The receptor tyrosine kinase (RTK)/phosphoinositide 3-kinase (PI3K) pathway is fundamental for cancer cell proliferation and is known to be frequently altered and activated in neoplasia, including embryonal tumors. Based on the high frequency of alterations, targeting components of the PI3K signaling pathway is considered to be a promising therapeutic approach for cancer treatment. Here, we have investigated the potential of targeting the axis of the insulin-like growth factor-1 receptor (IGF-1R) and PI3K signaling in two common cancers of childhood: neuroblastoma, the most common extracranial tumor in children and medulloblastoma, the most frequent malignant childhood brain tumor. By treating neuroblastoma and medulloblastoma cells with R1507, a specific humanized monoclonal antibody against the IGF-1R, we could observe cell line-specific responses and in some cases a strong decrease in cell proliferation. In contrast, targeting the PI3K p110α with the specific inhibitor PIK75 resulted in broad anti-proliferative effects in a panel of neuro- and medulloblastoma cell lines. Additionally, sensitization to commonly used chemotherapeutic agents occurred in neuroblastoma cells upon treatment with R1507 or PIK75. Furthermore, by studying the expression and phosphorylation state of IGF-1R/PI3K downstream signaling targets we found down-regulated signaling pathway activation. In addition, apoptosis occurred in embryonal tumor cells after treatment with PIK75 or R1507. Together, our studies demonstrate the potential of targeting the IGF-1R/PI3K signaling axis in embryonal tumors. Hopefully, this knowledge will contribute to the development of urgently required new targeted therapies for embryonal tumors.

Tämä insinöörityö tehtiin SEW Industrial Gears Oy:lle, joka valmistaa hammasvaihteita teollisuuden eri sovellutuksiin. Työn tavoitteena oli kehittää modulaarinen paisuntaastiasarja compact -sarjan teollisuusvaihteeseen. Työn alkuvaiheessa käytössä oli kahdenlaisia paisunta-astioita: kosteissa oloissa käytettävä suljettu astiamalli ja kuiviin olosuhteisiin soveltuva malli. Koska käytössä oleva suljettu astiamalli oli kallis ja kuivien olosuhteiden mallin käyttö oli rajoitettu, oli uudelle modulaariselle paisunta-astialle selkeä tarve. Uuden astiamallin tuli olla yhteensopiva vaihteen yleisimpien varusteiden ja asennusasentojen kanssa. Lisäksi tavoitteena oli puolittaa uuden konseptin hinta vanhaan verrattuna ja paikata vanhoissa konstruktioissa olleita puutteita. Työ aloitettiin analysoimalla olemassa olevat paisunta-astiatyypit, minkä jälkeen paisuntaastialle ideoitiin uusia ominaisuuksia. Toisessa vaiheessa perehdyttiin modulaariseen järjestelmään sekä öljynpaisunnan teoriaan teollisuusvaihteessa. Viimeisessä vaiheessa tehtiin konseptin varsinainen suunnittelutyö, valittiin astian valmistustapa ja materiaali sekä optimoitiin hinta. Uudesta mallista valmistettiin myös prototyyppi, jotta suunniteltua tuotetta päästiin kokeilemaan myös käytännössä. Näin voitiin varmistaa tuotteen toimivuus. Suunnittelutyön tuloksena saatiin uuden paisunta-astiasarjan piirustukset tarvittavista osista ja kokoonpanoista sekä komponenttistandardi ja myyntiä tukevaa materiaalia. Käytännön osuus antoi tietoa laskelmien oikeellisuudesta sekä öljyn täyttöön ja paisuntaan liittyvistä asioista.

Objective To analyze the methodological aspects used for the preparation of terminology subsets of the International Classification for Nursing Practice (ICNP®), in dissertations and theses in the Brazilian nursing. Method This is an integrative review of the Brazilian dissertations and theses defended in the period from 2007 to 2013, which were included seven dissertations. Results The increasing production of studies on the theme by Brazilian nurses shows a concern for a unified language for the profession. However, the results demonstrate the lack of uniformity in the conduct of studies, especially in relation to the stages of content validation. The initiatives of some authors to systematize alternative methods for creating these subsets also stood out. Conclusion We suggest the development of new terminology subsets, following standards of methodological rigor, as well as its application and validation by the selected clientele, to ensure greater reliability of results and desired changes for the profession.

