Isolated growth hormone deficiency type-2 (IGHD-2), the autosomal-dominant form of GH deficiency, is mainly caused by specific splicing mutations in the human growth hormone (hGH) gene (GH-1). These mutations, occurring in and around exon 3, cause complete exon 3 skipping and produce a dominant-negative 17.5 kD GH isoform that reduces the accumulation and secretion of wild type-GH (wt-GH). At present, patients suffering from IGHD-2 are treated with daily injections of recombinant human GH (rhGH) in order to reach normal height. However, this type of replacement therapy, although effective in terms of growth, does not prevent toxic effects of the 17.5-kD mutant on the pituitary gland, which can eventually lead to other hormonal deficiencies. Considering a well-known correlation between the clinical severity observed in IGHD-2 patients and the increased expression of the 17.5-kD isoform, therapies that specifically target this isoform may be useful in patients with GH-1 splicing defects. This chapter focuses on molecular strategies that could represent future directions for IGHD-2 treatment.

ABSTRACT Lime Roman mortars from the Iberian Peninsula has been analyzed in this reesearch. A group of samples were selected from some sites and all the samples come from industrial structures. All this samples show ceramic additives. From the start, the effort was centered in the pieces of pottery that were found in the mortars. The samples were treated and processed to develop a first phase of the research using macroscopy. With this technique, great results were achieved in the characterization of mortars, the microstratigraphy and the location of the ceramic pieces. While observing these pieces, it was seen that the fragments of pottery had a ring bordering the piece. The pieces with the bigger and more vivid rings were chosen and they were analyzed by a polarized light microscope. The mortars were characterized microscopically and it also showed that the rings were not a physical alteration or a visual defect. After some more tests, the rings were a chemical change associated with the hydraulicity of the mortar. The best samples were selected and mappings of their chemical elements were performed in order to know what could be happening in the interface between the ceramic matrix fragment and lime. With the results obtained it was found that there was a potential; both calcium accumulation within the ceramic grains and outside, just in the area of contact with the binder, forming spots longitudinally along the edge. These data were very useful for carrying out the next phase of study, which would meet promptly what was happening chemically in the area of contact. Another group of samples were taken, and this time focused on ceramic grains that met a statistical variables. Grains were chosen with two types of cooking as well as grains with different sizes and grains with ash additive in the matrix of lime, thinking that might be the most logical to be some sort of chemical change between the baked clay and lime array variables . Such variables were adapted to a multi-varial and geometric statistical system in order to synthesize the results and optimally display the data together, as mentioned several times in this work. After selecting the variables grains proceeded to perform a linear and spectral analysis SEM-EDX. This analysis led to determine that the chemical changes were graduals. These changes are summarized in an increase in the percentages of calcium inside the reaction rim of ceramics, from the edge to the outer ring. So percentage increasing is also observed at the interface of the ceramic matrix fragment with lime, confirming the results obtained by the mapping. Overall in all samples can be seen a shoulder in graphic calcium through the area of the ring reaction condition. The remaining percentages of magnesium, silicon and aluminum are usual. We have promptly confirmed that the increase of calcium is accentuated in samples where there is no ash and lime matrix. Cases for these grains suffer a greater increase than the rest. The second variable suffering more calcium is increased corresponding to good cooking grains with clay. Therefore, it appears that the variable size of the fragment is not critical. Therefore, considering the visual tests to the rings and their response to chemical results, we might think that increasing calcium inside the ceramic fragments was due to an injection of calcium inside clay in the run-up to the setting phase. It is at this point that would be the key to the chemical change that occurs at this interface, silica-calcium aluminate some authors have already investigated. This injection of calcium into the grain does not come alone, but generate a lime crust on the outside interface of ceramics, which we tested for mapping is real in our samples. The consequence of these results is the improvement of our understanding of historical hydraulic factor in building materials, such as mortar. For example, knowing that the incorporation of organic materials such as ash powder, may be detrimental to the injection of calcium inside the ceramic additives. Archaeologically speaking, it's very interesting to correlate different factories or structures located on a single site, or in the same regional area. Characterization studies and microstratigraphy not only provide new information to help restore mortars, but create the possibility of generating constructive patterns that serve as guide fossils to determinate the age of the structures. With regard to the results obtained in different archaeological sites it has seen a difference between mortars of pools or sinks with respect to industrial floors of undetermined use. The sample of the site of Las Arenas does not have any micro-stratigraphy, as if instead in the samples obtained in Rotonda de Mejorada, Val de la Viña and La Magdalena sites. In these settlements the structures are really similar, with different construction levels using lime mortars with aggregates and gravel in the inner layers, and ceramic aggregates as external additions. With regard to the grain size of the ceramic additions Val de la Viña and La Magdalena samples has several coincidences about the size of grains and distribution. Also, samples taken at La Magdalena, there is a difference between the MG1, MG2, MG3 and MG4 samples and the MG5 and MG6 samples, so the last corresponding to a mortar samples with larger ceramic fragments. The EMG1 and EMG2 structures, corresponding to a bucket and a pool of La Magdalena settlement, have similarities with regard to micro-stratigraphy and grain size. It has been determined that the function and manufacturing must be linked with a same chronocultural context.

