It is well known that a predator has the potential to regulate a prey population only if the predator responds to increases in prey density and inflicts greater mortality rates. Predators may cause such density-dependent mortality depending on the nature of the functional and numerical responses. Yet, few studies have examined the relationship between the addition of refuges and the characteristic of functional response fits. We investigated whether addition of a refuge changed the type of functional response exhibited by Dermestes ater on Musca domestica, comparing the inherent ability of D. ater to kill houseflies in the absence and in the presence of refuge. An additional laboratory experiment was also carried out to assess handling and searching times exhibited by D. ater. Logistic regression analyses revealed a type III functional response for predator-prey interaction without refuge, and results were described by the random predator equation. The mean number of prey killed did not differ between experimental habitats, indicating that the addition of refuge did not inhibit predation. However, predators that interacted with prey without refuge spent less time searching for prey at higher densities, increasing predatory interaction. We concluded that this interaction may be weak, because data from experiments with refuge fitted poorly to models. However, the high variability and the nonsignificance of the data from the experiment with refuge show the importance of refuge for promoting spatial heterogeneity, which may prevent prey extinction.

In the present study 21 young swine females, sexually mature, reared and kept under an industrial management system were assessed for cortisol and oestradiol-17 beta serum prolifes during the oestrus cycle. Blood sampling was performed always at the same interval, from 8:00 to 10:00 a.m. Each animal was submitted to 14 venal punctions at days 0, 2, 4, 6, 8, 10, 12, 14, 16, 18, 20, 21, 22 and 23 of the oestrous cycle. The first day of the oestrous phase was assumed to be day 0 and the 23(rd) as the first of the next cycle. Hormone determinations were performed using a solid phase radioimmunoassay (RIA). Regarding to cortisol concentrations, there was a variation from 3.5 to 8.0 mu g/dl and for oestradiol-17 beta the mean values detected varied from 3.5 to 14.9pg/ml.

Morphologic analysis of geographic strains of Musca domestica carried out on natural and laboratory experiments starting with 400 and 800 eggs showed phenotypic variations related with latitude. Females of the natural populations showed clines for several morphological traits of the wing, whereas male flies showed a reduction in the dispersion measures (s2 and CV) of wing width and length. The same reduction was obtained for males, females and total number of flies of the natural populations in dispersion of the number of bristles on the fourth abdominal sternite. A significant negative correlation was observed for the head width of females and for the total number of flies emerged in the laboratory experiments started with 400 eggs. All flies produced by the experiments starting with 800 eggs showed a reduction in variability of dispersion of the bristles on the fourth abdominal sternite in the strains obtained from locations south of the area analyzed. Evolutionary aspects of these correlation coefficients between morphometric traits and latitude are discussed.

We examined the variation in mitochondrial DNA by sequencing the D-loop region in wild and domestic (large-white breed) pigs, in hybrids between domestic and wild pigs, and in Monteiro pigs. A D-loop fragment of approximately 330 bp was amplified by PCR. Sequencing of DNA amplicons identified haplotypes previously described as European and Asian types. Monteiro pigs and wild pigs had European haplotypes and domestic pigs had both European and Asian haplotypes. ©FUNPEC-RP.

The roots' powder of timbo species collected in different regions of Amazonia that were tested in larvae groupings, didn't produce differential significative effects in two strains of Musca domestica. The two species with the greater number of plants used in the trial were Derris urucu and Derris nicou; the individuals from the species came from regions considered as "forestal refugies" during the Amazonian pleistocene. Among each species the plants varied since that inefficient to control, until plants lethals to the fies. This differential capacity for larvae control among plants of the same species, originated from different regions, suggests that both species had their populations isolated, during the quaternary epoch. In regions or "forestal refugies", where both species were represented, D. urucu was superior to D. nicou in the capacity to control larvae. While among plants from F region (Peruvian-East refuge) of the State of Acre, the two species had convergence in the values of damage to larvae groupings. Among the another species, Derris sp. (yellow timbo or watermelon timbo) didn't show differences in larvae control between samples from the two regions; while the species that was introduced in the Amazonia Denis elleptica showed damage in the larvae groupings similar to the most effective plants of D. nicou and D. urucu.

