Immunohistochemical analysis of the p53 gene protein and cytometric assessment of nuclear DNA were performed in a series of 51 cases of intraductal breast proliferation. The series included 22 cases of intraductal hyperplasia without atypia, 6 cases of intraductal hyperplasia with atypia, and 23 cases of pure intraductal carcinoma. Expression of p53 protein was detected in one case of intraductal hyperplasia without atypia (4.5 per cent), one case of intraductal hyperplasia with atypia (16.6 per cent) and six cases of intraductal carcinoma (26.0 per cent). No significant correlation was observed between p53 expression and histological subtype of intraductal carcinoma. Aneuploidy was demonstrated in two cases of intraductal hyperplasia with atypia (33.3 per cent) and in 18 cases of intraductal carcinoma (78.2 per cent). All cases of intraductal hyperplasia without atypia were euploid. No significant association was observed between p53 protein expression and ploidy in intraductal hyperplasia. The only case of intraductal hyperplasia without atypia positive for p53 was euploid, whereas the only p53-positive case of intraductal hyperplasia with atypia was aneuploid. Among the intraductal carcinomas, only the aneuploid cases showed positivity for p53, regardless of histological subtype. The results suggest that some of the changes observed in invasive breast carcinoma, such as p53 expression and aneuploidy, are already present in breast intraductal proliferation, especially in areas with atypia and in intraductal carcinoma. The expression of p53 in breast intraductal proliferation may reflect the acquisition of p53 gene mutations in cells unable adequately to repair DNA damage, with genomic instability which would lead to clonal expansion and putative evolution to invasive disease.

Tendon composition changes according to differentiation, mechanical load, and aging. In this study, we attempted to identify, localize, and quantify type VI collagen in bovine tendons. Type VI collagen was identified by the electrophoretic behavior of the alpha chains and Western blotting, and by rotary shadowing. Type VI collagen was extracted from powdered tendon with three sequential 24-h extractions with 4 M guanidine-HCl. The amount of type VI collagen was determined by enzyme-linked immunosorbent assay for purely tensional areas and for the compressive fibrocartilage regions of the deep flexor tendon of the digits, for the corresponding fetal and calf tendons, and for the extensor digital tendon. The distal fibrocartilaginous region of the adult tendon was richer in type VI collagen than the tensional area, reaching as much as 3.3 mg/g (0.33%) of the wet weight. Calf tendons showed an accumulation of type VI at the fibrocartilage site. Immunocytochemistry demonstrated that type VI collagen was evenly distributed in the tensional areas of tendons but was highly concentrated around the fibrochondrocytes in the fibrocartilages. The results demonstrate that tendons are variable with regard to the presence and distribution of type VI collagen. The early accumulation of type VI collagen in the region of calf tendon that will become fibrocartilage in the adult suggests that it is a good marker of fibrocartilage differentiation. Furthermore, the distribution of type VI collagen in tendon fibrocartilage indicates that it organizes the pericellular environment and may represent a survival factor for these cells.

1. Water intake induced by injection of 0.2 M-NaCl into the lateral preoptic area was increased by the injection of angiotensin II into the subfornical organ of rats. The injection of hypertonic saline solution into the subfornical organ increased water intake. However, the increase was lower than when the solution was injected into the lateral preoptic area. The injection of 4 μg angiotensin II into the lateral preoptic area further augmented this effect. 2. Injection of angiotensin II into the subfornical organ caused a rise in blood pressure which preceded the thirst-inducing effect. The injection of 0.2 M NaCl into the subfornical organ caused no changes in blood pressure, whereas the injection of angiotensin II into the lateral preoptic area caused some increase. 3. Dehydration of the lateral preoptic area by means of 0.2 M NaCl in combination with intravenous infusion of angiotensin II caused a summation of effects in terms of the water intake, without changing cardiovascular alterations induced by the infusion of angiotensin II. A summation of effects in the water intake, but not in blood pressure, was also observed when 0.5 M NaCl was infused intravenously in combination with the injection of angiotensin II into the subfornical organ and into the lateral preoptic area. 4. The results indicate that there are interactions between the subfornical organ and lateral preoptic area in the regulation of cardiovascular and thirst mechanisms.

