Salivary gland dysfunction is a feature in diabetes and hypertension. We hypothesized that sodium-glucose cotransporter 1 (SGLT1) participates in salivary dysfunctions through a sympathetic- and protein kinase A (PKA)-mediated pathway. In Wistar-Kyoto (WKY), diabetic WKY (WKY-D), spontaneously hypertensive (SHR), and diabetic SHR (SHR-D) rats, PKA/SGLT1 proteins were analyzed in parotid and submandibular glands, and the sympathetic nerve activity (SNA) to the glands was monitored. Basal SNA was threefold higher in SHR (P < 0.001 vs. WKY), and diabetes decreased this activity (similar to 50%, P < 0.05) in both WKY and SHR. The catalytic subunit of PKA and the plasma membrane SGLT1 content in acinar cells were regulated in parallel to the SNA. Electrical stimulation of the sympathetic branch to salivary glands increased (similar to 30%, P < 0.05) PKA and SGLT1 expression. Immunohistochemical analysis confirmed the observed regulations of SGLT1, revealing its location in basolateral membrane of acinar cells. Taken together, our results show highly coordinated regulation of sympathetic activity upon PKA activity and plasma membrane SGLT1 content in salivary glands. Furthermore, the present findings show that diabetic- and/or hypertensive-induced changes in the sympathetic activity correlate with changes in SGLT1 expression in basolateral membrane of acinar cells, which can participate in the salivary glands dysfunctions reported by patients with these pathologies.

Mandibular prognathism typically shows familial aggregation. Various genetic models have been described and it is assumed to be a multifactorial and polygenic trait, with a threshold for expression. Our goal was to examine specific genetic models of the familial transmission of this trait. The study sample comprised of 2,562 individuals from 55 families. Complete family histories for each proband were ascertained and the affection status of relatives were confirmed by lateral cephalograms, photographs, and dental models. Pedigrees were drawn using PELICAN and complex segregation analysis was performed using POINTER. Parts of some pedigrees were excluded to create one founder pedigrees, so the total N was 2,050. Analysis showed more affected females than males (P = 0.030). The majority of the pedigrees suggest autosomal dominant inheritance. Incomplete penetrance was demonstrated by the ratio of affected/unaffected parents and siblings. The heritability of mandibular prognathism was estimated to be 0.316. We conclude that there is a major gene that influences the expression of mandibular prognathism with clear signs of Mendelian inheritance and a multifactorial component. (C) 2007 Wiley-Liss, Inc.

Triatoma infestans (Hemiptera: Reduviidae) is a hematophagous insect that transmits the protozoan parasite Trypanosoma cruzi, the etiological agent of Chagas` disease. Its saliva contains trialysin, a protein that forms pores in membranes. Peptides based on the N-terminus of trialysin lyse cells and fold into alpha-helical amphipathic segments resembling antimicrobial peptides. Using a specific antiserum against trialysin, we show here that trialysin is synthesized as a precursor that is less active than the protein released after saliva secretion. A synthetic peptide flanked by a fluorophore and a quencher including the acidic proregion and the lytic N-terminus of the protein is also less active against cells and liposomes, increasing activity upon proteolysis. Activation changes the peptide conformation as observed by fluorescence increase and CD spectroscopy. This mechanism of activation could provide a way to impair the toxic effects of trialysin inside the salivary glands, thus restricting damaging lytic activity to the bite site.

Neste trabalho é apresentado um método para medição de deslocamentos sem contato, utilizando sensores magnetoresistivos, os quais são sensibilizados pela variação do campo magnético produzido por um imã permanente em deslocamento no espaço. Os sensores magnetoresistivos possuem, internamente, uma ponte de Wheathestone, onde a resistência varia conforme varia o campo magnético, de modo que o circuito mais indicado para este caso é um amplificador e um filtro para cada sensor. O principal objetivo do trabalho é a obtenção de uma técnica para medir deslocamentos sem contato, e estender os resultados para medida de movimentos mandibulares. A montagem consiste de duas placas de celeron, distantes 30mm uma da outra e unidas por parafusos de polietileno. Em cada uma destas placas foram dispostos quatro sensores, num total de oito, sendo que para cada um deles existe um circuito de amplificação e filtragem do sinal de saída. Sob uma chapa de alumínio foi fixado este equipamento e uma mesa de calibração em 3D, a qual, após a obtenção da matriz de calibração, foi substituída por um emulador de movimento mandibular. Os parâmetros do modelo foram estimados através do método dos mínimos quadrados com auxílio do software Matlab, Release 12. Este software também foi utilizado para o sistema de aquisição de dados durante a realização dos experimentos. A imprecisão dos resultados obtidos na determinação dos deslocamentos, está na ordem de décimos de milímetros. O trabalho apresenta, também, o mapeamento do campo magnético do magneto utilizado nos experimentos através do software FEM2000 – Método de elementos finitos aplicado ao eletromagnetismo.

