Cells of Mikania glomerata, Cephaelis ipecacuanha and Maytenus aquifolia were co-cultured in a two-phase system using filter paper as a solid support. The species were co-cultured in all possible paired combinations. Interaction between Mikania and Maytenus cells resulted in increased biomass production of Maytenus cells, but the friedelin content was reduced. Co-cultivation of Cephaelis and Mikania cells enhanced coumarin content, but inhibited the growth of Mikania cells. However, yield of emetine as well as Cephaelis biomass accumulation were positively stimulated by the co-cultivation. Results indicate a possible occurrence of allelopathy in such a system.

Alocação de biomassa e nutrientes em Myriophyllum aquaticum sob diferentes níveis de macronutrientes

O objetivo deste trabalho foi verificar o efeito de diferentes concentrações de macronutrientes no crescimento e na alocação de biomassa seca em folha, caule e raiz de Myriophyllulm aquaticum. As plantas foram cultivadas em vasos plásticos preenchidos com areia lavada e emersas em solução nutritiva. O experimento foi conduzido em um delineamento inteiramente ao acaso, com quatro níveis (0, 25, 50 e 75%) da concentração original de N, P, K Ca, Mg e S da solução nutritiva, além de uma testemunha (100% da solução nutritiva), com quatro repetições e por um período de 38 dias. A maior produção de biomassa total foi obtida na testemunha. A ausência de K afetou a alocação de biomassa em raízes. Quanto ao comprimento total dos ramos, a maior resposta ocorreu entre os níveis de Ca a 60 mg L-1. A solução a 100% dos macronutrientes propiciou o maior comprimento das plantas. O número de ramos entre os níveis de N e P seguiu uma tendência linear e crescente com as concentrações desses nutrientes, porém em maior proporção para o fósforo. As condições mais favoráveis ao desenvolvimento das plantas ocorreram a 100% dos macronutrientes da solução base. Entre os níveis de nitrogênio verificaram-se maiores teores de N nas folhas do que nos caules. Já para os demais nutrientes a alocação desses recursos foi destinada em maior proporção para o caule. A maior alocação de biomassa nesta espécie destinou-se ao caule.

The seasonal variation of the periphytic community attached to an artificial substratum (glass tubes) was studied during two different periods in a lagoon connected to the Paranapanema River, the main tributary of the Jurumirim Reservoir (São Paulo-Brazil). An analysis of dry weight, ash free dry weight, chlorophyll a, phaeophytin and primary productivity of periphyton was carried out. The first experiment lasted from August to December 1993, the second from February to June 1994. Tubes were removed after 7, 14, 21, 28, 60, 90 and 119 days of incubation. In the 1st experiment, the periphytic community reached a higher biomass after the 4th week of colonization (28th day), in the 2nd experiment after the 2nd week (14th day). This discrepancy is related to seasonal differences in environmental factors (water temperature, nutrients concentrations and water discharge) that determine initial colonization. After the first stages of colonization, the biomass and primary productivity of periphyton reached their maximum values after the 60th day of incubation. In both experiments, three developmental phases could be discerned. In the initial phase, an exponential growth was observed. In the second phase, the bioderm reached its maximum biomass and productivity. In the third phase, a decrease of biomass and productivity occurred.

Penicillium citrinum grown in orange juice processing wastes medium under continuous agitation was studied in order to establish optimal conditions (incubation period, incubation temperature, initial pH and nitrogen addition) for biomass and ribonuclease production, as well as, biological depuration of the wastes. Nitrogen addition to wastes medium increased the biomass and ribonuclease production and provides COD reduction. The soy meal shows to be the best nitrogen source. The conditions for a more favorable enzyme and biomass production and COD reduction were initial pH 6.0 and temperature of 27°C. The maximum value obtained for these parameters on optimal conditions of cultivation was 11 U/mL of enzyme, 4 mg/mL of biomass (dry matter) and 55% of COD reduction, in 96 hours of incubation.

