Realizou-se um estudo das deficiências minerais em vacas em lactação de rebanhos leiteiros pertencentes a 13 propriedades da bacia leiteira do município de Rondon do Pará, estado do Pará. Foram determinados os níveis de fósforo (P) no osso, e os níveis de cobre (Cu), cobalto (Co), selênio (Se) e zinco (Zn) no fígado de 47 vacas leiteiras no 2º terço da lactação. Estas amostras foram coletadas por meio de biópsias realizadas no terço superior da 12a costela do lado direito e no bordo caudal do lobo caudado do fígado, respectivamente. Os rebanhos eram formados por animais mestiços (Holandes x Zebu), mantidos em sistema de produção extensivo em pastos de Brachiaria brizantha cv Marandu e recebiam suplementação mineral. A mistura mineral em 12 propriedades era do tipo comercial, dita "completa", acrescida de quantidades de NaCl acima do recomendado pelos fabricantes em dez propriedades. Em sete propriedades as misturas minerais eram fornecidas em cochos sem cobertura e em oito, o fornecimento da mistura mineral não era realizado diariamente. Em 11 propriedades, havia históricos clínicos condizentes com deficiências minerais nos rebanhos. Nessas fazendas a retenção de placenta e a osteofagia foram as alterações mais relatadas. Após as análises minerais observou-se deficiência de P em cinco propriedades, de Co em três propriedades, de Se em nove propriedades e de Zn em dez propriedades. Conclui-se que ocorre a deficiência de P, Co, Se e Zn; a suplementação mineral realizada na maioria das propriedades não atendeu as exigências diárias de P, Se e Co, baseadas no consumo estimado de 30 g de NaCl/animal/dia; os cochos pouco adequados ou inadequados para a suplementação, assim como o fornecimento inconstante das misturas minerais possivelmente contribuíram para a deficiência de um ou mais minerais.

Current cattlemen must seek methods to maximum reproductive efficiency of their beef herds, making the business competitive and enhancing greater profitability. For the cow-calf producers, efficiency translates into more cows producing one calf every year. Fixed-time artificial insemination (TAI) is one of several technologies that producers utilize to reach this goal. Postpartum anestrus is one of the biggest obstacles to overcome in order to obtain suitable results at the end of the breeding season. The TAI protocols are efficient in re-establishing cyclicity in noncycling cows during the postpartum period, streamlining the use of labor on farms, allowing the use of artificial insemination (AI) on a large scale, and introducing superior genetics to the herd. The protocols that are most commonly used in Brazil are based on progesterone (P4) releasing devices that prevent premature estrus and ovulation; and estradiol (E2) to synchronize the initiation of a new follicular wave. In such protocols, administration of equine chorionic gonadotropin (eCG) helps the development of the follicle, increases ovulation rate, improves the endocrine and uterine environment during proestrus and diestrus, and improves fertility. The use of eCG in TAI protocols for suckled cows, non-lactating cows and heifers increases the pregnancy rate allowing more calves at the end of the breeding season and higher profitability to the cattlemen

Follicular estradiol triggers luteolysis in cattle. Therefore, the control of follicle growth and steroidogenesis is expected to modulate luteal function and might be used as an anti-luteolytic strategy to improve embryo survival. Objectives were to evaluate follicular dynamics, plasma concentrations of estradiol and luteal lifespan in Bos indicus and crossbred cows subjected to sequential follicular aspirations. From D13 to D25 of a synchronized cycle (ovulation = D1), Nelore or crossbred, non-pregnant and non-lactating cows were submitted to daily ultrasound-guided aspiration of follicles >6 mm (n = 10) or to sham aspirations (n = 8). Diameter of the largest follicle on the day of luteolysis (7.4 +/- 1.0 vs 9.7 +/- 1.0 mm; mean +/- SEM), number of days in which follicles >6 mm were present (2.3 +/- 0.4 vs 4.6 +/- 0.5 days) and daily mean diameter of the largest follicle between D15 and D19 (6.4 +/- 0.2 vs 8.5 +/- 0.3 mm) were smaller (p <0.01) in the aspirated group compared with the control group, respectively. Aspiration tended to reduce (p< 0.10) plasma estradiol concentrations between D18 and D20 (2.95 +/- 0.54 vs 4.30 +/- 0.55 pg/ml). The luteal lifespan was similar (p > 0.10) between the groups (19.6 +/- 0.4 days), whereas the oestrous cycle was longer (p <0.01) in the aspirated group (31.4 +/- 1.2 vs 21.2 +/- 1.3 days). Hyperechogenic structures were present at the sites of aspiration and were associated with increase in concentration of progesterone between luteolysis and oestrus. It is concluded that follicular aspiration extended the oestrous cycle and decreased the average follicular diameter on the peri-luteolysis period but failed to delay luteolysis.