In humans, only a small fraction (2-12%) of a sperm population can respond by chemoattraction to follicular factors. This recent finding led to the hypothesis that chemotaxis provides a mechanism for selective recruitment of functionally mature spermatozoa (i.e., of capacitated spermatozoa, which possess the potential to undergo the acrosome reaction and fertilize the egg). This study aimed to examine this possibility. Capacitated spermatozoa were identified by their ability to undergo the acrosome reaction upon stimulation with phorbol 12-myristate 13-acetate. Under capacitating conditions, only a small portion (2-14%) of the spermatozoa were found to be capacitated. The spermatozoa were then separated according to their chemotactic activity, which resulted in a subpopulation enriched with chemotactically responsive spermatozoa and a subpopulation depleted of such spermatozoa. The level of capacitated spermatozoa in the former was approximately 13-fold higher than that in the latter. The capacitated state was temporary (50 min < life span < 240 min), and it was synchronous with the chemotactic activity. A continuous process of replacement of capacitated/chemotactic spermatozoa within a sperm population was observed. Spermatozoa that had stopped being capacitated did not become capacitated again, which indicates that the capacitated state is acquired only once in a sperm's lifetime. A total sperm population depleted of capacitated spermatozoa stopped being chemotactic. When capacitated spermatozoa reappeared, chemotactic activity was restored. These observations suggest that spermatozoa acquire their chemotactic responsiveness as part of the capacitation process and lose this responsiveness when the capacitated state is terminated. We suggest that the role of sperm chemotaxis in sperm-egg interaction in vivo may indeed be selective recruitment of capacitated spermatozoa for fertilizing the egg.

We extend an earlier model of protein evolution on a rugged landscape to the case in which the landscape exhibits a variable degree of correlation (i.e., smoothness). Correlation is introduced by assuming that a protein is composed of a set of independent blocks or domains and that mutation in one block affects the contribution of that block alone to the overall fitness of the protein. We study the statistical structure of such landscapes and apply our theory to the evolution by somatic hypermutation of antibody molecules composed of framework and complementarity-determining regions. We predict the expected number of replacement mutations in each region.

