OBJECTIVES: To determine the clinical performance of a laser fluorescence device (DIAGNOdent pen, KaVo) to discriminate between different occlusal caries depths (D(0)-D(1-4); D(0-2)-D(3,4)) in permanent molars. METHODS: In this prospective, randomized two-centre-study 120 sound/uncavitated carious sites in 120 patients were measured after visual and radiographic caries assessment. In cases of operative intervention (n=86), the lesion depths after caries removal were recorded (reference). In cases of preventive intervention (n=34), the sites were reassessed visually/radiographically after 12 months to verify the status assessed before (reference). The discrimination performance was determined statistically (Mann-Whitney test, Spearman's rho coefficient, and areas under the receiver operating characteristic curves (AUCs)). Sensitivities (SE) and specificities (SP) were plotted as a function of the measured values and cut-off values for the mentioned thresholds suggested. RESULTS: Sound sites (n=13) had significantly minor fluorescence values than carious sites (n=107) (P<0.0001) as had sites with no/enamel caries (n=63) compared to dentinal caries (n=57). The AUCs for the same discriminations were 0.92 and 0.78 (P<0.001). For the D(0)-D(1-4) threshold, a cut-off at a value of 12 (SE: 0.88, SP: 0.85) and for the D(0-2)-D(3,4) threshold at 25 (SE: 0.67, SP: 0.79) can be suggested. A moderate positive correlation between the measurements and the caries depths was calculated (rho=+0.57, P=0.01). CONCLUSION: Within this study, the device's discrimination performance for different caries depths was moderate to very good and it may be recommended as adjunct tool in the diagnosis of occlusal caries.

A wealth of genetic associations for cardiovascular and metabolic phenotypes in humans has been accumulating over the last decade, in particular a large number of loci derived from recent genome wide association studies (GWAS). True complex disease-associated loci often exert modest effects, so their delineation currently requires integration of diverse phenotypic data from large studies to ensure robust meta-analyses. We have designed a gene-centric 50 K single nucleotide polymorphism (SNP) array to assess potentially relevant loci across a range of cardiovascular, metabolic and inflammatory syndromes. The array utilizes a "cosmopolitan" tagging approach to capture the genetic diversity across approximately 2,000 loci in populations represented in the HapMap and SeattleSNPs projects. The array content is informed by GWAS of vascular and inflammatory disease, expression quantitative trait loci implicated in atherosclerosis, pathway based approaches and comprehensive literature searching. The custom flexibility of the array platform facilitated interrogation of loci at differing stringencies, according to a gene prioritization strategy that allows saturation of high priority loci with a greater density of markers than the existing GWAS tools, particularly in African HapMap samples. We also demonstrate that the IBC array can be used to complement GWAS, increasing coverage in high priority CVD-related loci across all major HapMap populations. DNA from over 200,000 extensively phenotyped individuals will be genotyped with this array with a significant portion of the generated data being released into the academic domain facilitating in silico replication attempts, analyses of rare variants and cross-cohort meta-analyses in diverse populations. These datasets will also facilitate more robust secondary analyses, such as explorations with alternative genetic models, epistasis and gene-environment interactions.