Problem statement: The aim of the present study was to characterize and differentiate the effects of addition of flavomycin or monensin on ruminal fermentation and degradability as well as on total digestibility in bovine. Approach: Twelve non-pregnant and non-lactating cows (736 kg of BW) were randomly assigned to three treatments: control, flavomycin (20 mg animal-1 day-1) and monensin (300 mg animal-1 day-1). The trial lasted 21 days. The last 10 days were used for external marker administration (15 g of chromic oxide animal-1 day-1). The last 5 days of the trial were used for feces collection and evaluation of corn grain, soybean meal or sugarcane ruminal degradability and the 21st day was used for ruminal fluid sampling. Results: Monensin increased 27.2%, on average, propionate molar proportion at 0, 4, 6, 8, 10 and 12 h after feeding, compared to control and flavomycin groups. When compared to control, flavomycin reduced the degradation rate of soybean meal CP in 31.0%, decreasing the effective degradability when passage rates of 5 and 8% h-1 were used. Dry matter intake, pH, total Short Chain Fatty Acids (tSCFA) or ammoniacal Nitrogen (NH3-N) concentration were not influenced by the addition of either antibiotics. Effective degradability of sugarcane NDF was not influenced by the use of either antibiotic; neither were the TDN nor the digestibility of DM, CP, EE, NFE, ADF, NDF, GE or starch of the diet. Conclusion/Recommendations: In the present study, it was possible to show the beneficial effects of monensin but not of flavomycin, on rumen fermentation

Our aim was to develop an explant model to define more precisely the early response of bovine mammary epithelial cells to infection. Therefore we investigated the mRNA expression encoding for some soluble immunological factors in lipopolysaccharide (LPS)-treated bovine mammary gland explants. Explants were taken out from the mammary gland of eight lactating cows after slaughter then incubated with LPS (10 mug/ml) for 6 h. The mRNA expression of alpha-lactalbumin (alpha-la), various cytokines, tumour necrosis factor (TNF)-alpha, interleukin (IL)-1beta, IL-6, IL-8, and two immunoglobulin receptors, the neonatal Fc receptor (FcRn) and polymeric immunoglobulin receptor (pIGR), were assessed with qPCR before and after 3 h and 6 h of LPS challenge. Both immunoglobulin receptors and alpha-la increased at 3 h then recovered their initial level at 6 h whereas IL-1beta, IL-6 and IL-8 increased only after 6 h (P<0.05). Surprisingly, TNF-alpha transcripts did not show any regulation in response to the LPS treatment. We nevertheless concluded that our model was valid to examine the short-term response of mammary epithelial cell challenged with LPS.

Two synchronization protocols in lactating dairy and beef cows and in dairy heifers were tested for efficacy of breeding by artificial insemination (AI) with or without estrus detection. Controls received three prostaglandin F2a (PGF2a) injections 14 days apart before AI at observed estrus. Pregnancy rates were compared with animals on the Ovsynch protocol that combined gonadotropin releasing hormone (GnRH) and PGF2a treatments with a timed AI 16 to 20 hours after the second GnRH injection. The pregnancy rates were similar at synchronized ovulation to fixed-time AI in lactating cows, but not effective in heifers because of the lack of synchronization.