In this paper, we discuss two-dimensional failure modeling for a system where degradation is due to age and usage. We extend the concept of minimal repair for the one-dimensional case to the two-dimensional case and characterize the failures over a two-dimensional region under minimal repair. An application of this important result to a rnanufacturer's servicing costs for a two-dimensional warranty policy is given and we compare the minimal repair strategy with the strategy of replacement of failure. The electrical ceramic insulators industry, uses noble raw materials such as siliceous and aluminous clays of white burning, in order to provide plasticity of the mass and contribute to electrical and mechanical properties required of the product, and feldspar with the flux function In literature references the composition of the masses indicates that the clay participates in percentage between 20 and 32, and feldspar 8 to 35, these materials have significant cost. In this research was performed the total replacement of commercial clay, for white burning clay from Santa Luzia region in southern Bahia and partial replacement of feldspar by ash residue of husk conilon coffee burning, from extreme south of Bahia. The objective of replacement these raw materials is to aver its technical feasibility and call attention for the embryo pole of ceramic industry for the existing in the south and extreme south of Bahia, which has significant reserves of noble raw materials such as clay white burning, kaolin, quartz and feldspar, and generates significant volume of gray husk conilon coffee as alternate flux. Clay Santa Luzia is prima noble material whose current commercial application is the production of white roofing. The residue of coffee husk ash is discarded near of production sites and is harmful to the environment. Phase diagrams and statistic design of experiments, were used for optimization and cost savings in research. The results confirmed the expectations of obtaining electrical ceramic insulators, with white burning clay of Santa Luzia and partial replacement up to 35.4% of feldspar, by treaty residue of conilon ash coffee husk burning. The statistic design that showed best results was for formulation with percentages of: clay 26.4 to 30.4%; kaolin 14.85 to 17.1%; feldspar 12.92 to 16.96%; R2 residue 7.08 to 9.2% and Quartz 32.5 to 38.75%, relative to the total mass of the mixture. The best results indicated; 0.2 to 1.4% apparent porosity , water absorption 0.1 to 0.7%, flexural strength 35 to 45MPa , dielectric strength 35-41 kV/cm , the transverse resistivity 8x109 2.5x1010 Ω.cm and for the dielectric constant ε/ε0 7 to 10.4, specification parameters for manufacturing ceramic electrical insulators of low and medium voltage.

Scientific research in forest production technology area search techniques that increase production per unit area, with high economic viability and reducing environmental impacts. When dealing with forest plantations, studies are needed in the production of biomass and its nutrient content, and these are data parameters for planning the environmental implications of different intensities of forest harvesting. Given the above, this study aimed to elucidate the production and export of biomass and nutrients for two species of the genus Eucalyptus (E. grandis and E. urophylla) grown in the southwestern region of Parana. For this, it was evaluated: the stock of biomass and nutrients in eucalyptus (wood, bark, branches and leaves) at 60 months of age; the export rate of nutrients; the calorific value and economic viability. The biomass and the largest eucalyptus nutrient stocks are predominantly allocated to the stem (wood + bark). The components of biomass showed different chemical compositions, generally being higher in the leaves and bark and lower in wood and branches components. As for the calorific value, the leaves had its calorific value statistically superior than the other fractions, followed by branches, wood and bark. The organic carbon content (C.O.) is directly connected to the calorific value, and the calorific value increases as its content increase. The wood had the highest nutrient use efficiency values, something highly desirable and of great interest to forestry. The leaves showed smaller nutrient utilization efficiency values, with the exception of Ca and Mg that were smaller in the bark, indicating the importance of maintaining these components in the soil after harvest. The wood fraction presents the biomass lower cost when considering the replacement of nutrients exported by its biomass. On the other hand, the leaf fraction showed NPK higher cost of replacement.

Within the Kauffung Limestone dolomite bodies related to volcanic contacts occure and are interpreted as a post-Variscan, hydrothermal volcanogenic formation. Two genetic dolomite types are discernible: a pervasive replacement saddle dolomite and a cavity filling saddle dolomite cement. Micro texture and oxygen isotopy of both dolomite types refer to heightened temperatures of formation. The ocurrence of the dolomite in contact to cross-cutting rhyolithic dikes points to a close petrogenetic relation. Dolomite bodies and rhyolithic injections in contrast to the rock wall are characterized by a distinctive cavernous texture, so that a prekinematic genesis is excluded. The formation of replacement saddle dolomite and saddle dolomite cement altogether are considered as a concomitant phenomenon of the Permo-Carboniferous volcanism widespread in the Bober-Katzbach Mountains.

