Plant extracts are some of the most attractive sources of new drugs and have shown promising results for the treatment of gastric ulcers. Several folk medicinal plants and herbs have been used to treat gastrointestinal disorders, including gastric ulcers. Mammea americana L. (Guttiferae) fruit is very common in the diet of the population of northern South America. Our research interest in this plant arose because of its potential medicinal value as a tonic and against stomachache, as used in folk medicine. In this paper we evaluated three different extracts (ethanolic/EtOH, methanolic/MeOH and dichloromethane/DCM) obtained from M. americana L., for their ability to protect the gastric mucosa against injuries caused by necrotizing agents (0.3 M HCI/60% EtOH), hypothermic restraint stress, nonsteroidal anti-inflammatory drugs (NSAID, indomethacin) and pylorus ligation. In the HCI/EtOH-induced gastriculcer model, EtOH and DCM extracts demonstrated significant inhibition of the ulcerative lesion index by 54% (12.0 +/- 2.6 mm) and 86% (3.7 +/- 1.8 mm), respectively, in relation to the control value (26.0 +/- 1.4 mm) (p < 0.000 1). In the NSAID/cholinomimetic-induced lesion model, both EtOH and DCM extracts showed antiulcerogenic effects with significant reduction in the damage to these gastric lesions of 36% (8.3 +/- 2.0 mm) and 42% (7.5 +/- 1.4 mm), respectively, as compared to the control group (13.0 +/- 0.9 mm) (p < 0.0001). In the gastric ulcer induced by hypothermic-restraint stress, both extracts also showed significant activity, and inhibited the gastric lesion index by 58% and 75%, respectively. The EtOH and DCM extracts also changed gastric juice parameters as well as those of cimetidine, decreased gastric acid secretion significantly (p < 0.0001), increased pH values and promoted reduced acid output (p < 0.0001). In all gastric-ulcer-induced models, MeOH extract did not show any significant antiulcerogenic activity, nor did it change gastric-juice parameters (p > 0.05). The results suggest that EtOH and DCM extracts obtained from M. americana possess excellent antisecretory and/or gastrotective effect in all gastric ulcer models. These results suggest that the antiulcerogenic compound(s) present in M. americana may be clustered in the apolar fraction, which will be investigated by our group for the probable mechanisms of action. Several plants are used in folk medicine to treat gastrointestinal disorders. Mouriri pusa Gardn. (Melastomataceae) is a medicinal plant commonly used in the central region of Brazil against gastric ulcer. Two organic extracts methanolic (MeOH) and dichloromethane (DCM) obtained by sequential extraction from the leaves of Mouriri pusa were evaluated for their ability to protect the gastric mucosa against injuries caused by necrotizing agents (0.3 M HCl/60% EtOH, absolute ethanol, non-steroidal anti-inflammatory drug, stress and pylorus ligature) in mice and rats. The best results were obtained after pretreatment with MeOH extract whereas the DCM extract did not show the same significant antiulcerogenic activity. No acute toxicity was observed in animals treated with 5 g/kg, p.o. of MeOH extract. The mechanism involving the antiulcerogenic action of MeOH extract seemed to be related to NO generation and also suggested the effective participation of endogenous sulfhydryl group in the gastroprotective action. Phytochemical investigation of the MeOH extract of Mouriri pusa yielded tannins, flavonoids and (-)-epicatechin. The presence of these phenolic compounds probably would explain the antiulcerogenic effect of the polar extract of Mouriri pusa leaves. (c) 2006 Elsevier B.V.. All rights reserved.