Bovine mastitis is a frequent problem in Swiss dairy herds. One of the main pathogens causing significant economic loss is Staphylococcus aureus. Various Staph. aureus genotypes with different biological properties have been described. Genotype B (GTB) of Staph. aureus was identified as the most contagious and one of the most prevalent strains in Switzerland. The aim of this study was to identify risk factors associated with the herd-level presence of Staph. aureus GTB and Staph. aureus non-GTB in Swiss dairy herds with an elevated yield-corrected herd somatic cell count (YCHSCC). One hundred dairy herds with a mean YCHSCC between 200,000 and 300,000cells/mL in 2010 were recruited and each farm was visited once during milking. A standardized protocol investigating demography, mastitis management, cow husbandry, milking system, and milking routine was completed during the visit. A bulk tank milk (BTM) sample was analyzed by real-time PCR for the presence of Staph. aureus GTB to classify the herds into 2 groups: Staph. aureus GTB-positive and Staph. aureus GTB-negative. Moreover, quarter milk samples were aseptically collected for bacteriological culture from cows with a somatic cell count ≥150,000cells/mL on the last test-day before the visit. The culture results allowed us to allocate the Staph. aureus GTB-negative farms to Staph. aureus non-GTB and Staph. aureus-free groups. Multivariable multinomial logistic regression models were built to identify risk factors associated with the herd-level presence of Staph. aureus GTB and Staph. aureus non-GTB. The prevalence of Staph. aureus GTB herds was 16% (n=16), whereas that of Staph. aureus non-GTB herds was 38% (n=38). Herds that sent lactating cows to seasonal communal pastures had significantly higher odds of being infected with Staph. aureus GTB (odds ratio: 10.2, 95% CI: 1.9-56.6), compared with herds without communal pasturing. Herds that purchased heifers had significantly higher odds of being infected with Staph. aureus GTB (rather than Staph. aureus non-GTB) compared with herds without purchase of heifers. Furthermore, herds that did not use udder ointment as supportive therapy for acute mastitis had significantly higher odds of being infected with Staph. aureus GTB (odds ratio: 8.5, 95% CI: 1.6-58.4) or Staph. aureus non-GTB (odds ratio: 6.1, 95% CI: 1.3-27.8) than herds that used udder ointment occasionally or regularly. Herds in which the milker performed unrelated activities during milking had significantly higher odds of being infected with Staph. aureus GTB (rather than Staph. aureus non-GTB) compared with herds in which the milker did not perform unrelated activities at milking. Awareness of 4 potential risk factors identified in this study guides implementation of intervention strategies to improve udder health in both Staph. aureus GTB and Staph. aureus non-GTB herds.

Mycoplasma bovis causes mastitis in dairy cows and is associated with pneumonia and polyarthritis in cattle. The present investigation included a retrospective case–control study to identify potential herd-level risk factors for M. bovis associated disease, and a prospective cohort study to evaluate the course of clinical disease in M. bovis infected dairy cattle herds in Switzerland. Eighteen herds with confirmed M. bovis cases were visited twice within an average interval of 75 d. One control herd with no history of clinical mycoplasmosis, matched for herd size, was randomly selected within a 10 km range for each case herd. Animal health data, production data, information on milking and feeding-management, housing and presence of potential stress- factors were collected. Composite quarter milk samples were aseptically collected from all lactating cows and 5% of all animals within each herd were sampled by nasal swabs. Organ samples of culled diseased cows were collected when logistically possible. All samples were analyzed by real-time polymerase chain reaction (PCR). In case herds, incidence risk of pneumonia, arthritis and clinical mastitis prior to the first visit and incidence rates of clinical mastitis and clinical pneumonia between the two visits was estimated. Logistic regression was used to identify potential herd-level risk factors for M. bovis infection. In case herds, incidence risk of M. bovis mastitis prior to the first visit ranged from 2 to 15%, whereas 2 to 35% of the cows suffered from clinical pneumonia within the 12 months prior to the first herd visit. The incidence rates of mycoplasmal mastitis and clinical pneumonia between the two herd visits were low in case herds (0–0.1 per animal year at risk and 0.1-0.6 per animal year at risk, respectively). In the retrospective-case-control study high mean milk production, appropriate stimulation until milk-let-down, fore-stripping, animal movements (cattle shows and trade), presence of stress-factors, and use of a specific brand of milking equipment, were identified as potential herd-level risk factors. The prospective cohort study revealed a decreased incidence of clinical disease within three months and prolonged colonization of the nasal cavity by M. bovis in young stock.