The municipal management in any country of the globe requires planning and allocation of resources evenly. In Brazil, the Law of Budgetary Guidelines (LDO) guides municipal managers toward that balance. This research develops a model that seeks to find the balance of the allocation of public resources in Brazilian municipalities, considering the LDO as a parameter. For this using statistical techniques and multicriteria analysis as a first step in order to define allocation strategies, based on the technical aspects arising from the municipal manager. In a second step, presented in linear programming based optimization where the objective function is derived from the preference of the results of the manager and his staff. The statistical representation is presented to support multicriteria development in the definition of replacement rates through time series. The multicriteria analysis was structured by defining the criteria, alternatives and the application of UTASTAR methods to calculate replacement rates. After these initial settings, an application of linear programming was developed to find the optimal allocation of enforcement resources of the municipal budget. Data from the budget of a municipality in southwestern Paraná were studied in the application of the model and analysis of results.

Replacement of fishmeal by soybean meal in juvenile curimba (Prochilodus lineatus) diets was evaluated. Five isonitrogenous (26% crude protein) and isocaloric (4,000 kcal of gross energy kg(-1)) diets with 0, 25, 50, 75 and 100% of replacement were used. Fish (3.71 +/- 0.54 g) were distributed in 25 tanks (120 liters). The diets were provided for 84 days. Total replacement of fishmeal (100%) by soybean meal worsened weight gain, feed conversion and protein efficiency ratio. An increase in carcass fat was observed at the expense of reducing protein percentage, as well as an increase in hepatosomatic index. It was concluded that replacing 75.0% fish meal by soybean meal (33.2% of inclusion) did not affect the animal growth, although there been observed changes in corporal composition and hepatic hyperplasia.

Glass fibre-reinforced plastics (GFRP), nowadays commonly used in the construction, transportation and automobile sectors, have been considered inherently difficult to recycle due to both the cross-linked nature of thermoset resins, which cannot be remoulded, and the complex composition of the composite itself, which includes glass fibres, polymer matrix and different types of inorganic fillers. Hence, to date, most of the thermoset based GFRP waste is being incinerated or landfilled leading to negative environmental impacts and additional costs to producers and suppliers. With an increasing awareness of environmental matters and the subsequent desire to save resources, recycling would convert an expensive waste disposal into a profitable reusable material. In this study, the effect of the incorporation of mechanically recycled GFRP pultrusion wastes on flexural and compressive behaviour of polyester polymer mortars (PM) was assessed. For this purpose, different contents of GFRP recyclates (0%, 4%, 8% and 12%, w/w), with distinct size grades (coarse fibrous mixture and fine powdered mixture), were incorporated into polyester PM as sand aggregates and filler replacements. The effect of the incorporation of a silane coupling agent was also assessed. Experimental results revealed that GFRP waste filled polymer mortars show improved mechanical behaviour over unmodified polyester based mortars, thus indicating the feasibility of GFRP waste reuse as raw material in concrete-polymer composites.

In this study, the effect of incorporation of recycled glass fibre reinforced plastics (GFRP) waste materials, obtained by means of shredding and milling processes, on mechanical behaviour of polyester polymer mortars (PM) was assessed. For this purpose, different contents of GFRP recyclates, between 4% up to 12% in weight, were incorporated into polyester PM materials as sand aggregates and filler replacements. The effect of the addition of a silane coupling agent to resin binder was also evaluated. Applied waste material was proceeding from the shredding of the leftovers resultant from the cutting and assembly processes of GFRP pultrusion profiles. Currently, these leftovers as well as non-conform products and scrap resulting from pultrusion manufacturing process are landfilled, with additional costs to producers and suppliers. Hence, besides the evident environmental benefits, a viable and feasible solution for these wastes would also conduct to significant economic advantages. Design of experiments and data treatment were accomplish by means of full factorial design approach and analysis of variance ANOVA. Experimental results were promising toward the recyclability of GFRP waste materials as partial replacement of aggregates and reinforcement for PM materials, with significant improvements on mechanical properties of resultant mortars with regards to waste-free formulations.
