Gene expression is a quantitative trait that can be mapped genetically in structured populations to identify expression quantitative trait loci (eQTL). Genes and regulatory networks underlying complex traits can subsequently be inferred. Using a recently released genome sequence, we have defined cis- and trans-eQTL and their environmental response to low phosphorus (P) availability within a complex plant genome and found hotspots of trans-eQTL within the genome. Interval mapping, using P supply as a covariate, revealed 18,876 eQTL. trans-eQTL hotspots occurred on chromosomes A06 and A01 within Brassica rapa; these were enriched with P metabolism-related Gene Ontology terms (A06) as well as chloroplast-and photosynthesis-related terms (A01). We have also attributed heritability components to measures of gene expression across environments, allowing the identification of novel gene expression markers and gene expression changes associated with low P availability. Informative gene expression markers were used to map eQTL and P use efficiency-related QTL. Genes responsive to P supply had large environmental and heritable variance components. Regulatory loci and genes associated with P use efficiency identified through eQTL analysis are potential targets for further characterization and may have potential for crop improvement.

In this work we used the conversion process of a precursor polymer into polyparaphenylenevinylene (PPV) at low temperatures in order to control the effective conjugation degree of spin-casted PPV films. The absorption and emission spectra of the films were studied by following a partial substitution of chloride counterions from poly(xylylidene tetrahydrothiophenium chloride) (PTHT), used as a precursor, by sodium acid dodecyl benzenesulfonate (DBS), added as a surfactant salt. Upon controlling the DBS amount and conversion temperature (T-c) of PTHT/DBS to PPV films, the band gap of PPV changed from 409 to 506 nm, and 505 to 532 nm, values obtained from absorbance and emission measurements, respectively. Based on these experimental data, we proposed a physical model which represents the chemical structure of PPV as a distribution of conjugated chain segments (like oligomers) alternated by non-conjugated segments (structural defects and/or from the precursor polymer). In this work we studied the properties of absorption and emission line shape of layer-by-layer (LBL) poly(p-phenylene vinylene) (PPV) on indium-tin oxide (ITO) electrode. To minimize the PPV thermal conversion effects during the polymer processing, we used a less aggressive leaving group in the precursor polymer; minimizing electrode degradation. LBL ITO/PPV films showed the same absorption and emission line shape compared with LBL PPV films deposited on non-metallic substrates (glass). With this analysis we indirectly observe the decrease in the ITO degradation. Atomic force microscopy (AFM) technique was used to analyze quantitatively the microscopic morphology of the film surface. Results indicated that the substrate topology is not affected, to a large extent, by the use of dodecylbenzensulfonate (DBS) ion. (C) 2008 Elsevier B.V. All rights reserved.

Foram estudadas, neste trabalho, interacões por complexos de transferência de carga, através de medidas gravimétricas do grau de inchamento de géis de poli(isopreno) reticulado,contendo grupos aceptores de carga, em presença de grupos doadores de carga, de pequeno peso molecular, dissolvidos em tolueno. Para este fim, poli(isopreno) sintético proveniente da COPERBO - Companhia Pernambucana de Borracha Sintética, após purificação em clorofórmio, foi caracterizado por espectroscopia no infra-vermelho. O seu peso molecular médio foi determinado por viscosimetria em tolueno e o valor de Mv obtido foi 8,25 x 105. Através de epoxidação da dupla ligacão e hidrólise ácida do respectivo anel oxirano, foram introduzidos no poli(isopreno), previamente purificado, grupos aceptores de carga do tipo cloranil e ácido 3,5-dinitro-benzóico, e grupos doadores de carga do tipo carbazola. Obteve-se, desta forma, copolímeros aceptores do tipo poli (isopreno - co-cloranil) e poli(isopreno-co-(3,5-dinitro-benzoato)) e copolímero doador do tipo poli(isopreno- co-carbazola). A quantidade de cada espécie introduzida foi determinada por espectroscopia no ultra violeta. Poli(isopreno) e os copolímeros contendo grupos doadores ou aceptores foram reticulados em solução utilizando 4,4'-(4,4'-difenilmetileno)-bis- 1,2,4-triazolina-3,5-diona (BPMTD). Os filmes reticulados, após retirada a fase sol, e secos à vácuo, foram submetidos a inchamento em tolueno puro e em solução de tolueno contendo grupos aceptores ou doadores de carga, nas proporções 10:1, 1:1 e 1:10 (polímero: grupo doador ou polímero: grupo aceptor de carga), a 25, 30, 35 e 40ºC. Das medidas gravimétricas do grau de inchamento, foi verificado a formação de complexos por transferência de carga entre os copolímeros aceptores e carbazola. Foi verificado ainda que copolímeros contendo grupos aceptores do tipo cloranil possuem maior tendência a formar complexos por transferência de carga do que copolímeros aceptores contendo grupos 3,5-dinitro-benzoíla.

A presente dissertação tem por objetivo articular os conceitos de estratégia e políticas públicas, orientadas para o papel do BNDES no contexto brasileiro. A expressividade da atuação do BNDES, a origem de seus recursos e a relevância de seus desembolsos ratificam a importância dos financiamentos da instituição para a economia doméstica.

