The burden of foodborne disease has large economic and social consequences worldwide. Despite strict regulations, a number of pathogens persist within the food environment, which is greatly contributed to by a build-up of resistance mechanisms and also through the formation of biofilms. Biofilms have been shown to be highly resistant to a number of antimicrobials and can be extremely difficult to remove once they are established. In parallel, the growing concern of consumers regarding the use of chemically derived antimicrobials within food has led to a drive toward more natural products. As a consequence, the use of naturally derived antimicrobials has become of particular interest. In this study we investigated the efficacy of nisin A and its bioengineered derivative M21A in combination with food grade additives to treat biofilms of a representative foodborne disease isolate of Listeria monocytogenes. Investigations revealed the enhanced antimicrobial effects, in liquid culture, of M21A in combination with citric acid or cinnamaldehyde over its wild type nisin A counterpart. Subsequently, an investigation was conducted into the effects of these combinations on an established biofilm of the same strain. Nisin M21A (0.1 μg/ml) alone or in combination with cinnamaldehyde (35 μg/ml) or citric acid (175 μg/ml) performed significantly better than combinations involving nisin A. All combinations of M21A with either citric acid or cinnamaldehyde eradicated the L. monocytogenes biofilm (in relation to a non-biofilm control). We conclude that M21A in combination with available food additives could further enhance the antimicrobial treatment of biofilms within the food industry, simply by substituting nisin A with M21A in current commercial products such as Nisaplin® (Danisco, DuPont).

Nella mia sperimentazione ho studiato i meccanismi di risposta, in termini di modificazione a livello delle membrane citoplasmatiche e in termini di rilascio di molecole volatili, messi in atto da Listeria. monocytogenes (Scott A) quando sottoposta in fase stazionaria a diversi trattamenti ad alta pressioni di omogeneizzazione compresi tra 0.1 e 200 MPa. I dati della mia tesi hanno consentito di evidenziare i meccanismi di risposta di un importante patogeno alimentare quando sottoposto ad una tecnologia proposta in alternativa al trattamento termico al fine di ridurre il danno termico degli alimenti preservandone le caratteristiche nutrizionali, funzionali ed organolettiche, senza effetti negativi sulla loro sicurezza e shelf-life. Inoltre, hanno dimostrato che L. monocytogenes risponde al trattamento iperbarico modificando in prima istanza la composizione in acidi grassi e producendo delle ossilipine al crescere dello stress. Alle condizioni di stress più stringenti adottate diventano fondamentali in L. monocytogenes anche la produzione di molecole volatili quali alcoli ed esteri sia per bilanciare il potenziale redox cellulare sia per eliminare molecole estremamente tossiche quali gli acidi grassi rilasciati dai meccanismi messi in atto alle condizioni meno stressanti di pressione

Aims: To evaluate the probiotic properties of strains isolated from smoked salmon and previously identified as bacteriocin producers. Methods and Results: Strains Lactobacillus curvatus ET06, ET30 and ET31, Lactobacillus fermentum ET35, Lactobacillus delbrueckii ET32, Pediococcus acidilactici ET34 and Enterococcus faecium ET05, ET12 and ET88 survived conditions simulating the gastrointestinal tract (GIT) and produced bacteriocins active against several strains of Listeria monocytogenes, but presented very low activity against other lactic acid bacteria (LAB). Cell-free supernatants containing bacteriocins, added to 3-h-old cultures of L. monocytogenes 603, suppressed growth over 12 h. Auto-aggregation was strain-specific, and values ranged from 7 center dot 2% for ET35 to 12 center dot 1% for ET05. Various degrees of co-aggregation with L. monocytogenes 603, Lactobacillus sakei ATCC 15521 and Enterococcus faecalis ATCC 19443 were observed. Adherence of the bacteriocinogenic strains to Caco-2 cells was within the range reported for Lactobacillus rhamnosus GG, a well-known probiotic. The highest levels of hydrophobicity were recorded for Lact. curvatus (61 center dot 9-64 center dot 6%), Lact. fermentum (78 center dot 9%), Lact. delbrueckii (43 center dot 7%) and Ped. acidilactici (51 center dot 3%), which are higher than the one recorded for Lact. rhamnosus GG (53 center dot 3%). These strains were highly sensitive to several antibiotics and affected by several drugs from different generic groups in a strain-dependent manner. Conclusions: Smoked salmon is a rich source of probiotic LAB. All strains survived conditions simulating the GIT and produced bacteriocins active against various pathogens. Adherence to Caco-2 cells was within the range reported for Lact. rhamnosus GG, a well-known probiotic. In addition, the high hydrophobicity readings recorded define the strains as good probiotics. Significance and Impact of the Study: Smoked salmon contains a number of different probiotic LAB and could be marketed as having a potential beneficial effect.

