A distribuição de sementes para a instalação de uma cultura é efetuada com base nos resultados do teste de germinação e para complementar as informações sobre a qualidade das sementes são utilizados testes de vigor. Porém, como não há pesquisas relacionadas à avaliação do vigor das sementes de fumo (Nicotiana tabacum L.), essa pesquisa foi realizada como objetivo de estudar o teste de condutividade elétrica para essas sementes. Para isso, foram avaliadas sementes do grupo varietal 'Virgínia', cultivar 'CSC 439', representadas por cinco lotes de sementes nuas e revestidas, hidratadas em 25 mL de água destilada por 2, 4, 6, 8 e 24 h a 25 °C. Foram utilizadas 0,5; 0,8 e 1,0 g de sementes nuas e 2,5; 4,0 e 5,0 g de revesidas. Para complementar as análises as sementes foram avaliadas quanto à germinação (total e primeira contagem) e a emergência da plântula (total e velocidade). O delineamento experimental foi o inteiramente ao acaso, com quatro repetições; os dados foram submetidos à análise de variância e à comparação das médias pelo teste de Tukey (5%). É possível concluir que o teste de condutividade elétrica não é eficiente para classificar os lotes de sementes de fumo, nuas e revestidas, do cultivar 'CSC 439', considerando as quantidades de sementes e os tempos de hidratação estudados.

As informações disponíveis sobre beneficiamento de sementes pequenas e, em especial de tabaco, cujas sementes apresentam diferenças acentuadas no peso específico em virtude da desuniformidade de maturação, são reduzidas. Assim sendo, é importante estudar a influência da utilização da mesa densimétrica na qualidade de sementes dessa espécie. O presente trabalho foi conduzido com o objetivo de avaliar os atributos fisiológicos e sanitários de sementes de tabaco, beneficiadas em mesa densimétrica. Foram realizados ensaios independentes utilizando sementes de sete cultivares. Inicialmente, as sementes foram limpas em máquinas de ar e a seguir beneficiadas em mesa densimétrica. O eixo terminal de descarga da mesa densimétrica de largura 50 cm foi dividido em quatro partes mais a bica de saída de pedras. Os tratamentos constituíram-se em seis frações obtidas, uma no depósito da alimentação, quatro nas partes alta, intermediária alta, intermediária baixa e baixa e uma na bica de saída de pedras. As sementes de tabaco descarregadas na parte alta da zona de descarga da mesa densimétrica apresentaram qualidade fisiológica significativamente superior às sementes descarregadas nas partes baixas. O descarte das sementes descarregadas na parte baixa da mesa densimétrica contribui para o aprimoramento da qualidade fisiológica de lotes de sementes de tabaco.

Gamma-aminobutyric acid (GAB A) is a ubiquitous non-protein amino acid synthesized via the decarboxylation of L-glutamate in a reaction catalyzed by the cytosolic enzyme L-glutamate decarboxylase (GAD). In animals it functions as an inhibitory neurotransmitter. In plants it accumulates rapidly in response to various stresses, but its function remains unclear. The hypothesis that GABA accumulation in leaf tissue may function as a plant resistance mechanism against phytophagous insect activity was investigated. GABA accumulation in response to mechanical stimulation, mechanical damage and insect activity was demonstrated. In wt tobacco (Nicotiana tabacum cv Samsun), mechanical stimulation or damage caused GABA to accumulate within 2 min from mean levels of 14 to 37 and 1~9 nmol g-l fresh weight (FW), respectively. In the transgenic tobacco strain CaMVGAD27c overexpressing Petunia GAD, the same treatments caused GABA to accumulate from 12 to 59 and 279 nmol g-l FW, respectively. In the transgenic tobacco strain CaMVGADilC 11 overexpressing Petunia GAD lacking an autoinhibitory domain, mechanical stimulation or damage caused GABA to accumulate from 180 to 309 and 630 nmol g-l FW, respectively. Ambulatory activity by tobacco budworm (TBW) larvae (Heliothis virescens) on leaves of CaMVGAD27c tobacco caused GABA to accumulate from 28 to 80 nmol g-l FW within 5 min. Ambulatory and leaf-rolling activity by oblique banded leaf roller (OBLR) larvae (Choristoneura rosaceana cv Harris) on wt soybean leaves (Glycine max cv Harovinton) caused GABA to accumulate from 60 to 1123 nmol g-l FW within 20 min. Increased GABA levels in leaf tissue were shown to affect phytophagous preference in TBW larvae presented with wt and transgenic tobacco leaves. When presented with leaves of Samsun wt and CaMVGAD27c plants, TBW larvae consumed more wt leaf tissue (640 ± 501 S.D. mm2 ) than transgenic leaf tissue (278 ± 338 S.D. mm2 ) nine times out of ten. When presented with leaves of Samsun wt and CaMVGAD~C11 plants, TBW larvae consumed more transgenic leaf tissue (1219 ± 1009 S.D. mm2 ) than wt leaf tissue (28 ± 31 S.D. mm2 ) ten times out of ten. These results indicate that: (1) ambulatory activity of insect larvae on leaves results in increased GABA levels, (2) transgenic tobacco leaves with increased capacity for GABA synthesis deter feeding, and (3) transgenic tobacco leaves with constitutively higher GABA levels stimulate feeding.