Poly(p-phenylene vinylene) (PPV) derivatives are well known for their applications in polymer light emitting diodes (PLEDs). PPV derivatives are highly susceptible to photo-oxidation though, which is mainly caused by the scission of the vinyl double bond on the polymer backbone. In this work, we show that Langmuir-Blodgett (LB) films are less degraded than cast films of a PPV derivative (OC1OC6-PPV). Both films had similar thickness (similar to 50 nm) to allow for a more realistic comparison. Degradation was monitored with UV-vis and FTIR spectroscopies. The results indicated that cast films were completely degraded in ca. 400 min, while LB took longer time, i.e. about four times the values for the cast films. The differences can be attributed to the more compact morphology in the LB than in the cast films. With a compact morphology the diffusion of oxygen in the LB film is hampered and this causes a delay in the degradation process. (c) 2006 Elsevier Ltd. All rights reserved.

Paracoccidioides brasiliensis is a dimorphic fungus known to produce invasive systemic disease in humans. The 43-kDa glycoprotein of P, brasiliensis is the major diagnostic antigen of paracoccidioidomycosis and may act as a virulence factor, since it is a receptor for laminin. Very little is known about early interact-ions between this fungus and the host cells, so we developed in vitro a model system employing cultured mammalian cells (Vero cells), in order to investigate the factors and virulence mechanisms of P. brasiliensis related to the adhesion and invasion process. We found that there is a permanent interaction after 30 min of contact between the fungus and the cells. The yeasts multiply in the cells for between 5 and 24 h. Different strains of P, brasiliensis were compared, and strain 18 thigh virulence) was the most strongly adherent, followed by strain 113 (virulent), 265 (considered of low virulence) and 113M(mutant obtained by ultraviolet radiation, deficient in gp43). P. brasiliensis adhered to the epithelial cells by a narrow tube, while depressions were noticed in the cell surface, suggesting an active cavitation process. An inhibition assay was performed and it was verified that anti-gp43 serum and a pool of sera from individuals with paracoccidioidomycosis were able to inhibit the adhesion of P. brasiliensis to the Vero cells. Glycoprotein 43 (gp43) antiserum abolished 85 % of the binding activity of P. brasiliensis. This fungus can also invade the Vero cells, and intraepithelial parasitism could be an escape mechanism in paracoccidioidomycosis. (C) 2000 Editions scientifiques et medicales Elsevier SAS.

A simple model of electron transfer is adapted to explain fluorescence quenching in self-assembled films of poly( p-phenylene vinylene) (PPV) alternating with poly(thiophene acetic acid) (PTAA). Quenching is caused by a photo-induced electron transfer between the excited PPV (donor, D) and the PTAA (acceptor, A). The electron-transfer process can be mediated by insertion of electronically inert spacing bilayers between the D and A layers, As the number of bilayers is increased, the fluorescence is gradually recovered which is explained theoretically by assuming that the electron-transfer rate can be described as k = Z exp(- beta r) where r is the distance between D and A. Space-charge-limited currents measurements have been carried out on undoped amorphous poly p-phenylene sulfide. The scaling law is checked for different samples with varying thickness, and J-V data analyzed. The position of the quasi-Fermi level and the density of states was obtained.

Both films had similar thickness (∼50 nm) to allow for a more realistic comparison. Photodegradation experiments were carried out by illuminating the films with white light from a halogen lamp (50W, 12 V), placed at a fixed dstance from the sample. The decay was monitored by UV-Vis and FTIR spectroscopies. The results showed that cast films are completely degraded in ca. 300 min, while LB took longer times, ca. 1000 min, i.e. 3 times the values for the cast films. The degradation process occurs in at least two stages, the rates of which were calculated assuming that the reaction follows a first order kinetics. The characteristic times for the first stage were 3.6×10-2 and 1.3×10-3 min-1 for cast and LB films, respectively. For the second stage the characteristic times were 5.6×10-2 and 5.0×10 -3 min-1. The differences can be attributed to the more compact morphology in the LB than in the cast films. With a compact morphology the diffusion of oxygen in the LB film is hampered and this causes a delay in the degradation process.

A vespa social Polybia paulista (Hymenoptera, Vespidae) é bastante abundante e endêmica nos Estados de São Paulo e sul de Minas Gerais. Os indivíduos da espécie causam um elevado número de acidentes de importância médica. Após a ferroada a vítima pode experimentar reações imunológicas locais e/ou sistêmicas, que em alguns casos podem conduzir a anafilaxia e morte. O diagnóstico e terapia de alergia à ferroada de P. paulista é baseado no uso de extrato de veneno bruto o que se associa à ocorrência de reatividade cruzada e reações imunológicas adversas durante a imunoterapia. O uso de alérgenos recombinantes (r) tem-se mostrado como uma alternativa interessante para reduzir o impacto destas desvantagens. Neste trabalho, foram avaliadas diferentes condições para otimizar a expressão recombinante e solubilização dos corpúsculos de inclusão da fosfolipase A1 (Poly p 1) (70kDa) do veneno de P. paulista previamente obtida mediante expressão heteróloga no sistema procariótico, Escherichia coli. Os resultados aqui obtidos contribuirão para aumentar as quantidades do r Poly p 1 necessárias para sua avaliação bioquímica e imunológica, e finalmente para melhorar os resultados do diagnóstico e imunoterapia específica de alergia ao veneno de P. paulista
