Flüchtige organische Halogenverbindungen übernehmen in der Chemie der Troposphäre eine Schlüsselrolle. Photolytisch gebildete Halogenatome reagieren mit troposphärischem Ozon und können durch Oxidation, vor allem von Iod, zur Neubildung von Partikeln beitragen. Auf diese Weise beeinflussen Halogenalkane den Strahlungshaushalt der Atmosphäre. Aus analytischem Blickwinkel ist es wichtig die Konzentration der einzelnen Spezies zu untersuchen um Rückschlüsse auf deren biotische oder abiotische Quellen ziehen und die Emissionswege besser verstehen zu können. Im Rahmen der vorliegenden Arbeit wurde daher eine sensitive Methode zur Untersuchung von halogenierten Kohlenwasserstoffen entwickelt, basierend auf anreichernder Probenahme mit anschließender Thermodesorption und der Analyse mittels Massenspektrometrie mit negativer chemischer Ionisation. Die Kennwerte der Methode sind: Nachweisgrenzen zwischen 0.11 pg und 5.86 pg bzw. zwischen 1.0 ppqV und 44.7 ppqV, Linearität zwischen R2=0.993 und R2=1.000, Reproduzierbarkeit (Triplikate) RSD < 15 % und ein sicheres Probenahmevolumen von 10 L. Die Methode wurde im Anschluss im Rahmen von zwei Feldmessungen, in Mace Head, Irland und auf einer Schiffskampagne im antarktischen Amundsen-Meer, angewendet. Durch die Ergebnisse aus Irland kann gezeigt werden, dass die Mischungsverhältnisse der Iodalkane mit denen früherer Studien vergleichbar sind, und dass die verschiedenen untersuchten Algenarten deutlich unterschiedliche Emissionsraten zeigen. Die Ergebnisse der Kampagne im Amundsen-Meer zeigen einen großen Einfluss der Windrichtung auf die Halogenalkan-Konzentrationen. So sind die Mischungsverhältnisse der Halogenalkane deutlich höher, wenn der Wind zuvor über die antarktischen Eisflächen strömt. Für die biotischen Quellen wurden die Emissionsraten ausgewählter Makroalgen unter dem Einfluss von Ozon untersucht. Die Emissionsrate der Iodalkane zeigt einen exponentiellen Zusammenhang, sowohl zur I2-Emission als auch zum Gesamtiodgehalt der Algen. Unter oxidativen Bedingungen zeigt L. Digitata eine linear steigende Iodalkanemission. Mit diesem Verhalten wird die These der Bildung von Iodalkanen als Nebenprodukt beim Abbau reaktiver Sauerstoffspezies unterstützt. Neben den Makroalgen wurden auch Mikroalgen als biotische Quellen untersucht. Hierbei können zwei unterschiedliche Emissionsmuster der Halogenalkane für Diatomeen und Phaeocystis sp. gezeigt werden. Im Gegensatz zur Iodalkan-Emission hängt die I2 Emission der Mikroalgenproben von der Ozonkonzentration der Luft ab. Durch die lineare Korrelation der I2-Emission mit der Iodid-Konzentration der wässrigen Phase einerseits, und dem Ozonverbrauch andererseits, kann die Bildung von I2 durch Oxidation von Iodid durch Ozon bestätigt werden. Für das Emissionsverhalten der Mikroalgenprobe aus dem Sylter Wattenmeer, welche keine Korrelation mit dem verbrauchten Ozon zeigt, gibt es zwei Erklärungen: Zum einen kann I2 durch den hohen Gehalt an organischen Verbindungen an diesen adsorbiert bzw. chemisch gebunden werden und wird dann nicht mehr in die Gasphase emittiert. Zum anderen können aktive organische Verbindungen das Gleichgewicht zwischen HOI und I2 in Richtung HOI verlagern. Im Versuch zur abiotischen Bildung von Iodalkanen aus Partikeln, bestehend aus I2O5 und verschiedenen Alkoholen, kann gezeigt werden, dass die Bildung von Iodmethan und Diiodmethan abläuft, dass jedoch die Emission bis zu zwei Größenordnungen kleiner ist als die von I2. Somit trägt die Bildung von Iodalkanen nur in einem sehr eingeschränkten Rahmen zum Recycling des Iods in der Atmosphäre bei. Der vorgestellte abiotische Bildungsweg hängt sowohl vom pH-Wert als auch vom Mischungsverhältnis im Partikel ab.

