The coastal upwelling system off the coast of Peru is characterized by high biological activity and a pronounced subsurface oxygen minimum zone, as well as associated emissions of atmospheric trace gases such as N2O, CH4 and CO2. From 3 to 23 December 2012, R/V Meteor (M91) cruise took place in the Peruvian upwelling system between 4.59 and 15.4°S, and 82.0 to 77.5°W. During M91 we investigated the composition of the sea-surface microlayer (SML), the oceanic uppermost boundary directly subject to high solar radiation, often enriched in specific organic compounds of biological origin like chromophoric dissolved organic matter (CDOM) and marine gels. In the SML, the continuous photochemical and microbial recycling of organic matter may strongly influence gas exchange between marine systems and the atmosphere. We analyzed SML and underlying water (ULW) samples at 38 stations focusing on CDOM spectral characteristics as indicator of photochemical and microbial alteration processes. CDOM composition was characterized by spectral slope (S) values and excitation-emission matrix fluorescence (EEMs), which allow us to track changes in molecular weight (MW) of DOM, and to determine potential DOM sources and sinks. Spectral slope S varied between 0.012 to 0.043 1 nm-1 and was quite similar between SML and ULW, with no significant differences between the two compartments. Higher S values were observed in the ULW of the southern stations below 15°S. By EEMs, we identified five fluorescent components (F1-5) of the CDOM pool, of which two had excitation/emission characteristics of amino-acid-like fluorophores (F1, F4) and were highly enriched in the SML, with a median ratio SML : ULW of 1.5 for both fluorophores. In the study region, values for CDOM absorption ranged from 0.07 to 1.47 m-1. CDOM was generally highly concentrated in the SML, with a median enrichment with respect to the ULW of 1.2. CDOM composition and changes in spectral slope properties suggested a local microbial release of DOM directly in the SML as a response to light exposure in this extreme environment. In a conceptual model of the sources and modifications of optically active DOM in the SML and underlying seawater (ULW), we describe processes we think may take place (Fig. 1); the production of CDOM of higher MW by microbial release through growth, exudation and lysis in the euphotic zone, includes the identified fluorophores (F1, F2, F3, F4, F5). Specific amino-acid-like fluorophores (F1, F4) accumulate in the SML with respect to the ULW, as photochemistry may enhance microbial CDOM release by (a) photoprotection mechanisms and (b) cell-lysis processes. Microbial and photochemical degradation are potential sinks of the amino-acid-like fluorophores (F1, F4), and potential sources of reworked and more refractory humic-like components (F2, F3, F5). In the highly productive upwelling region along the Peruvian coast, the interplay of microbial and photochemical processes controls the enrichment of amino-acid-like CDOM in the SML. We discuss potential implications for air-sea gas exchange in this area.

Diatoms can occur as single cells or as chain-forming aggregates. These two strategies affect buoyancy, predator evasion, light absorption and nutrient uptake. Adjacent cells in chains establish connections through various processes that determine strength and flexibility of the bonds, and at distinct cellular locations defining colony structure. Chain length has been found to vary with temperature and nutrient availability as well as being positively correlated with growth rate. However, the potential effect of enhanced carbon dioxide (CO2) concentrations and consequent changes in seawater carbonate chemistry on chain formation is virtually unknown. Here we report on experiments with semi-continuous cultures of the freshly isolated diatom Asterionellopsis glacialis grown under increasing CO2 levels ranging from 320 to 3400 µatm. We show that the number of cells comprising a chain, and therefore chain length, increases with rising CO2 concentrations. We also demonstrate that while cell division rate changes with CO2 concentrations, carbon, nitrogen and phosphorus cellular quotas vary proportionally, evident by unchanged organic matter ratios. Finally, beyond the optimum CO2 concentration for growth, carbon allocation changes from cellular storage to increased exudation of dissolved organic carbon. The observed structural adjustment in colony size could enable growth at high CO2 levels, since longer, spiral-shaped chains are likely to create microclimates with higher pH during the light period. Moreover increased chain length of Asterionellopsis glacialis may influence buoyancy and, consequently, affect competitive fitness as well as sinking rates. This would potentially impact the delicate balance between the microbial loop and export of organic matter, with consequences for atmospheric carbon dioxide.