[GRAPHICS]This work proposes a combined swelling-electron paramagnetic resonance (EPR) approach aiming at determining some unusual polymer solvation parameters relevant for chemical processes occurring inside beads. Batches of benzhydrylamine-resin (BHAR), a copolymer of styrene-1% divinylbenzene containing phenylmethylamine groups were, labeled with the paramagnetic amino acid 2,2,6,6-tetramethylpiperidine-1-oxyl-4-amine-4-carboxylic acid (TOAC), and their swelling properties and EPR spectra were examined in DCM and DMF. By taking into account the BHARs labeling degrees, the corresponding swelling values, and some polymer structural characteristics, it was possible to calculate polymer swelling parameters, among them, the volume and the number of sites per bead, site-site distances and site concentration. The latter values ranged from 17 to 170 angstrom and from 0.4 to 550 mM, respectively. EPR spectroscopy was applied to validate the multistep calculation strategy of these swelling parameters. Spin-spin interaction was detected in the labeled resins at site-site distances less than approximately 60 A or probe concentrations higher than approximately 1 x 10(-2) M, in close agreement with the values obtained for the spin probe free in solution. Complementarily, the yield of coupling reactions in different resins indicated that the greater the inter-site distance or the lower the site concentration, the faster the reaction. The results suggested that the model and the experimental measurements developed for the determination of solvation parameters represent a relevant step forward for the deeper understanding and improvement of polymer-related processes.

An optimization study of the reaction conditions of Fe(TDCPP)Cl when it is used as catalyst in the hydroxylation of cyclohexane by iodosylbenzene (PhIO) has been carried out, It was found that Fe(TDCPP)Cl follows the classical PhIO mechanism described for Fe(TPP)Cl, which involves the monomeric active species Fe-IV(O)P-+. (I). In the optimized condition ([Fe(TDCPP) = 3.0 X 10(-4) mol l(-1) in 1,2-dichloroethane (DCE); ultrasound stirring at 0 degrees C; PhIO/FeP molar ratio = 100), this FeP led to a yield of cyclohexanol (C-ol) of 96% and a turnover number of 96, Therefore, Fe(TDCPP)Cl may be considered a good biomimetic model and a very stable, resistant and selective catalyst, which yields C-ol as the sole product. DCE showed to be a better solvent than dichloromethane (DCM), 1 DCE:1 MeOH mixture or acetonitrile (ACN). Since the Fe-IV(O)P-+. is capable of abstracting hydrogen atom from DCM, MeOH or ACN, the solvent competes with the substrate. Presence of O-2 lowers the yield of C-ol, as it can further oxidize this alcohol to carboxylic acid in the presence of radicals, Presence of H2O also causes a decrease in the yield, since it converts the active species I into Fe-IV(OH)P, which cannot oxidize cyclohexane. Addition of excess imidazole or OH- to the system results in a decrease in the yield of C-ol, due to the formation of the hexacoordinated complexes Fe(TDCPP)Im(2)(+) (low-spin, beta(2) = 2.5 X 10(8) mol(-2) l(2)) and Fe(TDCPP)(OH)(2)(-) (high-spin, beta(2) = 6.3 X 10(7) mol(-2) l(2)), the formation of both Fe(TDCPP)Im(2)(+) and Fe(TDCPP)(OH)(2)(-) complexes were confirmed by EPR studies. The catalytic activities of Fe(TDCPP)C and Fe(TFPP)Cl were compared, the unusually high yields of C-ol with Fe(TFPP)Cl obtained when ultrasound, DCM and O-2 atmosphere were used, suggest that a parallel mechanism involving the mu-oxo dimer form, O-2 and radicals may also be occurring with this FeP, besides the PhIO mechanism.

Electrically Detected Magnetic Resonance (EDMR) was used to investigate the influence of dye doping molecules on spin-dependent exciton formation in Aluminum (III) 8-hydroxyquinoline (Alq(3)) based OLEDs with different device structures and temperature ranges. 4-(dicyanomethylene)-2-methyl-6-{2-[(4-diphenylamino-phenyl]ethyl}-4H-pyran (DCM-TPA) and 5,6,11,12-tetraphenylnaphthacene (Rubrene) were used as dopants. A strong temperature dependence have been observed for doped OLEDs, with a decrease of two orders of magnitude in EDMR signal for temperatures above similar to 200 K. The signal temperature dependence were fitted supposing different spin-lattice relaxation processes. The results suggest that thermally activated vibrations of dopants molecules induce spin pair dissociation, reducing the signal.

