Among the main etiologies of secondary arterial hypertension figure out the tumorous affections of adrenal gland, located on cortex - primary aldosteronism (Conn’s syndrome) and Cushing’s syndrome - or at glandular medulla - pheocromocytoma. Although these tumors are at most benign the surgical resection is needed in order to eliminate the disturbances provided by them and to limit the mass growth, being curative in about 80-90% of the cases. In this paper some particularities above surgical treatment of these diseases will be focused emphasizing the pre-operative prepare of the patients and the currently preconized approach.

The authors present two cases of Primary Hiperaldosteronism, caused by functioning adenoma of the adrenal's cortex. The two females patients presented classic symptoms of the Conn's disease, especially hypocalemia and hypertension. Both were operated and unilateral adrenalectomy was done, with excelent outcome in one and satisfactory in the other one. It is discussed many aspects related to the incidence, the problems with the diagnosis, indication and surgery treatment.

Chaperone members of the protein disulfide isomerase family can catalyze the thiol-disulfide exchange reaction with pairs of cysteines. There are 14 protein disulfide isomerase family members, but the ability to catalyze a thiol disulfide exchange reaction has not been demonstrated for all of them. Human endoplasmic reticulum protein chaperone thio-oxidoreductase (ERp18) shows partial oxidative activity as a protein disulfide isomerase. The aim of the present study was to evaluate the participation of ERp18 in gonadotropin-releasing hormone receptor (GnRHR) expression at the plasma membrane. Cos-7 cells were cultured, plated, and transfected with 25 ng (unless indicated) wild-type human GnRHR (hGnRHR) or mutant GnRHR (Cys14Ala and Cys200Ala) and pcDNA3.1 without insert (empty vector) or ERp18 cDNA (75 ng/well), pre-loaded for 18 h with 1 µCi myo-[2-3H(N)]-inositol in 0.25 mL DMEM and treated for 2 h with buserelin. We observed a decrease in maximal inositol phosphate (IP) production in response to buserelin in the cells co-transfected with hGnRHR, and a decrease from 20 to 75 ng of ERp18 compared with cells co-transfected with hGnRHR and empty vector. The decrease in maximal IP was proportional to the amount of ERp18 DNA over the range examined. Mutants (Cys14Ala and Cys200Ala) that could not form the Cys14-Cys200 bridge essential for plasma membrane routing of the hGnRHR did not modify maximal IP production when they were co-transfected with ERp18. These results suggest that ERp18 has a reduction role on disulfide bonds in wild-type hGnRHR folding.

Este trabajo se soporta sobre una base conformada por dos conceptos, que constituyen el horizonte contextual de la investigación: Derecho Administrativo y Comunidad de integración. Es claro que al hablar de una Comunidad de integración específica como la Comunidad Andina (CAN), incide de forma directa en la actividad del derecho administrativo.Ha sido necesario que los países que conforman los grupos de integración económica cedan parte de sus competencias para crear un ordenamiento jurídico de orden supranacional, el cual modifica la dinámica social de las relaciones comerciales y afecta la estructura de la administración pública, haciendo que el derecho administrativo no se limite a interactuar solo con derecho nacional. Esta mutua dependencia de las políticas internas y las regionales se realiza, entonces, cada vez que existe participación y compromiso de los Estados frente a las decisiones, de carácter vinculante o no. En esa medida surge el problema de investigación tendiente a esclarecer cómo ha sido la adopción de las decisiones e interpretaciones, emanadas de los mecanismos oficiales del Derecho Comunitario Andino, en el ordenamiento jurídico colombiano. La existencia de varios puntos de vista con respecto a la obligatoriedad o no de dicha adopción, moldea un tema de suficiente amplitud y profundidad para ser estudiado en un trabajo de investigación.