The rotenone contents analysis in clones of three timbó species permited the classification of these plants based on their efficiency to control Musca domestica larvae. The results showed significant correlations between the rotenone contents in the Derris urucu and D. nicou clones with the capacity to control the larvae. The plants with high rotenone contents were more effective. The rotenone contents, the effect of Derris species clones on the flics and the localization of the plants origin, showed that among the plants could have happened a populational isolation in the Amazonia during the pleistocene epoch. In Derris sp., that presented lower rotenone contents and was ineffective to control of the larvae, these differences were not observed.

Musca domestica larvae present two different digestive chymotryptic activities found in the posterior midgut (PMG): one major soluble activity in the lumen and another minor present in cell membrane fractions. Both soluble and membrane-bound chymotryptic activities have different half lives of thermal inactivation (46 degrees C) in the presence and absence of 10 mM Triton X-100, indicating that they are two different molecular species. Purified soluble chymotryptic activity has pH optimum 7.4 and a molecular mass of 28 kDa in SDS-PAGE. It does not cleave short substrates, such as Suc-F-MCA, preferring longer substrates, such as Suc-AAPF-MCA, with a primary specificity (kcat/Km) for Phe rather than Tyr and Leu residues. In-gel activity revealed a unique band against S-AAPF-MCA with the same migration as purified chymotrypsin. One chymotrypsinogen-like sequence (MdChy1) was sequenced, cloned and recombinantly expressed in Escherichia coli (DE3) Star. MdChy1 is expressed in the proximal posterior midgut (PMG1), as seen by RT-PCR. Expression analysis of other chymotrypsin genes revealed genes expressed at the anterior midgut (AMG) and PMG. Western blot of M. domestica midgut tissues using anti-MdChy1 antiserum showed a single band in samples from AMG and PMG, co-migrating with recombinant and purified enzymes. Immunogold labeling corresponding to Mdchy1 was found in small vesicles (thus indicating exocytosis) and in the lumen of AMG and PMG, corroborating the existence of two similar groups of chymotrypsins. Transcriptomes of M. domestica AMG and whole midgut prepared by pyrosequencing disclosed 41 unique sequences of chymotrypsin-like enzymes (19 probably functional), from which MdChy1 is highly expressed. Phylogenetic reconstruction of Drosophila melanogaster and M. domestica chymotrypsin-like sequences revealed that the chymotrypsin genes expanded before the evolutionary separation of Musca and Drosophila. Kolumnare Apfelbäume (Malus x domestica) stellen aufgrund ihres auffälligen Phänotyps eine ökonomisch interessante Wuchsform dar. Diese extreme Form des Kurztriebwuchses zeichnet sich durch einen insgesamt sehr schlanken, säulenförmigen Habitus aus, welcher eine dichte Pflanzung und damit einhergehend Ertragssteigerungen im Vergleich zu normalwüchsigen Bäumen ermöglicht. Verursacht wird der Phänotyp durch die Anwesenheit eines einzelnen, dominanten Allels des Columnar (Co)-Gens. Bis auf die approximative Lokalisation des Gens auf Chromosom 10 ist über mögliche Identität und Funktion bislang nichts bekannt.rnIn der vorliegenden Arbeit wurde ein erster Versuch unternommen, mit Hilfe von Next Generation Sequencing (NGS) Technologien und RNA-Seq Einblicke in das Transkriptom des Sprossapikalmeristems (SAM) kolumnarer Apfelbäume zu gewinnen. So konnte gezeigt werden, dass unabhängig vom Zeitpunkt der Entnahme des Materials mehrere hundert Gene differentiell reguliert werden. Diese lassen sich funktional in mehrere überrepräsentierte Kategorien gruppieren, von denen sich einige wiederum mit dem kolumnaren Phänotyp assoziieren lassen. Durch den Einsatz weiterer Expressionsstudien (Microarrays, qRT-PCR) konnten frühere Ergebnisse bezüglich des Hormonhaushalts auf Genebene bestätigt und neue Erkenntnisse gewonnen werden, die eine mögliche Erklärung für den Phänotyp darstellen. Weiterhin ergab der Vergleich aller durchgeführten Expressionsstudien eine Anreicherung signifikant differentiell regulierter Gene auf Chromosom 10, was auf einen „selective sweep“ hindeutet. Eine potentielle epigenetische Regulation dieser Gene durch das Genprodukt von Co könnte daher möglich sein. Mehr als die Hälfte dieser Gene lassen sich darüber hinaus aufgrund ihrer Funktion direkt mit dem kolumnaren Phänotyp assoziieren.rnDiese Ergebnisse zeigen, dass die Anwesenheit des Co-Allels massive Veränderungen in der Genregulation des SAMs mit sich bringt, wobei einige dieser differentiell regulierten Gene mit großer Wahrscheinlichkeit an der Etablierung des kolumnaren Phänotyps beteiligt sind. Auch wenn die Funktion des Co-Genproduktes nicht abschließend geklärt werden konnte, sind doch anhand der Resultate schlüssige Hypothesen diesbezüglich möglich.rn