Apicomplexan parasites possess an apical complex that is composed of two secretory organelles recognized as micronemes and rhoptries. Rhoptry contents are secreted into the parasitophorous vacuole during the host cell invasion process. Several rhoptry proteins have been identified in Toxoplasma gondii and seem to be involved in host-pathogen interactions and some of them are considered to be important virulence factors. Only one rhoptry protein, NcROP2, has been identified and extensively characterized in the closely related parasite Neospora caninum, and this has showed immunoprotective properties. Thus, with the aim of increasing knowledge of the rhoptry protein repertoire in N. caninum, a subcellular fractionation of tachyzoites was performed to obtain fractions enriched for this secretory organelle. 2-D SDS-PAGE followed by MS and LC/MS-MS were applied for fraction analysis and 8 potential novel rhoptry components (NcROP1, 5, 8, 30 and NcRON2, 3, 4, 8) and several kinases, proteases and phosphatases proteins were identified with a high homology to those previously found in T. gondii. Their existence in N. caninum tachyzoites suggests their involvement in similar events or pathways that occur in T. gondii. These novel proteins may be considered as targets that could be useful in the future development of immunoprophylactic measures.

We describe the measurement, at 100 K, of the SIMS relative sensitivity factors (RSFs) of the main physiological cations Na+, K+, Mg2+, and Ca2+ in frozen-hydrated (F-H) ionic solutions. Freezing was performed by either plunge freezing or high-pressure freezing. We also report the measurement of the RSFs in flax fibers, which are a model for ions in the plant cell wall, and in F-H ionic samples, which are a model for ions in the vacuole. RSFs were determined under bombardment with neutral oxygen (FAB) for both the fibers and the F-H samples. We show that referencing to ice-characteristic secondary ions is of little value in determining RSFs and that referencing to K is preferable. The RSFs of Na relative to K and of Ca relative to Mg in F-H samples are similar to their respective values in fiber samples, whereas the RSFs of both Ca and Mg relative to K are lower in fibers than in F-H samples. Our data show that the physical factors important for the determination of the RSFs are not the same in F-H samples and in homogeneous matrixes. Our data show that it is possible to perform a SIMS relative quantification of the cations in frozen-hydrated samples with an accuracy on the order of 15%. Referencing to K permits the quantification of the ionic ratios, even when the absolute concentration of the referencing ion is unknown. This is essential for physiological studies of F-H biological samples.

Polybrominated diphenyl ethers (PBDEs) are considered persistent organic pollutants because of their ubiquity, persistence and bioaccumulation. Its harmful effects on human health and the environment, has led to its inclusion of the Stockholm Convention. Little information is found about PBDEs in abiotic systems of the South America in open literature. This paper reports the presence and concentration level of four PBDEs congeners in Mendoza River, Argentina. The selected PBDEs were: 2,2',4,4'-tetrabromodiphenyl ether (BDE-47), 2,2',4,4',5-pentabromodiphenyl ether (BDE-99), 2,2',4,4',6-pentabromodiphenyl ether (BDE- 100) and 2,2',4,4',5,5'-hexabromodiphenyl ether (BDE-153). The analytical methodology used was head space-solid phase micro extraction combined with gas chromatographymass spectrometry (HS-SPME-GC-MS/MS). Several variables, including pH, salting out, extraction technique type and extraction time were studied and optimized over the relative response the target analytes. The precision of HS-SPME-GC-MS/MS evaluated over five replicate, leading RSDs values <13%, detection limits (S/N=3) ranging from 0.03 pg ml-1 to 0.12 pg ml-1 and the calibration graph was linear with r2=0.9959. BDE-47 and BDE-100 were the predominant congeners found in the analyzed samples. Their concentrations ranged from not detected to 1.9 pg ml-1 and to 0.5 pg ml-1, respectively.

A multiresidue method was developed for the simultaneous determination of 31 emerging contaminants (pharmaceutical compounds, hormones, personal care products, biocides and flame retardants) in aquatic plants. Analytes were extracted by ultrasound assisted-matrix solid phase dispersion (UA-MSPD) and determined by gas chromatography-mass spectrometry after sylilation. The method was validated for different aquatic plants (Typha angustifolia, Arundo donax and Lemna minor) and a semiaquatic cultivated plant (Oryza sativa) with good recoveries at concentrations of 100 and 25 ng g-1 wet weight, ranging from 70 to 120 %, and low method detection limits (0.3 to 2.2 ng g-1 wet weight). A significant difference of the chromatographic response was observed for some compounds in neat solvent versus matrix extracts and therefore quantification was carried out using matrix-matched standards in order to overcome this matrix effect. Aquatic plants taken from rivers located at three Spanish regions were analyzed and the compounds detected were parabens, bisphenol A, benzophenone-3, cyfluthrin and cypermethrin. The levels found ranged from 6 to 25 ng g-1 wet weight except for cypermethrin that was detected at 235 ng g-1 wet weight in Oryza sativa samples.