We have analyzed the expression of T cell receptor (TcR) genes in the thymus using in situ RNA hybridizations with probes to the constant regions of the TcR alpha, beta, gamma and delta chains. Localization of transcripts revealed low TcR alpha mRNA levels in the thymus cortex and very low levels in the subcapsular region. In contrast, TcR beta message was very abundant in the cortex. TcR gamma or delta mRNA+ thymocytes showed a scattered, predominantly cortical localization. In contrast to gamma, TcR delta transcripts were abundant in the subcapsular region. Control experiments with sorted TcR alpha/beta or gamma/delta cells revealed a detection efficiency of 75%-85% for the respective TcR mRNA and data on TcR gene expression in mature, CD3+ thymocytes were consistent with previous reports. The analysis of immature, CD3- thymocyte subsets, however, revealed a virtual absence of TcR alpha transcripts and an unexpectedly high proportion of cells (14%-29%) expressing the gene for the TcR delta chain. The data are discussed in view of current models of lineage relationships in the thymus.

The choice network revenue management model incorporates customer purchase behavioras a function of the offered products, and is the appropriate model for airline and hotel networkrevenue management, dynamic sales of bundles, and dynamic assortment optimization.The optimization problem is a stochastic dynamic program and is intractable. A certainty-equivalencerelaxation of the dynamic program, called the choice deterministic linear program(CDLP) is usually used to generate dyamic controls. Recently, a compact linear programmingformulation of this linear program was given for the multi-segment multinomial-logit (MNL)model of customer choice with non-overlapping consideration sets. Our objective is to obtaina tighter bound than this formulation while retaining the appealing properties of a compactlinear programming representation. To this end, it is natural to consider the affine relaxationof the dynamic program. We first show that the affine relaxation is NP-complete even for asingle-segment MNL model. Nevertheless, by analyzing the affine relaxation we derive a newcompact linear program that approximates the dynamic programming value function betterthan CDLP, provably between the CDLP value and the affine relaxation, and often comingclose to the latter in our numerical experiments. When the segment consideration sets overlap,we show that some strong equalities called product cuts developed for the CDLP remain validfor our new formulation. Finally we perform extensive numerical comparisons on the variousbounds to evaluate their performance.

Asymptotic chi-squared test statistics for testing the equality ofmoment vectors are developed. The test statistics proposed aregeneralizedWald test statistics that specialize for different settings by inserting andappropriate asymptotic variance matrix of sample moments. Scaled teststatisticsare also considered for dealing with situations of non-iid sampling. Thespecializationwill be carried out for testing the equality of multinomial populations, andtheequality of variance and correlation matrices for both normal andnon-normaldata. When testing the equality of correlation matrices, a scaled versionofthe normal theory chi-squared statistic is proven to be an asymptoticallyexactchi-squared statistic in the case of elliptical data.

Na actual monografia intitulada “Sistemas de Tratamento e Disposição dos Resíduos Sólidos” adoptar-se-á as teorias clássicas de solos, para representar os resíduos sólidos urbanos. No que diz ao tratamento e disposição, diversos métodos e técnicas vão ser apresentados. Assim esta pesquisa tem como objectivo principal avaliar o Aterro Sanitário proposto para Santiago, 1999 e a tecnologia do Compact Power (pirólise/gasificação). Os dados dos resíduos sólidos ficaram restringidos aos resíduos urbanos, encontrando-se uma grande heterogeneidade e constantes questionamentos em diversas bibliografias. Analisa-se os elementos base para o dimensionamento do Aterro Sanitário abrangendo um estudo mais aprofundado sobre a estabilização dos taludes. Quanto ao Compact Power, far-se-á a apresentação da planta que incluiu duas linhas, uma para o resíduo misturado e o outro para resíduos separados, uma planta de compostagem e uma planta de conversão térmica avançada (empregando os processos de pirólises, gaseificação e oxidação a alta temperatura). Na compilação deste documento, encontra-se uma análise SWOT, com o intuito de melhor analisar as referidas tecnologias apresentadas.

