Cystic fibrosis (CF) lung disease is an inherited condition with an incidence rate of approximately 1 in 2500 new born babies. CF is characterized as chronic infection of the lung which leads to inflammation of the airway. Sputum from CF patients contains elevated levels of neutrophils and subsequently elevated levels of neutrophil serine proteases. In a healthy individual these proteases aid in the phagocytic process by degrading microbial peptides and are kept in homeostatic balance by cognate antiproteases. Due to the heavy neutrophil burden associated with CF the high concentration of neutrophil derived proteases overwhelms cognate antiproteases. The general effects of this protease/antiprotease imbalance are impaired mucus clearance, increased and self-perpetuating inflammation, and impaired immune responses and tissue. To restore this balance antiproteases have been suggested as potential therapeutics or therapeutic targets. As such a number of both endogenous and synthetic antiproteases have been trialed with mixed success as therapeutics for CF lung disease.

Background: Excessive activation of epithelial sodium channels (ENaC) contributes to CF lung pathophysiology due to the resultant dehydration of the airway surface liquid (ASL) and impaired mucociliary clearance. Regulated proteolysis of the endogenous α and γ subunits of ENaC by apical membrane-bound Channel Activating Proteases (CAPs) is a fundamental regulatory mechanism for channel activity. In the CF lung a stark imbalance between the levels of CAPs and their natural inhibitors drives the activation of normally inactive ENaC. On this basis inhibition of CAPs-ENaC signalling represents a potential therapeutic intervention. To this end we have developed a novel cell impermeable active-site directed compound (QUB-TL1) designed to inactivate key trypsin-like CAPs highly relevant in this regard. Objectives & Methods: Utilize differentiated non-CF and CF human airway epithelial cells to assess the impact of QUB-TL1 on a range of parameters including surface CAP activities, ENaC subunit processing/channel activity, ASL height and mucociliary clearance. Results: Treatment of airway epithelial cells with QUB-TL1 results in the significant downregulation of key endogenous CAP activities found to be excessively active at the surface of CF cultures. QUB-TL1-mediated CAP inhibition subsequently causes the internalisation of a pool of processed (active) ENaCγ prominent at the apical surface of CF cultures which correlates with a decline in channel activity. This downregulation of ENaC activity results in an increase in ASL height and improved mucociliary clearance in CF cells. We further find QUB-TL1 uniquely inhibits the ENaC activating enzyme furin, which is in contrast to the alternate trypsin-like CAP inhibitors camostat mesylate and aprotinin. QUB-TL1-mediated furin inhibition correlates with a reduction in neutrophil elastase-induced ENaC activation. Moreover we find QUB-TL1 treatment protects CF cultures from Pseudomonas aeruginosa exotoxin A-induced cytotoxicity. Pseudomonas aeruginosa exotoxin A is a major toxic product activated by furin and positively associated with mortality. Conclusion: The novel inhibitor (QUB-TL1) dampens CAPs-ENaC signalling which improves hydration status mucociliary clearance in CF airway epithelial cell cultures. Moreover this compound provides additional benefit by preventing Pseudomonas aeruginosa exotoxin A-induced cytotoxicity.

A identificação e caracterização de processos mediados por metaloproteinases de uma grande variedade de eucariotas está a avançar rapidamente, tanto a nível molecular como celular – e os muitos papéis que as proteases desempenham nestes organismos estão a ser trazidas para o foco. As metaloproteinases de matriz executam tarefas de ―housekeeping” comuns a muitos organismos, bem como funcionam a um nível muito mais específico no ciclo de vida dos parasitas, por exemplo. Papéis pivotais foram propostos em diferentes processos, como invasão e egressão de células, enquistação e desenquistação, catabolismo de proteínas, diferenciação, progressão do ciclo celular, citoaderência, e ainda estimulação e evasão ao sistema imune. No presente estudo, é feito o reconhecimento e a caracterização de metaloproteinases de dois diferentes extractos de parasitas protozoários: Trypanosoma brucei brucei e Leishmania infantum. A classificação de metaloproteases foi obtida a partir da inibição diferencial da actividade destas proteases sobre diferentes substratos (gelatina e caseína), tendo sido efectuada uma prospecção de inibidores selectivos com potencial para serem empregues em novas estratégias de quimioterâpeutica contra estes agentes. Adicionalmente, é feito um estudo bioinformático sobre uma metaloproteinase comum aos parasitas aqui abordados, alargando o estudo a outros organismos relacionados. Os resultados obtidos demonstram a presença de metaloproteinases capazes de degradar proteínas de matriz em ambos os extractos estudados, sendo possível inibir a sua actividade com concentrações relativamente moderadas de inibidores. Além disso, sugerem que em todos os eventos patológicos abordados neste estudo, a presença de metaloproteinases activas é estável no seu ciclo de vida e provavelmente na progressão da patologia provocada pelos diferentes agentes em estudo. A análise de todos os resultados e observações poderá possivelmente levar à identificação e integração de elementos comuns nos processos de invasão celular e progressão parasitária aqui abordados. Por essa razão, a compreensão destas interacções permanece um desafio importante.