Des preuves archéopalynologiques directes appuient maintenant l’hypothèse d’une lente adaptation horticole durant la néolithisation amérindienne de la région de Montréal. Les sites archéologiques Hector-Trudel (BhFl-1a) de Pointe-du-Buisson et Séminaire de Saint-Sulpice (BjFj-18) dans le Vieux-Montréal ont été retenus pour élaborer une méthodologie archéopalynologique d’étude des sols archéologiques. Cela a permis de caractériser l’impact de la présence humaine sur l'environnement végétal des sites et d’identifier des indices de culture et de gestion de plantes allogènes et indigènes. Un complexe horticole de production à petite échelle de maïs (Zea mays), de tournesol (Helianthus annuus) et de petit tabac (Nicotiana rustica) et une forme de gestion des arbustes à petits fruits sont identifiés au site Hector-Trudel durant le Sylvicole moyen tardif (500 à 1000 A.D.). Ces cultigènes sont aussi identifiés au site du Séminaire pour la fin du Sylvicole supérieur ancien (1200 à 1300 A.D.), dans des proportions toutefois plus importantes, et une activité de gestion forestière au profit des arbres à noix et du tilleul d’Amérique (Tilia americana), reflet des pratiques d’entretien des champs cultivés, témoignent d’une évolution dans les comportements

An in silico screen of 41 of the 81 coding regions of the Nicotiana plastid genome generated a shortlist of 12 candidates as DNA barcoding loci for land plants. These loci were evaluated for amplification and sequence variation against a reference set of 98 land plant taxa. The deployment of multiple primers and a modified multiplexed tandem polymerase chain reaction yielded 85–94% amplification across taxa, and mean sequence differences between sister taxa of 6.1 from 156 bases of accD to 22 from 493 bases of matK. We conclude that loci should be combined for effective diagnosis, and recommend further investigation of the following six loci: matK, rpoB, rpoC1, ndhJ, ycf5 and accD.

Ultrastructural features of embryogenic pollen in Datura innoxia are described, just prior to, during, and after completion of the first division of the presumptive vegetative cell. In anther cultures initiated towards the end of the microspore phase and incubated at 28 degrees C in darkness, the spores divide within 24 h and show features consistent with those of dividing spores in vivo. Cytokinesis is also normal in most of the spores and the gametophytic cell-plate curves round the presumptive generative nucleus in the usual highly ordered way. Further differentiation of the 2 gametophytic cells does not take place and the pollen either switches to embryogenesis or degenerates. After 48-72 h, the remaining viable pollen shows the vegetative cell in division. The cell, which has a large vacuole and thin layer of parietal cytoplasm carried over from the microspore, divides consistently in a plane parallel to the microspore division. The dividing wall follows a less-ordered course than the gametophytic wall and usually traverses the vacuole, small portions of which are incorporated into the daughter cell adjacent to the generative cell. The only structural changes in the vegetative cell associated with the change in programme appear to be an increase in electron density of both plastids and mitochondria and deposition of an electron-dense material (possibly lipid) on the tonoplast. The generative cell is attached to the intine when the vegetative cell divides. Ribosomal density increases in the generative cell and exceeds that in the vegetative cell. A thin electron-dense layer also appears in the generative-cell wall. It is concluded that embryogenesis commences as soon as the 2 gametophytic cells are laid down. Gene activity associated with postmitotic synthesis of RNA and protein in the vegetative cell is switched off. The data are discussed in relation to the first division of the embryogenic vegetative cells in Nicotiana tabacum.