The biological activity of interleukin (IL)-2 and other cytokines in vivo can be augmented by binding to certain anti-cytokine monoclonal antibodies (mAb). Here, we review evidence on how IL-2/anti-IL-2 mAb complexes can be used to cause selective stimulation and expansion of certain T-cell subsets. With some anti-IL-2 mAbs, injection of IL-2/mAb complexes leads to expansion of CD8 T effector cells but not CD4 T regulatory cells (Tregs); these complexes exert less adverse side effects than soluble IL-2 and display powerful antitumor activity. Other IL-2/mAb complexes have minimal effects on CD8 T cells but cause marked expansion of Tregs. Preconditioning mice with these complexes leads to permanent acceptance of MHC-disparate pancreatic islets in the absence of immunosuppression.

Certains pathogènes tels que le virus de l'hépatite C ainsi que le virus de l'immunodéficience humaine sont capables de détourner les mécanismes de défense du système immunitaire adaptatif afin d'établir des infections chroniques chez l'homme. La souche clone-13 du virus de la chorioméningite lymphocytaire est utilisée comme modèle d'études d'infection virale chronique chez la souris. Les raisons qui expliqueraient la persistance de certains virus ne sont pas encore bien définies. Toutefois, il a été montré qu'une exposition prolongée à un environnement inflammatoire ainsi que la présence prolongée de l'antigène sont des facteurs qui vont déclencher un procédé de différentiation particulier des lymphocytes T CD8+. Ces cellules sur-expriment alors des récepteurs inhibiteurs tels que PD-1 tandis que leur capacité à produire des cytokines diminue. Ce procédé de différentiation est appelé « exhaustion » et serait à l'origine de la génération de réponses lymphocytaires non ou peu fonctionnelles entraînant de ce fait la persistance de l'infection. Néanmoins, il a également été démontré que ces lymphocytes maintiennent des fonctions effectrices et qu'ils permettent de limiter la réplication du virus. Afin d'étudier la fonction effectrice résiduelle de ces lymphocytes T, nous avons transféré des cellules provenant de souris infectées chroniquement dans des souris receveuses naïves qui ont à leur tour été infectées avec le virus. Grâce à ces expériences, nous avons démontré que les cellules transférées contiennent des cellules qui sont capables de i) re-proliférer, ii) de protéger les souris contre une infection virale, et de iii) survivre en l'absence d'antigène. Nous avons remarqué que les cellules stimulées de façon chronique maintiennent le même phénotype lorsqu'elles sont transférées dans des souris naïves soumises à une infection virale aiguë. Nous avons de ce fait conclu que les cellules stimulées chroniquement contiennent une sous-population de cellules qui comporte des attributs de cellules T mémoire. D'autre part, nous avons pu identifier le facteur de transcription Tcf-1 comme l'élément essential pour la génération des cellules T ressemblant à des cellules mémoires. D'autre part, nous avons étudié l'impact du niveau de stimulation via le récepteur des cellules T (TCR) sur le phénotype adopté par les lymphocytes T au cours d'une infection chronique. Dans ce but, nous avons généré des souches de virus recombinants qui expriment un épitope modifié de manière à réduire le niveau de stimulation via le TCR. D'autre part, nous avons utilisé un mélange de deux souches virales de manière à moduler spécifiquement la quantité d'un épitope tout en conservant la même charge virale. Nous avons montré que la quantité d'antigène avait plus d'influence sur le phénotype des lymphocytes T que la force d'interaction entre le complexe peptide-CMH et le TCR. De plus, l'apparition de ce phénotype ne semble pas avoir d'impact sur la prolifération des cellules en réponse à une infection primaire ou secondaire. Ainsi, nous proposons un modèle par lequel le procédé d'« exhaustion » des cellules T correspond à une différentiation cellulaire particulière qui est indépendante de la capacité de prolifération des cellules. De manière générale, ces découvertes apportent de nouvelles connaissances sur les sous-catégories de lymphocytes T CD8+ qui sont générés pendant une infection virale chronique. Nous pensons que la réponse effectrice du système immunitaire est maintenue pendant de longues périodes grâce à la présence de cellules par partagent certaines caractéristiques avec des cellules mémoires. L'étude approfondie de ces cellules peut avoir des implications importantes sur l'optimisation des stratégies utilisant l'immunothérapie pour combattre les infections chroniques et cancers.

