There are no adequate vaccines against some of the new or reemerged infectious scourges such as HIV and TB. They may require strong and enduring cell-mediated immunity to be elicited. This is quite a task, as the only known basis of protection by current commercial vaccines is antibody. As DNA or RNA vaccines may induce both cell-mediated and humoral immunity, great interest has been shown in them. However, doubt remains whether their efficacy will suffice for their clinical realization. We look at the various tactics to increase the potency of nucleic acid vaccines and divided them broadly under those affecting delivery and those affecting immune induction. For delivery, we have considered ways of improving uptake and the use of bacterial, replicon or viral vectors. For immune induction, we considered aspects of immunostimulatory CpG motifs, coinjection of cytokines or costimulators and alterations of the antigen, its cellular localization and its anatomical localization including the use of ligand-targeting to lymphoid tissue. We also thought that mucosal application of DNA deserved a separate section. In this review, we have taken the liberty to discuss these enhancement methods, whenever possible, in the context of the underlying mechanisms that might argue for or against these strategies.

Predator-prey relationships are an important aspect of the natural world, and, because of its relevance to survival and natural selection, is an interesting relationship to study. In amphibian larvae, level of activity and landscape use are often what determines the survival as prey. I studied the anti-predator behavior of the North American bullfrog (Rana catesbeiana) tadpoles when presented with dragonfly (Aeshna) larvae, a known predator of tadpoles. Tadpoles were acclimated to four different habitats with varying degrees of habitat cover, and were transferred to a new habitat with a degree of cover equal to one of the acclimation tanks. A restrained predator, and thus its chemical cue, was introduced, and the behavior, particularly the use of the habitat cover to hide from the perceived risk of predation was observed. A significantly higher frequency of inactivity was found in tank I than in II and III, and inactivity followed a general trend of decreasing with increasing habitat cover. Difference in tank cover was not found to have a significant effect on swimming behavior, but did have a significant effect on hiding behavior, which increased with higher availability. Foraging decreased significantly with the addition of a predator, but did not vary significantly with different levels of cover. Hiding behavior and reducing conspicuous behaviors (like foraging) are probably the behaviors that afford the tadpole the most success at eluding a predator in their natural environment.

Cnidarians display a diverse range of reproductive tactics including sexual and asexual modes of reproduction. Although few studies have looked for intraspecific variation in reproductive tactics, flexible expression of such life-history traits may be favoured in species that occupy a range of habitats. We tested this in the sea anemone Actinia tenebrosa by comparing cycles of reproductive activity and the mode of production of brooded larvae in local populations occupying boulder fields and stable rock platforms. We determined the mode of production of broods from eight rock platforms (separated by up to 1600 km) and two boulder shores on the south east coast of Australia using a combination of allozyme data and four newly characterised microsatellite markers.

We determined seasonal patterns of brooding and gonad development by monthly dissection of 15–30 adults from each of two boulder fields and two stable platforms. Previous genetic studies have shown that populations of A. tenebrosa on rock platforms can be highly clonal, whereas anemones on more heterogeneous boulder habitats display levels of genotypic diversity similar to that expected for sexual reproduction. We genotyped a total of 221 juveniles from 37 brooding adults including 11 broods and 80 juveniles from boulder shores. We did not detect any evidence of sexual production of broods. All brooded juveniles displayed identical multi-locus genotypes to their brood parent irrespective of habitat of origin or location, including 28 broods (200 juveniles) that were heterozygous at one or more locus. Similarly, we found that temporal patterns of gonad formation and brooding were consistent across habitats and locations. We detected 346 mature males, 234 non-reproductive or immature individuals, and no mature females within a total of 580 dissected individuals. These data suggest that the reproductive tactics of A. tenebrosa are essentially fixed and that variation in the genotypic diversity of populations may reflect variation in factors such as the input of sexually derived planktonically dispersed recruits or post-settlement processes. However, the apparent lack of females paradoxically implies that sexual reproduction, and hence recruitment, must be rare or no longer possible within some populations, and highlights the need for long-term studies of these populations.

Naval training communities use a systems approach to training. This study considers the question "what is effectiveness in naval training?" Instructor courses of five navies were observed and naval instructors surveyed. Conclusions and recommednations are drawn from the data. It is proposed that most problems of effectiveness identified can be overcome without investment in technology or significant system changes.

Courtship displays are often important in determining male mating success but can also be costly. Thus, instead of courting females indiscriminately, males might be expected to adjust their signalling effort strategically. Theory, however, predicts that such adjustments should depend on the rate with which males encounter females, a prediction that has been subject to very little empirical testing. Here, we investigate the effects of female encounter rate on male courtship intensity by manipulating the time interval between sequential presentations of large (high quality) and small (low quality) females in a fish, the Australian desert goby Chlamydogobius eremius. Males that were presented with a small female immediately after a large female reduced their courtship intensity significantly. However, males courted large and small females with equal intensity if the interval between the sequential presentations was longer. Our results suggest that mate encounter rate is an important factor shaping male reproductive decisions and, consequently, the evolutionary potential of sexual selection.

The sharing of music files has been the focus of a massive struggle between representatives of major record companies and artists in the music industry, on one side, and peer-to-peer (p2p) file-sharing services and their users, on the other. This struggle can be analysed in terms of tactics used by the two sides, which can be classified into five categories: cover-up versus exposure, devaluation versus validation, interpretation versus alternative interpretation, official channels versus mobilisation, and intimidation versus resistance. It is valuable to understand these tactics because similar ones are likely to be used in ongoing struggles between users of p2p services and representatives of the content industries.

In this study, we repeatedly observed individually marked Brook Trout in a field setting to determine if the bimodal variation in foraging tactics previously observed in young salmonid populations (without distinct morphological differences) is generated by individuals specializing at different foraging tactics. We found significant but low repeatability in several foraging movement parameters. This indicated that while individuals did have tendencies to be sedentary versus highly active, there was considerable variation in foraging activity within individuals. These results suggest that relatively consistent differences among individuals may facilitate selection for specialized morphology and that there may be a heritable component to activity.

This essay reconceptualizes “social capital” as it relates to scholarship regarding the traditional news media. Much academic attention links the news media to Robert Putnam's view which focuses on social capital as enhancing “civic pride” and collective/community involvement. I suggest Putnam's perspective is often adopted without wider exploration of what the theory may offer the future of the commercial news media in western societies. This essay proposes the term “mediated social capital” may be a more suitable lens through which to consider this theory, taking a cue from Pierre Bourdieu who views social capital as a resource of power that may be utilized to maintain or build a position of advantage.

Vivemos um momento de grande crescimento da construção naval no mundo, que é impulsionado pelo crescimento do comércio entre as nações, em um mundo cada vez mais globalizado. O mesmo se repete no Brasil. O principal objetivo deste trabalho é propor uma metodologia para o cálculo objetivo do valor a ser garantido ao cliente da indústria naval, de modo a criar os incentivos econômicos corretos para os artífices da relação principal-agente. Inicialmente descreverei a Tecnologia da Construção Naval e os problemas econômicos que encontramos, passando a seguir a expor o Mercado Naval, estando aí incluídos o lado consumidor e os produtores de navios, sendo eles nacionais ou internacionais. Finalmente passaremos ao detalhamento da solução de seguro proposta, com a formalização do tipo de seguro, nível de cobertura e monitoramento. Para tal utilizaremos a metodologia de Vector-Autorregression combinada a uma simulação de Monte Carlo. Os resultados encontrados são checados e apontamos os caminhos para aperfeiçoar a metodologia e possíveis usos alternativos para ela.