Listeria monocytogenes is a bacterial pathogen that represents a serious threat during pregnancy and several cases of listeriosis have been linked to the consumption of contaminated foods worldwide. In Brazil, there is no report of foodborne listeriosis, despite some sporadic cases of infection by this bacterium occur. In general in our country, there is no awareness of medical personnel to instruct moms-to-be to avoid high risk foods. In the present study, a total of 141 samples were surveyed for the presence of Listeria spp., including cervicovaginal samples of patients, foods and home refrigerators. No clinical sample was positive for Listeria spp., but it was isolated from two refrigerators. L. monocytogenes was detected in two food samples out of five positive ones for Listeria spp. In conclusion, it was shown the presence of contaminated food items at home level and the lack of information on the risks of listeriosis, indicating the need of implementation of food safety education programs. The objective of this research was to evaluate the effects of 2 levels of raw milk somatic cell count (SCC) on the composition of Prato cheese and on the microbiological and sensory changes of Prato cheese throughout ripening. Two groups of dairy cows were selected to obtain low-SCC (<200,000 cells/mL) and high-SCC (>700,000 cells/mL) milks, which were used to manufacture 2 vats of cheese. The pasteurized milk was evaluated according to the pH, total solids, fat, total protein, lactose, standard plate count, coliforms at 45 degrees C, and Salmonella spp. The cheese composition was evaluated 2 d after manufacture. Lactic acid bacteria, psychrotrophic bacteria, and yeast and mold counts were carried out after 3, 9, 16, 32, and 51 d of storage. Salmonella spp., Listeria monocytogenes, and coagulase-positive Staphylococcus counts were carried out after 3, 32, and 51 d of storage. A 2 x 5 factorial design with 4 replications was performed. Sensory evaluation of the cheeses from low- and high-SCC milks was carried out for overall acceptance by using a 9-point hedonic scale after 8, 22, 35, 50, and 63 d of storage. The somatic cell levels used did not affect the total protein and salt: moisture contents of the cheeses. The pH and moisture content were higher and the clotting time was longer for cheeses from high-SCC milk. Both cheeses presented the absence of Salmonella spp. and L. monocytogenes, and the coagulase-positive Staphylococcus count was below 1 x 10(2) cfu/g throughout the storage time. The lactic acid bacteria count decreased significantly during the storage time for the cheeses from both low- and high-SCC milks, but at a faster rate for the cheese from high-SCC milk. Cheeses from high-SCC milk presented lower psychrotrophic bacteria counts and higher yeast and mold counts than cheeses from low- SCC milk. Cheeses from low- SCC milk showed better overall acceptance by the consumers. The lower overall acceptance of the cheeses from high-SCC milk may be associated with texture and flavor defects, probably caused by the higher proteolysis of these cheeses.

Mice transgenic for the E7 tumor Ag of human papillomavirus type 16, driven from a keratin 14 promoter, express E7 in keratinocytes but not dendritic cells. Grafted E7-transgenic skin is not rejected by E7-immunized mice that reject E7-transduced transplantable tumors. Rejection of recently transplanted E7-transgenic skin grafts, but not of control nontransgenic grafts or of established E7-transgenic grafts, is induced by systemic administration of live or killed Listeria monocytogenes or of endotoxin. Graft recipients that reject an E7 graft reject a subsequent E7 graft more rapidly and without further L. monocytogenes exposure, whereas recipients of an E7 graft given without L. monocytogenes do not reject a second graft, even if given with L. monocytogenes. Thus, cross-presentation of E7 from keratinocytes to the adaptive immune system occurs with or without a proinflammatory stimulus, but proinflammatory stimuli at the time of first cross-presentation of Ag can determine the nature of the immune response to the Ag. Furthermore, immune effector mechanisms responsible for rejection of epithelium expressing a tumor Ag in keratinocytes are different from those that reject an E7-expressing transplantable tumor. These observations have implications for immunotherapy for epithelial cancers.