The aim of this work is to evaluate the emissions of the main pollutants of a pellet stove, by trying to simulate the real use in domestic operations. All the operating phases of this system were considered: ignition, partial load, increase in power, and nominal load. In each phase, quantity and type of some pollutants in emissions were determined: the main pollutant gases (CO, NOx, SO2, H2S and volatile organic compounds (VOCs)), total dust (PM) and its content of polycyclic aromatic hydrocarbons (PAHs), regulated heavy metals (Ni, Cd, As and Pb), main soluble ions and Total Carbon (TC). Results show that emission factors of TSP, CO, and of the main determined pollutants (TC, Cd and PAHs) are higher during ignition phase. In particular, this phase prevalently contributes to PAHs emissions. During increase in power phase, gas and particulate emissions do not appreciably differ from nominal load ones; nevertheless, PAH emission factors are higher than steady state ones, but lower than ignition phase. Moreover, during not-steady state phases, PAH mixture is more toxic than during steady state phases. In conclusion, this study allowed to go deeper in pellet stove environmental impact, by pointing out how the different operating conditions can modify the emissions. These are different from certificated data, which are based exclusively on measurements in steady state conditions.

Questo lavoro si è posto l’obiettivo di valutare come differenti qualità di pellet di legno, in accordo con la norma EN 14961-2, e la diversa potenza termica di una moderna stufa domestica influenzino le relative emissioni prodotte. La norma EN 14961-2 prevede una serie di proprietà per il pellet di legno con valori caratteristici per ogni classe di qualità, A1, A2 e B. Per simulare le condizioni cui sono sottoposte le emissioni in aria ambiente, il campionamento del particolato è stato effettuato per mezzo di un tunnel di diluizione. Per valutare e confrontare le emissioni prodotte della stufa a pellet, i dati ottenuti sono stati espressi come fattori di emissione cioè il rapporto tra la quantità di inquinante emesso e i MJ sviluppati dalla combustione. Dallo studio emerge che il pellet di più scarsa qualità mostra emissioni maggiori di CO, NOx, PM, ioni solubili, e la formazione di IPA con alta tossicità: questo implica un maggior impatto sulla salute dell’uomo e sull’ambiente. Inoltre la combustione di pellet di bassa qualità di tipo B causa frequenti problemi di combustione dovuti all’alta percentuale di ceneri, portando a maggiori emissioni di prodotti da combustione incompleta (TC, IPA, PM). La maggior potenza di funzionamento della stufa limita l’emissione di prodotti di combustione incompleta, TC, CO, PM e IPA, ma aumenta la tossicità di quest’ultimi.
