THE STORY OF HOW FEATHERS EVOLVED IS FAR FROM OVER. IN 1868, THOMAS HUXLEY declared that dinosaurs gave rise to birds. He based his claim on Compsognathus, a 150-million-year-old dinosaur fossil from Solnhofen, Germany, whose delicate hind legs were remarkably similar to those of table fowl. The discovery seven years earlier of Archaeopteryx, a fossil bird with a long bony tail, toothed jaws and clawed fingers, had convinced many people that birds were somehow related to reptiles. But Compsognathus was the fossil that placed dinosaurs firmly in the middle of this complex evolutionary equation. Wings, claimed Huxley, must have grown out of rudimentary forelimbs. And feathers? Whether Compsognathus had them, Huxley could only guess. Nevertheless, his theory clearly required that scales had somehow transformed into feathers. The question was not just how, but why?

The spermatozoon of Apus apus is typical of non-passerines in many respects. Features shared with palaeognaths and the Galloanserae are the conical acrosome, shorter than the nucleus; the presence of a proximal as well as distal centriole; the elongate midpiece with mitochondria grouped around an elongate distal centriole; and the presence of a fibrous or amorphous sheath around the principal piece of the axoneme. The perforatorium and endonuclear canal are lost in A. apus as in some other non-passerines. All non-passerines differ from palaeognaths in that the latter have a transversely ribbed fibrous sheath whereas in non-passerines it is amorphous, as in Apus, or absent. The absence of an annulus is an apomorphic but homoplastic feature of swift, psittaciform, gruiform and passerine spermatozoa. The long distal centriole, penetrating the entire midpiece, is a remarkably plesiomorphic feature of the swift spermatozoa, known elsewhere only in palaeognaths. The long centriole of Apus, if not a reversal, would be inconsistent with the former placement of the Apodiformes above the Psittaciformes from DNA-DNA hybridization. In contrast to passerines, in A. apus the microtubules in the spermatid are restricted to a transient single row encircling the cell. The form of the spermatozoon fully justifies the exclusion of swifts from the passerine family Hirundinidae.

This study describes the identification of outer membrane proteins (OMPs) of the bacterial pathogen Pasteurella multocida and an analysis of how the expression of these proteins changes during infection of the natural host. We analysed the sarcosine-insoluble membrane fractions, which are highly enriched for OMPs, from bacteria grown under a range of conditions. Initially, the OMP-containing fractions were resolved by 2-DE and the proteins identified by MALDI-TOF MS. In addition, the OMP-containing fractions were separated by 1-D SDS-PAGE and protein identifications were made using nano LC MS/MS. Using these two methods a total of 35 proteins was identified from samples obtained from organisms grown in rich culture medium. Six of the proteins were identified only by 2-DE MALDI-TOF MS, whilst 17 proteins were identified only by 1-D LC MS/MS. We then analysed the OMPs from P. multocida which had been isolated from the bloodstream of infected chickens (a natural host) or grown in iron-depleted medium. Three proteins were found to be significantly up-regulated during growth in vivo and one of these (Pm0803) was also up-regulated during growth in iron-depleted medium. After bioinformatic analysis of the protein matches, it was predicted that over one third of the combined OMPs predicted by the bioinformatics sub-cellular localisation tools PSORTB and Proteome Analyst, had been identified during this study. This is the first comprehensive proteomic analysis of the P. multocida outer membrane and the first proteomic analysis of how a bacterial pathogen modifies its outer membrane proteome during infection.

The sperm of Caprimulgus europaeus is typical of other nonpasserines in many respects. Features shared with Paleognathae and Galloanserae are the conical acrosome, shorter than the nucleus; the presence of a perforatorium and endonuclear canal; the presence of a proximal as well as distal centriole; the elongate midpiece with mitochondria grouped around a central axis (here maximally six mitochondria in similar to 10 tiers); and the presence of a fibrous or amorphous sheath around the principal piece of the axoneme. A major (apomorphic) difference from paleognaths and galloanserans is the short distal centriole, the midpiece being penetrated for most of its length by the axoneme and for only a very short proximal portion by the centriole. Nonpasserines differ from paleognaths in that the latter have a transversely ribbed fibrous sheath, whereas in nonpasserines it is amorphous, as in Caprimulgus, or absent. The absence of an annulus is an apomorphic feature of Caprimulgus, apodiform, psittaciform, gruiform, and passerine sperm, homoplastic in at least some of these. In contrast to passerines, in Caprimulgus the cytoplasmic microtubules in the spermatid are restricted to a transient longitudinal manchette. The structure of the spermatid and spermatozoon is consistent with placement of the Caprimulgidae near the Psittacidae, but is less supportive of close proximity to the Apodidae, from DNA-DNA hybridization and some other analyses.

A total of 36 tonsil swab samples were collected from healthy swine prior to slaughter at the abattoirs in Can tho and Tien giang provinces of Southern Vietnam, The presence of Pasteurella multocida in these samples was detected by the combination of direct cultivation and isolation, mouse inoculation and the polymerase chain reaction (PM-PCR). P. multocida was detected in 16 samples by PCR, with 17 strains ultimately isolated. All samples were negative for serogroup B by HSB-PCR and conventional serotyping, with isolates identified as A:3, D:1 or D:3. In addition, all samples were determined to be negative for the P. multocida toxin (PMT). Characterisation of isolated P, multocida by REP-PCR and biotyping revealed nine distinct REP profiles and seven biotypes among the 17 isolates. Some correlation was seen with P. multocida isolated from a previous Australian outbreak of acute swine pasteurellosis, and those isolated from fowl cholera outbreaks in Vietnamese poultry. Enzyme products did not have a significant effect (P>0.05) on weekly fed intake and weight gain of birds. But feed intake tended to drop and weight gain tended to increase in response to supplementation of the three enzymes. O chumbo é um metal pesado que constitui um dos grandes problemas ambientais em termos de poluição atmosférica, aquática e terrestre. O impacto da exposição ao chumbo tem consequências nas características morfológicas e bioquímicas deem aves, porém são escassos os estudos sobre os efeitos no sistema reprodutivo das aves. O objetivo deste trabalho é avaliar os efeitos do acetato de chumbo em parâmetros de integridade, histopatologia e bioquímica emnas células espermáticas. Foram coletadas 36 aves silvestres (Chrysomus ruficapillus) adultos, machos e expostas em gaiolas. Foi administrada uma dose única de 50 e 100 mg/kg de acetato de chumbo através de uma injeção intraperitoneal e o grupo controle recebeu uma injeção de solução salina. Após sete dias da administração das doses, foi realizada a coleta dos ductos deferentes e testículos para as análises nas células espermáticas. Os resultados mostraram que houve deterioração na integridade da membrana e DNA, e diminuição da funcionalidade mitocondrial nos testículos das aves expostas ao acetato de chumbo nas duas doses do estudo (P<0,05). Na histopatologia foi observada diminuição na quantidade de células dos estágios de desenvolvimento da espermatogênese, além de patologias nas mesmas. Observou-se danos oxidativos nas aves tratadas com 100mg/kg e um aumento da peroxidação lipídica nos testículos. Portanto, o acetato de chumbo causou efeitos negativos no aparelho reprodutivo de Chrysomus ruficapillius