Aims: To quantify Listeria levels on the shell and flesh of artificially contaminated cooked prawns after peeling, and determine the efficacy of Listeria innocua as a model for L. monocytogenes in this system. Methods and Results: A L. monocytogenes and L. innocua strain were inoculated separately onto cooked black tiger prawns using two protocols ( immersion or swabbing with incubation). Prawns were peeled by two methods ( gloved hand or scalpel and forceps) and numbers of Listeria on shells, flesh and whole prawn controls were determined. Prawns were exposed to crystal violet dye to assess the penetration of liquids. Regardless of preparation method or bacterial strain there were ca 1log(10) CFU more Listeria per shell than per peeled prawn. Dye was able to penetrate to the flesh in all cases. Conclusions: Shell-on prawns may be only slightly safer than shell-off prawns. Listeria innocua is an acceptable model for L. monocytogenes in this system. Significance and Impact of the Study: Reduced risk from L. monocytogenes on prawns can only be assured by adequate hygiene or heating.

A total of 130 Listeria strains were tested in order to evaluate lecithinase production and capacity for Congo red adsorption as markers of pathogenicity. The strains were identified according to acid production from sugars and by the CAMP test and the data were correlated with the ability to produce keratoconjunctivitis in guinea pigs. L. monocytogenes cultures presented 51.8% and 88.8% positivity rates for Congo red adsorption and lecithinase production, respectively, whereas 80.8% and 100% for L. innocua cultures were negative for the two test, respectively

Visando complementar as informações sobre a qualidade microbiológica do leite comercializado na cidade do Recife, foram analisadas 250 amostras de leite pasteurizado tipo C e 50 amostras de leite cru para a pesquisa de Yersinia enterocolitica e Listeria monocytogenes, bactérias patogênicas capazes de se desenvolverem em temperatura de refrigeração. Y. enterocolitica não foi encontrada em nenhuma das amostras analisadas, entretanto foi detectada a presença de Y. intermedia e Y. frederiksenii, espécies ambientais que se comportam como patógenos oportunistas. L. monocytogenes também não foi encontrada, mas, através da metodologia empregada para seu isolamento foi obtido um isolamento de Salmonella Montevideo em uma amostra de leite pasteurizado e outro em leite cru. Além dessas, várias outras bactérias foram encontradas, supondo-se que a ampla microbiota crescida nos meios empregados pode ter interferido no isolamento da Y. enterocolitica e L. monocytogenes.

A microbiologia preditiva é a conjugação de conhecimentos provenientes de disciplinas como a matemática, estatÃstica e os sistemas de informação e tecnologia que pretende providenciar modelos preditivos que prevejam o comportamento microbiano em ambientes alimentares, de forma a poder prevenir a deterioração dos géneros alimentares bem como as doenças de origem alimentar. Os modelos preditivos primários, secundários e terciários são aplicados no intuito de melhorar a qualidade e segurança alimentar e particularmente os terciários, podem ser utilizados como ferramentas auxiliadoras na área de HACCP; é necessário ter em conta que estes modelos são uma representação muito simplificada da realidade, que possuem limitações devido á complexidade do comportamento microbiano e dos ambientes alimentares, podendo por isto estimar previsões que se desviem das situações reais. O presente trabalho tem como principal objectivo averiguar a aplicabilidade da microbiologia preditiva, particularmente, dos modelos terciários ou softwares preditivos, na análise de amostras de carne de vaca e porco armazenadas a 5ºC e 10ºC, comparando os resultados obtidos através da análise microbiológica clássica, realizada no laboratório de microbiologia da empresa SGS, com os resultados obtidos das previsões provenientes de dois softwares preditivos, nomeadamente, ComBase Predictor e PMP (Pathogen Modeling Program). Para tal, foram realizadas análises microbiológicas, por forma a realizar contagens de E.coli, S.aureus, L.monocytogenes e pesquisa de Salmonella e análises quÃmicas para analisar o pH, aw e NaCl de 20 amostras de carne de vaca e 20 amostras de carne de porco Adicionalmente foram efetuadas contagens de microrganismos totais a 30ºC. Os resultados demonstraram que a ferramenta preditiva ComBase conseguiu efectuar melhores previsões para o crescimento de E. coli, S. aureus e L. monocytogenes em amostras de carne de vaca e de porco do que a ferramenta preditiva PMP. Contudo, mesmo sendo melhor, as previsões efectuadas pelo programa apresentaram desvios em relação à s contagens reais que muito provavelmente se relacionam com a existência da flora de decomposição. Os resultados estimados pela ferramenta PMP foram sempre muito mais elevados do que os resultados obtidos na análise microbiológica laboratorial, o que demonstrou a sua não aplicabilidade a este tipo de amostras.