Objective - To evaluate plication of the free wall of the left ventricle, which reduces the left ventricular area and volume, as a method to improve the left ventricular systolic function without cardiopulmonary bypass. Animals - 8 mixed-breed adult dogs. Procedure - Dilated cardiomyopathy (DCM) was induced in each dog by administration of doxorubicin (30 mg/m2, IV, q 21 d for 168 days). Two dogs died during induction of cardiomyopathy. Plication surgery was performed in 4 dogs. Two dogs did not ondergo to surgery (control group). Values for cardiac output (CO), 2-dimensional and M-mode echocardiography, arterial blood pressure, electrocardiography, blood cell counts, and serum biochemical analyses were recorded after induction of DCM (baseline) and 1, 2, 7, 15, 21, 30, 60, 90, 120, 150, and 180 days after plication surgery. Ambulatory ECG (Holter) recordings were conducted for 24 hours on the day of surgery. Results - 1 dog died after plication surgery. The remaining dogs undergoing ventricular plication had a significant improvement in CO, ejection fraction, and fractional shortening and reductions of left ventricular area and volume after surgery. Electrocardiographic and Holter recordings revealed premature ventricular complexes, which resolved without treatment during the first week after surgery. Clinical condition of the control dogs declined, and these 2 dogs died approximately 40 days after induction of cardiomyopathy. Conclusions and Clinical Relevance - Plication of the free wall of the left ventricle improved left ventricular systolic function in dogs with doxorubicin-induced cardiomyopathy. Additional studies are needed to evaluate its application in dogs with naturally developing DCM.

This paper proposes a bridgeless boost interleaved PFC (power factor correction) converter with variable duty cycle control. The application of bridgeless technique causes reduction of conduction losses, while the interleaving technique of converters cells allows division of efforts in semiconductor devices and reduction of weight and volume of the input EMI filter. The use of variable duty cycle control has the functions of regulating the output voltage and eliminating the low order harmonic components that appears in the input current of the common interleaved power factor converters working in Discontinuous Conduction Mode (DCM). The simulation results of the proposed converter presented high power factor and a good transient response in relation to the output voltage regulation in presence of high load variations and supply voltage variations. © 2011 IEEE.

This paper presents theoretical evaluation and experimental results to the proposed bridgeless interleaved boost PFC (power factor correction) converter. The application of bridgeless technique causes reduction of conduction losses, while the interleaving technique of the converter cells allows division of the current stress in semiconductor devices and reduction of weight and volume of the input EMI filter. In each cell of the converter, the inductor current operates in discontinuous conduction mode (DCM), which eliminates turn-on switching losses and the effects of reverse recovery in semiconductors, increasing the efficiency of the converter. The experimental results show the power factor of 0.96 for employed voltage ratio and an efficiency of 95.2 % for nominal load conditions. © 2012 IEEE.

Dilated cardiomyopathy (DCM) is characterized by chamber dilation and cardiac dysfunction. Because of the poor prognosis, models are needed for the investigation of and development of new therapeutic approaches, as well as stem cell therapy. Doxorubicin (DOX), used as chemotherapeutic agent, is reported to be cumulative cardiotoxic causing DCM. The aim of the study was to investigate the onset of systolic dysfunction using echocardiography in rabbits receiving two different doses of DOX (1. mg/kg twice a week and 2. mg/kg once a week). Twenty rabbits were treated with doxorubicin in two different doses for 6. weeks and compared with a control group treated with NaCl 0.9%. The effect of doxorubicin on the myocardium was investigated with histological analysis and scanning electron microscopy of left ventricle (LV), as well as in the interventricular septum (IVS) and right ventricle (RV). The results showed a high mortality rate for rabbits receiving 2. mg/kg once a week. A significant reduction in systolic function was present in animals treated with DOX after 6. weeks, with decreased ejection fraction and shortening fraction. Histology and electron microscopy revealed vacuolization, intracytoplasmic granulation, necrosis and interstitial fibrosis in LV, as well as in the IVS and RV. Doxorubicin induced changes are present in the LV, RV and IVS, and the administration at the dose of 1. mg/kg twice a week for only 6. weeks is safe and sufficient to induce DCM in rabbits. © 2012 Elsevier Ltd.