In Brazil, the degradation of soil and landscape by urban and agricultural frontiers expansion leads to the need for comprehensive studies and consider the diverse biological activities generated from different interventions in the landscape, becoming an instrument for assessing the impacts and the decision for its environmental management. The objective of this study was to evaluate the influence of different forms of occupation of the landscape, considering ecological elements and their interactions. The work was carried out on the Instituto Agronômico in the county of Jundiai, in the state of Sao Paulo, Brazil. The area under study has been subjected to different use and occupancy for a period of about 40 years. During this period the landscape has been transformed, with the current scenario can be classified as a degraded area mining; grassy area; Araucaria forest and pasture. These areas were evaluated by means of a transect, from which ten sampling sites were selected for the description of diverse biological activities, which included: evaluation and description of ground cover, identifying the presence of fungus and insect species. Furthermore, we evaluated in these points the pH, fertility and porosity of the topsoil (0-0.10 m). The results showed a variation of the elements analyzed and a relationship between the use and occupation of land in the different scenarios of the current landscape. The biological activity was more diverse in the Araucaria forest, reflected by the abundance of litter, higher content of organic matter and soil nutrients, demonstrating the effectiveness of the technique for assessing the level of degradation of the landscape used, which is expeditious and inexpensive.

This study is aimed at evaluating the effect of placing the top soil cover areas, such as tailings degraded by tin mining, in the Amazon rainforest in Brazil. The evaluations of the planting sites occurred in areas where tin mining was carried out, basically planting native trees over a period of seven years. This work did not come from a pre-decreed methodology of experimental design, and data was collected only seven years after planting. Thus, it was not possible to identify all variables that contributed to a better recovery of the areas. Sampling was done about seven years after placing the top soil and is determined: pH, organic matter content, P, K, Ca, Mg, Al, cation exchange capacity (T), base saturation (V%), B, Fe, Zn, Cu and Mn. The native forest species existing at the site were evaluated in relation to height (in meters) and diameter of the base. For all sites where the surface layer of soil was applied, there were significant differences in the growth of native species. A fundamental aspect in the rehabilitation of areas degraded by mining, in general, is the knowledge about the soil where that recovery must be conducted. The specific procedures in the rehabilitation of those areas depend essentially on the physical, chemical, biological and mineralogical properties of the soil, which must present conditions for the adequate development of the plants. The initial idea of implanting a project of recovery of soils degraded by mining in the Amazon Forest emerged from a first visit to the field, carried out in 1998. The conditions of the already mined areas, in comparison to the exuberant forest of the surroundings caught our attention. The mining company that acts in the area had already been trying for some time to implant a plan of recovery of these mining areas, however without reaching any significant positive results. The loss of organic matter is one of the main problems of degraded areas in Brazil. The storage and reuse of a blanket of soil (topsoil) produce excellent results, but most of the miners consider this technique expensive and difficult because of operational costs and the sharp topographical condition of the mine site. Therefore, a research project was elaborated prioritizing the recovery of the soil degraded by the tin mining as a prior step to the recovery activities with native forest species. The formation of a superficial pseudo-horizon that supported the vegetation and the time that it would take for its establishment became the main objective of this research. The objective of this work is to verify the levels of elements and their traces in areas where top soils were applied for the remediation of degraded areas with local re-vegetation. © 2011 WIT Press.

Predicting and mapping productivity areas allows crop producers to improve their planning of agricultural activities. The primary aims of this work were the identification and mapping of specific management areas allowing coffee bean quality to be predicted from soil attributes and their relationships to relief. The study area was located in the Southeast of the Minas Gerais state, Brazil. A grid containing a total of 145 uniformly spaced nodes 50 m apart was established over an area of 31. 7 ha from which samples were collected at depths of 0. 00-0. 20 m in order to determine physical and chemical attributes of the soil. These data were analysed in conjunction with plant attributes including production, proportion of beans retained by different sieves and drink quality. The results of principal component analysis (PCA) in combination with geostatistical data showed the attributes clay content and available iron to be the best choices for identifying four crop production environments. Environment A, which exhibited high clay and available iron contents, and low pH and base saturation, was that providing the highest yield (30. 4l ha-1) and best coffee beverage quality (61 sacks ha-1). Based on the results, we believe that multivariate analysis, geostatistics and the soil-relief relationships contained in the digital elevation model (DEM) can be effectively used in combination for the hybrid mapping of areas of varying suitability for coffee production. © 2012 Springer Science+Business Media New York.