T cell factor-1 (TCF-1) and lymphoid enhancer-binding factor 1, the effector transcription factors of the canonical Wnt pathway, are known to be critical for normal thymocyte development. However, it is largely unknown if it has a role in regulating mature T cell activation and T cell-mediated immune responses. In this study, we demonstrate that, like IL-7Ralpha and CD62L, TCF-1 and lymphoid enhancer-binding factor 1 exhibit dynamic expression changes during T cell responses, being highly expressed in naive T cells, downregulated in effector T cells, and upregulated again in memory T cells. Enforced expression of a p45 TCF-1 isoform limited the expansion of Ag-specific CD8 T cells in response to Listeria monocytogenes infection. However, when the p45 transgene was coupled with ectopic expression of stabilized beta-catenin, more Ag-specific memory CD8 T cells were generated, with enhanced ability to produce IL-2. Moreover, these memory CD8 T cells expanded to a larger number of secondary effectors and cleared bacteria faster when the immunized mice were rechallenged with virulent L. monocytogenes. Furthermore, in response to vaccinia virus or lymphocytic choriomeningitis virus infection, more Ag-specific memory CD8 T cells were generated in the presence of p45 and stabilized beta-catenin transgenes. Although activated Wnt signaling also resulted in larger numbers of Ag-specific memory CD4 T cells, their functional attributes and expansion after the secondary infection were not improved. Thus, constitutive activation of the canonical Wnt pathway favors memory CD8 T cell formation during initial immunization, resulting in enhanced immunity upon second encounter with the same pathogen.

Using phenotype techniques, characterization was made to species and serovar of 3,112 strains of Listeria, isolated from different sources of infection such as human (247-7.9%) and animals (239-7.6%), as well as from various routes of infection, including food (2,330-74.8%) and environmental constituents (296-9.5%), all coming from different regions of the country and collected during the period 1971-1997. The following species were recovered in the cultures analysed: L. monocytogenes (774-24.8%), L. innocua (2,269-72.9%), L. seeligeri (37-1.1%), L. welshimeri (22-0.7%), L. grayi (9-0.2%), and L. ivanovii (1-0.03%). L. monocytogenes was represented by ten serovars, the most prevalent being 4b (352-11.3%), 1/2a (162-5.2%), and 1/2b (148-4.7%). The predominant serovar in L. innocua was 6a (2,093-67.2%). Considerations about laboratory methods for diagnosis and epidemiological aspects are presented on the basis of the results obtained.

L’aplicació de tecnologies innovadores per a l’anà lisi de la qualitat (proteòmica) i per al processat de productes carnis (envasament actiu i altes pressions hidrostà tiques) amb la finalitat d’optimitzar la qualitat i la seguretat de productes carnis llestos per al consum fou evaluat. Els resultats obtinguts amb l’anà lisi proteòmic van permetre la detecció de pèptids/ proteïnes candidats a marcadors proteics de la qualitat dels lloms i dels pernils. La detecció d’aquests marcadors a la matèria primera (llom i pernil fresc) ajudaria a predir la qualitat final dels productes carnis processats (llom cuit i pernil curat), i proporcionaria una eina per al control de la qualitat de la carn de porc. No obstant, la validació del paper d’aquestes proteïnes a la qualitat final dels productes carnis és necessà ria abans de poder-los considerar marcadors proteics. Per altra banda, es va estudiar la possiblitat de millorar la seguretat alimentà ria de llonganissa sense sal afegida obtinguda amb el procés QDS® process a través l’ús de tecnologies innovadores (envasament actiu i altes pressions hidrostà tiques). La llonganissa sense sal afegida no va permetre el creixement de L. monocytogenes. No obstant, el patogen seria capaç de sobreviure durant la vida útil del producte en cas de recontaminació. L’envasament antimicrobià amb la inclusió de nisina com a antimicrobià natural es pot considerar un mètode efectiu per a millorar la seguretat de la llonganissa estudiada. L. monocytogenes va sobreviure al tractament d’alta pressió hidrostà tica (600 MPa, 5 min, 12ºC) grà cies a les caracterÃstiques del producte de baixa activitat d’aigua i presència de lactat a la seva formulació. Per aquest motiu, la APH no es consideraria un tractament apropiat per a reduir la presència de L. monocytogenes en aquest tipus de producte.