Parahancornia fasciculata (Poir.) Benoist (Apocynaceae), também conhecida como Parahancornia amapa (Hub.) Ducke, é uma espécie vegetal empregada popularmente no tratamento da malária, infecções no útero, gastrite, anemia, problemas respiratórios, entre outros. Os objetivos do presente trabalho foram realizar o estudo fitoquímico, avaliar a toxicidade oral aguda e a atividade antimalárica in vitro e in vivo de extratos, frações e substância isolada obtidas a partir de cascas do caule de P. fasciculata. Foram realizados dois tipos de extrações com o pó das cascas de P. fasciculata, por maceração / percolação, com etanol 96°GL e diclorometano, esta última tendo sido realizada a com o pó das cascas alcalinizado com hidróxido de amônio, obtendo-se os extratos secos EEPF e EDAPF, respectivamente. Uma terceira extração foi realizada a partir do EEPF por aquecimento sob refluxo, sucessivamente, com Hex:DCM (1:1), AcOEt:DCM (1:1) e AcOEt. EEPF foi, também, submetido a fracionamento por extrações ácido-base resultando nas frações de neutros (EEPFN) e de alcalóides (EEPFA). A prospecção fitoquímica realizada com o EEPF foi desenvolvida por CCD em cromatoplacas de sílica gel tendo sido detectada a presença de triterpenos, esteróides, heterosídeos flavônicos, saponinas, polifenóis, taninos, heterosídeos antracênicos e heterosídeos cardiotônicos. EDAPF foi submetido à cromatografia em coluna de sílica gel. Foram recolhidas 30 frações sendo que as frações Fr1-3, Fr4, Fr5-7 e Fr11 concentraram a maior parte da massa do extrato cromatografado. Da Fr5-7 foi isolada uma mistura de ésteres do lupeol que representam os componentes majoritários do EDAPF. Esta fração passou por um processo de hidrólise alcalina e o produto obtido (Fr5-7Hid) foi analisado por espectrometrias no IV, RMN de 1H e 13C e foi identificado como o triterpeno lupeol. A fração insolúvel em AcOEt obtida a partir do EEPF, por aquecimento sob refluxo, apresentou resultado positivo para o teste de proantocianidinas e foi submetido a doseamento desta classe de metabólitos. Os resultados foram expressos em porcentagem dos teores para a amostra não diluída (10,46±0,3419%), amostra diluída a 1:10 (9,94± 0,1598%) e amostra diluída a 1:100 (10,55± 0,9299%). A avaliação da atividade antiplasmódica in vitro em culturas de cepas W2 de Plasmodium falciparum foi realizada pelo teste da Proteína II Rica em Histidina (HRP-II) tendo sido testados EEPF, EEPFN, EEPFA, Fr1-3, Fr4, Fr5-7(ésteres do lupeol), Fr11 e o Fr5-7Hid (lupeol). Os melhores resultados obtidos foram para EEPF, EEPFA E EEPFN (CI50= ~ 50 μg/mL) sendo considerados moderadamente ativos. As demais amostras apresentaram CI50 > 50 μg/mL e foram consideradas inativas. Realizou-se também a avaliação da atividade antimalárica in vivo em camundongos fêmeas suíços infectados com cepas ANKA de P. berghei com o EEPF e o EEPF-HEX:DCM (1:1) em concentrações de 500, 250 e 125mg/kg de peso. EEPF foi parcialmente ativo, somente no 8° dia, em todas as concentrações. Já EEPF-HEX:DCM (1:1) foi parcialmente ativo na dose de 500mg/kg de peso e nas demais doses foi inativo. O teste de toxicidade oral aguda foi realizado em camundongos fêmeas suíços, pelo método da dose fixa (5.000mg/kg), com EEPF e não apresentou nenhum sinal de toxicidade evidente, o que foi confirmado pela ausência de alterações nos exames anátomohistopatológicos realizados.