A sociedade está cada vez mais exigente com relação à qualidade dos produtos consumidos e se preocupa com os benefícios para a saúde. Neste contexto, objetivou-se avaliar o efeito da inclusão de níveis de óleo de canola na dieta de vacas sobre amanteiga e muçarela, buscando produtos mais saudáveis para o consumo humano. Foram utilizadas 18 vacas Holandesas, em estágio intermediário de lactação, com produção média de 22 (± 4) Kg de leite/ dia, as quais foram distribuídas em dois quadrados latinos 3x3 contemporâneos e receberam as dietas experimentais: T1- Controle (0% de inclusão de óleo); T2- 3% de inclusão de óleo de canola e T3- 6% de inclusão de óleo de canola. O perfil lipídico foi determinado através de cromatografia gasosa, além da avaliação de qualidade nutricional, realizada através de equações utilizando os ácidos graxos obtidos no perfil lipídico, análises físico-químicas determinadas pela metodologia do Instituto Adolfo Lutz e análises microbiológicas. Houveram problemas durante processamento do leite, gerando alterações de tecnologia de fabricação do produto manteiga, obtendo-se outro produto, o creme de leite, ao invés de manteiga, além de prejuízos na qualidade microbiológicas do creme de leite e muçarela. A inclusão de óleo de canola na dieta em lactação reduziu quadraticamente os ácidos graxos de cadeia curta e proporcionou aumento quadrático dos ácidos graxos de cadeia longa, dos ácidos graxos insaturados e ácidos graxos monoinsaturados na muçarela. A relação ácidos graxos saturados/ ácidos graxos insaturados (AGS/ AGI) e a relação ômega-6/ômega-3, assim como os índices de aterogenicidade e trombogenicidade, na muçarela, reduziram linearmente 25,68%, 31,35%; 32,12% e 21,78%, respectivamente, quando comparando T1 e T3. No creme de leite, houve redução linear dos ácidos graxos de cadeia curta e média, bem como, os ácidos graxos saturados e a relação ácidos graxos saturados/ ácidos graxos insaturados (AGS/ AGI) em 41,07%; 23,82%; 15,91% e 35,59%, respectivamente, enquanto os ácidos graxos de cadeia longa, ácidos graxos insaturados e ácidos graxos monoinsaturados aumentaram linearmente 41,40%; 28,24% e 32,07%, nesta ordem, quando comparando T1 com T3. Os índices de aterogenicidade e trombogenicidade reduziram de forma linear, enquanto o índice h/H (razão ácidos graxos hipocolesterolêmicos e hipercolesterolêmicos) aumentou linearmente. A composição físico-química de ambos derivados e o rendimento da muçarela não apresentaram efeito significativo com a inclusão do óleo de canola, exceto a proteína bruta da muçarela que apresentou aumento linear e a gordura do creme de leite que apresentou efeito quadrático. As análises microbiológicas mostram contagens muito elevadas de microrganismos, sugerindo que os produtos não apresentam qualidade microbiológica, decorrente da ausência do processo de pasteurização do creme e da baixa eficiência do tratamento térmico aplicado ao leite destinado a produção da muçarela. Conclui-se que a adição de óleo de canola na dieta de vacas lactantes proporciona muçarela e creme de leite mais saudáveis para o consumo humano, pois apresentaram perfil lipídico mais rico em ácidos graxos insaturados, além da série ômega-3 e ácido oleico, entretanto, devido a problemas de processamento, estes produtos obtidos, não estão aptos ao consumo devido à ausência de qualidade microbiológica.

Objetivou-se avaliar a utilização de doses crescentes de enzima fibrolítica exógena na alimentação de vacas leiteiras e seus efeitos sobre o consumo e digestibilidade aparente total da matéria seca e nutrientes, cinética ruminal, fermentação e síntese de proteína microbiana ruminal, produção e composição do leite, perfil metabólico e balanço de energia e nitrogênio. Foram utilizadas 24 vacas da raça Holandesa, multíparas, em delineamento Quadrado Latino 4x4, com 646,75 ± 77,54 kg de peso corporal, 3,02 ± 0,56 de escore de condição corporal, com 176 ± 82,27 dias em lactação e produção de leite de 33,72 ± 7,63 kg/dia, no início do estudo. Os animais foram distribuídos aleatoriamente para receber os seguintes tratamentos: 1) Controle (0), composta por dieta basal sem a inclusão de enzima fibrolítica; 2) com inclusão de 8 g/vaca/dia de enzima fibrolítica; 3) com inclusão de 16 g/vaca/dia da enzima fibrolítica; 4) com inclusão de 24 g/vaca/dia da enzima fibrolítica (Fibrozyme® - Alltech Inc., Nicholasville, KY). A utilização de enzima fibrolítica nas dietas resultou em aumento linear no consumo de matéria seca, matéria orgânica e da fibra em detergente neutro. Foi detectado aumento linear no consumo de partículas longas com a suplementação de enzima. Houve efeito quadrático na ruminação e na atividade mastigatória. O aumento no consumo de matéria seca refletiu no aumento linear de consumo de energia líquida e no balanço de energia líquida. Houve efeito quadrático na concentração de N-NH3 ruminal e aumento linear na quantidade de ácido acético, propiônico e butírico com o aumento da dose de enzima suplementada. Houve efeito quadrático na síntese de proteína microbiana com a inclusão de enzima fibrolítica. Não foram observadas diferenças na produção de leite e na produção de seus componentes, entretanto houve aumento linear no ganho de peso corporal com utilização de enzima fibrolítica. Houve efeito quadrático positivo na excreção via urina e efeito quadrático negativo no balanço de nitrogênio mostrando maior retenção de nitrogênio com a suplementação intermediária de enzimas fibrolíticas. Conclui-se que a enzima fibrolítica exógena é efetiva em aumentar o consumo de matéria seca e FDN e também melhorar a eficiência fermentativa de vacas leiteiras melhorando o balanço energético, entretanto não foi efetiva em aumentar a produção de leite de vacas da raça holandesa no terço médio da lactação