The cassava processing industry generates wastewater named manipueira with a high organic content. Although considered a pollutant, manipueira can be used as substrate for fermentative processes including the cultivation of Geotrichum fragrans. This aerobic microorganism isolated from cassava wastewater has cyanide resistant respiration. Under cassava wastewater cultivation, G. fragrans produced fruit aroma volatile compounds. This study evaluated volatile compounds produced by G. fragrans in cassava liquid waste. The waste had a sugar composition composed of dextrin (2.6%), maltose (1.4%), sucrose (32.1%), glucose (38.3%), and fructose (25.6%). The average value of total sugars was 58.2 g l(-1), composed of 38.0 g l(-1) reducing and 20.2 g l(-1) non-reducing sugars. The chemical oxygen demand (COD) average value was 60 000 mg l(-1). G. fragrans used sugars (fructose and glucose) for energy generation reducing the COD value of the cassava wastewater by 40%. Biomass production of G. fragrans cultivated for 12 h in natural cassava liquid waste was 12.8 g l(-)1. The volatile compounds identified in the cassava liquid waste after 72 h cultivation were: 1-butanol, 3-methyl 1-butanol (isoamylic alcohol), 2-methyl 1-butanol, 1-3 butanodiol and phenylethanol; ethyl acetate, ethyl propionate, 2-methyl ethyl propionate and 2-methyl propanoic. The effect of substrate supplementation with glucose (50 g l(-1)), fructose (50 g l(-1)) and aqueous yeast extract (200 ml l(-1)) did not affect the qualitative and quantitative profiles of volatile compounds. These results indicate that the carbon (C) source utilized by microorganism was glucose or fructose, while nitrogen (N) supplementation was not necessary because the agent did not exhaust all the nitrogen of the wastewater. Cell cultures of Mikania glomerata Sprengel were established with leaf segments cultured on White medium supplemented with 1 mg/L BA and 3 mg/L NAA. Different types and concentrations of growth regulators were tested for callus maintenance. Determination of coumarin content was performed in HPLC using authentic coumarin standard. Growth regulator concentration affected biomass and coumarin accumulation. Cultures developed in semisolid medium containing both BA and NAA exhibited enhanced biomass production as well as coumarin accumulation. In the most favorable conditions tested, cells accumulated 25 μg/g of dry weight what is much inferior to the yield already reported in intact plants (5 mg/g of dry weight). However, results obtained so far suggest several alternatives for culture manipulation in order to optimize the productivity of coumarin by M. glomerata cultured cells.

The growth and the extracellular amylase production by Aspergillus ochraceus were studied in a stationary culture medium. Maximum growth rate of this fungus was found after 5 days of incubation at 30° C, but maximum amylase production was obtained after 2 days. The highest amylase production were attained with lactose, maltose, xylose and starch as carbon sources. The extracellular amylase production and mycelial growth were influenced by the concentration of starch. Other carbohydrates supported growth but did not induce amylase synthesis and glucose repressed it, indicating catabolite repression in this microorganism. The presence of both mechanisms of induction and repression suggests that at least these multiple forms of regulation are present in A. ochraceus. Of the nitrogen sources tested, casaminoacids, ammonium nitrate and sodium nitrate stimulated the highest yield of amylase. Optimal amylase production was obtained at pH 5.0, but enzyme activity was found only in the 4.0-6.0 pH range. These results were probably due to the inhibitory effect of NH 4 +-N in the culture medium.

The objective of this research was to investigate the potential of xylanase production by Aspergillus japonicus and to determine the effects of cultivation conditions in the process, aiming toward optimization of enzyme production. The best temperature, as well as the best carbon source, for biomass production was determined through an automated turbidimetric method (Bioscreen-C). The enzyme activity of this fungus was separately evaluated in two solid substrates (wheat and soybean bran) and in Vogel medium, adding other carbon sources. Temperature effects, cultivation time, and spore concentrations were also tested. The best temperature for enzyme and biomass production was 25°C; however, the best carbon source for growth (determined by the Bioscreen C) did not turn out to be a good inducer of xylanase production. Maximum xylanase activity was achieved when the fungus was cultivated in wheat bran (without the addition of any other carbon source) using a spore concentration of 1 × 107 spores/mL (25°C, pH 5.0, 120 h). A. japonicus is a good xylanase producer under the conditions presented in these assays. © 2006 Academic Journals.
Fatty acid production by four strains of Mucor hiemalis grown in plant oil and soluble carbohydrates