Esta investigación se centra en la Fédération Internationale de Football Association (FIFA) como organización política. Intenta responder dos interrogantes primordiales: 1) ¿cómo la FIFA ha constituido el poder que tiene actualmente y, así, hacerse del monopolio indiscutido del fútbol? Y 2) ¿cómo ha cambiado en el tiempo la política interna de FIFA y su vínculo con la política internacional? Para lograr esto, se realiza un estudio histórico, basado principalmente en documentos, que intenta caracterizar y analizar los cambios de la organización en el tiempo. Se enfatizan las últimas dos presidencias de FIFA, de João Havelange y Joseph Blatter, como casos de estudio.

In the past two decades, the geometric pathways involved in the transformations between inverse bicontinuous cubic phases in amphiphilic systems have been extensively theoretically modeled. However, little experimental data exists on the cubic-cubic transformation in pure lipid systems. We have used pressure-jump time-resolved X-ray diffraction to investigate the transition between the gyroid Q(II)(G) and double-diamond Q(II)(D) phases in mixtures of 1-monoolein in 30 wt% water. We find for this system that the cubic-cubic transition occurs without any detectable intermediate structures. In addition, we have determined the kinetics of the transition, in both the forward and reverse directions, as a function of pressure-jump amplitude, temperature, and water content. A recently developed model allows (at least in principle) the calculation of the activation energy for lipid phase transitions from such data. The analysis is applicable only if kinetic reproducibility is achieved, at least within one sample, and achievement of such kinetic reproducibility is shown here, by carrying out prolonged pressure-cycling. The rate of transformation shows clear and consistent trends with pressure-jump amplitude, temperature, and water content, all of which are shown to be in agreement with the effect of the shift in the position of the cubic-cubic phase boundary following a change in the thermodynamic parameters.

In this paper, we give an overview of our studies by static and time-resolved X-ray diffraction of inverse cubic phases and phase transitions in lipids. In 1, we briefly discuss the lyotropic phase behaviour of lipids, focusing attention on non-lamellar structures, and their geometric/topological relationship to fusion processes in lipid membranes. Possible pathways for transitions between different cubic phases are also outlined. In 2, we discuss the effects of hydrostatic pressure on lipid membranes and lipid phase transitions, and describe how the parameters required to predict the pressure dependence of lipid phase transition temperatures can be conveniently measured. We review some earlier results of inverse bicontinuous cubic phases from our laboratory, showing effects such as pressure-induced formation and swelling. In 3, we describe the technique of pressure-jump synchrotron X-ray diffraction. We present results that have been obtained from the lipid system 1:2 dilauroylphosphatidylcholine/lauric acid for cubic-inverse hexagonal, cubic-cubic and lamellar-cubic transitions. The rate of transition was found to increase with the amplitude of the pressure-jump and with increasing temperature. Evidence for intermediate structures occurring transiently during the transitions was also obtained. In 4, we describe an IDL-based 'AXCESS' software package being developed in our laboratory to permit batch processing and analysis of the large X-ray datasets produced by pressure-jump synchrotron experiments. In 5, we present some recent results on the fluid lamellar-Pn3m cubic phase transition of the single-chain lipid 1-monoelaidin, which we have studied both by pressure-jump and temperature-jump X-ray diffraction. Finally, in 6, we give a few indicators of future directions of this research. We anticipate that the most useful technical advance will be the development of pressure-jump apparatus on the microsecond time-scale, which will involve the use of a stack of piezoelectric pressure actuators. The pressure-jump technique is not restricted to lipid phase transitions, but can be used to study a wide range of soft matter transitions, ranging from protein unfolding and DNA unwinding and transitions, to phase transitions in thermotropic liquid crystals, surfactants and block copolymers.