Las sociedades desarrolladas generan una gran cantidad de residuos, que necesitan una adecuada gestión. Esta problemática requiere, de este modo, una atención creciente por parte de la sociedad, debido a la necesidad de proteger el medio ambiente. En este sentido, los esfuerzos se centran en reducir al máximo la generación de residuos y buscar vías de aprovechamiento de aquellos que son inevitables, soluciones mucho más aconsejables desde el punto de vista técnico, ecológico y económico que su vertido o destrucción. Las industrias deben adoptar las medidas precisas para fomentar la reducción de estos residuos, desarrollar tecnologías limpias que permitan el ahorro de los recursos naturales que poseemos, y sobre todo buscar métodos de reutilización, reciclado, inertización y valorización de los residuos generados en su producción. La industria de la construcción es un campo muy receptivo para el desarrollo de nuevos materiales en los que incorporar estos residuos. La incorporación de diferentes residuos industriales en matrices cerámicas se plantea como una vía barata de fijar las diferentes especies metálicas presentes en transformación de rocas ornamentales, lodos de galvanización o metalúrgicos, etc. En todos los casos, la adición de estos residuos requiere su caracterización previa y la optimización de las condiciones de conformado y cocción en el caso de su incorporación a la arcilla cocida. Entre los residuos incorporados en materiales de construcción se encuentran las escorias de aluminio. La industria metalúrgica produce durante sus procesos de fusión diferentes tipos de escorias. Su reciclado es una de las líneas de interés para estas industrias. En el caso de las escorias de aluminio, su tratamiento inicial consiste en una recuperación del aluminio mediante métodos mecánicos seguido de un tratamiento químico, o plasma. Este método conduce a que la escoria final apenas contenga aluminio y sea rica en sales solubles lo que limita su almacenamiento en escombreras. La escoria es una mezcla de aluminio metal y productos no metálicos como óxidos, nitruros y carburos de aluminio, sales y otros óxidos metálicos. En este estudio se ha analizado la posibilidad de la adición de escorias de aluminio procedentes de la metalurgia secundaria en materiales de construcción, de forma que tras un procesado de las mismas permita la obtención de materiales compuestos de matriz cerámica. En la presente Tesis Doctoral se ha analizado la viabilidad técnica de la incorporación de las escorias de aluminio procedentes de la metalurgia secundaria en una matriz de arcilla cocida. Para ello se han aplicado diferentes tratamientos a la escoria y se han aplicado diferentes variables en su procesado como la energía de molienda o la temperatura de sinterizacion, además del contenido de escoria. Su compactación con agua entre el 5-10 %, secado y sinterización permite obtener piezas rectangulares de diverso tamaño. Desde el punto de vista del contenido de la escoria, se incorporó entre un 10 y 40% de escoria TT, es decir sometida una calcinación previa a 750ºC en aire. Los mejores resultados alcanzados corresponden a un contenido del 20% ESC TT, sinterizada a 980ºC, por cuanto altos contenidos en escoria condicen a piezas con corazón negro. Los productos obtenidos con la adición de 20% de escoria de aluminio a la arcilla, presentan una baja expansión tras sinterización, mejores propiedades físicas y mecánicas, y mayor conductividad térmica que los productos obtenidos con arcilla sin adiciones. Aumenta su densidad, disminuye su absorción y aumenta sus resistencias de flexión y compresión, al presentar una porosidad cerrada y una interacción escoria-matriz. En todos los casos se produce una exudación superficial de aluminio metálico, cuyo volumen está relacionado con la cantidad de escoria adicionada. Mediante la incorporación de este contenido de escoria, tras un tratamiento de disolución de sales y posterior calcinación (ESC TTQ), se mejoran las propiedades del material compuesto, no sólo sobre la de la escoria calcinada (ESC TT), sino también, sobre la escoria sin tratamiento (ESC). Si además, la adición del 20% de escoria añadida, está tratada, no sólo térmicamente sino también químicamente (ESC TTQ), éstas mejoran aún más las propiedades del material compuesto, siendo el producto más compacto, con menos poros, por lo que los valores de densidad son más elevados, menores son las absorciones y mayores resistencias de flexión y compresión, que los productos obtenidos con la adición de escoria sólo tratada térmicamente. Alcanzando valores de resistencias características a compresión del orden de 109 MPa. Los valores de conductividad térmica obtenidos también son mayores. Los ensayos tecnológicos con piezas de 160 x 30 x 5 mm y el material compuesto optimizado de arcilla+ 20%ESCTTQ, consistieron en la determinación de su expansión por humedad, eflorescencia y heladicidad, mostrando en general un mejor comportamiento