Este estudo tem como tema de pesquisa a Educação do campo no Brasil. Ele resulta de nosso envolvimento político e profissional com a educação. Objetivamos com ele compreender a participação dos movimentos sociais organizados na Articulação Nacional por Uma Educação do Campo na elaboração das Diretrizes Operacionais para a Educação Básica nas Escolas do Campo (DOEBEC), por meio da análise do conteúdo das referidas Diretrizes e das Resoluções da I Conferência Nacional por Uma Educação Básica do Campo. Para tanto, levantamos duas questões gerais, que deram origem a outras específicas. As questões gerais são: Qual a contribuição dos movimentos sociais organizados na Articulação Nacional por Uma Educação do Campo na elaboração das Diretrizes Operacionais para a Educação Básica nas Escolas do Campo (DOEBEC)? Das Resoluções aprovadas na I Conferência Nacional por Uma Educação Básica do Campo, quais os princípios que se destacam no sentido de contribuir para a superação das desigualdades e melhoria da qualidade de vida e da educação dos sujeitos do campo? Metodologicamente realizamos análise de conteúdo do corpus, composto de dois grupos de documentos: as DOEBEC e as Resoluções da I Conferência Nacional por Uma Educação Básica do Campo. O aporte teórico eleito está composto de vários autores que pesquisam sobre educação do campo no Brasil, dos quais destacamos: Salomão Hage, Mônica Molina, Roseli Caldart e Miguel Arroyo. No campo da compreensão da sociedade e do Estado, também trabalhamos com vários autores, dentre os quais destacamos especialmente Boaventura de Sousa Santos. A análise ocorreu em três momentos: identificação dos eixos analíticos; escolha de indicadores de qualidade e articulação entre os eixos e indicadores de qualidade que geraram categorias de análise. Como resultado, identificamos que as categorias estão interligadas e expressam ações e práticas indispensáveis para a implementação da política de educação do campo na concepção proposta pela Articulação Nacional por uma Educação do Campo, que, de fato, influenciou na elaboração das DOEBEC no que se refere à democratização da gestão, no reconhecimento das especificidades da escola do campo e da necessidade de uma proposta político-pedagógica referenciada em um projeto de desenvolvimento sustentável e nos saberes e realidade dos sujeitos do campo. Entretanto, esta contribuição foi limitada por condicionantes institucionais ligados à legislação educacional vigente, marcada pela concepção neoliberal, e, de forma mais ampla, pela ordem estrutural econômica, social e política do capitalismo na sociedade brasileira, uma vez que as condições concretas para a efetivação de uma educação do campo, estão para além das DOEBC.

O solo desempenha importante papel no ciclo do C, porém a substituição da floresta tropical por áreas cultivadas altera a dinâmica e o estoque desse elemento. Em uma frente pioneira de colonização no município de Itupiranga (PA), na Amazônia Oriental, foi desenvolvido este estudo com o objetivo de avaliar as consequências da substituição de floresta nativa por pastagens de Brachiaria brizantha no conteúdo de C de um Latossolo Amarelo distrófico. As amostras de solo foram coletadas em área de floresta nativa (FN), floresta secundária de 8–10 anos (FS), pastagens de 1–2 anos (P1-2), de 5–7 anos (P5–7) e de 10–12 anos (P10–12), nas camadas de 0–2, 2–5 e 5–10 cm, para avaliar os teores e o estoque de C e realizar um fracionamento granulométrico da matéria orgânica. Após o desmatamento, a densidade do solo aumentou até a profundidade de 5 cm, sendo esse aumento maior nas pastagens mais antigas. As maiores mudanças no conteúdo de C ocorreram na camada superior do solo, havendo aumento nesse conteúdo com o tempo de implantação das pastagens. Nas camadas de 2–5 e 5–10 cm, o conteúdo de C se mostrou estável entre os tipos de cobertura vegetal avaliados. As maiores concentrações de C foram encontradas na fração silte, mas os maiores conteúdos de C ocorreram na fração argila, independentemente do tipo de cobertura vegetal. Um aumento da quantidade de C associado à fração areia, na forma de resíduos orgânicos pouco decompostos, foi observado nas pastagens, confirmando a maior sensibilidade dessa fração às mudanças de uso do solo.

Regeneration microsites are characterized by diverse combinations of attributes which assure the best conditions for seed germination and seedling establishment. By understanding these attributes, we can contribute to determining better management methodologies for reestablishing ecological process in sites under restoration. Thus, we sought to characterize and differentiate the micro-site conditions of restoration plantings to indentify likely physical-chemical limitations for the establishment of native tree species in the forest understory. This study was carried out in reforestation plantings with different ages (10, 22 and 55 years). The physical-chemical characterization of the micro-site of regeneration of the study areas was carried out by evaluating the soil compression level, porosity, humidity, organic matter and nutrients content and granulometry, as well as litter dry mass and canopy cover. An increase on the canopy cover and soil porosity, humidity, clay and organic matter content were observed in the oldest restored areas, as well as a decrease in soil compression. Thus, these findings demonstrated that the evaluated microsite properties are in process of restoration. Therefore, microsite conditions for seedling establishment become even more similar to reference ecosystems as restoration planting evolve.

A comparative study of the primary properties of six cocoa butter samples, representative of industrial blends and cocoa butter extracted from fruits cultivated in different geographical areas in Brazil is presented. The samples were evaluated according to fatty acid composition, triacylglycerol composition, regiospecific distribution, melting point, solid fat content and consistency. The results allowed for differentiating the samples according to their chemical compositions, thermal resistance properties, hardness characteristics, as well as technological adequacies and potential use in regions with tropical climates.