A frequency-dependent compact model for inductors in high ohmic substrates, which is based on an energy point-of-view, is developed. This approach enables the description of the most important coupling phenomena that take place inside the device. Magnetically induced losses are quite accurately calculated and coupling between electric and magnetic fields is given by means of a delay constant. The later coupling phenomenon provides a modified procedure for the computation of the fringing capacitance value, when the self-resonance frequency of the inductor is used as a fitting parameter. The model takes into account the width of every metal strip and the pitch between strips. This enables the description of optimized layout inductors. Data from experiments and electromagnetic simulators are presented to test the accuracy of the model.

Whereas numerical modeling using finite-element methods (FEM) can provide transient temperature distribution in the component with enough accuracy, it is of the most importance the development of compact dynamic thermal models that can be used for electrothermal simulation. While in most cases single power sources are considered, here we focus on the simultaneous presence of multiple sources. The thermal model will be in the form of a thermal impedance matrix containing the thermal impedance transfer functions between two arbitrary ports. Eachindividual transfer function element ( ) is obtained from the analysis of the thermal temperature transient at node ¿ ¿ after a power step at node ¿ .¿ Different options for multiexponential transient analysis are detailed and compared. Among the options explored, small thermal models can be obtained by constrained nonlinear least squares (NLSQ) methods if the order is selected properly using validation signals. The methods are applied to the extraction of dynamic compact thermal models for a new ultrathin chip stack technology (UTCS).

We propose a new method of operating laser interferometric gravitational-wave detectors when observing chirps of gravitational radiation from coalescing compact binary stars. This technique consists of the use of narrow-band dual recycling to increase the signal but with the tuning frequency of the detector arranged to follow the frequency of a chirp. We consider the response of such an instrument to chirps, including the effect of inevitable errors in tracking. Different possible tuning strategies are discussed. Both the final signal-to-noise ratio and timing accuracy are evaluated and are shown to be significantly improved by the use of dynamic tuning. This should allow an accurate and reliable measurement of Hubble's constant.

To determine the phenotype of peripheral blood lymphocytes during the time-course of adjuvant arthritis (AA) to detect alterations that could be involved in the pathogenesis of the arthritic process. METHODS--Phenotype analysis was performed on days 7, 14, 21, 28, 42, 56 and 70 after arthritis induction using monoclonal antibodies to CD5, CD4 and CD8 subsets, and flow cytometry. The proportion of activated lymphocytes and lymphocytes was also assessed with monoclonal antibodies to IL-2R (CD25), to Ia antigen and by polyclonal antibodies to rat Ig. RESULTS--Adjuvant arthritis produced leukocytosis with neutrophilia. Rats with AA showed a marked increase in the number of both CD4+ and CD8+ cells. The ratio CD4/CD8 decreased because the rise in CD8+ cells was more pronounced than the increase in CD4+ cells. Changes in lymphocyte counts showed two well-defined periods: the first, from day 14 to day 28, during which the inflammation of the joints reached a maximum and changes in lymphocyte subsets were more pronounced, that is, there was a threefold increase in CD8+ lymphocytes over normal counts, and the second, from day 42 to day 70, in which modified parameters improved considerably but remained different from controls. CONCLUSION--Alterations were detected in the phenotype of peripheral blood lymphocytes in AA, which provides an additional marker of disease activity.