que la arcilla sin adiciones. Así, se han obtenido nuevos materiales compuestos de matriz cerámica para la construcción, mejorando sus propiedades físicas, mecánicas y térmicas, utilizando escorias de aluminio procedentes de la metalurgia secundaria, como opción de valorización de estos residuos, evitando así, que se viertan a vertederos y contaminen el medio ambiente. ABSTRACT Developed societies generate a lot of waste, which need proper management. Thus, this problem requires increased attention from the society, due to the need to protect the environment. In this regard, efforts are focused on to minimize the generation of waste and find ways of taking advantage of those who are inevitable, much more advisable solutions from the technical, ecological and economic viewpoint to disposal or destruction. Industries should adopt precise measures to promote waste reduction, develop clean technologies that allow the saving of natural resources that we possess, and above all seek methods of reuse, recycling, recovery and valorisation of the waste generated in their production. The industry of the construction is a very receptive field for the development of new materials in which to incorporate these residues. The incorporation of different industrial residues in ceramic counterfoils appears as a cheap route to fix the different metallic present species in transformation of ornamental rocks, muds of galvanization or metallurgical, etc. In all the cases, the addition of these residues needs his previous characterization and the optimization of the conditions of conformed and of baking in case of his incorporation to the baked clay. Residues incorporated into construction materials include aluminium slag. The metallurgical industry produces during their fusion processes different types of slags. Recycling is one of the lines of interest to these industries. In the case of aluminium slag, their initial treatment consists of a recovery of the aluminium using mechanical methods followed by chemical treatment, or plasma. This method leads to that final slag just contains aluminium and is rich in soluble salts which limits storage in dumps. The slag is a mixture of aluminium metal and non-metallic such as oxides, nitrides and carbides of aluminium salts products and other metal oxides. The present Doctoral thesis has analysed the technical viability of the incorporation of aluminium slag from secondary Metallurgy in an array of baked clay. So they have been applied different treatments to the slag and have been applied different variables in its processing as the temperature of sintering, in addition to the content of slag or energy grinding. Its compaction with water between 5-10%, drying and sintering allows rectangular pieces of different size. From the point of view of the content of the slag, it is incorporated between 10 and 40% slag TT, that is to say, submitted a calcination prior to 750 ° C in air. The best results achieved correspond to 20% ESC TT, sintered at 980 ° C, as high levels of slag in accordance to pieces with black heart. The products obtained with the addition of 20% of slag from aluminium to clay, present a low expansion after sintering, better physical properties and mechanical, and higher thermal conductivity than the products obtained with clay, without addictions. Its density increases, decreases its absorption and increases its resistance to bending and compression, introducing a closed porosity and slag-matrix interaction. In all cases there is a superficial exudation of metallic aluminium, whose volume is related to the amount of slag added. By incorporating this content of slag, following a treatment of salt solution and subsequent calcination (ESC TTQ), are improved the properties of composite material not only on the calcined slag (ESC TT), but also in the slag without treatment (ESC). If the addition of 20% of slag added, is also treated, not only thermally but also chemically (ESC TTQ), they further improve the properties of the composite material, the product is more compact, less porous, so the values are higher density, minors are absorptions and greater resistance in bending and compression, to the products obtained with the addition of slag only treated thermally. Reaching values of compressive resistance characteristic of the order of 109 MPa. The thermal conductivity values obtained are also higher. Testing technology with pieces of 160 x 30 x 5 mm and optimized composite material of clay 20% ESCTTQ, consisted in the determination of its expansion by moisture, efflorescence and frost resistance, in general, showing a better performance than the clay without additions. Thus, we have obtained new ceramic matrix composite materials for construction, improving its physical, mechanical and thermal properties, using aluminium slag secondary metallurgy, as an option Valuation of these wastes, thus preventing them from being poured to landfills and pollute environment.