In the present study,heterotrophic protease producing bacterial isolates were screened for protease activity and a potent protease producing bacterial isolate was selected,identified and coded as Pseudomonas aeruginosa MCCB 123.The organism was capable of producing three different types of enzymes each having potential industrial applications.The non-toxic nature of the bacterial strain and the relatively non-toxic nature of three enzymes suggested their poetential application in various industries.Application of LasA protease and beta-1,3 glucanase in DNA extraction is a promising area for commercial utilization. LasB protease can find its potential application in detergent and tanning industries.As on today Bacillus sp.has been the source of commercial proteases,and the ones produced form P.aeruginosa 123 can pave way for making the industrial and biomedical processes more cost effective and refined.

Microorganisms distributed in the marine and brackish environments play an important role in the decomposition of organic matter and mineralisation in the system (Seshadri and lgnacimuthu, 2002). Estuary is one of the most productive ecosystems, at the same time one among the least explored ecosystems on earth, which has immense potential as a source of potent microorganisms that produce valuable compounds particularly, enzymes such as proteases. In this scenario, it is very appropriate to embark on finding novel alkaline protease producers from the estuarine system. The area where the present investigation was carried out is a part of the extensive estuarine system of South India viz. Cochin Estuary. There is meagre knowledge regarding the microbial composition, particularly the protease producers of Cochin Estuary. Hence, the present study has been undertaken with the objective of finding novel alkaline protease producing bacteria from Cochin Estuary

Protease inhibitors can be versatile tools mainly in the fields of medicine, agriculture and food preservative applications. Fungi have been recognized as sources of protease inhibitors, although there are only few such reports on mushrooms. This work reports the purification and characterization of a trypsin inhibitor from the fruiting body of edible mushroom Pleurotus floridanus (PfTI) and its effect on the activity of microbial proteases. The protease inhibitor was purified up to 35-fold by DEAE-Sepharose ion exchange column, trypsin-Sepharose column and Sephadex G100 column. The isoelectric point of the inhibitor was 4.4, and its molecular mass was calculated as 37 kDa by SDS-PAGE and 38.3 kDa by MALDI-TOF. Inhibitory activity confirmation was by dot-blot analysis and zymographic activity staining. The specificity of the inhibitor toward trypsin was with Ki of 1.043×10−10 M. The inhibitor was thermostable up to 90 °C with maximal stability at 30 °C, active over a pH range of 4–10 against proteases from Aspergillus oryzae, Bacillus licheniformis, Bacillus sp. and Bacillus amyloliquefaciens. Results indicate the possibility of utilization of protease inhibitor from P. floridanus against serine proteases

Os produtos naturais são uma importante fonte de possíveis novos fármacos para uso clínico. O Plectranthus ornatus Codd. pertence a um género associado ao isolamento de diterpenos que são compostos responsáveis ??por diversas actividades biológicas. Os três diterpenos em estudo foram isolados no passado de P. ornatus e são agora avaliados como potenciais inibidores da COX-1. Os resultados obtidos sugerem que o ácido rinocerotinoico (2), mas não a plectrornatina C (1) e o halimano (3), apresenta uma ligeira actividade inibidora da COX-1. Estudos em desenvolvimento visam entender se estes compostos têm efeito como inibidores da COX-2. Deste modo, pretendese verificar o seu possível interesse como agentes anti-inflamatórios.