Background and aims Unilateral incompatibility (UI) occurs when pollinations between species are successful in one direction but not in the other. Self-incompatible (SI) species frequently show UI with genetically related, self-compatible (SC) species, as pollen of SI species is compatible on the SC pistil, but not vice versa. Many examples of unilateral incompatibility, and all those which have been studied most intensively, are found in the Solanaceae, particularly Lycopersicon, Solanum, Nicotiana and Petunia. The genus Capsicum is evolutionarily somewhat distant from Lycopersicon and Solanum and even further removed from Nicotiana and Petunia. Unilateral incompatibility has also been reported in Capsicum; however, this is the first comprehensive study of crosses between all readily available species in the genus. Methods All readily available (wild and domesticated) species in the genus are used as plant material, including the three genera from the Capsicum pubescens complex plus eight other species. Pollinations were made on pot-grown plants in a glasshouse. The number of pistils pollinated per cross varied (from five to 40 pistils per plant), depending on the numbers of flowers available. Pistils were collected 24 h after pollination and fixed for 3-24 h. After staining, pistils were mounted in a drop of stain, squashed gently under a cover slip and examined microscopically under ultra-violet light for pollen tube growth. Key results Unilateral incompatibility is confirmed in the C. pubescens complex. Its direction conforms to that predominant in the Solanaceae and other families, i.e. pistils of self-incompatible species, or self-compatible taxa closely related to self-incompatible species, inhibit pollen tubes of self-compatible species. Conclusions Unilateral incompatibility in Capsicum does not seem to have arisen to prevent introgression of self-compatibility into self-incompatible taxa, but as a by-product of divergence of the C. pubescens complex from the remainder of the genus. P>A cDNA encoding a small lysine-rich protein of unknown function was identified in a tobacco (Nicotiana tabacum) stigma/style suppression subtractive hybridization cDNA library. After its characterization, the corresponding gene was designated stigma/style cell cycle inhibitor 1 (SCI1). Fluorescence microscopy with an SCI1-GFP protein fusion demonstrated its nuclear localization, which was confined to the interchromatic region. Real-time RT-PCR and in situ hybridization experiments showed that SCI1 is stigma/style-specific and developmentally regulated. SCI1 RNAi knockdown and overexpression plants had stigmas/styles with remarkably enlarged and reduced areas, respectively, which was attributable to differences in cell numbers. These results indicate that SCI1 is a tissue-specific negative cell cycle regulator. The differences in cell division had an effect on the timing of the differentiation of the stigmatic papillar cells, suggesting that their differentiation is coupled to stigma cell divisions. This is consistent with a role for SCI1 in triggering differentiation through cell proliferation control. Our results revealed that SCI1 is a novel tissue-specific gene that controls cell proliferation/differentiation, probably as a component of a developmental signal transduction pathway.

Nas últimas décadas o conhecimento florístico e taxonômico sobre a família Solanaceae no Rio Grande do Sul tem recebido incremento significativo de informações. Dos gêneros com espécies nativas, Bouchetia, Calibrachoa, Nicotiana, Nierembergia, Petunia e Solanum já foram alvo de revisões. Visando complementar o estudo desta família no Estado, foram estudados os gêneros Acnistus, Athenaea, Aureliana, Brunfelsia, Capsicum, Cestrum, Dyssochroma, Grabowskia, Lycianthes, Solandra e Vassobia, além das espécies do gênero Solanum seção Pachyphylla (Cyphomandra sensu lato). Foram catalogadas 22 espécies nativas, sendo três destas, novas citações de ocorrência. Para os onze primeiros gêneros foram identificados 19 espécies, dos quais cinco são de ocorrência restrita (Athenaea picta, Dyssochroma longipes, Grabowskia duplicata, Lycianthes rantonnei e Solandra grandiflora). A seção Pachyphylla do gênero Solanum está representada por três espécies. Dessa forma, foram atualizados os dados quali e quantitativos sobre a participação da família Solanaceae na flora sul-riograndense. Para cada gênero e para cada táxon específico foram elaboradas descrições, e acrescentados dados sobre fenologia, hábitat, comentários pertinentes e mapas de ocorrência. Chaves analíticas para identificação das espécies, quando mais de uma, também foram elaboradas. A revisão de doze herbários regionais possibilitou a elaboração de uma sinopse taxonômica e de uma chave analítica para identificação dos gêneros presentes no Estado. Dos 27 gêneros confirmados, 22 têm espécies nativas. O número de espécies nativas é de 114 e o de introduzidas, cultivadas ou assilvestradas, é até o momento 26.