We present a kinetic model for transformations between different self-assembled lipid structures. The model shows how data on the rates of phase transitions between mesophases of different geometries can be used to provide information on the mechanisms of the transformations and the transition states involved. This can be used, for example, to gain an insight into intermediate structures in cell membrane fission or fusion. In cases where the monolayer curvature changes on going from the initial to the final mesophase, we consider the phase transition to be driven primarily by the change in the relaxed curvature with pressure or temperature, which alters the relative curvature elastic energies of the two mesophase structures. Using this model, we have analyzed previously published kinetic data on the inter-conversion of inverse bicontinuous cubic phases in the 1-monoolein-30 wt% water system. The data are for a transition between QII(G) and QII(D) phases, and our analysis indicates that the transition state more closely resembles the QII(D) than the QII(G) phase. Using estimated values for the monolayer mean curvatures of the QII(G) and QII(D) phases of -0.123 nm(-1) and -0.133 nm(-1), respectively, gives values for the monolayer mean curvature of the transition state of between -0.131 nm(-1) and -0.132 nm(-1). Furthermore, we estimate that several thousand molecules undergo the phase transition cooperatively within one "cooperative unit", equivalent to 1-2 unit cells of QII(G) or 4-10 unit cells of QII(D).

A proteína ácida fibrilar glial (GFAP) é uma proteína da classe dos filamentos intermediários, exclusivamente expressa em astrócitos no sistema nervoso central (SNC). A função específica da fosforilação desta proteína é ainda desconhecida. No entanto, tem sido demonstrado que o equilíbrio dinâmico entre o estado fosforilado e desfosforilado de sítios específicos da GFAP pode regular a polimerização e despolimerização dos filamentos intermediários durante eventos de estruturação do citoesqueleto glial. Nosso grupo de pesquisa demonstrou que a fosforilação da GFAP em hipocampo de ratos jovens (P12-P16) é estimulada no mesmo nível por glutamato, via um receptor glutamatérgico metabotrópico do grupo II (mGluR II), e pela ausência de Ca2+ externo (presença de EGTA). Entretanto, o tratamento simultâneo com glutamato e EGTA não resulta em efeito sinergístico, sugerindo um mesmo mecanismo de ação para estas duas situações estimulatórias da fosforilação da GFAP (WofchuK & Rodnight, 1994; Kommers et al., 1999; Rodnight et al., 1997). Este mecanismo provavelmente não envolve reservas intracelulares de Ca2+ associadas a receptores de IP3, uma vez que mGluRs II estão envolvidos com o mecanismo de transdução de sinal via adenilato ciclase e não via hidrólise de fosfoinositídios. Uma hipótese proposta é de que o glutamato, via mGluR, bloqueia canais de Ca2+ tipo L, inibindo uma cascata de desfosforilação dependente de Ca2+, associada a GFAP (Rodnight et al., 1997). Interessantemente, os receptores rianodina (RyRs) presentes nas reservas intracelulares de Ca2+ reguladas por tais receptores estão associados com canais de Ca2+ tipo L (Chavis et al., 1996). Com base nestes dados, buscou-se neste trabalho avaliar se a modulação glutamatérgica da fosforilação da GFAP em fatias de hipocampo de ratos jovens envolve as reservas intracelulares de Ca2+ reguladas por RyRs e se o Ca2+ proveniente destas reservas atua de maneira semelhante ao Ca2+ oriundo do espaço extracelular. Nossos resultados mostraram que há uma evidente participação do Ca2+ proveniente das reservas intracelulares reguladas por RyRs no mecanismo modulatório da fosforilação da GFAP via ativação de mGluRs em fatias de hipocampo de ratos jovens, uma vez que a cafeína e a rianodina (agonistas de RyRs) revertem totalmente o efeito estimulatório do agonista glutamatérgico metabotrópico 1S,3R-ACPD sobre a fosforilação da proteína e este efeito da cafeína é inibido por dantrolene (antagonista de RyRs). Talvez o Ca2+ oriundo das reservas reguladas por RyRs tenha o mesmo papel do Ca2+ proveniente do espaço extracelular, ou seja, desencadeia uma cascata de desfosforilação associada à GFAP mediada pela calcineurina, uma vez que quelando o Ca2+ intracelular livre com BAPTA-AM, após a mobilização destas reservas, tal efeito não ocorre. A participação de receptores adenosina (AdoRs) e do AMP cíclico (AMPc) ainda permanece a ser estudada. Entretanto, é sabido que em ratos jovens a ativação de mGluRs aumenta a formação de AMPc potenciando o efeito de outros tipos de receptores, como os AdoRs e, provavelmente, isto é mediado por um mGluR II (Schoepp & Johnson, 1993; Winder & Conn, 1996). Neste trabalho mostrou-se justamente o possível envolvimento de tais mecanismos de transdução de sinal na modulação da fosforilação da GFAP, pois a adenosina deaminase (enzima que metaboliza adenosina endógena) e a forscolina (agente que estimula a enzima adenilato ciclase) alteraram o nível de fosforilação da GFAP. Estes resultados evidenciam o envolvimento das reservas intracelulares de Ca2+ reguladas por RyRs no mecanismo de transdução de sinal que modula o estado de fosforilação GFAP mediado pela ativação de mGluRs.