Volemitol (d-glycero-d-manno-heptitol, α-sedoheptitol) is an unusual seven-carbon sugar alcohol that fulfills several important physiological functions in certain species of the genus Primula. Using the horticultural hybrid polyanthus (Primula × polyantha) as our model plant, we found that volemitol is the major nonstructural carbohydrate in leaves of all stages of development, with concentrations of up to 50 mg/g fresh weight in source leaves (about 25% of the dry weight), followed by sedoheptulose (d-altro-2-heptulose, 36 mg/g fresh weight), and sucrose (4 mg/g fresh weight). Volemitol was shown by the ethylenediaminetetraacetate-exudation technique to be a prominent phloem-mobile carbohydrate. It accounted for about 24% (mol/mol) of the phloem sap carbohydrates, surpassed only by sucrose (63%). Preliminary 14CO2 pulse-chase radiolabeling experiments showed that volemitol was a major photosynthetic product, preceded by the structurally related ketose sedoheptulose. Finally, we present evidence for a novel NADPH-dependent ketose reductase, tentatively called sedoheptulose reductase, in volemitol-containing Primula species, and propose it as responsible for the biosynthesis of volemitol in planta. Using enzyme extracts from polyanthus leaves, we determined that sedoheptulose reductase has a pH optimum between 7.0 and 8.0, a very high substrate specificity, and displays saturable concentration dependence for both sedoheptulose (apparent Km = 21 mm) and NADPH (apparent Km = 0.4 mm). Our results suggest that volemitol is important in certain Primula species as a photosynthetic product, phloem translocate, and storage carbohydrate.

Al-resistant (alr) mutants of Arabidopsis thaliana were isolated and characterized to gain a better understanding of the genetic and physiological mechanisms of Al resistance. alr mutants were identified on the basis of enhanced root growth in the presence of levels of Al that strongly inhibited root growth in wild-type seedlings. Genetic analysis of the alr mutants showed that Al resistance was semidominant, and chromosome mapping of the mutants with microsatellite and random amplified polymorphic DNA markers indicated that the mutants mapped to two sites in the Arabidopsis genome: one locus on chromosome 1 (alr-108, alr-128, alr-131, and alr-139) and another on chromosome 4 (alr-104). Al accumulation in roots of mutant seedlings was studied by staining with the fluorescent Al-indicator dye morin and quantified via inductively coupled argon plasma mass spectrometry. It was found that the alr mutants accumulated lower levels of Al in the root tips compared with wild type. The possibility that the mutants released Al-chelating organic acids was examined. The mutants that mapped together on chromosome 1 released greater amounts of citrate or malate (as well as pyruvate) compared with wild type, suggesting that Al exclusion from roots of these alr mutants results from enhanced organic acid exudation. Roots of alr-104, on the other hand, did not exhibit increased release of malate or citrate, but did alkalinize the rhizosphere to a greater extent than wild-type roots. A detailed examination of Al resistance in this mutant is described in an accompanying paper (J. Degenhardt, P.B. Larsen, S.H. Howell, L.V. Kochian [1998] Plant Physiol 117: 19–27).

Symptoms associated with pistachio dieback in Australia include decline (little or no current season growth), xylem staining in shoots two or more years old, trunk mu and limb lesions (often covered by black, superficial fungal growth), excessive exudation of resin, dieback and death of the tree. Bacteria belonging to the genus Xanthomonas have been suggested as the causal agent. To confirm the constant association between these bacteria and the disease syndrome, the absence of other pathogens and the identity of the pathogen, we performed a series of isolations and pathogenicity tests. The only microorganism consistently isolated from diseased tissue was a bacterium that produced yellow, mucoid colonies and displayed morphological and cultural characteristics typical of the genus Xanthomonas. Database comparisons of the fatty acid and whole-cell protein profiles of five representative pistachio isolates indicated that they all belonged to X. translucens, but it was not possible to allocate the isolates to pathovar. Pathogenicity tests on cereals and grasses supported this identification. However, Koch's postulates have been only partially fulfilled because not all symptoms associated with pistachio dieback were reproduced on inoculated two-year-old pistachio trees. While discolouration was observed, dieback, excessive resinous exudate and trunk and limb lesions were not produced; expression of these symptoms may be delayed, and long-term monitoring of a small number of inoculated trees is in progress.