O presente trabalho descreve a análise fitoquímica e avaliação da atividade antiplasmódica em Plasmodium falciparum (W2) de Himatanthus articulatus (Vahl) Woodson. O extrato etanólico (EEHS) obtido por percolação do pó das cascas, após concentração, forneceu um precipitado (EEHSP) e um resíduo pastoso que foi submetido a liofilização (EEHS). Fracionou-se este por: re-extração sob refluxo, partição ácido-base e coluna cromatográfica de sílica gel. Além disso, submeteu-se o pó das cascas a extração com ácido clorídrico 1 N para separação de alcalóides. A prospecção fitoquímica, em CCD, foi realizada com EEHS e EEHSP, enquanto que EEHS, frações (FrDCM EEHS, FrAcOET EEHS e FrMeOH EEHS) e FAHS2 DCM foram analisadas por cromatografia líquida de alta eficiência acoplada a DAD (arranjo de diiodo-CLAE-DAD). FrAcOET EEHS foi fracioanda por coluna cromatográfica fornecendo uma substância isolada (S1). Analisou-se o EEHS, a fração majoritária obtida na coluna cromatográfica (F71), FAHS2 DCM e S1 por cromatografia liquida acoplada a espectrometria de massas (CL-EM). O espectro no infravermelho foi obtido para F71 e S1. A estrutura química de S1 foi definida como sendo o plumierídeo por ressonância magnética nuclear. A avaliação de atividade antiplasmódica foi realizada com EEHS, EEHSP, FrDCM EEHS, FrAcOET EEHS, FrMeOH EEHS, FAHS1 DCM, FAHS2 DCM, FAEEHS, FNEEHS, FNHS DCM e S1 pelo método da lactato desidrogenase parasitária (pLDH). Na prospecção fitoquímica, EEHS e EEHSP apresentaram resultados positivos para polifenóis e taninos, saponinas, triterpenos e esteroides, alcaloides e geninas flavônicas. EEHS e frações analisadas por CLAE-DAD mostraram os picos mais intensos sugestivos de iridoides, comprovados por CL-EM de EEHS que mostrou como substância majoritária o iridóide plumierídeo, coerente com o espectro no IV de F71 e S1 que revelou absorções de grupos funcionais presentes em iridóides. Nas análises destas por CL-EM, observou-se um pico a m/z 471 (M+H), atribuído ao íon pseudomolecular do plumierídeo e/ou isoplumierídeo. S1 foi identificada como plumierídeo. Até o presente momento não foi relatada a presença de alcaloides para a espécie, porém nas frações alcaloídicas (FAHS1 DCM, FAHS2 DCM, FAEEHS) e de neutros (FNEEHS e FNHS DCM) em análise por CCD revelada com reagente Dragendorff observaram-se manchas sugestivas de alcalóides, coerente com análise por CLAE-DAD de FAHS2 DCM, a qual mostrou pico majoritário com cromóforo sugestivo de alcaloide β-carbolínico. Em análises por CL-EM de FAHS2 DCM observou-se pico majoritário sugestivo do alcalóide 10-hidroxi-antirina-N-óxido com massa molecular 328 u. Os ensaios de atividade antiplasmódica foram negativos para EEHS, EEHSP, FrDCM EEHS, FrAcOET EEHS, FrMeOH EEHS, S1, FAEEHS, FAHS1 DCM, FNEEHS, FNHS DCM e moderadamente ativo (CI50 22,89 μg/mL) para FAHS2 DCM. Estes resultados indicam que a atividade antiplasmódica da planta pode se atribuída aos alcaloides, cuja presença em H. articulatus é descrita pela primeira vez.