The genome of all organisms constantly suffers the influence of mutagenic factors from endogenous and/or exogenous origin, which may result in damage for the genome. In order to keep the genome integrity there are different DNA repair pathway to detect and correct these lesions. In relation to the plants as being sessile organisms, they are exposed to this damage frequently. The Base Excision DNA Repair (BER) is responsible to detect and repair oxidative lesions. Previous work in sugarcane identified two sequences that were homologous to Arabidopsis thaliana: ScARP1 ScARP3. These two sequences were homologous to AP endonuclease from BER pathway. Then, the aim of this work was to characterize these two sequence using different approaches: phylogenetic analysis, in silico protein organelle localization and by Nicotiana tabacum transgenic plants with overexpression cassette. The in silico data obtained showed a duplication of this sequence in sugarcane and Poaceae probably by a WGD event. Furthermore, in silico analysis showed a new localization in nuclei for ScARP1 protein. The data obtained with transgenic plants showed a change in development and morphology. Transgenic plants had slow development when compared to plants not transformed. Then, these results allowed us to understand better the potential role of this sequence in sugarcane and in plants in general. More work is important to be done in order to confirm the protein localization and protein characterization for ScARP1 and ScARP3

Flowering is controlled by several environmental and endogenous factors, usually associated with a complex network of metabolic mechanisms. The gene characterization in Arabidopsis model has provided much information about the genetic and molecular mechanisms that control flowering process. Some of these genes had been found in rice and maize. However, in sugarcane this processe is not well known. It is known that early flowering may reduce its production up to 60% at northeast conditions. Considering the impact of early flowering in sugarcane production, the aim of this work was to make the gene characterization of two cDNAs previously identified in subtractive cDNA libraries: scPKCI and scSHAGGY. The in silico analysis showed that these two cDNAs presented both their sequence and functional catalytic domains conserved. The results of transgenic plants containing the overexpression of the gene cassette scPKCI in sense orientation showed that this construction had a negative influence on the plant development as it was observed a decrease in plant height and leaf size. For the scPKCI overexpression in antisense orientation it was observed change in the number of branches from T1 transgenic plants, whereas transgenic T2 plants showed slow development during germination and initial stages of development. The other cDNA analyzed had homology to SHAGGY protein. The overexpression construct in sense orientation did not shown any effect on development. The only difference observed it was an increase in stigma structure. These results allowed us to propose a model how these two genes may be interact and affect floweringdevelopment.

Foram estudados os grãos de pólen de 12 gêneros e 41 espécies de Solanaceae ocorrentes na Reserva do Parque Estadual das Fontes do Ipiranga: Acnistus arborescens (L.) Schlecht., Athenaea picta (Mart.) Sendtn., Brunfelsia latifolia Benth., Brunfelsia pauciflora (Cham. & Schlecht.) Benth., Capsicum flexuosum (L.) Sendtn., Capsicum villosum (L.) Sendtn., Cestrum amictum (L.) Schlecht., Cestrum corymbosum (L.) Schlecht., Cestrum lanceolatum (L.) Miers, Cestrum schlechtendalii (L.) G. Don, Cestrum sendtnerianum (L.) Mart. ex Sendtn., Cyphomandra diploconos Sendtn., Cyphomandra velutina Sendtn., Dyssochroma viridiflora (Sims) Ducke, Nicotiana langsdorffii (Weinm.)