Foi realizado um estudo comparativo entre os resultados das medidas obtidas segundo dois traçados (Altonen, Haavikko e Mattila x Puricelli), efetuadas sobre radiografias panorâmicas, verificando-se a presença ou ausência de compatibilidade entre os mesmos. Avaliou-se também a direção de crescimento mandibular (vertical ou sagital) na amostra composta por pacientes com maloclusão de Classe II esquelética e ainda a existência de uma relação desta direção com o grau de inclinação dos terceiros molares. Desta forma, as seguintes medidas foram comparadas: ângulo β de Altonen, Haavikko e Mattila correspondente ao γ de Puricelli, o qual indicou o relacionamento dos terceiros molares inferiores em relação aos segundos molares; ângulo γA de Altonen, Haavikko e Mattila correspondente ao M2L3 de Puricelli, o qual indicou o ângulo de inclinação do segundo molar com uma linha de base e ângulo α de Altonen, Haavikko e Mattila correspondente ao Pc-Go-B de Puricelli, proporcionando o ângulo mandibular. Baseando-se nos resultados obtidos, apresentados e discutidos no presente trabalho e de acordo com a metodologia estudada, pode-se concluir que: 1) Os traçados diferem entre si, com relação a todas as medidas. Entretanto, apresentam uma correlação positiva para todas as medidas estudadas. 2) Há um crescimento mandibular predominantemente vertical (condilar) segundo o traçado proposto por Puricelli. 3) O grau de inclinação do terceiro molar não se correlaciona com a direção do crescimento mandibular indicada através do ângulo mandibular em ambos os traçados. Desta forma, é imprescindível que ao se compararem os resultados de diferentes estudos analisem-se apenas aqueles que utilizaram o mesmo traçado.

The purpose of this research was to study the mammary lymphatic drainage under a macroscopic and mesoscopic view, comparing the vascular pattern of healthy and neoplasic mammary glands injected with drawing ink alcoholic and fluorescein solutions, in 46 mongrel female dogs. The results pointed out that the thoracic gland is drained by the axillary lymph centre, but in mammary neoplasia either superficial cervical or ventral thoracic lymph centres can be involved. Cranial and caudal abdominal glands may be drained by the axillary, inguinofemoral and popliteal lymph centres. However, the popliteal drainage is specific for the healthy caudal abdominal mammary gland. The inguinal gland can be drained by both inguinofemoral and popliteal lymph centres in both neoplasic and healthy conditions. Regarding the mammary lymphatic communications, this research demonstrated that neoplasic glands present more types of anastomosis (40.9%), than healthy glands (33.33%), and an increase in contralateral anastomosis (50%) compared with healthy ones (33%). Given the data, the mammary neoplasia can change the lymphatic drainage pattern in terms of lymph centres and vascular arborization, thus forming new drainage channels and recruiting a larger number of lymph nodes. Lastly, some comments were made about the severity of a specific neoplasic mammary gland and conditions to be considered before making a decision in terms of the most adequate operative procedure, and suggestions for further investigations.

We investigated the changes of minor salivary glands during 4NQO-induced rat tongue carcinogenesis. Histopathological examinations of serous and mucous tongue salivary glands of 30 male Wistar rats were performed after 4, 12 and 20 weeks of 50 ppm 4NQO chronicle administration in drinking water. Ten rats were used as control. Morphometric results were expressed as volume density (Vv %) of each of the components. Histopathological and morphometric changes in the salivary glands were evident only at 20 weeks following 4NQO administration and they included a significant (P < 0.05) decreased in the mean Vv of the serous acini compared with the control group accompanied by abnormal acini (Vv=14 %). Neither mucous acini nor ducts demonstrated significant changes. In conclusion, minor salivary glands are involved in the progression of 4NQO-induced carcinoma.