A new approach to the analysis of metabolic pathways involving poorly water-soluble intermediates is proposed. It relies upon the ability of the hydrophobic intermediates formed by a sequence of intracellular reactions to cross the membrane(s) and partition between aqueous and organic phases, when cells are incubated in the presence of a nonpolar and nontoxic organic solvent. As a result of this thermodynamically driven efflux of the formed intermediates from the cell, they accumulate in the organic medium in sufficient quantities for GC-MS analysis and identification. This enables direct determination of the sequence of chemical reactions involved with no requirement for the isolation of each individual metabolite from a cell-free extract. The feasibility of the proposed methodology has been demonstrated by the elucidation of the biosynthesis of (R)-gamma-decalactone from (R)-ricinoleic acid catalyzed by the yeast Sporidiobolus ruinenii grown in the presence of decane. The corresponding 4-hydroxy-acid intermediates, formed in the course of beta-oxidation of (R)-ricinoleic acid, were simultaneously observed in a single experiment on the same chromatogram. Potential applications of this proposed methodology are briefly discussed.

Neste trabalho é proposto pela primeira vez, o desenvolvimento e validação de um método analítico baseado no emprego da dispersão da matriz em fase sólida (MSPD) modificada, para extração das espécies CH3Hg+ e Hg2+ em amostras de peixe e determinação por cromatografia em fase gasosa acoplada à espectrometria de massas (GC-MS). O método de extração utilizando a MSPD combina o rompimento da estrutura física da amostra, através da maceração e do uso de SiO2 como suporte sólido, com o método da extração ácida, utilizando uma solução de HCl 4,2 mol L-1 e NaCl 0,5 mol L-1. Para otimização da MSPD, foram avaliados parâmetros como massa de amostra, massa de suporte sólido, concentração de HCl, concentração de NaCl, tipo de suporte sólido e o tempo de agitação, com auxílio da metodologia de superfície de resposta. Além disso, a etapa de derivatização e a separação cromatográfica também foram otimizadas na determinação de CH3Hg+ e Hg2+ por GC-MS. O método mostrouse adequado para extração e determinação de espécies de mercúrio através da aplicação em materiais de referência certificados de fígado de peixe (DOLT-3) e músculo de peixe (DORM-2), apresentando boas concordâncias com os valores certificados e desvio padrão relativo inferior a 9,5%. Os limites de detecção foram de 0,06 e 0,12 µg g-1 para CH3Hg+ e Hg2+, respectivamente. Além disso, foi observado um significativo efeito de matriz e, por isso, a calibração foi feita com curvas preparadas com o extrato da MSPD. O método mostrou boa concordância na comparação entre a soma da concentração das espécies e a concentração de mercúrio total determinada por espectrometria de massas com plasma indutivamente acoplado com geração de vapor frio (CVG-ICP-MS), após digestão assistida por micro-ondas (MAD) em peixes do tipo atum (Thunnus thynnus), cação anjo (Squatina squatina) e cação viola (Rhinobatos blochii.).

Tomato (Lycopersicon esculentum L.) is the second most important vegetable crop worldwide and a key component in the so-called “Mediterranean diet”. In the Northeastern region of Portugal, local populations still prefer to consume traditional tomato varieties which they find very tasty and healthy, as they are grown using extensive farming techniques. A previous study of our research team described the nutritional value of the round (batateiro), long (comprido), heart (coração) and yellow (amarelo) tomato varieties [1], but the phenolic profile was unknown until now. Thus, the objective of this study was to characterize the phenolic profiles of these four tomato farmers’ varieties by using HPLC-DAD-ESI/MS and evaluate its antioxidant capacity through four in vitro assays based on different reaction mechanisms. A cis p-coumaric acid derivative was the most abundant compound in yellow and round tomato varieties, while 4-O-caffeolyquinic acid was the most abundant in long and heart varieties. The most abundant flavonoid was quercetin pentosylrutinoside in the four tomato varieties. Yellow tomato presented the highest levels of phenolic compounds, including phenolic acids and flavonoids, but the lowest antioxidant activity. In turn, the round tomato gave the best results in all the antioxidant activity assays. This study demonstrated that these tomato farmers’ varieties are a source of phenolic compounds, mainly phenolic acid derivatives [2], and possess high antioxidant capacity [1]; being thus key elements in the diet to prevent chronic degenerative diseases associated to oxidative stress, such as cancer and coronary artery disease.