Four Mucor hiemalis strains (M1, M2, M3 and M4), isolated from soil at a depth of 0 - 15 cm in the Juréia-Itatins Ecology Station (JIES), in the state of São Paulo, Brazil and were evaluated for the production of γ-linolenic (GLA) and other unsaturated fatty acids. Five growth variables (temperature, pH, carbon source, nitrogen source, and vegetable oils) were studied. Liquid media containing 2% vegetable oil (palm oil, canola oil, soybean oil, sesame oil, or sunflower oil) or 2% carbohydrate (fructose, galactose, glycerol, glucose, lactose, maltose, sucrose, sorbitol or xylose) and 1% yeast extract as a nitrogen source were used. The greatest biomass production was observed with M3 and M4 strains in palm oil (91.5 g l -1) and sunflower oil (68.3 g l -1) media, respectively. Strain M4 produced greater quantities of polyunsaturated acids in medium containing glucose. The GLA production in the M4 biomass was 1,132.2 mg l -1 in glucose medium. Plant oils were inhibitors of fatty acid production by these strains. © 2007 Academic Journals.

Many plants utilized in the urban center shows substances considered toxic whose production could be influenced by some factors, like hydric stress, including ornamental Nerium oleander L., widely used in gardens in various parts of the world, which presents production of cardioative glucosides, considered toxic. This study had the objective to evaluate the effect of field capacity in the biomass and the level of cardioative glucosides in seedlings of Nerium oleander. The experiment was carried at UNIDERP, in Campo Grande City, Mato Grosso do Sul State, Brazil, at the University for Development of State and Pantanal Region, using the experimental delineation in randomized blocks. There were 4 treatments (25%; 50%; 75% and 100% of the field capacity), 5 replications and 4 plants by parcel, totalling 80 plants. The evaluations were realized 60 days after the seedlings were planted. The quantitative analysis of the cardioative glucosides was realized by gravimetric test, after selective extraction of the glucosides. Were conclude that increase of the quantity of water in the soil raised the biomass production until 75% of the field capacity and increased the level of cardioative glucosides, showing that water management is very important and should be provided only the necessary to development of the plant.

Upland rice (Oryza sativa L.) cultivation has been increasing in importance in Asia while water availability for irrigation has been decreasing because of rapid growth in industry and urban centers. Therefore, the development of technologies that increase upland rice yields under aerobic conditions, thereby saving water, would be an effective strategy to avoid a decrease in global rice grain production. The use of the no-tillage system (NTS) and cover crops that maintain soil moisture would prove advantageous in the move toward sustainable agriculture. However, upland rice develops better in plowed soil, and it has been reported that this crop does not perform well under the NTS. Therefore, the aim of this study was to investigate the effect of cover crops on upland rice grain yield and yield components sowed in a NTS. A field experiment was conducted during two growing seasons (2008-2009 and 2009-2010), and treatments consisted of growing rice under five cover crops in a NTS and two control treatments under the conventional tillage system (plowing once and disking twice). Treatments were carried out in a randomized block design with three replications. Our findings are as follows: On average, Brachiaria brizantha (12.32Mgha-1), Brachiaria ruziziensis (11.08Mgha-1) and Panicum maximum (11.62Mgha-1) had outstanding biomass production; however, these grasses provided the worst upland rice yields (2.30, 2.04, and 2.67Mgha-1, respectively) and are not recommended as cover crops before upland rice. Millet and fallow exhibited the fastest straw degradation (half-lives of 52 and 54 days, respectively), and millet exhibited the fastest nitrogen release (N half-life of 28 days). The use of a NTS was promising when millet was used as a cover crop; this allowed the highest upland rice yield (3.94Mgha-1) and did not statistically differ from plowed fallow (3.52Mgha-1). © 2012 Elsevier B.V.
