to PRSV on transgenic plants expressing the PRSV coat protein (cp) gene was shown to be dependent on the sequence homology between the cp transgene expressed in the plant genome and the cp gene from the incoming virus, in an isolate-specific fashion. Therefore, knowledge of the degree of homology among the cp genes from distinct PRSV isolates which are present in a given area is important to guide the development of transgenic papaya for the control of PRSV in that area. The objective of the present study was to assess the degree of homology among the PRSV cp genes of several Brazilian isolates of this virus. Papaya and PRSV are present in many different ecosystems within Brazil. Twelve PRSV isolates, collected in eight different states from four different geographic regions, were used in this study. The sequences of the cp gene from these isolates were compared among themselves and to the gene used to generate transgenic papaya for Brazil. An average degree of homology of 97.3% at the nucleotide sequence was found among the Brazilian isolates. When compared to 27 isolates from outside Brazil in a homology tree, the Brazilian isolates were clustered with Australian, Hawaiian, and Central and North American isolates, with an average degree of homology of 90.7% among them.

This study compared three mild and three severe strains of Papaya ringspot virus - type W (PRSV-W), based on nucleotide and amino acid sequences of the capsid protein (CP) gene. The CP nucleotide sequences of the mild strains shared 98% to 100% identity. When compared to the severe strains the identity ranged from 93% to 95%, except in the case of PRSV-W-2R, which resulted from reversion of the mild strains PRSV-W-2. The CP sequence of the reverting strain showed 100% identity with the sequence of its parental strain. An insertion of six nucleotides in the core region of the CP gene, which reflected the addition of two amino acids (Asn and Asp) in the deduced sequence of the protein, was found in all mild strains. These sequence comparisons were used to design strain-specific primers that were used to specifically amplify regions for either the mild or severe strains.

The chemical treatment evaluation in the field to control post-harvest fruit anthracnose (Colletotrichum gloeosporioides) requires a suitable disease incidence assessment on harvested papaya (Carica papaya) fruits. The minimum number of papaya fruit harvests was determined for valid treatment comparison in field trials for anthracnose chemical control. Repeatability analysis was done using previously published data. The coefficient determination (R²) estimate range, using four methods, and based on means of 12 assessment times, was 92.58 < R² < 94.45%. The number of assessment times required for R²=90% varied from seven to nine. The R² values of 85.1 < R² < 91.3% estimated by ANOVA suggested that any seven successive assessment times were sufficient for treatment comparison.

Este trabalho teve como objetivo inicial selecionar uma estirpe fraca do Papaya ringspot virus - type W (PRSV-W) de bolhas de folhas de melancia (Citrullus lanatus) com mosaico. A infetividade dessa estirpe, PRSV-W-3, em plantas de melancia foi comparada com a das estirpes fracas PRSV-W-1 e PRSV-W-2, previamente selecionadas de bolhas de folhas de abobrinha de moita (Cucurbita pepo) cv. Caserta com mosaico. Além do efeito da origem da estirpe fraca na infetividade de plantas de melancia, avaliaram-se, ainda, os efeitos da concentração do inóculo, da espécie da planta fonte do inóculo e da idade da planta de melancia inoculada. Como controle, foi utilizada uma estirpe severa do vírus obtida de abobrinha de moita (PRSV-W-C). A avaliação do efeito da concentração e da espécie da planta fonte do inóculo foi feita com extratos de quatro, oito, 12 e 16 discos de folhas de abobrinha de moita e de melancia, infetadas separadamente com as estirpes fracas e severa, e diluídos em 2 ml de tampão fosfato. O efeito da idade da planta-teste de melancia foi estudado comparando-se plantas inoculadas em três estádios de desenvolvimento, com inóculos das estirpes do PRSV-W extraídos de 12 discos foliares/2 ml de tampão. Em todos os testes de infetividade, independente da concentração do inóculo, da planta fonte do inóculo e do estádio de desenvolvimento da planta-teste, a estirpe fraca PRSV-W-3 apresentou taxas de infetividade semelhantes as das estirpes PRSV-W-1 e PRSV-W-2, chegando a 100% em alguns casos. A infetividade da estirpe severa PRSV-W-C foi de 100%. A infetividade das três estirpes fracas está mais diretamente associada à intensidade de fricção das folhas no momento da inoculação do que às outras variáveis estudadas.