DESIGN. Retrospective analysis PURPOSE. Macular oedema is not directly visible on two dimensional digital photographs such that surrogate markers need to be used. In the English National Screening Programme these are exudate within one optic disc diameter (DD) of the fovea, group of exudates within two DD of the fovea and haemorrhages or microaneurysms (HMA) within one DD of the fovea with best corrected visual acuity (VA) worse than 6/9. All patients who present with any of these surrogate markers at screening are referred to an ophthalmology clinic for slit lamp examination. The purpose of this audit was to determine how many patients with positive maculopathy diagnosis on photography were truly identified by optical coherence tomography (OCT) with macular oedema. METHODS. Data was collected from patients attending digital diabetic retinopathy screening. Patients who presented with surrogate markers for macular oedema also had an OCT scan. The fast macula scan on the Stratus OCT was used and an ophthalmologist reviewed the scans to determine whether macular oedema was present. RESULTS. Maculopathy by exudates: Of 155 patients 45 (29%) showed thickening on the OCT of these 12 required laser. Those who also had pre-proliferative retinopathy (n=20) were more likely to have macular oedema (75%) than those with background diabetic retinopathy. Maculopathy by HMA and VA worse than 6/9: Of 66 patients 11 (16.7%) showed thickening on the OCT. 5 (7.6%) of these had macular oedema, 5 (7.6%) epi-retinal membrane, and 1 (1.5%) age related macular degeneration. None of these patients required laser. CONCLUSIONS. The likelihood of the presence of macular oedema and requiring laser treatment is greater with macular exudation than HMA within one DD and reduced VA. Overall the surrogate markers used show low specificity for macular oedema, however combining OCT with photography does identify those with macular oedema who require a true referral for an ophthalmological slit lamp examination.

The exponential growth of studies on the biological response to ocean acidification over the last few decades has generated a large amount of data. To facilitate data comparison, a data compilation hosted at the data publisher PANGAEA was initiated in 2008 and is updated on a regular basis (doi:10.1594/PANGAEA.149999). By January 2015, a total of 581 data sets (over 4 000 000 data points) from 539 papers had been archived. Here we present the developments of this data compilation five years since its first description by Nisumaa et al. (2010). Most of study sites from which data archived are still in the Northern Hemisphere and the number of archived data from studies from the Southern Hemisphere and polar oceans are still relatively low. Data from 60 studies that investigated the response of a mix of organisms or natural communities were all added after 2010, indicating a welcomed shift from the study of individual organisms to communities and ecosystems. The initial imbalance of considerably more data archived on calcification and primary production than on other processes has improved. There is also a clear tendency towards more data archived from multifactorial studies after 2010. For easier and more effective access to ocean acidification data, the ocean acidification community is strongly encouraged to contribute to the data archiving effort, and help develop standard vocabularies describing the variables and define best practices for archiving ocean acidification data.

The filamentous and diazotrophic cyanobacterium Nodularia spumigena plays a major role in the productivity of the Baltic Sea as it forms extensive blooms regularly. Under phosphorus limiting conditions Nodularia spumigena has a high enzyme affinity for dissolved organic phosphorus (DOP) by production and release of alkaline phosphatase. Additionally, it is able to degrade proteinaceous compounds by expressing the extracellular enzyme leucine aminopeptidase. As atmospheric CO2 concentrations are increasing, we expect marine phytoplankton to experience changes in several environmental parameters including pH, temperature, and nutrient availability. The aim of this study was to investigate the combined effect of CO2-induced changes in seawater carbonate chemistry and of phosphate deficiency on the exudation of organic matter, and its subsequent recycling by extracellular enzymes in a Nodularia spumigena culture. Batch cultures of Nodularia spumigena were grown for 15 days aerated with three different pCO2 levels corresponding to values from glacial periods to future values projected for the year 2100. Extracellular enzyme activities as well as changes in organic and inorganic compound concentrations were monitored. CO2 treatment-related effects were identified for cyanobacterial growth, which in turn was influencing exudation and recycling of organic matter by extracellular enzymes. Biomass production was increased by 56.5% and 90.7% in the medium and high pCO2 treatment, respectively, compared to the low pCO2 treatment and simultaneously increasing exudation. During the growth phase significantly more mucinous substances accumulated in the high pCO2 treatment reaching 363 µg Gum Xanthan eq /l compared to 269 µg Gum Xanthan eq /l in the low pCO2 treatment. However, cell-specific rates did not change. After phosphate depletion, the acquisition of P from DOP by alkaline phosphatase was significantly enhanced. Alkaline phosphatase activities were increased by factor 1.64 and 2.25, respectively, in the medium and high compared to the low pCO2 treatment. In conclusion, our results suggest that Nodularia spumigena can grow faster under elevated pCO2 by enhancing the recycling of organic matter to acquire nutrients.