A Segurança Alimentar tem assumido uma importância cada vez maior a nÃvel mundial, devido essencialmente ao aumento das trocas internacionais de alimentos e da consciencialização dos consumidores face ao direito de obter alimentos seguros. Tendo em conta este panorama, diversos paÃses encaram a Segurança Alimentar como uma prioridade a nÃvel de gestão governamental, onde se destaca os paÃses em vias de desenvolvimento, como é o caso de Cabo Verde. No domÃnio do setor de produção de alimentos, têm sido implementadas ações que visam, principalmente, o aumento da qualidade. Em Cabo Verde existe uma grande tradição relacionada com a produção e consumo de queijo, sendo esta atividade possivelmente herdada dos antepassados coloniais. A maioria do queijo comercializado provém de fabricação artesanal e sua quantificação não consta em estatÃsticas oficiais, embora seja conhecida a existência de unidades de produção caseira e unidades semi-industriais. O presente trabalho, afeto ao curso de Mestrado em Segurança Alimentar da Faculdade de Farmácia – Universidade de Coimbra, teve como objetivo avaliar as condições sanitárias de produção e a qualidade microbiológica e fÃsico-quÃmica de queijo fresco de leite de cabra produzidos em Cabo Verde. No total foram analisados 60 amostras de queijo, sob o ponto de vista microbiológico (30 amostras) e sob o ponto de vista fÃsico-quÃmico (30 amostras). Da avaliação sanitária conclui-se que ainda persistem algumas deficiências, como por exemplo, a realização de ordenha de forma inadequada e a produção de queijo sem tratamento térmico do leite. De acordo com os resultados microbiológicos, constataram-se elevadas contagens de microrganismos a 30ºC, na totalidade das amostras, e de E. coli, em 63% das amostras. Não se encontraram amostras positivas à presença de L. monocytogenes e Salmonella spp. As análises fÃsico-quÃmicas apresentaram teores médios, por 100g de produto, de 53% de humidade, 24% de matéria-gorda, 19% de proteÃna bruta, 3% de cinzas, 1,5% de açúcares (lactose) e 1,4% de sal, conduzindo a teores médios de 69% de humidade no queijo isento de matéria-gorda e de 51% de matéria gorda no resÃduo-seco; o pH médio foi de 5,88 e o aw médio situou-se em 0,847. Foram identificadas variações inter-ilhas, a nÃvel dos X parâmetros analisados, embora não muito marcantes, mas suficientes para conduzir a alguma diferenciação em função da proveniência. Recomendou-se o uso contÃnuo das boas práticas de higiene e fabrico na elaboração dos queijos, com o objetivo de melhorar a qualidade sanitária da produção e dos produtos finais.

Listeria monocytogenes was inoculated on the surface of sliced fermented sausages with no added sodium salt. The pathogen was progressively inactivated during the product shelf life (90 days). Antimicrobial packaging of fermented sausages with PVOH films containing nisin induced a more pronounced reduction of L. monocytogenes counts during refrigerated storage. HPP alone (600 MPa, 5 min, 12 °C) had no antimicrobial effect against L. monocytogenes at the studied conditions. Combination of HPP with antimicrobial packaging did not produce any extra protection against L. monocytogenes compared to antimicrobial packaging alone. The lack of effect of HPP on L. monocytogenes was attributed to a protective effect exerted by the low water activity of the product and its lactate content. These results reflect that antimicrobial packaging with the inclusion of nisin as a natural antimicrobial could be considered as an effective method to reduce the levels of L. monocytogenes in sliced fermented sausages with no added sodium salt