Tumor specific immunity is mediated by cytotoxic T lymphocytes (CTL) that recognize peptide antigen (Ag) in the context of major histocompatibility complex (MHC) class I molecules and by helper T (Th) lymphocytes that recognize peptide Ag in the context of MHC class II molecules. The purpose of this study is (1) to induce or augment the immunogenicity of nonimmunogenic or weakly immunogenic tumors by genetic modification of tumor cells, and (2) to use these genetically altered cells in cancer immunotherapy. To study this, I transfected a highly tumorigenic murine melanoma cell line (K1735) that did not express constitutively either MHC class I or II molecules with syngeneic cloned MHC class I and/or class II genes, and then determined the tumorigenicity of transfected cells in normal C3H mice. K1735 transfectants expressing either $\rm K\sp{k}$ or $\rm A\sp{k}$ molecules alone produced tumors in normal C3H mice, whereas most transfectants that expressed both molecules were rejected in normal C3H mice but produced tumors in nude mice. The rejection of K1735 transfectants expressing $\rm K\sp{k}$ and $\rm A\sp{k}$ Ag in normal C3H mice required both $\rm CD4\sp+$ and $\rm CD8\sp+$ T cells. Interestingly, the $\rm A\sp{k}$ requirement can be substituted by IL-2 because transfection of $\rm K\sp{k}$-positive/A$\sp{\rm k}$-negative K1735 cells with the IL-2 gene also resulted in abrogation of tumorigenicity in normal C3H mice but not in nude mice. In addition, 1735 $(\rm I\sp+II\sp+)$ transfected cells can function as antigen presenting cells (APC) since they could process and present native hen egg lysozyme (HEL) to HEL specific T cell hybridomas. Furthermore, the transplantation immunity induced by K1735 transfectants expressing both $\rm K\sp{k}$ and $\rm A\sp{k}$ molecules completely cross-protected mice against challenge with $\rm K\sp{k}$-positive transfectants but weakly protected them against challenge with parental K1735 cells or $\rm A\sp{k}$-positive transfectants. Finally, I demonstrated that MHC $(\rm I\sp+II\sp+)$ or $\rm K\sp{k}$-positive/IL-2-positive cells can function as anti-cancer vaccines since they can abrogate the growth of established tumors and metastasis.^ In summary, my results indicate that expression of either MHC class I or II molecule alone is insufficient to cause the rejection of K1735 melanoma in syngeneic hosts and that both molecules are necessary. In addition, my data suggest that the failure of $\rm K\sp{k}$-positive K1735 cells to induce a primary tumor-rejection response in normal C3H mice may be due to their inability to induce the helper arm of the anti-tumor immune response. Finally, the ability of MHC $(\rm I\sp+II\sp+)$ or $\rm K\sp{k}$-positive/IL-2-positive cells to prevent growth of established tumors or metastasis suggests that these cell lines can serve as potential vaccines for the immunotherapy of cancer. Cretaceous sediments were recovered at all four sites (Sites 463-466) of the central North Pacific drilled during Leg 62 of the Deep Sea Drilling Project. One of the objectives was to get more information about the development of ocean plankton communities and early evolution of planktonic groups of the Mesozoic. In this article, the Cretaceous calcareous nannofossils from two areas of the central North Pacific (Mid-Pacific Mountains and Hess Rise) are listed and discussed. (The Cenozoic calcareous nannofossils are discussed by R. Schmidt 1981). Coring was continuous at all sites. Mesozoic calcareous nannoplankton assemblages range on the Mid-pacific Mountains from Barremian to Early Maastrichtian, and on Hess Rise from Albian to Late Maastrichtian. (No calcareous nannofossils older than Barremian or Albian respectively were found).

Benthic foraminifers were studied in 99 samples collected from the lower 200 m of Hole 765C. The studied section ranges from the Tithonian to Aptian, and benthic foraminifers can be subdivided into five assemblages on the basis of faunal diversity and stratigraphic ranges of distinctive species. Compared with deep-water assemblages from Atlantic DSDP sites and Poland, assemblages from the Argo Abyssal Plain display a higher diversity of agglutinated forms, which comprise the autochthonous assemblages. Assemblages at the base of Hole 765C are wholly composed of agglutinated forms, reflecting deposition beneath the carbonate compensation depth (CCD). Most calcareous benthic species are found in turbidite layers, and the presence of an upper Valanginian Praedorothia praehauteriviana Assemblage may indicate deposition at or just below the CCD. The P. praehauteriviana Assemblage from Hole 765C is the temporal equivalent of similar assemblages from DSDP Holes 534A, 416A, 370, 105, and 101 in the Atlantic Ocean and Hole 306 in the Pacific Ocean. Stratigraphic ranges of cosmopolitan agglutinated species at Site 765 generally overlap with their reported ranges in the Atlantic and in the bathyal flysch sequences of the Carpathians; however, several species from Hole 765C have not been previously reported from Uppermost Jurassic to Lower Cretaceous abyssal sediments.

Palynological data from offshore Costa Rica, allow us to investigate the relationship between dinoflagellate cyst assemblages and changes in regional oceanic primary productivity. From Miocene to Pleistocene, productivity at ODP Site 1039 was influenced by tectonic drift, as Site 1039 approached the continent, from the Equator to its current position at ~10°N. In addition, dinoflagellate abundance is modulated by regional productivity events, which modified primary productivity, as also indicated by available data on calcareous nannofossils, diatoms, TOC, and CaCO3 content. Five palynomorph intervals are defined. The early-late Miocene one, dominated by Batiacasphaera, represents relatively stable, productive oceanic conditions before the closure of the Indonesian and Panama Seaways. The late Miocene decrease in palynomorph recovery is related to the Carbonate Crash Event. The high abundance and diversity of the assemblages at the end of the late Miocene to early Pliocene indicate increased productivity related to the Global Biogenic Bloom, and a change in dominance from Batiacasphaera to Impagidinium to Nematosphaeropsis. The low abundance of the late Pliocene interval is related to El Niño-like conditions, and there is another change related to the disappearance of Batiacasphaera and dominance of Impagidinium, Nematosphaeropsis, and Operculodinium. The abundant Pleistocene assemblages represent increased marine productivity, and a high influx of continental palynomorphs and bissacate pollen, associated with the proximity of the Costa Rica Dome. Pleistocene dinoflagellates are characterized by Spiniferites and Selenopemphix, together with rare Impagidinium and Nematosphaeropsis.