A lean muscle line (L) and a fat muscle line (F) of rainbow trout were established (Quillet et al., 2005) by a two-way selection for muscle lipid content performed on pan-size rainbow trout using a non-destructive measurement of muscle lipid content (Distell Fish Fat Meter®). The aim of the present study was to evaluate the consequences of this selective breeding on flesh quality of pan size (290 g) diploid and triploid trout after three generations of selection. Instrumental evaluations of fillet color and pH measurement were performed at slaughter. Flesh color, pH, dry matter content and mechanical resistance were measured at 48 h and 96 h postmortem on raw and cooked flesh, respectively. A sensorial profile analysis was performed on cooked fillets. Fillets from the selected fatty muscle line (F) had a higher dry matter content and were more colorful for both raw and cooked fillets. Mechanical evaluation indicated a tendency of raw flesh from F fish to be less firm, but this was not confirmed after cooking, neither instrumentally or by sensory analysis. The sensory analysis revealed higher fat loss, higher intensity of flavor of cooked potato, higher exudation, higher moisture content and a more fatty film left on the tongue for flesh from F fish. Triploid fish had mechanically softer raw and cooked fillets, but the difference was not perceived by the sensorial panel. The sensorial evaluation also revealed a lower global intensity of odor, more exudation and a higher moisture content in the fillets from triploid fish. These differences in quality parameters among groups of fish were associated with larger white muscle fibers in F fish and in triploid fish. The data provide additional information about the relationship between muscle fat content, muscle cellularity and flesh quality.

Aluminium (Al) toxicity and drought are the two major abiotic stress factors limiting common bean production in the tropics. Using hydroponics, the short-term effects of combined Al toxicity and drought stress on root growth and Al uptake into the root apex were investigated. In the presence of Al stress, PEG 6000 (polyethylene glycol)-induced osmotic (drought) stress led to the amelioration of Al-induced inhibition of root elongation in the Al-sensitive genotype VAX 1. PEG 6000 (>> PEG 1000) treatment greatly decreased Al accumulation in the 1 cm root apices even when the roots were physically separated from the PEG solution using dialysis membrane tubes. Upon removal of PEG from the treatment solution, the root tips recovered from osmotic stress and the Al accumulation capacity was quickly restored. The PEG-induced reduction of Al accumulation was not due to a lower phytotoxic Al concentration in the treatment solution, reduced negativity of the root apoplast, or to enhanced citrate exudation. Also cell-wall (CW) material isolated from PEG-treated roots showed a low Al-binding capacity which, however, was restored after destroying the physical structure of the CW. The comparison of the Al(3+), La(3+), Sr(2+), and Rb(+) binding capacity of the intact root tips and the isolated CW revealed the specificity of the PEG 6000 effect for Al. This could be due to the higher hydrated ionic radius of Al(3+) compared with other cations (Al(3+) >> La(3+) > Sr(2+) > Rb(+)). In conclusion, the results provide circumstantial evidence that the osmotic stress-inhibited Al accumulation in root apices and thus reduced Al-induced inhibition of root elongation in the Al-sensitive genotype VAX 1 is related to the alteration of CW porosity resulting from PEG 6000-induced dehydration of the root apoplast.