Translatable and nontranslatable versions of the coat protein (cp) gene of a Papaya ringspot virus (PRSV) isolate collected in the state of Bahia, Brazil, were engineered for expression in Sunrise and Sunset Solo varieties of papaya (Carica papaya). The biolistic system was used to transform secondary somatic embryo cultures derived from immature zygotic embryos. Fifty-four transgenic lines, 26 translatable and 28 nontranslatable gene versions, were regenerated, with a transformation efficiency of 2.7%. Inoculation of cloned R0 plants with PRSV BR, PRSV HA or PRSV TH, Brazilian, Hawaiian and Thai isolates, respectively, revealed lines with mono-, double-, and triple-resistance. After molecular analysis and a preliminary agronomic evaluation, 13 R1 and R2 populations were incorporated into the papaya-breeding program at Embrapa Cassava and Tropical Fruits, in Cruz das Almas, Bahia, Brazil.

The natural occurrence of Pseudomonas syringae pv. tabaci causing leaf spot symptoms in papaya seedlings is reported. The pathogen was identified through biochemical, physiological, serological, and molecular assays and artificial inoculations in papaya plants. It was also shown that the strains were pathogenic to bean and tobacco plants. The restriction patterns obtained with Afa I, Alu I, Dde I, Hae III, Hpa II, Hinf I, Sau 3A I and Taq I of the PCR-RFLP of 16S-23S DNAr were identical to the P. s. pv. tabaci patterns. Primers corresponding to hrpL gene of P. syringae were also tested and the results grouped the papaya strains with P s. pv. tabaci. Bacterial strains were deposited at Coleção de Culturas IBSBF, Instituto Biológico, Campinas, Brazil, under access numbers 1687 and 1822.

Este trabalho teve por objetivo avaliar, em condições de casa de vegetação e de campo, os danos causados pelo PRSV-W e ZYMV em abobrinha-de-moita (Cucurbita pepo cv. Caserta). As plantas em casa de vegetação foram inoculadas com os vírus individualmente e em mistura aos 12 e 22 dias após emergência (DAE) e aos 5, 15 e 25 DAE no campo. Em casa de vegetação, as infecções com PRSV-W + ZYMV, PRSV-W e ZYMV, na primeira época de inoculação, ocasionaram reduções de área foliar de 39,6%, 36,8% e 12,1%, respectivamente. As massas fresca e seca também foram significativamente afetadas na primeira época de inoculação. No campo, as plantas com infecções individuais ou mistas dos potyvírus produziram frutos não comerciais em quantidades que variaram de 14 a 861 g/planta, dependendo da idade que foram inoculadas. As plantas tratadas com tampão fosfato aos 5, 15 e 25 DAE produziram em média 573 g, 937 g e 1172 g de frutos comerciais e 282 g, 221 g e 192 g de frutos não comerciais, respectivamente. A redução na massa fresca das plantas foi diretamente relacionada com a época de inoculação, com médias de 60,7% para aquelas inoculadas aos 5 DAE e de 22,7% para aquelas inoculadas aos 15 DAE. Na terceira época de inoculação não houve diferença significativa de massa fresca entre os tratamentos.

Se colectaron en diferentes regiones de Cuba semillas de las accesiones de papayo Tallo Morado de Nava, Amarilla de Duaba, Amarilla de Nava y Sapote de Pilón. Estas se plantaron en condiciones de campo en Jagüey Grande, Matanzas - Cuba, donde se evaluó la presencia e intensidad de síntomas de mancha anular, siguiendo una escala graduada del 1 al 5. Los resultados mostraron un incremento temporal de la proporción de plantas afectadas en todas las accesiones. La accesión Amarilla de Duaba mostró los síntomas a los dos meses después de la primera evaluación (MDPE), mientras que el resto lo hizo a los cinco MDPE, aunque con proporciones que variaron entre ellas. A los siete MDPE las accesiones Amarilla de Duaba y Amarilla de Nava mostraron proporciones idénticas de plantas enfermas, mientras que Tallo morado de Nava y Sapote de Pilón mostraron proporciones inferiores. La intensidad de los síntomas se incrementó en los diferentes órganos siguiendo el siguiente orden: tallo (1,7 a 2,66), pecíolos (2,21 a 3,03) y follaje (3,44-4,03). En los frutos la intensidad de los síntomas observados fue inferior a 2,5, considerados como leves de acuerdo a su intensidad y distribución. Se ofrecen los primeros resultados sobre la susceptibilidad a la mancha anular de estas accesiones, siendo de gran utilidad para los productores, así como para los programas de mejoramiento genético en el país.