Relato de coletas de anofelinos realizadas em Belém, Pará, Brasil, de 1995 a 2004, comparando os dados obtidos com os de levantamentos anteriores, feitos a partir da década de 1930. Nesses, vinte espécies haviam sido identificadas: Anopheles albitarsis s.l., An. aquasalis, An. argyritarsis, An. braziliensis, An. darlingi, An. eiseni, An. evansae, An. galvaoi, An. intermedius, An. kompi, An. mediopunctatus, An. nimbus, An. nuneztovari, An. oswaldoi, An. peryassui, An. punctimacula, An. shannoni, An. strodei, An. thomasi e An. triannulatus. Sete (An. argyritarsis, An. eiseni, An. galvaoi, An. kompi, An. nimbus, An. punctimacula e An. thomasi) não são agora registradas. A permanência de tantas outras espécies provavelmente decorre da preservação de áreas de mata no âmbito urbano. Duas delas são consideradas de importância vetorial (An. darlingi e An. aquasalis). Esta última continua sendo a de maior densidade nas coletas (46,26% dos adultos e 99,21% das larvas) e é a única registrada em todos os distritos administrativos. Existe, portanto, risco potencial de transmissão de malária em todo o município.

The major Neotropical malaria vector, Anopheles darlingi, was reintroduced into the Iquitos, Loreto, Peru area during the early 1990s, where it displaced other anophelines and caused a major malaria epidemic. Since then, case numbers in Loreto have fluctuated, but annual increases have been reported since 2012. The population genetic structure of An. darlingi sampled before and after the introduction of long-lasting insecticidal nets (LLINs) was investigated to test the hypothesis of temporal population change (2006 vs. 2012). Current samples of An. darlingi were used to test the hypothesis of ecological adaptation to human modified (highway) compared with wild (riverine) habitat, linked to forest cover. In total, 693 An. darlingi from nine localities in Loreto, Peru area were genotyped using 13 microsatellite loci. To test the hypothesis of habitat differentiation in An. darlingi biting time patterns, HBR and EIR, four collections of An. darlingi from five localities (two riverine and three highway) were analysed. Analyses of microsatellite loci from seven (2006) and nine settlements (2012-2014) in the Iquitos area detected two distinctive populations with little overlap, although it is unclear whether this population replacement event is associated with LLIN distribution or climate. Within the 2012-2014 population two admixed subpopulations, A and B, were differentiated by habitat, with B significantly overrepresented in highway, and both in near-equal proportions in riverine. Both subpopulations had a signature of expansion and there was moderate genetic differentiation between them. Habitat and forest cover level had significant effects on HBR, such that Plasmodium transmission risk, as measured by EIR, in peridomestic riverine settlements was threefold higher than in peridomestic highway settlements. HBR was directly associated with available host biomass rather than forest cover. A population replacement event occurred between 2006 and 2012-2014, concurrently with LLIN distribution and a moderate El Niño event, and prior to an increase in malaria incidence. The likely drivers of this replacement cannot be determined with current data. The present-day An. darlingi population is composed of two highly admixed subpopulations, which appear to be in an early stage of differentiation, triggered by anthropogenic alterations to local habitat.
